Growth Characteristics of Juvenile Abalone, Haliotis discus hannai by Reared Methods in order to High Density Intermediate Culture in Land-based Tank (육성수조 내 북방전복, Haliotis discus hannai 치패 고밀도 중간양성 사육방식별 성장특성)

  • The effect of different intermediated rearing method by expanding the attached floor space in order to highly density culture on the growth characteristics and survival rate of the juvenile abalone, Haliotis discus hannai, were investigated in land-based tanks. The intermediated culture methods was determined thru the shelter counts and layer for 10 month with two replicates : the single layer shelter (SLS), the double layer shelter (DLS), the triple layer shelter (TLS) and the single layer shelter under net cage (SLSNC). In addition, the culture on shallow race way tank had to set up as culture of the ditch raceway tank (CDRT) and the floor race way tank (CFRT). In the growth performance of reared abalone (initial mean shell length $54.18{\pm}7.39mm$ and weight $1.93{\pm}0.14g$) at experimental tanks, that the absolute growth rate (ARG), daily growth rate (DGR) and specific growth rate (SGR) to the shell length and shell breadth was not significant at each experimental tanks except SLSNC. As well as too, weight gain (WG), daily weight gain (DWG) and specific weight gain (SWG) to weight was not significant at each experimental tanks except SLSNC, too. Survival rates of CDRT and CFRT was lower than those of different experimental tanks (P < 0.05). Therefore, these results is showed that high density different intermediated rearing method by expanding the attached floor space for juvenile H. discus hannai was not have difference as growth performance and survival rate both one layer shelter and multi layer shelter. Also, it is considered that shallow race way tank was not useful rearing for the juvenile intermediate culture of H. discus hannai in land based.

  • This study was conduct to investigate the effect of intermediate culture types on the growth and survival rate of the juvenile abalone, Haliotis discus hannai fed seaweed and artificial diet. Intermediate cultures were to determine there that was to fed seaweed (SW) of artificial diet (A) of floor culture (FC), net floor culture (NFC), double shelter culture (DSC) and indoor net cage culture (INCC) in land-based tank, in two replicate. In the growth performance of juvenile abalone reared through intermediate culture to fed SW of A, that the absolute growth rate ($AGR_{SL}$, $AGR_{SB}$), daily growth rate ($DGR_{SL}$, $DGR_{SB}$), and specific growth rate ($SGR_{SL}$, $SGR_{SB}$) to the shell length (SL) and shell breadth (SB) of experimental groups were not significant. As weight gain (WG), daily weight gain (DWG) and specific weight gain (SWG) to body weight through intermediate culture types in land-based tank was not significant. However, as to survival rate to experimental groups, A-FC was higher than those of different groups (P < 0.05). Therefore, these results is showed that was not difference to growth of juvenile abalone over 2 cm fed seaweed diet and artificial diet according to intermediate culture types. But floor culture with artificial diet indicate that was highest to survival rate, therefore, it is beneficial for higher productivity in floor culture with artificial diet among intermediate culture types.

Acute Effects of Ethanol on Blood Pressure and Renal Function in Rat (Ethanol 급성투여(急性投與)가 흰쥐의 혈압(血壓)과 신기능(腎機能)에 미치는 영향(影響))

  • In order to determine the dose-response relationship of ethanol on blood pressure and renal function, 2 doses of ethanol were intubated into albino rats. For a direct measurement of arterial blood pressure, a polyethylene catheter(PE 10) was implanted in the abdominal aorta, and the other end of the catheter was pulled out of the back of the neck. The experiment was conducted after the rats recovered from the surgery. After emptying their bladders, the rats were placed in a metabolism cage. Mean arterial pressure (MAP) was measured and arterial blood samples were collected through the catheter. Following the collection of the control urine sample, 1 ml of 10 g% (low dose), or 30 g% (high dose) of ethanol/100 g BW was intubated. 1 ml of water/100 g BW was intubated into the control group. MAP and blood samples were taken every hour, and urine samples were collected every 90 min for 3 hours. Blood alcohol concentrations reached a peak at 1 hour (low dose: $105.0{\pm}7.5$, high dose: $214.7{\pm}20.2\;mg%$) and decreased linearly thereafter. Following alcohol ingestion, MAP began to decrease at 15 min and remained at a significantly low level thoughout the 3 hours experimental period(low dose: $112{\pm}2{\rightarrow}102{\pm}4$, high dose: $117{\pm}2{\rightarrow}100{\pm}8\;mmHg$). Urine Flow increased markedly during the first 90 min of ethanol ingestion (low dose: $0.88{\pm}0.20{\rightarrow}1.04{\pm}0.22$, high dose: $0.56{\pm}0.11{\rightarrow}1.35{\pm}0.18\;ml/1.5\;hr$) and decreased during the second 90 min period(low dose: $0.25{\pm}0.06$, high dose: $0.22{\pm}0.06\;ml/1.5\;hr$). Urine flow of the control group decreased gradually during the experiment $(0.88{\pm}0.10{\longrightarrow}0.59{\pm}0.09{\rightarrow}0.45{\pm}0.09\;ml/1.5\;hr)$. These results indicate that the blood-pressure-lowering and diuretic effects of ethanol are dose-related: higher doses of ethanol produce a greater decrease in blood pressure and greater diuresis.

