Cloning of Geranylgeranyl Pyrophosphate Synthase (CrtE) Gene from Kocuria gwangalliensis and Its Functional Co-expression in Escherichia coli (코쿠리아 광안리엔시스의 제라닐제라닐 피로인산염 합성 효소의 클로닝과 대장균에서 공발현을 통한 효소 활성에 관한 연구)

  • A gene encoding a novel geranylgeranyl pyrophosphate (GGPP) synthase from Kocuria gwangalliensis has been cloned and expressed in Escherichia coli. The deduced amino acid sequence showed 59.6% identity with a putative GGPP synthase (CrtE) from K. rhizophila. An expression plasmid containing the crtE gene was constructed, and E. coli cells containing this plasmid produced a recombinant protein with a theoretical molecular mass of 41 kDa, corresponding to the molecular weight of GGPP synthase. Due to the lack of crtE, crtB, and crtI in E. coli, the biosynthesis of lycopene was only obtained when the plasmid pCcrtE was co-transformed into E. coli expressing the pRScrtBI-carrying carotenoid biosynthesis crtB and crtI genes, which were sub-cloned from Paracoccus haeundaensis. The biochemical studies on the expressed proteins were performed via HPLC. The results obtained from this study will provide a wider base of knowledge regarding the primary structure of CrtE cloned from K. gwangalliensis at the molecular level.

DCT Domain Zero-Watermarking based on CRT (CRT 기반의 DCT 영역 제로-워터마킹)

  • Zero-watermarking is a digital watermarking technique that can escape from data distortion and quality degradation by no watermark insertion into digital images to be watermarked. This paper proposes DCT0CRT, a CRT-based zero-watermarking technique in the domain of DCT. One of the DC and low-frequency AC coefficients of each DCT block chosen in chaotic way from an image is selected by testing whether it satisfies the CRT-based condition matching with the watermark bit to be embedded. Such selection information forms a key to extract the watermark from the watermarked image. Experimental results show that the image quality watermarked by DCT0CRT is better than that watermarked by any other CRT-based watermarking techniques and the inserted watermark is robust against some common attacks such as sharpening, blurring, and JPEG lossy compression.

Molecular Cloning of a cDNA Encoding Putative Calreticulin from the Silkworm, Bombyx mori

  • We describe here the cloning of a cDNA encoding putative calreticulin (CRT) from the silkworm, Bombyx mori. The CRT cDNA comprised of 1,194 bp encoding 398 amino acid residues. B. mori. CRT has a HDEL sequence at the end of the C-domain. The B. morl, CRT showed 88% protein sequence identity to the G. mellonella CRT, 71 % to A. aegypti CRT, and 63% to H. sapiens CRT, Phylogenetic analysis revealed that the deduced amino acid sequences of the B. mori CRT formed a highly inclusive subgroup with other insect CRTs. Northern blot analysis exhibited an expression of the B. mori CRT gene in the fat body, evidencing the fat body as a major site for CRT synthesis.

Secure RSA with CRT Protected Against Fault Attacks without using Checking Procedure (비교연산을 사용하지 않는 오류주입 공격에 안전한 CRT 기반의 RSA)

  • Because Chinese Remainder Theorem based RSA (RSA CRT) offers a faster version of modular exponentiation than ordinary repeated squaring, it is promoting with standard. Unfortunately there are major security issues associated with RSA CRT, since Bellcore announced a fault-based cryptanalysis against RSA CRT in 1996. In 1997, Shamir developed a countermeasure using error free immune checking procedure. And soon it became known that the this checking procedure can not effect as the countermeasures. Recently Yen proposed two hardware fault immune protocols for RSA CRT, and this two protocols do not assume the existence of checking procedure. However, in FDTC 2006, the method of attack against the Yen's two protocols was introduced. In this paper, the main purpose is to present a countermeasure against the method of attack from FDTC 2006 for CRT-RSA. The proposed countermeasure use a characteristic bit operation and dose not consider an additional operation.

Cloning and Characterization of the Zeaxanthin Glucosyltransferase Gene (crtX) from the Astaxanthin-Producing Marine Bacterium, Paracoccus haeundaensis

  • Zeaxanthin glucosyltransferase (CrtX) mediates the formation of zeaxanthin to zeaxanthin diglucoside. Here, we report cloning of the crtX gene responsible for zeaxanthin diglucoside biosynthesis from Paracoccus haeundaensis and the production of the corresponding carotenoids in transformed cells carrying this gene. An expression plasmid containing the crtX gene (pSTCRT-X) was constructed, and Escherichia coli cells containing this plasmid produced the recombinant protein of approximately 46 kDa. Biosynthesis of zeaxanthin diglucoside was obtained when the plasmid pSTCRT-X was co-transformed into E. coli containing the pET-44a(+)-CrtEBIYZ carrying crtE, crtB, crtI, crtY, and crtZ genes required for zeaxanthin $\beta$-D-diglucoside biosynthesis.

