This study is conducted to investigate the influence of addition of quarternary ammonium salt(cetyl trimethyl ammonium bromide: CTAB) when polyester(PET) fabric is treated with sodium hydroxide(NaOH), depending on experimental variables such as CTAB concentration, NaOH concentration, time & temperature, and the change in physical & chemical properties of alkaline-hydrolyzed PET fabrics depending on their weight loss. The results are as follows: 1. By adding CTAB in aqueous NaOH, the weight loss of PET fabric is increased remarkably and until the concentration of CTAB is reached at its cmc, and the higher the concentration of CTAB are, the more weight loss on PET fibrics are. 2. The addition of CTAB in aqueous NaOH is most effective at lower NaON concentration(2%) among various NaOH concentration, on increasing the amount of weight loss, while there are almost similar results through various treatment time and temperature. 3. As the amount of weight of weight loss on PET fabric is increased, the increase of void space in the PET yarn, of softness & dyeability of PET fabric and the decrease of tensile strength are found. On the other hand, the moisture regain shows a little increase by alkaline-hydrolysis on PET fabric while vertical absorption test & water retention value are not sufficiently sensitive to distinguish between the hydrophillicity of untreated and treated PET fabric. The shrinkage of PET fabric is induced by swelling in hot aqueous NaOH regardless of NaOH concentration & addition of CTAB.
The inhibition performance of sodium gluconate (SG), cetyltrimethylammonium bromide (CTAB) and their mixture (SG/CTAB) on the corrosion behavior of ${\alpha}$-brass in 0.5 M $H_2SO_4$ solution has been investigated by potentiodynamic polarization, electrochemical impedance spectroscopy (EIS), Scanning Electron Microscope with Energy-Dispersive Spectrometer (SEM-EDS), Inductively Coupled Plasma Spectrometry (ICPS) and molecular dynamics (MD) simulation techniques. The results reveal that SG with 5ppm CTAB, noted SG/CTAB, acts as a good corrosion inhibitor and its inhibition efficiency reached 89% after 24 h immersion in sulfuric acid solution, but slightly decreased at higher temperatures. The polarization curves displayed that SG/CTAB acts as a cathodic-kind inhibitor. Electrochemical impedance spectroscopy (EIS) studies revealed that the addition of 5ppm CTAB to different concentrations of SG considerably increases the corrosion resistance of ${\alpha}$-brass. The SEM-EDS and ICPS analyses support the experimental results. Further, molecular dynamics (MD) simulations were used to understand the adsorption profiles of SG/CTAB on Cu(111) and Zn(111) surfaces.
본 연구에서는 견과류로부터 식품 분석에 사용될 DNA를 4가지 방법으로 추출하고 그 효율을 비교하였다. 동일한 양의 시료를 사용하여 추출된 DNA의 양은 CTAB법이 가장 우수한 것으로 확인되었지만, 추출 시간이 수배이상 오래 걸리고 유기용매를 사용한다는 한계점이 있다. 다른 방법들과 DNA 추출 양의 차이가 큰 잣, 캐슈너트, 피스타치오 너트, 땅콩의 시료는 CTAB법이 가장 효율적인 방법으로 판단되며, 호두, 헤이즐넛, 아몬드의 시료는 변형 CTAB법과 실리카 막법이 CTAB법을 대체할 수 있을 것으로 판단된다. 추출된 DNA를 식물 내재유전자 및 각 견과류에 특이적인 유전자를 사용하여 PCR을 진행하였으며, 모든 추출 방법에서 DNA가 정상적으로 증폭되는 것을 확인하였다.
Quercus infectoria (nutgall) has been reported to possess antimicrobial activities against a wide range of pathogens. Nevertheless, the biofilm removal effect of nutgall extract has not been widely investigated. In this study, we therefore evaluated the effect of nutgall extract in combination with cetrimonium bromide (CTAB) against preformed biofilm of Salmonella Typhimurium on polypropylene (PP) and stainless steel (SS) coupons in comparison with other sanitizers. The minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) of nutgall extract and surfactants (CTAB and sodium dodecyl sulfate; SDS) were assessed. CTAB showed a more efficient antimicrobial activity than SDS and was selected to use in combination with nutgall extract for removing biofilm. To determine the biofilm removal efficacy, the PP and SS coupons were individually submerged in 2x MBC of nutgall extract (256 mg/ml) + 2x MBC of CTAB (2.5 mg/ml), nutgall extract alone (256 mg/ml), CTAB alone (2.5 mg/ml), distilled water, and 100 ppm sodium hypochlorite for 5, 15, and 30 min. The remaining sessile cells in biofilm were determined. Overall, the greatest biofilm removal efficacy was observed with nutgall extract + CTAB; the biofilm removal efficacy of sanitizers tended to increase with the exposure time. The SEM analysis demonstrated that S. Typhimurium biofilm on PP and SS coupons after exposure to nutgall extract + CTAB for 30 min displayed morphological alterations with wrinkles. This study suggests nutgall extract + CTAB may be an alternative to commonly used sanitizers to remove biofilm from food contact surfaces in the food industry and household.
A simplified but reliable method was developed for the polymerase chain reaction (PCR)-based detection of genetically modified (GM) plants. The modified CTAB (mCTAB) method enabled us to prepare a high quality of genomic DNA from several hundred plant leaf samples in one day. Using DNA samples prepared from seven dicots and two monocots, approximately 1.75-kb regions spanning 17 S to 25 S ribosomal RNA genes were successfully amplified in a 2X PCR pre-mix containing BLOTTO. Further fidelity assessment of the mCTAB method by PCR analysis with Roundup Ready soybean (RRS) and non-RRS plants showed that the DNA samples prepared alternately from each of two lines were evidently free of cross-contamination. These results demonstrate that the mCTAB method is highly recommended for the rapid detection of transgenes in large numbers of leaf samples from diverse transgenic plants.
