This study were performed to investigate effect of dietary cadmium(Cd) and protein levels on growth, body protein metabolism and Cd toxicity in growing rats. Forty eight male rats of Sprague-Dawley weighing 113$\pm$2g were blocked into 6 groups accoridng to body weight. Dietary protein were given at the levels of 7, 15 and 40% of diet and Cd (200ppm)were either added or not. The result obtained were summerized as follow; 1) Food intake, weight gain, FER PER, liver and kidney weight, weight and length of bones, hematocrit, and hemoglobin content in Cd-added groups were low than those in Cd-free groups. 2) Serum total protein showed no significant difference with Cd addition, but it was significantly lower in low protein diet groups. Liver protein in Cd-added groups was lower than Cd-free groups, and was tend to be increased with increasing dietary protein level. 3) Daily urinary and fecal nitrogen excretions in Cd-added groups were lower than Cd-free groups, and were increased with increasing dietary protein level. 4) Cadmium contents in blood, liver, kidney, and femur were tend to be decreased with increasing dietary protein level. Especially, Cd content in kidney of Cd-added groups was significantly decreased with increasing dietary protein level. 5) Daily urinary and fecal Cd excretions were tend to be increased with increasing dietary protein level, and Cd-added-high protein diet group showed the highest Cd excretion among the Cd-added groups, Cd absorption ration and Cd retention ratio were tend to be decreased with increasing dietary protein level.

This study was performed to investigate the effect of Aloe arborescens on the cadmium toxicity in rats. Thirty male Sprague-Dawley strains were divided into five groups consisting of a control group, a cadmium treatment group and 3 aloe(0.5%, 0.75%, 1%) treatment groups and observed for 9 weeks. The weight increment of the cadmium and 0.75% aloe group was higher than that of the cadmium treatment group(p<0.01). The food intake did not show the consistency rule among the experimental groups and the decrement tendency of food intake affected by cadmium feeding group. The decrement tendency of water intake affected by cadmium appeared to be suppressed by aloe treatment, especially cadmium and 0.75% aloe treatment group showed the remarkable increment of water intake. The diet efficiency of the control group was the highest among the experimental groups and that of cadmium and 0.75% aloe group was higher than other aloe treatment groups. The weight of each organ did not show consistency among the experimental groups but only the testicle of cadmium and 0.75% aloe treatment group was heavier than that of the control group. The cadmium accumulation was high in order of kidney>liver>spleen>heart>lung>testicle>brain. The cadmium content of the cadmium treatment group was more than that of cadmium and 0.5% aloe group, cadmium and 0.75% aloe group, cadmium and 1% aloe group. The cadmium content of cadmium and 0.75% aloe group was the lowest among other aloe treatment groups. Therefore, cadmium and 0.75% aloe is the most recommendable aloe treatment to eliminate the cadmium accumulated in organ.
This study was to investigate the effects of chitin, chitosan , NOCC and cellulose on cadmium toxicity and lipid metabolism in rats. Fifty male rats of Sprague-Dawley strain weighing 155$\pm$17g were divided into 10 groups according to body weight, and were raised for 5 weeks. Levels of 0 and 400 ppm of cadmium chloride were in the diets. And chitin, chitosan , NOCC and cellulose were given at the level of 0 and 4%(w/w) of the diets. Chitosan and NOCC decreased Cd concentration of liver and kidney, and increased the fecal excretion of Cd. Although cellulose was less effective, cellulose also decreased Cd concentration of liver and increased fecal Cd excretion . However chitin had little effect on alleviating Cd toxicity. In addition, chitosan and NOCC lowered total lipid, cholesterol and triglyceride contents in serum and liver by decreasing the lipid absorption. The lowering of cholesterol especially had a remarkable effect. Although it was less effective than chitosan and NOCC, chitin decreased the lipid absorption ratio and lowered cholesterol contents in serum and liver. In conclusion , chitosan and NOCC showed effects of decreasing the absorption of Cd and lipid.
