Cloning of CAB cDNA encoding chlorophyll a/b binding protein of photosystem II in Korean ginseng and Use in Plant (고려인삼 광계 II Chlorophyll a/b binding Protein 유전자(CAB)의 cloning 및 식물에의 활용연구)

  • A CAB cDNA clone(pKGCAB) encoding the light harvesting chlorophyll a/b binding protein of the semi-shade plant, Korean ginseng(Panax ginseng C. A. Meyer) was isolated by the one-way path random sequencing of ginseng cDNA library clones and transgenic tobacco plants(Nicotiana tabacum NC82) were produced by the transformation of this ginseng CAB gene in use of Agrobacterium tumefaciens LBA4404. The CAB gene showed type 1 structure of LHCP-II, 84% similarity in nucleotide sequence and 92% in amino acid sequence to that of Nicotiana tabacum CAB40, respectively. Seed germination and initial growth of the transgenic tobacco plants transformed with the cDNA fragment were accelerated under low light intensity compared with those of normal tobacco plant, that may result from the higher light sensitivity of the transgenic plants than that of the normal.

  • Abnormal cab vibrations in the Y direction in commercial vehicles during driving(70~90kph) are not common vibrations that happen to vehicles during driving and can be an obstacle to normal driving. This study conducted Operation Deflection Shape(ODS) testing to identify the causes of those abnormal cab vibrations and find solutions for them and also a sine sweep test to find resonance and frequency in the cab suspension system and set directions for improvement. The study also altered the shape of the bush inner part for changes to the rigidity features of the cab bush in the Y direction and revised the design with optimal rigidity in the Y direction, thus improving abnormal cab vibrations in the Y direction during driving.

  • In the first part of this paper, an analysis of the cab noise of a selected agricultural tractor was presented. In this study, using the results of the previous analysis, two passive noise control measures of the sound insulation and absorption were conducted to reduce the noise level inside the cab. These measures of noise control reduced the total noise level by 6.2㏈(A) at the operator position inside the cab. In order to further reduce the cab noise, particularly, of lower frequencies than 630Hz, stiffness and damping of the floor panel should be enforced. It was also suggested that a proper suspension for the cab mounting is necessary to reduce the level of structure-born noises.

  • Photo system II (PSII) is one of the two photosynthetic reaction centers in the chloroplast of higher plants. The chlorophyll a/b-light harvesting complex serves primarily as an antenna for PSII. We isolated a cDNA that encodes a chlorophyll a/b-binding protein (Cab) from Panax ginseng. The small subunit consists of 935 nucleotides long and has an open reading frame of 795 bp with the deduced amino acid of 265 residues (pI 5.63), 28.6 kDa. The deduced amino acid sequence matched to the previously reported Cab genes. Their degree of amino acid identity ranged from 68 to $92\%$. Phylogenetic analysis based on the amino acid residues was showed that the ginseng Cab gene was grouped with P. persica (AAC34983), A. thaliana (AAD28771), G. hirsutum (CAA38025), G. max (AAL29886), and V. radiate (AAF89205).

Biological Control using Bacillus toyonensis Strain CAB12243-2 against Soft Rot on Chinese Cabbage (Bacillus toyonensis CAB12243-2 균주를 이용한 배추 무름병의 생물적 방제)

  • Pectobacterium carotovorum subsp. carotovorum was found to be highly virulent to various vegetables, including Chinese cabbage. The antibacterial isolate CAB12243-2 was tested in a field bioassay for suppressing soft rot disease. The nucleotide sequencing of the 16S rRNA gene identified, the CAB12243-2 strain used in this study as Bacillus toyonensis. B. toyonensis CAB12243-2 inhibited the pectate lyase process by soft rot pathogens, and used trehalose and glucose as carbon sources. In field tests, the antibacterial isolate B. toyonensis CAB12243-2 suppressed soft rot disease with 73.0% control efficacy on the spring cultivar "Norangbom" and with 68.9% efficacy on the fall cultivar "Bulam 3". These results suggest that B. toyonensis CAB12243-2 can be used as a biological control agent for the control of soft rot diseases on vegetables.

