꾸지뽕 신초 엽위별 잎 추출물의 항비만 효과 (Leaves of Cudrania tricuspidata on the Shoot Positional Sequence Show Different Inhibition of Adipogenesis Activity in 3T3-L1 Cells)

  • 본 연구는 꾸지뽕 신초에 부착된 잎을 6단계 즉 정아의 잎(L0), 정아부에서 하부의 순서대로 L1, L2, L3, L4 및 기부의 잎을 L5로 분류하여 채취하였다. 신초의 위치에 따라서 분류한 잎을 70% 에틸알콜로 추출한 다음 3T3-L1 cell line을 이용하여 지방분화 억제 효과를 스크린하고, 가장 효과가 좋았던 꾸지뽕 신초 정단잎(CTL0) 추출물의 작용기작을 규명하였다. 꾸지뽕 신초 정단잎(CTL0) 추출물의 지방분화 억제효과가 가장 좋았으며, 신초의 중간 잎인 CTL2까지는 정단 잎에서부터 중간 잎 추출물일수록 효과가 감소하였으며, 신초의 중간 하부 잎인 CTL3부터 기부 잎인 CTL5 추출물은 100 ㎍/ml의 고농도에서 효과가 없었다. 꾸지뽕 신초 정단잎(CTL0) 추출물을 12.5, 25 및 50 ㎍/ml의 농도로 처리하여 Oil Red O를 분석한 결과, 농도 의존적으로 3T3-L1세포의 지방분화를 억제시켰다. 3T3-L1 세포에 지방분화물질인 insulin, dexamethasone 및 rosiglitazone을 첨가한 분화배지(DM)에서는 배지내에 glucose의 함량이 낮았으나, CTL0 추출물의 처리는 배지에 glucose함량이 많이 남아있었기 때문에 세포내로 glucose의 흡수를 억제시켰으며, 세포내 중성지방의 함량도 감소하였다. CTL0처리는 전구지방세포에서 지방세포로 분화될 때에 주요 전사인자로 알려진 PPARγ와 PPARγ의 target 유전자인 LPL, A-FABP 및 Glut4 단백질의 발현을 농도 의존적으로 억제하였다. 따라서 꾸지뽕 신초 정단잎(CTL0) 추출물은 항비만 효과가 우수한 소재로서 활용가치가 높을 것으로 사료된다.

Effects of Annatto (Bixa orellana L.) Seeds Powder on Physicochemical Properties, Antioxidant and Antimicrobial Activities of Pork Patties during Refrigerated Storage

  • This study investigated the effect of the powder produced by ball-milling the outer layer of annatto (Bixa orellana L.) seeds on the physicochemical properties as well as the antioxidant and antimicrobial activities of pork patties over 14 d of refrigerated storage (4±1℃). Five pork patty treatments were produced containing three different concentrations of annatto seeds, 0.1, 0.25 and 0.5% (ANT0.1, ANT0.25, ANT0.5), 0.1% ascorbic acid (AA0.1), and a control (CTL). Based on the results, annatto seed powder appeared to show antioxidant activity. The Hunter color values of pork patties were affected by the addition of annatto seed powder, which increased the redness and yellowness values, but decreased the lightness of the patties (p<0.05). To evaluate the antioxidative effects of annatto on pork patties, thiobarbituric acid reactive substances (TBARS) and peroxide values (POV) were analyzed over 14 d of refrigerated storage. Treatments containing annatto seed showed lower TBARS and POV than control (CTL) samples (p<0.05). The volatile basic nitrogen (VBN) of the pork patties containing annatto seeds were lower than that of CTL at the end of storage (p<0.05). Although no differences in total bacterial counts were observed between control and treated patties, those containing annatto seeds had lower microbial counts for Enterobacteriacease than CTL or AA 0.1%. Therefore, annatto seed powder might be a good source of natural antioxidants for the production of meat products.

