In vitro Anti-oxidant and Anti-inflammatory Effects of Moringa Folium and Eucommiae Cortex 2:1 (g/g) Mixed Formula (모링가:두충 2:1 (g:g) 복합 조성물의 in vitro 항산화 및 항염 효능 연구)

  • The purpose of study is to evaluate in vitro anti-oxidant and anti-inflammatory effects of Moringa Folium and Eucommiae Cortex 2:1 (g/g) mixtures (MEMix). HaCaT and human normal dermal fibroblast were treated with 0.01-1 mg/mL of MEMix to monitor cytotoxicity. Radical scavenging activities of MEMix were examined by DPPH assay. To explore anti-inflammatory effect, Raw 264.7 cells were pretreated with MEMix for 1h and subsequently exposed to LPS for 18h. NO release and cytotoxicity of Raw 264.7 cells were measured by adding Griess and MTT reagents, respectively. TNF-α, IL-1β, IL-6, and PGE2 productions were examined by ELISA. Immunoblot analysis was conducted to examine COX-2 expression in MEMix pretreated Raw 264.7 cells. Up to 1 mg/mL concentration, treatment of MEMix for 24 h did not affect normal dermal fibroblast viability and significantly reduced cell viability of HaCaT cells with no concentration dependency. MEMix increased DPPH radical scavenging activity with concentration dependency. Radical scavenging activities by 1 mg/mL of MEMix was comparable with 30 µM of trolox. Pretreatment of MEMix did not change the reduction of Raw 264.7 cell viability. Exposure of LPS in Raw 264.7 cells significantly increased NO, TNF-α, IL-1β, IL-6, and PGE2 productions, and MEMix pretreatment attenuated these productions by LPS concentration dependently. However, pretreatment with MEMix did not change COX-2 expression by LPS in Raw 264.7 cells. MEMix showed in vitro anti-oxidant and anti-inflammatory activities. MEMix would be useful candidate agent against inflammation.

Complete Genome and Calcium Carbonate Precipitation of Alkaliphilic Bacillus sp. AK13 for Self-Healing Concrete

  • Bacteria that are resistant to high temperatures and alkaline environments are essential for the biological repair of damaged concrete. Alkaliphilic and halotolerant Bacillus sp. AK13 was isolated from the rhizosphere of Miscanthus sacchariflorus. Unlike other tested Bacillus species, the AK13 strain grows at pH 13 and withstands 11% (w/v) NaCl. Growth of the AK13 strain at elevated pH without urea promoted calcium carbonate (CaCO3) formation. Irregular vaterite-like CaCO3 minerals that were tightly attached to cells were observed using field-emission scanning electron microscopy. Energy-dispersive X-ray spectrometry, confocal laser scanning microscopy, and X-ray diffraction analyses confirmed the presence of CaCO3 around the cell. Isotope ration mass spectrometry analysis confirmed that the majority of CO32- ions in the CaCO3 were produced by cellular respiration rather than being derived from atmospheric carbon dioxide. The minerals produced from calcium acetate-added growth medium formed smaller crystals than those formed in calcium lactate-added medium. Strain AK13 appears to heal cracks on mortar specimens when applied as a pelletized spore powder. Alkaliphilic Bacillus sp. AK13 is a promising candidate for self-healing agents in concrete.

Eugenol Inhibits ATP-induced P2X Currents in Trigeminal Ganglion Neurons

  • Eugenol is widely used in dentistry to relieve pain. We have recently demonstrated voltage-gated $Na^+$ and $Ca^{2+}$ channels as molecular targets for its analgesic effects, and hypothesized that eugenol acts on $P2X_3$, another pain receptor expressed in trigeminal ganglion (TG), and tested the effects of eugenol by whole-cell patch clamp and $Ca^{2+}$ imaging techniques. In the present study, we investigated whether eugenol would modulate 5'-triphosphate (ATP)-induced currents in rat TG neurons and $P2X_3$-expressing human embryonic kidney (HEK) 293 cells. ATP-induced currents in TG neurons exhibited electrophysiological properties similar to those in HEK293 cells, and both ATP- and $\alpha$, $\beta$-meATP-induced currents in TG neurons were effectively blocked by TNP-ATP, suggesting that $P2X_3$ mediates the majority of ATP-induced currents in TG neurons. Eugenol inhibited ATP-induced currents in both capsaicin-sensitive and capsaicin-insensitive TG neurons with similar extent, and most ATP-responsive neurons were IB4-positive. Eugenol inhibited not only $Ca^{2+}$ transients evoked by $\alpha$, $\beta$-meATP, the selective $P2X_3$ agonist, in capsaicin-insensitive TG neurons, but also ATP-induced currents in $P2X_3$-expressing HEK293 cells without co-expression of transient receptor potential vanilloid 1 (TRPV1). We suggest, therefore, that eugenol inhibits $P2X_3$ currents in a TRPV1-independent manner, which contributes to its analgesic effect.