Changes in Catch Rate of Monochamus saltuarius (Coleoptera: Cerambycidae) Relation to Sexual Maturation (북방수염하늘소(딱정벌레목: 하늘소과)의 성적 성숙에 따른 포획 효율의 변화)

  • In this study, we aimed to investigate the changes in the catch rate of Monochamus saltuarius, based on sexual maturation by using aggregation-sex pheromone traps. Ovariole development of caught M. saltuarius females was compared to that of the ones not caught using traps. In a mesh cage set up at the Hongneung experimental forest, we placed a multi-funnel trap with or without an aggregation-sex pheromone lure. M. saltuarius adults, which emerged from pine logs, were grouped in four according to the emergence dates (0, 1, 7, and 10 days after emergence [DAE]). We released beetles into the mesh cage to investigate the catch rate using the traps. In each group, a total of 80 beetles (20 beetles × 4 replications) were tested, making it a total of 320 beetles. Among the four groups, M. saltuarius adults in the 7 DAE group were caught more frequently using the traps, especially with a pheromone lure; the other groups showed a low catch rate. A similar number of female and male beetles were caught using the traps. Regarding ovariole development, all the female adults in the 0 and 1 DAE groups were immature, while those in the other two groups were completely developed. Therefore, aggregation-sex pheromone traps might have a limitation in the prevention of pine wilt disease because of the transmission of pine wood nematode during maturation feeding of newly emerged M. saltuarius adults. However, aggregation-sex pheromone traps can be effective for collecting sexually mature M. saltuarius adults, for the investigation of seasonal occurrence of beetles in forests.

Origin and Source Appointment of Sedimentary Organic Matter in Marine Fish Cage Farms Using Carbon and Nitrogen Stable Isotopes (탄소 및 질소 안정동위원소를 활용한 어류 가두리 양식장 내 퇴적 유기물의 기원 및 기여도 평가)

  • We investigated physicochemical properties and isotopic compositions of organic matter (δ13CTOC and δ 15NTN) in the old fish farming (OFF) site after the cessation of aquaculture farming. Based on this approach, our objective is to determine the organic matter origin and their relative contributions preserved at sediments of fish farming. Temporal and spatial distribution of particulate and sinking organic matter(OFF sites: 2.0 to 3.3 mg L-1 for particulate matter concentration, 18.8 to 246.6 g m-2 day-1 for sinking organic matter rate, control sites: 2.0 to 3.5 mg L-1 for particulate matter concentration, 25.5 to 129.4 g m-2 day-1 for sinking organic matter rate) between both sites showed significant difference along seasonal precipitations. In contrast to variations of δ13CTOC and δ15NTN values at water columns, these isotopic compositions (OFF sites: -21.5‰ to -20.4‰ for δ13 CTOC, 6.0‰ to 7.6‰ for δ15NTN, control sites: -21.6‰ to -21.0‰ for δ13CTOC, 6.6‰ to 8.0‰ for δ15NTN) investigated at sediments have distinctive isotopic patterns(p<0.05) for seawater-derived nitrogen sources, indicating the increased input of aquaculture-derived sources (e.g., fish fecal). With respect to past fish farming activities, representative sources(e.g., fish fecal and algae) between both sites showed significant difference (p<0.05), confirming predominant contribution (55.9±4.6%) of fish fecal within OFF sites. Thus, our results may determine specific controlling factor for sustainable use of fish farming sites by estimating the discriminative contributions of organic matter between both sites.