Elaeagnus umbellata leaves have been reported to suppress inflammation, allergic responses, lung cancer proliferation and oral bacterial growth. Cadmium (Cd) is a heavy metal that has been found to cause many toxicities, including liver toxicity. The aim of this study was to evaluate the capacity of 70% ethanol extract of E. umbellata leaves (EUL) to protect human hepatocytes from Cd toxicity. After exposure of HepG2 cells to Cd at 10 𝜇M for 24 h, cell viability, expression levels of apoptosis- and antioxidant-related proteins, reactive oxygen species (ROS) accumulation and Cd uptake were assessed. EUL protected HepG2 cells from Cd-induced apoptosis as determined by MTT assay. A decrease in caspase-3 and p-p53 protein levels was observed in cells pretreated with EUL prior to Cd exposure. Furthermore, the Cd-induced increase in intracellular DCF fluorescence was attenuated by EUL, indicating that the Cd-induced apoptosis preventing effect was associated with the suppression of ROS accumulation. Moreover, EUL's effects on the inhibition of p38, JNK, and AKT phosphorylation also appear to be associated with protection against Cd toxicity. Moreover, EUL upregulated Cd-depressed expression of Nrf2, HO-1, catalase, and MT-1,2 proteins, suggesting that Cd uptake-induced apoptosis in HepG2 cells may be inhibited by EUL's antioxidative potential.

For the experiment of the effects between cadmium aerosol inhalation toxicity and ethyl acetate extracts of Folium Mori, 4 inhalation exposure groups of rat were exposed to cadmium aerosol in air by whole-body inhalation exposure for 6 hours/day, 5 days/week, and 4 weeks. Cadmium concentration in the air was 0.96㎎/㎥ and mass median diameter (MMD) was 2.48㎛ with 1.85 of geometric standard deviation(GSD). Intraperitoneal injections of ethyl acetate extracts of Folium Mori to inhalation exposure groups were performed for 4 weeks and the results were as follows: The highest body weight gain for 4 weeks and food intake per day were 159.29/4 weeks in treated group III and 18.45g/day in treated group I, respectively. The highest lung and liver weights were 1.31 g in treated group I and 9.42g in treated group III, respectively. The highest kidney weight was 2.21g from treated group I. The lowest cadmium content in lung was 86.39㎍/g from treated group III and the lowest cadmium concentration in blood was 2.72㎍/㎗ from treated group II. Cadmium concentrations of 22.09㎍/g in liver and 24.82㎍/g in kidney were the lowest from inhalation exposure group I and III, respectively. For weekly cadmium concentration in urine, the value of the fourth week from treated group III was the highest, 1.35㎍/㎖. For weekly cadmium concentration in feces, the values of the second and fourth week from treated group I were the highest, 1.11㎍/g. The highest metallothionein concentration in lung was 31.85㎍/g from treated group III and the highest metallothionein concentration in liver was 205.77㎍/g from treated group III. The highest metallothionein concentration in kidney was 206.55㎍/g from treated group III. The highest Hct and Hb values were 38.26% and 11.63g/㎗ from treated group III, respectively. The highest RBC and WBC values were 7.68×106/㎣ and 9.85×10³/㎣ from treated group I, respectively.
This study was conducted to investigate the metallothionein (MT) induction by Lonicerae rios in cadmium chloride intoxication. The results were as follows: Generally, detoxication effects by Lonicerae fios extract increased to the increase of extract concentrations. When 90 mg/kg dosage of Lonicerae fios extract was administered, it showed the highest antitoxic effects in metallothionein induction. From the above results, Lonicerae rios extract increased metallothionein concentration and decreased the toxicity of cadmium in liver.
This study was conducted to investigate the metallothionein (MT) induction by Panax ginseng C. A. Meyer in cadmium chloride intoxication. The results were as follows: Generally, detoxicatlon effects by Panax ginseng C. A. Meyer extract increased proportionally to the increase of cadmium concentrations. When a 8 mg/g dosage of cadmium was administered, formation of the cadmium and EDTA complex showed the highest antitoxic effect. Also, when a 4 mg/g dosage of cadmium was administered, Panax ginseng C. A. Meyer extract showed the highest antitoxic effect in metallothionein induction. According to the above results, Panax ginseng C. A. Meyer extract administered with cadmium increased metallothionein concentration and decreased the toxicity of cadmium in liver.
