Analysis of Gene Expression in Mouse Spinal Cord-derived Neural Precursor Cells During Neuronal Differentiation

  • The differentiation of neural precursor cells (NPCs) into neurons and astrocytes is a process that is tightly controlled by complicated and ill-defined gene networks. To extend our knowledge to gene networks, we performed a temporal analysis of gene expression during the differentiation (2, 4, and 8 days) of spinal cord-derived NPCs using oligonucleotide microarray technology. Out of 32,996 genes analyzed, 1878 exhibited significant changes in expression level (fold change>2, p<0.05) at least once throughout the differentiation process. These 1878 genes were classified into 12 groups by k-means clustering, based on their expression patterns. K-means clustering analysis revealed that the genes involved in astrogenesis were categorized into the clusters containing constantly upregulated genes, whereas the genes involved in neurogenesis were grouped to the cluster showing a sudden decrease in gene expression on Day 8. Functional analysis of the differentially expressed genes indicated the enrichment of genes for Pax6- NeuroD signaling.TGFb-SMAD and BMP-SMAD.which suggest the implication of these genes in the differentiation of NPCs and, in particular, key roles for Nova1 and TGFBR1 in the neurogenesis/astrogenesis of mouse spinal cord.

Distribution of PFOA and PFOS in Maternal Blood, Cord Blood and Breast Milk in Busan (부산 지역 임산부의 모체혈, 제대혈, 모유에서 PFOA, PFOS의 농도)

  • Objectives: Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) are man-made, persistent global pollutants widely diffused throughout the environment. They have been even found in the cord blood and breast milk of humans. Furthermore evidence of developmental toxicity in animals exists. To assess the distribution of maternal and fetal exposure to PFOS and PFOA, we analyzed paired maternal blood, cord blood and breast milk samples. Methods: Maternal blood, cord blood and breast milk were collected from 150 volunteers from the general population (aged 20-40, mean $30.5{\pm}2.9$) of the city of Busan in 2009-2010. The samples were extracted using the weak anion exchange and solid-phase extraction methods and quantified by high-performance liquid chromatograph (HPLC, Agilent 1200 Series) coupled with an Triple Quad LC-MS/MS system (Agilent 6410). Results: Median PFOA and PFOS concentrations in maternal blood were 2.18 and 3.32 ng/ml, in cord blood were 0.83 and 0.58 ng/ml, and in breast milk were 0.13 and 0.11 ng/ml, respectively. PFOS and PFOA concentrations were significantly correlated among matrices (Spearson's ${\rho}=0.226$, p = 0.05 for maternal blood; ${\rho}=0.736$, p < 0.01 for cord blood; ${\rho}=0.493$ p < 0.01 for breast milk). The ratio of cord blood/maternal blood was 0.39 for PFOA and 0.19 for PFOS. The ratio of breast milk/maternal blood was 0.07 for PFOA and 0.06 for PFOS. Conclusions: Our findings suggest that PFOA and PFOS exposure through the placenta was more prominent than through breast milk among Korean neonates born in Busan. The transfer efficiency of maternal blood to breast milk was similar between PFOA and PFOS, but that of maternal blood to cord blood was higher in PFOA than PFOS.

Auto-Walking Training After Incomplete Spinal Cord Injury (불완전 척수손상 후의 자동보행훈련)

  • This study was conducted to assess the effects of the gait training method in incomplete spinal cord injured persons using an auto-walking machine. Persons with incomplete spinal cord injury level C or D on the American Spinal Injury Association impairment scale participated for eight weeks in an auto-walking training program. The gait training program was carried out for 15 minutes, three times per day for 8 weeks with an auto-walking machine. The foot rests of the auto-walking machine can be moved forward, downward, backward and upward to make the gait pattern with fixed on crank. The patient's body weight is supported by a harness during waking training. We evaluated the gait speed, physiologic cost index, motor score of lower extremities and the WISCI (walking index for spinal cord injury) level before the training and after the forth and eighth week of walking training. 1. The mean gait speed was significantly increased from .22 m/s at pre-training to .28 m/s after 4 weeks of training and .31 m/s after 8 weeks of training (p=.004). 2. The mean physiologic cost index was decreased from 4.6 beats/min at pre-training to 3.0 beats/min after 4 weeks and 2.0 beats/min after 8 weeks of training, but it was not statistically significant (p=.140). 3. The mean motor score of lower extrernities was significantly increased from 29.8 to 35.8 after 8 weeks of training (p=.043). 4. The mean WISCI level was significantly increased from level 10 to level 19 after 8 weeks of training (p=.007). The results of this study suggest that the gait training program using the auto-walking machine increased the gait speed, muscle strength and galt pattern (WISCI level) in persons with incomplete spinal cord injury. A large, controlled study of this technique is warranted.