Gut Microbiome and Gut Immunity in Broiler Chickens Fed Allium hookeri Root Powder from Day 10 to 28 (육계 사료 내 삼채뿌리분말 첨가가 장내 미생물 및 장관면역에 미치는 영향)

  • This study was conducted to investigate the effects of supplementation of Allium hookeri (AH) root powder on the gut microbiome, immunity, and health in broiler chickens fed experimental diets from d 10 to 28. A total of 60 10-day-old Ross 308 broilers were weighed and assigned to two dietary treatments with 5 birds per cage in a randomized complete block design based on body weight. The two experimental diets consisted of a control diet based on corn-soybean meal and the control diet supplemented with 0.3% AH root powder. All birds were fed ad libitum with experimental diets and water for 18 d. At 28 d, two birds near the median weight from each cage were selected for cecal content and small intestinal tissue sample collection. The addition of AH changed the gut microbiome by increasing probiotic candidate beneficial bacteria such as Enterococcaceae, Lactobacillaceae, Limosilactobacillus, Cuneatibacter, and Ruminoccoides. Regarding gut immunity, the supplementation of AH resulted in changes in intestinal immune cells, including reduced CD3+CD4+ T cells, which are a type of helper T cell, in the small intestine of birds (P=0.049). Additionally, there was a tendency to increase the expression of antioxidant function-related gene such as GPX2 (P=0.060), but no significant changes were observed in cytokines such as IL1b, IL6, and IL10. Overall, the addition of AH root powder may have positive effects on the microbiome of the chickens. This may help promote gut health in broiler chickens at the age of d 10 to 28.

Effects of Fermented Diets Including Liquid By-products on Nutrient Digestibility and Nitrogen Balance in Growing Pigs (착즙부산물을 이용한 발효사료가 육성돈의 영양소 소화율 및 질소균형에 미치는 영향)

  • This study was conducted to evaluate the effects of fermented diets including liquid by-products on nutrient digestibility and nitrogen balance in growing pigs. Treatments were 1) CON (basal diet), 2) F (fermented diet with basal diet), 3) KF (fermented diet with basal diet including 30% kale pomace), 4) AF (fermented diet with basal diet including 30% angelica keiskei pomace), 5) CF (fermented diet with basal diet including 30% carrot pomace) and 6) OF (fermented diet with basal diet including 30% grape pomace). A total of 24 pigs (41.74kg average initial body weight, Landrace $\times$ Yorkshire $\times$ Duroc), were assigned to 6 treatments, 4 replicates and 1 pig per metabolic cage in a randomized complete block (RCB) design. Pigs were housed in $0.5\times1.3m$ metabolic cage in a 17d digestibility trial. During the entire experimental period, Digestibility of dry matter (p<0.05) of treatment CON, F and CF were higher than other treatments. In crude protein digestibility, treatment F was higher than treatment AF and GF (p<0.05). Treatment GF showed the lowest digestibility of crude fiber among all treatments (p<0.05). In ether extract digestibility, treatment AF and CF showed higher than other treatments (p<0.05) except KF treatment. CF treatment showed the best digestibility of ash among all treatments (p<0.05). Whereas, For Ca and P digestibility, CF and OF treatments were improved than other treatments (p<0.05). Energy digestibility (p<0.05) of CON, F and CF treatments were higher than KF, AF and GF treatments. In total essential amino acid digestibility, F treatment was improved than AF, CF and GF treatments (p<0.05). In total non-essential amino acid digestibility, F treatment was higher than CON, AF and GF treatments (p<0.05). In total amino acid digestibility, F treatment was higher than AF and CF treatments (p<0.05) and GF treatment showed the lowest digestibility (p<0.05). In fecal nitrogen excretion ratio, GF treatment was greatest among all treatments (p<0.05) and F treatment was decreased than other treatments (p<0.05). In urinary nitrogen excretion ratio, CON and GF treatments showed the lowest among all treatments (p<0.05). In nitrogen retention ratio, CON treatment showed the high and KF treatment showed the lost among all treatments (p<0.05). Therefore, this experiment suggested that fermented diet could improve nutrient and amino acid digestibilities of growing pigs.

Effects of Glucuronic Acid Derivative Isolated from Xylan on Antioxidative Defense System in Rat Red Gastrocnemius after Aerobic Exercise

