손바닥선인장의 항염증 활성 (Anti-inflammatory Activity of Opuntia ficus-indica)

  • Cactus (Opuntia ficus-indica var saboten Makino) is a tropical or subtropical plant, which is widely used as folk medicine for burned wound, edema and indigestion. We previously found that the ethanol extract of cactus stem showed anti-inflanunatory action. This investigation was designed to isolate the active fraction of anti-inflanimatory action from cactus stem by solvent extraction and colunm chromatography. Carrageenan-induced paw edema in rats and acetic acid-induced writhing test in mice were used as animal models to search anti-inflammatory and analgesic activities. respectively. The ethanol extract of cactus stem was consecutively extracted with hexane, ethyl acetate, and n-butanol. The hexane fraction was the most effective in carrageenan-induced paw edema, and then was separated in colunm chromatography of silica gel by the elution with hexane/ethyl acetate mixture. The most effective fraction 1 was separated in a second colunm chromatography by eluting with hexane/diethyl ether mixture. The most effective fraction 1-5 was obtained, and separated in a third column chromatography by eluting with hexane/chloroform mixture. It produced the most effective fraction 1-5-1. Moreover, fraction 1-5-1 showed an inhibitory effect on acetic acid-induced writhing in the doses of 30mg/kg and 60mg/kg,p.o.,indicating that it also contained analgesic activity.

손바닥 선인장의 항산화활성 및 분말 첨가 쿠키의 품질특성 연구 (The Antioxidant Activity of Korean Cactus (Opuntia humifusa) and the Quality characteristics of Cookies with Cactus Powder added)

  • In order to better understand the quality of Korean cactus, Opuntia humifusa, as the functional foodstuffs, it's seeds, stems and fruits were analyzed. In addition, we performed quality evaluation on cookies manufactured with cactus powder added. The results showed the soluble nitrogen-free component levels in the cactus to be very high. The highest component levels were crude fiber(48.75%) and crude fat(7.38%) in the seeds, crude ash(15.39%) in the stems, and moisture(11.26%) and crude protein(8.79%) in the fruits. The antioxidant activities of the cactus were evaluated based on peroxide values(POV), thiobarbituric acid reactive substances(TBARS value), and electron donating ability(EDA) using the DPPH method. The POVs for soybean oil containing a 0.05% concentrated water extract from each part of the cactus revealed a remarkable antioxidant effects on peroxidation as storage time increased. The TBA values were 63.65% in BHT, 60.67% in the stems, 57.31% in the fruits and 49.16% in the seeds. The EDAs were 30.26% in the stems, 25.89% in the fruits, and 25.65% in the seeds. The total polyphenol contents of the water soluble materials from the cactus were 3.14 g/100 g in the fruits, 2.93 g in the stems and 2.11 g in the seeds. For the cookies, as the amounts of fruit and stem powder increased, the spreadness was significantly lowered, but moisture content was increased compared to the control. The color degree of the cookies changed from dim to intense as more fruit powder was added, resulting from a decrease in the L- and b-values and a significantly increased a-value. With the addition of stem powder, the L- and a-values revealed no significant differences, while the b-value was lowered compared to the control as more stem powder added. For texture changes of the fruit cookies, there were significant differences in hardness, gumminess, cohesiveness, springiness and brittleness. The stem cookies, however, showed only differences in hardness, gumminess, and brittleness, and not in cohesiveness and springiness. Finally, an overall sensory preference was found with the 1% fruit powder and 3% stem powder additions.