Characterization of CTL Clones Specific for Single Antigen, H60 Minor Histocompatibility Antigen

  • Background: Disparities of Minor H antigens can induce graft rejection after MHC-matched transplantation. H60 has been characterized as a dominant antigen expressed on hematopoietic cells and considered to be an ideal model antigen for study on graft-versus-leukemia effect. Methods: Splenocytes from C57BL/6 mice immunized with H60 congenic splenocytes were used for establishment of H60-specific CTL clones. Then the clones were characterized for proliferation capacity and cytotoxicity after stimulation with H60. Clone #14, #15, and #23 were tested for the TCR binding avidity to H60-peptide/$H-2K^b$ and analyzed for TCR sequences. Results: H60-specific CTL clones showed different levels of proliferation capacity and cytotoxic activity to H60-stimulation. Clones #14, #15, and #23 showed high proliferation activity, high cytotoxicity, and low activities on both aspects, respectively, and have TCRs with different binding avidities to H60-peptide/$H-2K^b$ with $t_{1/2}$ values of 4.87, 6.92, and 13.03 minutes, respectively. The TCR usages were $V{\alpha}12D-3-01+J{\alpha}11-01$ and $V{\beta}12-1-01+D{\beta}1-01+J2-7-01$ for clone #14, $V{\alpha}13D-1-02+J{\alpha}34-02$ and $V{\beta}13-1-02+D{\beta}2-01+J{\beta}2-7-01$ for clone #15, and $V{\alpha}16D+J{\alpha}45-01$ and $V{\beta}12-1-01+D{\beta}1-01+J{\beta}2-5-01$ for clone #23. Conclusion: The results will be useful for modeling GVL and generation TCR transgenic mouse.

Immunomodulatory activity of phytoprotein isol Acanthopanax senticosus : Regulation of CTL responses and activation of macrop

  • We previously reported that the phytoprotein extracted from Acanthopanax senticosus (PA) had anti-carcinogenic anti-metastatic activity via increase of inhibition of gap junctional intercellular communication. In the present study investigated the immunomodulatory mechanism of phytoprotein isolated from the stem bark of Acanthopanax sentic (PA). PA was found to significantly stimulate macrophages producing TNF-α and IL-1β in vitro. It also showed tumori activity indicating that PA had the ability to stimulate macrophage directly. Moreover, PA induced the CDB/sup +/ CTL cy responses to recognize antigen on the B16-BL6 melanoma cells. Treatment of PA with B16-BL6 melanoma cells increased the proliferation of splenocytes compared with untreated control. These results demonstrate that PA immunomodulatory activity suggesting a useful anti-tumor agent applicable to treatment and prevention of cancer.

산화적 손상에 대한 꾸지뽕나무 잎, 열매 및 줄기 추출물의 간세포 보호효과 (Hepatoprotective Effect of Curdrania tricuspidata Extracts against Oxidative Damage)

  • 본 연구에서는 산화적 스트레스에 의한 간 손상 개선 소재 개발을 위하여 꾸지뽕나무 각 부위별(잎, 줄기, 열매), 용매별(80% 에탄올, 10% 에탄올, 물) 추출물의 항산화 활성 및 간세포 보호효과를 측정하였다. 총 폴리페놀 함량과 플라보노이드 함량은 꾸지뽕 잎 80% 에탄올 추출물에서 가장 높게 나타났으며, 부위별로는 잎>줄기>열매 순이었고, 추출용매별로는 80% 에탄올>10% 에탄올>물 추출물 순으로 나타났다. DPPH 라디칼 소거능과 ABTS 라디칼 소거능 또한 잎80% 에탄올 추출물이 가장 높았으며, HepG2 세포에서 $H_2O_2$로 유도된 산화적 손상에 대해서는 꾸지뽕 잎 80% 에탄올 추출물만 유의적으로 높은 세포보호활성을 나타내었으며, HepG2/2E1 세포에서 알코올로 유도된 산화적 손상에 대한 각 부위별, 용매별 추출물의 간세포보호효과 또한 꾸지뽕잎 80% 에탄올 추출물이 가장 높게 나타났다. 부위별로는 잎>줄기>열매 순이었고, 추출 용매별로는 꾸지뽕 잎의 경우 80% 에탄올>10% 에탄올>물 순이었으며, 줄기와 열매의 경우는 용매별로 유의적인 차이가 나타나지 않았다. 이상의 결과로부터 꾸지뽕나무 잎 추출물은 우수한 항산화활성을 가질 뿐만 아니라 $H_2O_2$와 알코올로 유도된 간 손상으로부터 간세포 보호활성을 보임을 확인하였다. 이에 꾸지뽕나무 잎 추출물은 산화적 스트레스에 의한 간 손상으로부터 간세포 보호효과를 갖는 기능성 소재로 활용될 수 있을 것으로 사료된다.

