Studied on Enzymes Responsible for Polyamine Level Effects of IAA on the activities of lysine decarboxylase and diamine oxidase and polyamine level in seedlings of soybean (Polyamine 농도에 미치는 효소에 관한 연구 대두의 유묘에서 IAA가 Lysine Decarboxylase와 Diamine Oxidase의 활성과 Polyamine함량에 미치는 영향)

  • The effect of IAA on lysine decarboxylase(LDC)activity, diamine oxidase (DAO) activity and the polyamine level in seedlings of soybean were investigated. The seedlings of soybean were treated with IAA (10-8M, 10-6M, 10-4M) and LDC activity, DAO activity and polyamine levels of cotyledon, elongating hypocotyl, elongated hypocotyl and the root were measured after 4, 6, 8, 10 days. Increased concentration of IAA stimulated the LDC activity and increased the level of polyamine in the seedlings, which reducing the DAO activity and polyamine level was increased in the seedlings. The light also stimulated the IAA effect on polyamine level and DAO activity, and polyamine level was higher in the seedlings grown in the light than those in the dark. Cadaverine was the most abundant polyamine in the seedlings of soybean. The levels of diamines, cadaverine, putrescine were higher in the elongated hypocotyl and root than in the elongating hypocotyl, but the levels of spermidine and spermine were relatively higher in the cotyledon than in the other parts. Accumulation of diamines by high concentration of IAA is postulated to be a phenomenon of inhibition in seedlings growth.

A Sensitive Fluorescent Detection Method for Prostaglandins by High Performance Liquid Chromatography (고속액체크로마토그라피에 의한 프로스타글란딘류의 고감도 형광 분석법)

  • The Prostaglandins were derivatized rapidly with monodansyl cadaverine as a fluorophore in mild conditions. The carboxylic moiety of prostaglandins was activated with diethyl phosphorocyanidate and successively coupled with fluorophore in dimethylformamide at room temperature. The labeling yield was reached about 95% at 15 min using arachidic acid $(C_{20:0})$ as a test sample. This derivative showed constant fluorescent intensity at $4^{\circ}C$ for 180 days. The derivatives of prostaglandins were shown high solvent selectivity with tetrahydrofuran in reversed-phase column. therefore, these derivatives could be successfully separated on YMC pack A-212(S-5 120A C8) column in tetrahydrofuran-based eluents. The detection limits of these derivatives was ca. 500 fmol and determination limits was ca. 5 pmol as injected amount in fluorescent detection $({\lambda}ex.\;340\;nm,\;{\lambda}em.\;520\;nm)$. In this method, the ranges of recovery and coefficient of variation were $93.6{\sim}102.7%$ and $4.3{\sim}5.8%$, respectively.

Characterization of cadC and cadR Mutants in Mediating the Expression of the Salmonella typhimurium cadBA Operon (Salmonella typhimurium cadBA 오페론의 발현에 관여하는 돌연변이체의 선별 및 그 특성)

  • It has been well known that the expression of S. typhimurium cadBA operon requires at least two extracellular signals: low pH and high concentration of lysine. To better understand the nature of pH-dependent and lysine dependent signal transduction, mutants were isolated in JF2238(cadA-lacZ) by Tn10 insertion, spontaneous mutagenesis, and EMS treatment. Two mutants were isolated from JF2238, expressed as a cadA-lacZ operon fusion in various growth conditions, and analyzed to have mutations in cadC, a gene encoding a function necessary for transcriptional activation of cadBA. One isolate (cadC6) conferred pH-independent and lysine-independent cadBA expression and the other(cadC4) showed pH-independent and lysine-dependent cadBA expression. cadR::Tn10 and cadR4 mutants were expressed in the absence of exogenously added lysine. They were also resistant to thiosine and complemented by lysP clone from E. coli. Thus, in the absence of exogenous lysine, cadR is a negative regulator of cadBA expression. Cadaverine, the product of lysine decarboxylation, was shown to inhibit expression of cadA-lacZ fusion in cad $C^+$ cell.

Formation of Biogenic Amines by Lactobacillus plantarum Isolated from Makgeolli (막걸리에서 분리한 Lactobacillus plantarum의 biogenic amine 생성능)

  • We examined biogenic amine (BA) production as well as the diversity of bacterial flora in 11 types of commercial makgeolli stored at 4 and $20^{\circ}C$. Moreover, we studied the BA-producing activity of three L. plantarum strains isolated from makgeolli. At $20^{\circ}C$, the BA content was highly increased and the denatured DNA bands were more variable in non-sterilized makgeolli compared to sterilized makgeolli. The major BAs produced in commercial makgeolli were histamine and putrescine. Histamine, tyramine, putrescine, and cadaverine were produced in excess by inoculation of the three L. plantarum isolates to makgeolli stored at $20^{\circ}C$ for 21 days. These results suggest that some L. plantarum strains in makgeolli can produce different types of BAs, depending on the extent of degradation of makgeolli.