Potential Biotypes in Korean Isolates of Bipolaris cactivora Associated with Stem Rot of Cactus

  • A total of 62 isolates of Bipolaris cactivora causing cactus stem rots were isolated from major cactus-growing areas in Korea. Colony morphology of the isolates on potato-dextrose agar was differentiated into aerial (CA) and non-aerial mycelial types (CB). CA had profound aerial mycelium with grayish brown (CA-l), light brownish (CA-2), and brownish (CA-3) pigmentations; respectively, while CB had dark brownish pigmentations. CA had conidia of less dark pigmentation and acute terminal end. CB had darker and more round-end conidia. Twenty-eight amplified fragments were produced by polymerase chain reaction (PCR) with a set of 2 random primers. The sizes of amplified DNA fragments ranged approximately from 0.1 to 2.3 kb. The isolates were classified into 2 major genomic DNA random amplified polymorphic DNA (RAPD) groups at the genomic similarity of 97.7% and 95.1%, respectively. Cluster analysis of genetic similarity among the isolates generated a dendrogram that clearly separated all isolates into SA or SB. This result suggests that there may be two morphotypes of B. cactivora in Korea that may differ in their genetic constitutes.

  • Crassulacean acid metaholism (CAM) was investigated in leaves and stems of the succulent $C_4$dicot Portulaca oleracea L. Under 14-hour days, stem tissues showed much greater fluctuation of acidity than leaf tissues. But leaf and stem tissues showed almost same CAM-like pattern of acid fluctuation under 8-hour days. Stem tissues of R oleracea grown under the naturai environment showed high CAM activity, but no CAM activity was seen in leaves of those plants. In the naturally growing plants, the rapid acidification was seen in intact stems at dawn, but defoliated stems showed only a gradual increase. RuBP carlboxylase activity was very high at 2:00 P.M. in both leaves and stems. However, its activity at 1:00 A.M. and 5:30 AM. was hardly detected. particularly, activity of PEP carboxylase in leaves was very high in the early morning, though that in stem tissues was little. These results indicate that $CO_2$ passed through open stomata at dawn may be assimilated by PEP carboxylase in leaves, and then $C_4$ products move to stems. The levels of nitrate concentration and of nitrate reductase were higher in stems than in leaves. The levels were also higher in the light than in the dark. It would be suggested that considerable amount of nitrate absorbed from roots ho assimilated in stems, and nitrate transferred to leaves via stem tissues be reduced there. First Report of Black Spot Caused by Alternaria alternata on Grafted Cactus

  • A stem spot disease was observed on Gymnocalycium mihanovichii (Korean name: Bimoran), a scion of graftcactus, in major growing areas of Goyang and Eumseong, Korea during 2008 and 2009. Typical symptoms were initial blackish brown lesions produced mainly on areoles and scars of injured stem tissues, gradually becoming large black spots. A causal organism isolate CD2-7A isolated from the infected stems was identified as Alternaria alternata based on its morphological characteristics and confirmed by the DNA sequencing analysis of ITS, glyceraldehydes-3-phosphate dehydrogenase (gpd), and allergen Alt a1 (Alt a 1) genes. Artificial inoculation of the fungal isolate CD2-7A produced the same rot symptoms on the cactus stems, from which the same fungus was isolated and identified. This is the first report of the black spot caused by A. alternata in the grafted cactus.

Occurrence of Anthracnose on Indian Fig Cactus Caused by Glomerella cingulata and Colletotrichum gloeosporioides

  • Anthracnose symptoms were frequently found on stems of Indian fig cactus in Cheju island of Korea in 1998 and 1999. Typical symptoms were gray to black dry rot of stems with concentric arrays of dot-like spots. A Glomerella sp. or a Colletotrichum sp. was frequently isolated from the symptoms, both of which were identified as Glomerella cingulata and its anamorph, Colletotrichum gloeosporioides based on their morphological and cultural characteristics. Out of 31 isolates obtained from the symptoms, 12 isolates were the anamorph producing only conidia, four isolates the telemorph producing only ascospores, and 15 isolates the holomorph producing both spores on potato dextrose agar. Stem rots similar to the original anthracnose symptoms were induced by wound inoculation of conidia and ascospores but not by non-wound inoculation. The anamorphic isolates caused more extensive stem rot than the telemorphic and holomorphic ones.