Casapse-3 protease is known as a key role of apoptotic enzyme, and caspase-3 activity is a central event that occurs upstream of DNA fragmentation during apoptosis. This study demonstrates that chitosan pretreatment inhibits cadmium-induced apoptosis by attenuating the activity of caspase-3. We also analyzed the protective effect of chitosan on DNA fragmentation induced by cadmium. Cadmium toxicity was examined by DNA fragmentation and nuclear condensation with Hoechst stain. Caspase-3 activities were increased cadmium treated group for 3 hours compared with control. When chitosan (150 mg/ml) was pretreated at 30 min before cadmium treatment, cadmium cytotoxicity was suppressed in a dose-dependent manner evaluated by DNA fragmentation and caspase activity. From these results, it is suggest that the protective effect of chitosan pretreatment against cadmium-induced cytotoxicity is mediated through inhibition of caspase-3 protease activation and DNA fragmentation.
The rate of metallothionein synthesis on cadmium-poisoned rats reflects the level of toxicity, and also it reduces the toxicity which is caused by the uptake of cadmium. Chlorella supplementation in the diets of the cadmium-poisoned rats decreased the concentration of cadmium in blood and urine compared with the control group. Although the liver and kidneys of rats are major target organs of cadmium and coherence of metallothionein and cadmium, no previous study has determined the correlation between the rate of metallothionein synthesis in the liver and kidneys of rats and dietary supplementation of chlorella with cadmium uptake. This study analyzed total metallothionein level on the tissue of the liver and kidneys, the concentration of cadmium bound to the metallothionein, and the total concentration of cadmium on the tissue of the liver and kidneys after dietary supplementation with 1%, 5%, and 10% dried chlorella and 40 ppm of cadmium to 46 male SD rats (mean weight: $150\pm20\;g$) for 4 weeks. According to the data analysis of the total rate of metallothionein synthesis in the liver and kidneys, the group of SD rats on the supplementation with 1% chlorella and 40 ppm of cadmium showed a rate of $93.2\pm8.9\;ng/g$, a significant decrease of 58.8% compared to that of the control group of SD rats on the supplementation with cadmium only, which showed a rate of $227.3\pm32.5 ng/g$ (P=0.0001). In contrast, no significant difference was observed through the changing of chlorella concentrations between 5% and 10% chlorella supplementation with cadmium. The group supplemented with 1% or greater chlorella levels represented a greater decrease in the total cadmium concentration of the kidney and liver tissues, the amount of total metallothionein synthesis, the amount of metallothionein with binding to cadmium, and the concentration of free cadmium without binding to metallothionein. Consequently, the supplementation of 1% and 5% chlorella was effective in reducing the synthesis of metallothionein for cadmium uptake, but increased the rate of binding of cadmium to metallothionein.
The short term (24-hr) and long term (21 days) effects of copper, cadmium, fenbendazole and sulfathiazole on the survival of the Korean fairy shrimp Branchinella kugenumaensis were evaluated. The 24-hr median lethal concentrations ($LC_{50}$) of copper, cadmium, fenbendazole, and sulfathiazole were 39, 512, 182, and 31,818 ${\mu}g/L$, respectively. The toxicity of copper is highest among 4 chemicals used in this study, while sulfathazole the lowest. After the long term (21 days) exposure experiment, the $LC_{50}$ copper, cadmium, fenbendazole, and sulfathiazole were 1.12, 2.1, 0.1, 6.6 ${\mu}g/L$, respectively. The long term effects of antibiotics were highly enhanced while the short-term effects were not strong. The sensitivities of B. kugenumaensis to copper and cadmium were higher than or comparable to those of other freshwater branchiopods (Streptocephalus spp., Thamnocephalus sp.), and far higher than the marine species (Artemia sp.). There were significant effects on the survival of B. kugenumaensis after long term exposure to relatively lower concentrations of copper, cadmium, fenbendazole and sulfathiazole. Therefore, B. kugenumaensis seems quite a good candidate species for the ecotoxicological assessments of freshwater environments.