Serum Iron Concentration of Maternal and Umbilical Cord Blood during Pregnancy (임신기 모체 혈청과 신생아 제대혈청의 철분함량)

  • Anemia diagnosed early in pregnancy is associated with increased risks of low birth weight and preform delivery. The purposes of this study were to assess the maternal iron status during pregnancy and to evaluate the relationships between the iron indices of maternal-umbilical cord serum iron and ferritin levels and pregnancy outcomes. Dietary intakes of the pregnant women were estimated by 24 hour-recall (3 times). Serum iron and ferritin levels in maternal blood and umbilical cord were measured at 1st-, 2nd-, 3rd- trimester and delivery, respectively. The mean of maternal se겨m iron levels of the trimester and delivery were $124.27\;{\mu}g/dl,\;97.03\;{\mu}g/dl,\;94.32\;{\mu}g/dl,\;and\;145.53\;{\mu}g/dl$. Those maternal levels were significantly lower than that of umbilical cord blood ($222.59\;{\mu}g/dl$). Serum ferritin levels of maternal trimester and delivery were 22.68 $22.68\;{\mu}g/l,\;11.09\;{\mu}g/l,\;14.18\;{\mu}g/l,\;and\;\;24.54\;{\mu}g/l$, which were significantly lower than those of umbilical cord blood ($184.35\;{\mu}g/l$) (p < 0.0001). This prevalence of anemia of total subjects was $30.3\%$ by WHO criteria (Hb < 11.0 g/dl, Hct < $33\%$). Iron levels of 2nd-trimester was significantly higher in the normal group than in the anemia group. And ferritin levels of 3rd-trimester and delivery was significantly higher in the normal group than in the anemia group. Therefore, we suggest for successful pregnancy outcome and delivery differential iron supplementation programs will be carried out with individual Pregnant women on the basis of pre-Pregnancy nutrition. Effect of Thermal Aging on the Change of Interfacial Adhesion between Polyketone Cord and Rubber by RFL Primer Treatment (RFL 프라이머 처리에 따른 폴리케톤 코드사와 고무 간의 계면접착성 변화에 열노화가 미치는 영향)

  • In the case of fiber/rubber composites for tire applications, the interfacial adhesion between fiber and rubber significantly affects the physical properties of the finished products. Generally, organic synthetic fibers used for tire cords are treated with resorcinol formaldehyde latex(RFL) primer on the surface of the fiber to improve the adhesion to rubber. Changes of adhesion between rubber and tire cords might weaken as temperature rises due to overheating of car engine and friction with road. In this study, the effects of temperature on the primer treated polyketone cord/rubber composites and the changes in interfacial adhesion were investigated. Polyketone cord/rubber composites were prepared after RFL solution treatment on the surface of polyketone fibers. After that, composites was thermally aged at different temperature conditions(60, 80, 100, $120^{\circ}C$) and times(1, 5, 10, 15days). The adhesion strength of polyketone cord/rubber composite treated with RFL primer was higher than untreated composite by more than 3 times. After heat aging, the adhesion strength of untreated polyketone cord/rubber composites increased while the RFL treated polyketone cord/rubber composites decreased somewhat.