Vanilloid Receptor 1 Agonists, Capsaicin and Resiniferatoxin, Enhance MHC Class I-restricted Viral Antigen Presentation in Virus-infected Dendritic Cells

  • DCs, like the sensory neurons, express vanilloid receptor 1 (VR1). Here we demonstrate that the VR1 agonists, capsaicin (CP) and resiniferatoxin (RTX), enhance antiviral CTL responses by increasing MHC class I-restricted viral antigen presentation in dendritic cells (DCs). Bone marrow-derived DCs (BM-DCs) were infected with a recombinant vaccinia virus (VV) expressing OVA (VV-OVA), and then treated with CP or RTX. Both CP and RTX increased MHC class I-restricted presentation of virus-encoded endogenous OVA in BM-DCs. Oral administration of CP or RTX significantly increased MHC class I-restricted OVA presentation by splenic and lymph node DCs in VV-OVA-infected mice, as assessed by directly measuring OVA peptide SIINFEKL-Kb complexes on the cell surface and by performing functional assays using OVA-specific CD8 T cells. Accordingly, oral administration of CP or RTX elicited potent OVA-specific CTL activity in VV-OVA-infected mice. The results from this study demonstrate that VR1 agonists enhance anti-viral CTL responses, as well as a neuro-immune connection in anti-viral immune responses.

Effects of Drying Temperature on Antioxidant Activities of Tomato Powder and Storage Stability of Pork Patties

  • This study was performed to evaluate the antioxidant activity of oven-dried tomato powder (OTP) as affected by drying temperature and the effect of OTP on the product quality of pork patties. Three OTP products were obtained by drying of fresh tomato at 60, 80 and 100℃ oven until constant weight was obtained. Total phenolic content of three kinds of OTPs ranged from 1.95 to 5.94 g/100 g. The highest amount of total phenolic compound was observed in OTP dried at 100℃. Antioxidant activity of three kinds of OTPs was measured by 1,1-diphenyl-2-pycrylhydrazyl (DPPH)-radical scavenging activity, iron chelating ability, reducing power and measurement of lipid peroxide in linoleic acid emulsion system. In all parameters, OTP at 100℃ showed the higher antioxidant activity than other temperatures (p<0.05). Based on the model study, the physicochemical properties, and antioxidant and antimicrobial activities of pork patties containing 1% OTP were measured. Redness of pork patties were increased with the addition of OTPs (p<0.05). Thiobarbituric acid reactive substances (TBARS) values of raw pork patties containing OTPs were lower than those of control (CTL) until 7 d of storage, regardless of drying temperatures (p<0.05). Peroxide values of pork patties made with OTP (1%) were lower than those of CTL until the end of storage time (p<0.05). However, no antimicrobial activities were observed among the treatments (p>0.05). Therefore, OTPs could be used as a natural antioxidant in meat products.