  • The degradation of non-volatile-amines by microorganisms were investigated. The degra-ding activity could be noted in four strains isolated from fermented sardine sauce, and those were Pseudomonas aeruginosa, Pseudomonas fluorescens P-2, Pseudomonas fluorescens P-3 and Enterobacter aerogenes. The strongest degrading activity of non-volatile-amines was showed in Pseudomonas fluorescens P-3 among the four strains isolated. The optimum temperature for degradation by Pseudomonas fluorescens P-3 was $35^{\circ}C$, corresponding to the optimum temperature for growth of this strain, pH between 7.0 and 7.5 could gave effective degradation and the optimum concentration of NaCl was 0 and/or $1\%$.

  • A liquid-based colorimetric assay using a pH indicator was introduced for high-throughput monitoring of lysine decarboxylase activity. The assay is based on the color change of bromocresol purple, measured at 595 nm in liquid reaction mixture, due to an increase of pH by the production of cadaverine. Bromocresol purple was selected as the indicator because it has higher sensitivity than bromothymol blue and pheonol red within a broad range and shows good linearity within the applied pH. We applied this for simple determination of lysine decarboxylase reusability using 96-well plates, and optimization of conditions for enzyme overexpression with different concentrations of IPTG on lysine decarboxylase. This assay is expected to be applied for monitoring and quantifying the liquid-based enzyme reaction in biotransformation of decarboxylase in a high-throughput way.

Selection and Characterization of Bacillus Strains Harboring the Gene for Biogenic Amine Degradation (바이오제닉아민 분해 유전자 보유 Bacillus 균주 선발 및 특성)

  • Ten Bacillus strains possessing amine oxidase activity were selected from 126 Bacillus isolates from meju and doenjang (two traditional Korean soybean foods). Among the isolates, two B. licheniformis strains (8MI05 and 8MS03) harbored the amine oxidase gene yobN. By conducting a gene search against published B. licheniformis genomes, the possession of yobN was proved to be a strain-specific property. Both strains degraded four types of biogenic amines (cadaverine, histamine, putrescine, and tyramine) supplemented in tryptic soy broth during their culture. A recombinant harboring yobN also degraded the four types of biogenic amines during cultivation. Both Bacillus strains could grow at a NaCl concentration of 14% and exhibited strain-specific protease and lipase activities.

Isolation of Biogenic Amines-Degrading Strains of Bacillus subtilis and Bacillus amyloliquefaciens from Traditionally Fermented Soybean Products (전통장류에서 Biogenic Amines 분해 능력을 가지는 Bacillus subtilis 및 Bacillus amyloliquefaciens 균주의 분리)

  • In order to reduce harmful biogenic amines in the traditionally fermented soybean products, five isolates with biogenic amines-degrading property were obtained from 83 traditionally fermented soybean products. The strains were found to reduce biogenic amines including histamine, tyramine, putrescine, and cadaverine by 27 to 92% in the cooked soybean containing 5.3% of each biogenic amine over 10 days of incubation. The morphological and biochemical tests and the phylogenetic relationships based on 16S rRNA gene sequences indicated that the five isolates were most closely related to Bacillus subtilis or B. amyloliquefaciens. The use of selected strains would be a potential control measure in manufacturing traditionally fermented soybean products that are difficult to control biogenic amine levels.

Survey of Biogenic Amine Contents in Commercial Soy Sauce (시판 간장 중의 biogenic amine의 함량 조사)

  • Nine commercial soy sauces $(A{\sim}I)$ were investigated for their biogenic amine (BAs) levels. Detected biogenic amines were putrescine (PUT), cadaverine (CAD), trytamine (TRP), ${\beta}-phenylethylamine$ (PHE), spermine (SPM), histamne (HIS), and tyramine (TYR). All products tested had biogenic amines as detected level. PUT was the major biogenic amines detected in six products, and difference between the highest and the lowest among products was more than 16 mg/kg. Six products had all seven biogenic amines tested, while one product had only five. Results indicate that soy sauces commercially available in Korea contain biogenic amines at various levels. Studies related to biogenic amines including survey of contents must be performed continuously.

Comparison of the Physiochemical Compositions and Biogenic Amine Contents of Salt-fermented Fishery Products fromy Different Raw Materials (원료에 따른 젓갈류의 이화학적 성분 및 Biogenic Amine류의 함량 비교)

  • This study was conducted to understand the quality characteristics of 89 types of commercial salt-fermented fishery products by measuring their physiochemical compositions and biogenic amine contents. All samples had the following measurements; 41.59-89.20 g/100 g of moisture, 1.71-25.70 g/100 g of salinity, 3.21-7.05 of pH, 0.80-2.93 g/100 g of total nitrogen, 87.02-1,296.78 mg/100 g of amino nitrogen, and 7.30-926.34 mg/100 g of volatile basic nitrogen. The physicochemical components differed extensively between samples from different raw materials, including, fish, shellfish, and others. Histamine (0 to 1,072.2 mg/kg), putrescine (0 to 2,536.4 mg/kg), cadaverine (0 to 545.2 mg/kg of cadaverine), tryptamine (0 to 1,287.9 mg/kg), and tyramine (11.3 to 563.3 mg/kg) were the major biogenic amines detected in the samples. These findings suggest that salt-fermented fishery products meet the domestic criteria but have different ingredient compositions and most had high biogenic amine contents. The results suggest that it is necessary to establish criteria for evaluating the quality characteristics using the ingredient composition and biogenic amine contents of commercial salt-fermented fishery products.