  • Prickly pear(Opuntia ficus-indica var saboten Makino, Cactaceae) is widely used as folk medicine for burned wound, edema and indigestion. To confirm the hypolipidemic action of the extract from Cactus stem and fruit in dietary hyperlipidemic rats, variations of the lipid- or cholesterol-related components were investigated. As results these changes were significantly prevented by treatments with Cactus extracts. The effects of the extract of fruit was better than those of stem.

Studies on the Pharmacological Actions of Cactus:Identification of Its anti-Inflammatory Effect

  • The ethanol extracts of Opuntia ficus-indica fructus (EEOF) and Opuntia ficus-indica stem (EEOS) were prepared and used to evaluate the pharmacological effects of cactus. Both the extracts inhibited the writhing syndrome induced by acetic acid, indicating that they contains analgesic effect. The oral administrations of EEOF and EEOS suppressed carrageenan-induced rat paw edema and also showed potent inhibition in the leukocyte migration of CMC-pouch model in rats. Moreover, the extracts suppressed the release of $\beta$-glucuronidase, a lysosomal enzyme in rat neutrophils. It was also noted that the extracts showed the protective effect on gastric mucosal layers. From the results it is suggested that the cactus extracts contain anti-inflammatory action having protective effect against gastric lesions.

수출용 접목선인장 재배시 간이양액재배 연구 (Studies on Simple Hydroponic Culture in Cultivation of Grafted Cactus for Export)

  • This study was carried out to investigate the effect of simple hydroponic culture in cultivation of grafted cactus. The results obtained are as follows: Growth indexes of grafted cactus cultivars, such as scion diameter, fresh weight, and scion hardness were improved in simple hydroponic culture compared with soil culture regardless of cultivars. Stem rot and rate of non-graft take were lower in simple hydroponic culture than in soil culture, but no significant difference was monitored between cultivars. Time for harvesting was fast in simple hydroponic culture compared with soil culture. Marketable yield increased in simple hydroponic culture.

선인장 추출물이 인체 장내미생물에 미치는 영향 (Effects of Cactus Extracts on Human Intestinal Microflora)

  • 본 연구에서는 인체 내 장내 세균 및 병원성 식중독 세균에 선인장 추출물이 미치는 영향과 항산화, 항고혈압 활성에 대하여 분석하였다. 항균활성은 96well-plate법과 paper disc법을 이용하여 실험하였으며 줄기 추출물의 경우 96well-plate 및 paper disc법의 결과, 장내유해균인 Eubacterium limosum 및 Clostridium perfringens, C. butyricum, C. difficile과 Staphylococcus aureus에 대하여 높은 억제율을 보였으나 Bacteroides fragilis, Bifidobacterium bifidum, Lactobacillus acidophilis, Streptococcus thermophilus의 경우 억제율을 나타내지 않았다. 열매 추출물의 경우 장내유해균 및 장내유익균에 Bacteroides fragilis, Bifidobacterium bifidum, Lactobacillus acidophilis, and Streptococcus thermophilis에 대하여 억제율을 나타내지 않았다. 또한 줄기 추출물의 경우 열 및 pH에서도 장내미생물에 대한 항균활성에 안정함을 나타내었다. 선인장 추출물의 폴리페놀과 플라보노이드 함량 및 DPPH 전자공여능을 측정한 결과 열처리하지 않은 열매와 줄기 추출물의 폴리페놀은 3.02 mg/g, 5.85 mg/g, 플라보노이드는 1.04 mg/g, 2.1 mg/g, 전자공여능은 87, 88%로 높은 수율을 나타내었다. 선인장 추출물의 항고혈압활성은 줄기 추출물의 경우 88.8% 열매 추출물의 경우 69.2%의 항고혈압활성을 보였으며 상기 결과를 종합할 때, 선인장 줄기추출물은 장내 유해균의 억제 및 유익균의 증진을 통한 장내 균총 개선을 위한 기능성 식품소재로 활용할 수 있다고 판단된다.