CTAB/hexanol/isooctane 역미셀을 이용하여 단백질을 선택적으로 분리할 수 있는 분리조건을 밝히기 위해서 먼저 분자량과 등전점이 다른 여러 단백질의 용해와 회수에 미치는 pH, 이온의 종류 및 이온 강도, 계면활성제의 종류 등 system parameters에 대해 연구 검토하였다. pH에 따른 단백질의 용해는 등전점이 낮은 pepsin이 pH5-7 이상, 등전점이 높은 lisozyme이나 ribonuclease-a는 pH11-12 이상으로 단백질이 계면활성제의 (+)전하와 반대전하인 (-)를 띠는 등전점 이상의 pH역역에서 용해가 가능하였다.
The purpose of this study is to investigate the change of the chemical properties of wool treated with six kinds of alkali (NaOH, Naac03, NH40H, NH2CH3CH30H, TMAH and BTMAH) with or without CTAB. Content of bound fatty acid liberated from wool surface, elemental composition and allw6rden time were measured to compare the surface modification of untreated and alkali treated wool. Also, the chemical degradation of the fiber was investigated by measuring cystine contents and urea-hydrogensulfite solubility. The result were as follows: 1. By the alkali treatment of wool, the covalently bound fatty acid of the epicuticle was removed and the allworden time was shortened, and in the case of wool treated with TMAH, BTMAH, the allw6rden sacs were formed unevenly and rarely. Also, cystine contents and urea-hydrogensulfite solubility were decreased by alkali treatment on wool. 2. The modification of epicuticle and the chemical degradation of wool were occurred due to alkaline hydrolysis in the order of TMAH, BTMAH > NaOH, Na3c03> NH2CH3CHaOH, NH40H. 3. As a treating time increased, the modification of epicuticle and chemical degradation of wool were accelerated. By the addition of CTAB to the alkali solution, the modification of epicuticle was increase, and the cystine contents and urea-hydrogensulfite solubility were reduced than that of wool teated with alkli without CTAB due to reduction of negative charge on the wool surface by the adhesion of CTAB.
The simple and convenient determination of trace Hg(II) with its 2-mercaptobenzothiazole (MBT) complex in cetyltrimethylammonium bromide (CTAB) media has been studied. The UV-visible spectrum of Hg(II)-MBT complex in CTAB media had a good sensitivity and reproducibility. The Hg(II)-MBT complex in CTAB solution was very stable at pH 10.0 and could be quantitatively complexed if MBT were added to the sample solution more than 10 equivalent of Hg(II). The optimum concentration of CTAB was 0.001%. The calibration curve of Hg(II)-MBT complex with good linearity ($R^2$ = 0.9985) was obtained at the concentration range between $1.0{\times}10^-7$ and $1.0{\times}10^-5$ M in 0.001% CTAB media. The detection limit was $3.1{\times}10^-8$ M (6.2 ng m$L^{-1}$). Hg(II) in the synthetic samples and in the wastewater of the university's wastewater tank and the industrial wastewater tank could also be determined. Based on the experimental results, this proposed technique could be applied to the simple and convenient determination of trace Hg(II) in real samples.
본 연구에서 분자육종을 위한 신속 DNA추출법을 개발하고자, 2% CTAB 추출법을 시스템적으로 개량한 Modified CTAB법에 대한 결과를 요약하면 아래와 같다. 1. DNA 추출용 샘플마쇄는 Voltex mixer, 3개 텅스텐 구슬 및 96 deep well block을 이용하였으며, 5분 이내에 96개 샘플을 분쇄할 수 있었다. 2. Modified CTAB DNA 추출법은 1.5 ml E-tube 대신에 1.2 ml 8연식 tube를 사용하였으며 DNA 추출, PCR작업 및 전기영동까지 일련의 작업이 8채널 멀티피펫을 이용할 수 있는 시스템적으로 개선하였다. 3. Modified CTAB법은 어린잎 뿐 만 아니라 이앙 후 30일째 채취한 잎에서도 DNA 추출이 가능하기 때문에 기존의 육종포장에 재식된 재료를 활용할 수 있는 장점이 있다.
CTAB/TTAB 혼합계면활성제 용액에서 Phenoxide 음이온의 가용화를 UV/Vis분광광도법을 이용하여 연구하였으며, 혼합계면활성제의 몰분율조성($$\alpha_{CTAB}$)에 따른 가용화상수(Ks)와 임계마이셀농도(CMC)의 변화를 측정하고 분석하였다. 또한 Phenoxide 음이온의 가용화에 영향을 미치는 n-펜탄을과 NaBr의 효과에 대하여 조사하였으며, 이러한 첨가제는 $K_s$와 CMC 값을 큰 폭으로 감소시키는 경향을 나타내었다·열역학적 고찰을 위하여 $K_s$ 값의 온도에 따른 변화를 측정하였으며, 온도에 따른 $K_s$ 값의 변화로부터 Phenoxide 음이온의 가용화에 대한 열역학 함수값(${\Delta}G^0_s$, ${\Delta}H^0_s$ 및 ${\Delta}S^0_s$)을 계산하였다·그 결과 ${\Delta}G^0_s$와 ${\Delta}H^0_s$은 측정한 범위 내에서 모두 음의 값을 그리고 ${\Delta}S^0_s$ 은 모두 양의 값을 나타내었다.