Sprouted Black Rice Oligopeptide Induces Expression of Hyaluronan Synthase in HaCaT Keratinocytes and Improves Skin Elasticity (발아 검은쌀 올리고펩타이드의 각질형성세포에서 Hyaluronan Synthase 발현과 피부 탄력 개선 효과)

  • In this study, the effect of oligopeptide ($Oligosproutin^{(R)}$) from sprouted black rice was evaluated for possible improvement in skin elasticity. We examined the changes in gene expression on oligopeptide-treated HaCaT cells using DNA microarray analysis. As a result, oligopeptide treatment showed a differential expression ratio of more than 2-fold : 745 genes were activated and 1011 genes were repressed. One of the most interesting findings is a 2-fold increase in hyaluronan synthase 2 (HAS 2) gene expression by oligopeptide. We also found that oligopeptide increased cell proliferation, HAS2 mRNA expression and intracellular ROS scavenging activity in HaCaT cells. A human clinical study which oil-in-water emulsion with oligopeptide was topically applied showed significant increase in skin elasticity. These results suggest that the sprouted black rice oligopeptide ($Oligosproutin^{(R)}$) can be effective anti-aging ingredient for cosmetics.

Effects of fermented black ginseng on wound healing mediated by angiogenesis through the mitogen-activated protein kinase pathway in human umbilical vein endothelial cells

  • Background: Fermented black ginseng (FBG) is produced through several cycles of steam treatment of raw ginseng, at which point its color turns black. During this process, the original ginsenoside components of raw ginseng (e.g., Re, Rg1, Rb1, Rc, and Rb2) are altered, and less-polar ginsenosides are generated (e.g., Rg3, Rg5, Rk1, and Rh4). The aim of this study was to determine the effect of FBG on wound healing. Methods: The effects of FBG on tube formation and on scratch wound healing were measured using human umbilical vein endothelial cells (HUVECs) and HaCaT cells, respectively. Protein phosphorylation of mitogen-activated protein kinase was evaluated via Western blotting. Finally, the wound-healing effects of FBG were assessed using an experimental cutaneous wounds model in mice. Results and Conclusion: The results showed that FBG enhanced the tube formation in HUVECs and migration in HaCaT cells. Western blot analysis revealed that FBG stimulated the phosphorylation of p38 and extracellular signal-regulated kinase in HaCaT cells. Moreover, mice treated with $25{\mu}g/mL$ of FBG exhibited faster wound closure than the control mice did in the experimental cutaneous wounds model in mice.

  • In this study, urethane acrylate macromer was synthesized and it was used in a gel polymer electrolyte (GPE), and then its electrochemical performances were evaluated. LiCoO$_2$/GPE/graphite cells were Prepared and their performances depending on discharge currents and temperatures were evaluated. The precursor consisting of urethane acrylate (UA), hexanediol dimethacrylate (HDDA) and benzoyl peroxide (BPO) had a low viscosity relatively ionic conductivity of the gel polymer electrolyte with UA at room temperature and -20$\^{C}$ was ca. 4.5 $\times$ 10$\^$-3/S$.$cm$\^$-1/ and 1.7 x 10$\^$-3/ S$.$cm$\^$-1/, respectively GPR was stable electrochemically up to potential of 4.i V vs. Li/Li$\^$+/. LiCoO$_2$/GPE/graphite cells showed good a high-rate and a low-temperature performance.

  • In this study, gel polymer electrolytes (GPE) with semi-interpenerating network of poly (methyl methacrylate) and hexanediol dimethacrylate were synthesized and their electrochemical performances were evaluated. LiCoO$_2$/GPE/graphite cells were prepared and their performances depending on discharge currents and temperatures were evaluated. The precursor containing 5 vol% curable mixture had a low viscosity relatively. GPE showed good electrochemical stability up to potential of 4.8 V vs. Li/Li$\^$+/. Ionic conductivity of the gel polymer electrolyte at room temperature and -20$^{\circ}C$ was ca. 5.9 and 1.4${\times}$10$\^$-3/ Scm$\^$-1/, respectively. LiCoO$_2$/GPE/graphite cells showed good rate capability, low-temperature performance and cycleability.