Computational Study to Understand the Cardiac Electromechanical Responses in LBBB and RBBB to the Application of CRT and LVAD

  • the aim of this study was to observe the combined effect of the CRT and LVAD on electromechanical cardiac behavior under LBBB and RBBB conditions computationally. We performed simulation by using advanced electromechanics model of failing ventricle combined with lumped model represents circulatory system, CRT and LVAD. We analyzed seven failing ventricle model including normal sinus rhythm, LBBB, LBBB coupled with CRT, LBBB coupled with CRT and LVAD, RBBB, RBBB coupled with CRT, and RBBB coupled with CRT and LVAD. We compared the effect from CRT and the effect from combined CRT and LVAD to both under LBBB and RBBB conditions. The results showed that the combined CRT and LVAD contributed a better hemodynamic compared to single CRT. This combined system synchronized the electrical activation greatly under LBBB and slightly under RBBB. It also shortened mechanical activation time which resulted short electromechanical delay. More importantly, the combined system produced better mechanical responses under both LBBB and RBBB conditions.

Molecular Cloning and Characterization of the Gene Encoding Phytoene Desaturase from Kocuria gwangalliensis (Kocuria gwangalliensis 유래 phytoene desaturase 유전자의 cloning과 특성 연구)

  • Carotenoids such as phytoene, lycopene, and ${\beta}-carotene$ are used as food colorants, animal feed supplements, and for human nutrition and cosmetic purposes. Previously, we reported the isolation of a novel marine bacterium, Kocuria gwangalliensis, which produces a pink-orange pigment. Phytoene desaturase (CrtI), encoded by the gene crtI, catalyzes lycopene formation from phytoene and is an essential enzyme in the early steps of carotenoid biosynthesis. CrtI is one of the key enzymes regulating carotenoid biosynthesis and has been implicated as a rate-limiting enzyme of the pathway in various carotenoid synthesizing organisms. Here, we report the cloning of the crtI gene responsible for lycopene biosynthesis from K. gwangalliensis. The gene consisted of 1,584 bases encoding 527 amino acid residues. The nucleotide sequence of the crtI gene was compared with that of other species, including Kocuria rhizophila and Myxococcus xanthus, and was found to be well conserved during evolution. An expression plasmid containing the crtI gene was constructed (pCcrt1), and Escherichia coli cells were transformed with this plasmid to produce a recombinant protein of approximately 57 kDa, corresponding to the molecular weight of phytoene desaturase. Lycopene biosynthesis was confirmed when the plasmid pCcrtI was co-transformed into E. coli containing the plasmid pRScrtEB carrying the crtE and crtB genes required for lycopene biosynthesis. The results from this study will provide valuable information on the primary structure of K. gwangalliensis CrtI at the molecular level.

Revisiting the Faking Issues of the CRT-A with Koreans

  • The Conditional Reasoning Test for Aggression (CRT-A) is an implicit assessment for aggression. The CRT-A is known to be resistant to response distortion. We revisited LeBreton et al.'s (2007) studies on the faking issues of the CRT-A with the Korean population. We divided 488 Korean college students into three groups and asked them to take the CRT-A under different instructions. Results showed that Koreans were able to identify aggressive alternatives when the purpose of the test was fully disclosed, and the students were less likely to select aggressive alternatives if they were told that the CRT-A was a personality test.

The Effects of Panel Convexity on Visual Performance and Fatigue in Using Cathode-Ray Tube (CRT) Displays (CRT 디스플레이의 패널곡률이 시각작업 수행도와 안피로도에 미치는 영향)

  • An experiment was carried out to compare the suitability in visual tasks between flat and conventional (convex) cathode-ray tube (CRT) displays. The subjects performed visual search tasks during 2-h for detecting target words among distracters presented on the screen. The subjects' visual performance was evaluated with average time and number of errors made to complete the tasks. Visual fatigue after the search tasks was also evaluated in terms of degradations in accommodative power and subjective ratings. Difference was not found in task time between the two displays, but flat CRT showed a lower number of errors than conventional CRT. The difference in number of errors was statistically significant at 0=0.05. Although there was no difference between the displays in degradations of accommodative power, results from the subjective ratings showed that flat CRT yields less fatigue than conventional CRT. The results partially support the hypothesis that panel convexity of CRT displays has a significant effect on the performance and fatigue during visual tasks and thus flat CRT is the better display than conventional one.

A Countermeasure Resistant to Fault Attacks on CRT-RSA using Fault Infective Method (오류 확산 기법을 이용한 CRT-RSA 오류 주입 공격 대응 방안)

  • Recently, the straightforward CRT-RSA was shown to be broken by fault attacks through many experimental results. In this paper, we analyze the fault attacks against CRT-RSA and their countermeasures, and then propose a new fault infective method resistant to the various fault attacks on CRT-RSA. In our CRT-RSA algorithm, if an error is injected in exponentiation with modulo p or q, then the error is spreaded by fault infective computation in CRT recombination operation. Our countermeasure doesn't have extra error detection procedure based on decision tests and doesn't use public parameter such as e. Also, the computational cost is effective compared to the previous secure countermeasures.