Investigation of Transmission Characteristics of Tractor Seat Vibrations Using Vibration Path Analysis Method (VPA를 이용한 트랙터 좌석 진동의 전달 특성 구명)

  • This work was intended to investigate the effect of vibration transmission paths on the ride vibration of tractor during the rotavating and transporting operations by applying the vibration path analysis method. Accelerations at the cab mounts were measured during the rotavating and transporting operations. Ride vibrations at the sear were than calculated using the measured accelerations at the cab mounts, and the frequency response functions and inertances between the seat and cab mounts, which were derived experimentally by the impact hammer test in static condition. The human sensitivity to vibration frequency was also taken into consideration for the calculation of ride vibrations at the 1/3 octave center frequencies in the frequency domain. Vibrations transmitted through rear cab mounts affected more significantly the ride vibration of tractor. The peak accelerations at the seat occurred at the frequencies of the engine and crank speed, and the frequency induced by tire lugs on the road transportation. It was found that the rear cab mounts should be improved in order to reduce the ride vibrations more effectively.

Optimum Design of Cab Suspension for Agricultural Tractors (농용 트랙터 안전캡 현가 장치의 최적 설계)

  • This work was intended to determine the optimum values of the cab suspension parameters by a simulation method in order to minimize the seat accelerations of agricultural tractors in the frequencies lower than 50Hz. A dynamic model of cab motions was developed and verified using a tractor excited over half-sine bumps on a concrete test road. A simulation program based on the model was also developed. A method was proposed to determine the optimum values of the suspension parameters. It was found that the natural frequencies of the cab and seat suspensions must be apart as far as possible until the sum of seat and cab accelerations is minimized, which also reduces the seat accelerations maximally.

  • When a truck cab is conveyed at a constant speed by a hanger and immersed into the painting reservoir, it may fall off from the hanger by buoyancy. In order to reduce the buoyancy, on the bottom of a cab panel are holes placed, which allow paint to flow into the inside of a cab. In this study, a differential equation is derived which can be solved numerically by using 4th-Order Runge-Kutta method to calculate transient behavior of the buoyant force with sizes and locations of the holes given. The solution is utilized to optimally determine sizes and locations of the holes.

Study on drag reduction of commercial vehicle using flow control device (유동 제어 장치를 이용한 상용차량의 항력저감 연구)

  • The primary challenge in improving fuel efficiency and reducing air pollution for commercial vehicles is reducing their aerodynamic drag. Various flow control devices, such as cab-roof fairing, gap fairing, cab extender, and side skirt have been introduced to reduce drag, however, the drag reduction effect and applicability are different depending on each commercial vehicle model. To evaluate the fuel consumption of heavy vehicles, a comprehensive research approach, including drag force measurement, flow field analysis is required. This study investigated the effect of a cab extender, which installed rear region of cab, on a drag coefficient of commercial vehicle through wind tunnel experiments and CFD. The results showed that the cab extender significantly modified the flow structure around the vehicle, leading to 8.2% reduction in drag coefficient compared to the original vehicle model. These results would provide practical application for enhancing the aerodynamic performance and fuel efficiency of heavy vehicle.

Bioactive Characterization of Bacillus thuriniensis subsp. kurstaki CAB133 Isolated from Domestic Soil (국내 토양으로 분리된 Bacillus thuriniensis subsp. kurstaki CAB133균주의 생물학적 특성)

  • To screen highly active Bacillus thuringiensis isolates against Spodoptera litura (Lepidoptera, Noctuidae), 46 B. thuringiensis was isolated from 115 samples obtained from several crop soils. Especially, B. thuringiensis subsp. kurstaki CAB133 and CAB162 isolates showed 100% mortality against S. litura. $LD_{50}$ values of CAB 133, CAB162 and HD-1 strains of B. thuringiensis subsp. kurstaki were 0.089, 3.144 and $0.513{\mu}g/ml$ against 2nd larva of S. litura, respectively. The weight of 3rd larva of S. litura which were fed crystal inclusion protein $(1.267{\mu}g/ml)$ with B. thuringiensis subsp. kurstaki CAB133 was about 30 times lass than control group. CAB133 and CAB 162 strains of B. thuringiensis subsp. kurstaki which were taken a highly toxity against S. litura were analyzed by SDS-PAGE, and estimated the molecular weight of the Cry proteins. Their serological identification by H serotypes were showed B. thuringiensis subsp. kurstaki (3abc) type.