Protective Antitumor Activity through Dendritic Cell Immunization is Mediated by NK Cell as Well as CTL Activation

  • Dendritic cells (DCs) are potent professional antigen-presenting cells (APC) capable of inducing the primary T cell response to antigen. Although tumor cells express target antigens, they are incapable of stimulating a tumor-specific immune response due to a defect in the costimulatory signal that is required for optimal activation of T cells. In this work, we describe a new approach using tumor-DC coculture to improve the antigen presenting capacity of tumor cells which does not require a source of tumor-associated antigen. Immunization of a weakly immunogenic and progressive tumor cocultured with none marrow-derived DCs generated an effective tumor vaccine. Immunization with the cocutured DCs was able to induce complete protectiv immunity against tumor challenges and was effective for the induction of tumor-specific CTL (cytotoxic T lymphocyte) activity. Furthermore, high NK cell activity was observed in mice in which tumors were rejected. In addition, immunization with tumor-pulsed DC s induced delayed tumor growth, but not tumor eradication in tumor-bearing mice. Our results demonstrate that coculture of DCs with tumors generated antitumor immunity due to the NK cell activation as well as tumor-specific T cell. This approach would be used for designing tumor vaccines using DCs when the information about tumor antigens is limited.

  • Hepatitis C virus (HCV) is a positive strand RNA virus of the Flaviviridae family and the major cause of post-transfusion non-A, non-B hepatitis. Vaccine development for HCV is essential but has been slowed by poor understanding of the type of immunity that naturally terminates HCV infection. The DNA-based immunization technique offers the potential advantage of including cellular immune responses against conserved internal proteins of a virus, as well as the generation of antibodies to viral surface proteins. Here, we demonstrate that cell lines expressing the HCV core and/or NS3 proteins can induce a specific CTL response in mice, and these results suggest a possibility that the HCV core and NS3 DNA can be used to induce CTL activity against the antigen in mice and can be further developed as a therapeutic and preventive DNA vaccine.

마우스 EAE, GVHD 질환에서 CTLA4Ig 융합단백의 면역치료 효과 (Immunotherapeutic Effects of CTLA4Ig Fusion Protein on Murine EAE and GVHD)

  • Background: CTLA4 (CD152), which is expressed on the surface of T cells following activation, has a much higher affinity for B7 molecules comparing to CD28, and is a negative regulator of T cell activation. In contrast to stimulating and agonistic capabilities of monoclonal antibodies specific to CTLA-4, CTLA4Ig fusion protein appears to act as CD28 antagonist and inhibits in vitro and in vivo T cell priming in variety of immunological conditions. We've set out to confirm whether inhibition of the CD28-B7 costimulatory response using a soluble form of human CTLA4Ig fusion protein would lead to persistent inhibition of alloreactive T cell activation. Methods: We have used CHO-$dhfr^-$ cell-line to produce CTLA4Ig fusion protein. After serum free culture of transfected cell line we purified this recombinant molecule by using protein A column. To confirm characterization of fusion protein, we carried out a series of Western blot, SDS-PAGE and silver staining analyses. We have also investigated the efficacy of CTLA4Ig in vitro such as mixed lymphocyte reaction (MLR) & cytotoxic T lymphocyte (CTL) response and in vivo such as experimental autoimmune encephalomyelitis (EAE), graft versus host disease (GVHD) and skin-graft whether this fusion protein could inhibit alloreactive T cell activation and lead to immunosuppression of activated T cell. Results: In vitro assay, CTLA4Ig fusion protein inhibited immune response in T cell-specific manner: 1) Human CTLA4Ig inhibited allogeneic stimulation in murine MLR; 2) CTLA4Ig prevented the specific killing activity of CTL. In vivo assay, human CTLA4Ig revealed the capacities to induce alloantigen-specific hyporesponsiveness in mouse model: 1) GVHD was efficiently blocked by dose-dependent manner; 2) Clinical score of EAE was significantly decreased compared to nomal control; 3) The time of skin-graft rejection was not different between CTLA4Ig treated and control group. Conclusion: Human CTLA4Ig suppress the T cell-mediated immune response and efficiently inhibit the EAE, GVHD in mouse model. The mechanism of T cell suppression by human CTLA4Ig fusion protein may be originated from the suppression of activity of cytotoxic T cell. Human CTLA4Ig could not suppress the rejection in mouse skin-graft, this finding suggests that other mechanism except the suppression of cytotoxic T cell may exist on the suppression of graft rejection.