  • The purpose of this study was to investigate the effects of glucuyonic acid (isolated from xylan) on the antioxidative defense systems of red gastrocnemius in rats after aerobic exercise. The glucuronic acid was isolated from xylan. Male Sprague-Dawley vats weighing 150$\pm$10 g were randomly assigned to one normal group and three exercise training groups. The exercise training groups were classified as T (glucuronic acid-free diet), TU (250mg glucuronic acid/kg bw) and 2TU (500mg glucuronic acid /kg bw) according to the level of glucuvonic acid supplementation. The rats in the normal group were confined to a cage for 4 weeks. The rats in the exercise training groups ran on a treadmill for 30 min/day, 5 days/week at a speed of 28 m/min (7% incline) for 4 weeks. Glutamate oxaloacetate transaminase (GOT) activity in the exercise training groups increased significantly compared with that of the normal group. That of the TU and 2TU groups decreased significantly compared with that of the T group. Xanthine oxidase (XOD) activity in the T group increased significantly to 74% compared with that of the normal group. That of the 2TU group decreased to 42% compared with that of the T group, thus recovering to a normal level. Superoxide dismutase (SOD) activity in the T group decreased to 32% compared with that of the normal group. That of the TU and 2TU groups increased to 28% and 34%, respectively, compared with that of the T group. Glutathione peroxidase (GSHpx) activity in the T group decreased to 16% compared with that of the normal group, but that of the TU group increased to 17% compared with that of the T group. Glutathiones transferase (GST) activity in the T group decreased to 11% compared with that of the normal group, but that of the TU and 2TU groups Increased to 28% and 31%, respectively, compared with that of the T group. The contents of thiobarbituric acid reactive substances (TBARS) in the T group increased to 81% compared with that of the normal group, but the glucuronic supplementation group recovered to the normal level. In conclusio, the effects of glucuronic acid on red gastrocnemius in rats engaged in exercise training would appear to be to reduced lipid peroxidation of tissue as an antioxidative defense mechanism.

The Effects of Complex Motor Training on Motor Function and Synaptic Plasticity After Neonatal Binge-like Alcohol Exposure in Rats (복합운동훈련이 신생 흰쥐의 알코올성 소뇌손상 후 운동기능 및 신경연접가소성에 미치는 영향)

  • The purposes of this study were to test that complex motor training enhance motor function significantly, to test change in cerebellum, and to test the synaptic plasticity into the immunohistochemistry response of synaptophysin. Using an animal model of fetal alcohol syndrome - which equates peak blood alcohol concentrations across developmental period - the effects of alcohol on body weight during periods were examined. The effect of complex motor training on motor function and synaptic plasticity of rat exposed alcohol on postnatal days 4 through 10 were studied. Newborn rats were assigned to one of two groups: (1) normal group (NG), via artificial rearing to milk formula and (2) alcohol groups (AG), via 4.5 g/kg/day of ethanol in a milk solution. After completion of the treatments, the pups were fostered back to lactating dams, where they were raised in standard cages (two-and three animals per cage) until they were postnatal 48 days. Rats from alcohol group of postnatal treatment then spent 10 days in one of two groups: Alcohol-experimental group was had got complex motor training (learning traverse a set of 6 elevated obstacles) for 4 weeks. The alcohol-control group was not trained. Before consider replacing with "the experiment/study", (avoid using "got" in writing) the rats were examined during four behavioral tests and their body weights were measured, then their coronal sections were processed in rabbit polyclonal antibody synaptophysin. The synaptophysin expression in the cerebellar cortex was investigated using a light microscope. The results of this study were as follows: 1. The alcohol groups contained significantly higher alcohol concentrations than the normal group. 2. The alcohol groups had significantly lower body weights than the normal group. 3. In alcohol groups performed significantly lower than the normal group on the motor behavioral test. 4. In alcohol-control group showed significantly decreased immunohistochemistric response of the synaptophysin in the cerebellar cortex compared to the nomal group. These results suggest that improved motor function induced by complex motor training after postnatal exposure is associated with dynamically altered expression of synaptophysin in cerebellar cortex and that is related with synaptic plasticity. Also, these data can potentially serve as a model for therapeutic intervention.

Hatchability and Temperature-dependent development of Overwintered Eggs of Ricania sp. (Hemiptera: Ricaniidae) (갈색날개매미충 월동 알의 부화율과 온도발육기간)

  • Ricania sp. lay eggs into the new twigs of blueberry and suck nutrients causing growth retardation and a sooty mold symptom. This study was conducted to investigate the emergence success and developmental period of overwintered eggs of Ricania sp. under the seven constant temperatures from 10 to $34^{\circ}C$ by $4^{\circ}C$ intervals (14L:10D). We also monitored the emergence time of the Ricania sp. nymph at blueberry by 3 ~ 4 day interval using a modified leaf clip cage. Development times from the overwintered egg to $1^{st}$ nymph were 107.1, 54.5, 33.9, 25.3, 25.1 and 16.7 days and the emergence successes were 23.1, 30.8, 13.8, 21.7, 11.9, and 0.6% at 14, 18, 22, 26, 30 and $34^{\circ}C$, respectively except at $10^{\circ}C$. The developmental periods were decreased with increasing temperatures. First emergence date of the nymphs in the field was between 19 and 22 May, and the average emergence success was 19.6%. The present study might be helpful to establish the management strategy of Ricania sp. based on the biological characteristic.