Water Extract of Kudzu Root (Pueraria radix) Decreases Apolipoprotein B100 and B48 Production in Vitro

  • We have previously demonstrated that kudzu root extracts have a hypocholesterolemic effect on rats fed diets high in fat and cholesterol. To further elucidate the mechanism involved, in this study we investigated the effect of water extracts of kudzu root, Pueraria radix, on the production of apolipoprotein B$_{100}$ (APo B$_{100}$) in HepG$_2$ liver cells and secretion of apolipoprotein B$_{48}$ (Apo B$_{48}$) in Caco$_2$ cells. Human cell lines, HepG$_2$ liver cells and Caco$_2$ intestinal epithelial cells, were grown with various concentrations (0%, 0.5%, 1.0%, 1.5%, 2.0%) of water extracts of kudzu root in the media. The kudzu root extract decreased Apo B$_{100}$ production and secretion. Treatment of HeP G$_2$ cells with the kudzu root extract also significantly decreased the intracellular total and free cholesterol concentration, and also decreased esterified cholesterol but was only significant at the highest dose of 2%. Apo B$_{48}$ production, but not secretion, from enterocytes was lowered by the kudzu root extracts. This research provided evidence that the hypocholesterolemic properties of kudzu root may be a consequence of decreased production and secretion of Apo B$_{100}$ in the liver and Apo B$_{48}$ in the intestine.

Activation of G Proteins by Aluminum Fluoride Enhances RANKL-Mediated Osteoclastogenesis

  • Receptor activator of NF-${\kappa}B$ ligand (RANKL)-induced osteoclastogenesis is accompanied by intracellular $Ca^{2+}$ mobilization in a form of oscillations, which plays essential roles by activating sequentially $Ca^{2+}$/calmodulin-dependent protein kinase, calcineurin and NFATc1, necessary in the osteoclast differentiation. However, it is not known whether $Ca^{2+}$ mobilization which is evoked in RANKL-independent way induces to differentiate into osteoclasts. In present study, we investigated $Ca^{2+}$ mobilization induced by aluminum fluoride ($AlF_4^-$), a G-protein activator, with or without RANKL and the effects of $AlF_4^-$ on the osteoclastogenesis in primary cultured mouse bone marrow-derived macrophages (BMMs). We show here that $AlF_4^-$ induces intracellular $Ca^{2+}$ concentration ($[Ca^{2+}]_i$) oscillations, which is dependent on extracellular $Ca^{2+}$ influx. Notably, co-stimulation of $AlF_4^-$ with RANKL resulted in enhanced NFATc1 expression and formation of tartrate-resistant acid phosphatase (TRAP) positive multinucleated cells. Additionally, we confirmed that mitogen-activated protein kinase (MAPK) is also activated by $AlF_4^-$. Taken together, these results demonstrate that G-protein would be a novel modulator responsible for $[Ca^{2+}]_i$ oscillations and MAPK activation which lead to enhancement of RANKL-mediated osteoclastogenesis.

Effects of Rubus coreanus and Artemisia princeps Extracts on the Ultraviolet B-Induced DNA Damage Responses in HaCaT Cells (HaCaT 세포에서 자외선 B에 의해 유도된 DNA 상해반응에 대한 복분자와 쑥 추출물의 효과)

  • We investigated the effects of extracts from Rubus coreanus (RC) and Artemisia princeps var. orientalis (AP) on DNA damage response in ultraviolet B (UVB)-exposed HaCaT cells. Cell activity upon treatment for 24 h with RC or AP alone was similar to or greater than that of the nontreated control. When UVB-exposed cells were postincubated for 24 h in medium containing RC or AP, cell activity increased in a concentration-dependent manner. Nuclear fragmentation analysis showed that postincubation with RC or AP decreased UVB-induced apoptosis by about 20% and 15%, respectively, of that in cells postincubated with growth medium. When UVB-exposed cells were postincubated for 24 h in medium containing RC or AP, the level of cyclobutane pyrimidine dimer decreased in a concentration- dependent manner. Western blot analysis showed that treatment of cells not exposed to UVB with RC or AP alone did not significantly change the levels of phospho-p53 and GADD45 protein. Interestingly, when UVB-exposed cells were postincubated for 24 h in medium containing RC or AP, phospho-p53 and GADD45 levels decreased in a concentration dependent manner. Our results suggest that RC and AP extract assist the survival of UVB-exposed cells in parallel with a decrease in levels of UVB-induced DNA damage and damage-response proteins, such as p53 and GADD45.