Single Stage Circumferential Cervical Surgery (Selective Anterior Cervical Corpectomy with Fusion and Laminoplasty) for Multilevel Ossification of the Posterior Longitudinal Ligament with Spinal Cord Ischemia on MRI

  • Objective : Anterior cervical corpectomy with fusion (ACF) or laminoplasty may be associated with substantial number of complications for treating multilevel cervical ossification of the posterior longitudinal ligament (OPLL) with significant cord compression. For more safe decompression and stabilization in multilevel cervical OPLL with prominent cord compression, we propose circumferential cervical surgery (selective ACF and laminoplasty) based on our favorable experience. Methods : Twelve patients with cervical myelopathy underwent circumferential cervical surgery and all patients showed multilevel OPLL with Signal change of the spinal cord on magnetic resonance imaging (MRI). A retrospective review of clinical, radiological. and surgical data was conducted. Results : There were 9 men and 3 women with mean age of 56.7 years and a mean follow up period of 15.6 months. The average corpectomy level was 1.16 and laminoplasty level was 4.58. The average Japanese Orthopedic Association score for recovery was 5.1 points and good clinical results were obtained in 11 patients (92%) (p < 0.05). The average space available for the cord improved from 58.2% to 87.9% and the average Cobb's angle changed from 7.63 to 12.27 at 6 months after operation without failure of fusion (p < 0.05). Average operation time was 8.36 hours, with an estimated blood loss of 760 mL and duration of bed rest of 2.0 days. There were no incidences of significant surgical complications, including wound infection. Conclusion : Although the current study examined a small sample with relatively short-term follow-up periods, our study results demonstrate that circumferential cervical surgery is considered favorable for safety and effectiveness in multilevel OPLL with prominent cord compression.

The Study of the Driving Characteristics in Persons With Spinal Cord Injury (척수손상 장애인의 자가운전 특성에 관한 연구)

  • The purpose of this study was to provide information on driving characteristics in persons with spinal cord injury through basic statistic analysis of the survey results. The survey was administered to 44 drivers with spinal cord injury. The subjects' general, neurologic and driving characteristics were analyzed, as well as the degree of difficulty in using their vehicles between tetraplegia and paraplegia. The results were as follows: thirty-five (79.6%) of forty-four respondents was men. The average age was 35.0 years old and the age at the time of injury was 29.0 years old. Their neurologic characteristics were tetraplegics 12 (27.3%) and paraplegics 32 (72.2%). Among complete lesions, the highest level those who could drive independently was C7. All the vehicles were equipped with special devices, including "power steering", "automatic transmission" and "hand controls". The vehicles for cervical cord injury were equipped with "grip bars" as well as for the degree of difficulty in using their vehicles, all the subjects felt that "moving the wheelchair in and out of their vehicles" was too difficult for them to do. We suggest that the driver training should be an essential part of the rehabilitation program for patients with spinal cord injuries to maximize their mobility in the community. This training seems to be essential in order to modify the standards of the Handicapped Drivers Ability Test and to aid the driver rehabilitation program in the health insurance payment system. Also, the driver rehabilitation training program should include instruction in that moving wheelchairs in and out of vehicles.

  • As Network Slicing functionality gets applied to mobile networking, a mechanism that enables the selection of network slices becomes indispensable. Following the 3GPP Technical Specification for the 5G Architecture, the inclusion of the Network Slice Selection Function (NSSF) in order to leverage the process of slice selection is apparent. However, actual implementation of this network function needs to deal with the dynamic changes of network instances, due to this, a platform that supports the orchestration of Virtual Network Functions (VNF) is required. Our proposed solution include the use of the Central Office Rearchitected as a Data Center (CORD) platform, with the specified profile for mobile networks (M-CORD) that integrates a service orchestrator (XOS) alongside solutions oriented to Software Defined Networking (SDN), Network Function Virtualization (VNF) and virtual machine management through OpenStack, in order to provide the right ecosystem where our implementation of NSSF can obtain slice information dynamically by relying on synchronization between back-end services and network function instances.

$\beta-Carotene,\;\alpha$-Tocopherol Concentrations in Maternal-Umbilical Cord Plasma and Their Relationships to Pregnancy Outcome (임신 말 모체와 제대혈의 Retinol, $\beta-Carotene,\;\alpha$-Tocopherol의 농도 및 임신결과와의 상관성)

  • The purpose of this study was to evaluate the intake of antioxidant vitamins and plasma concentrations of those in 60 maternal-infant pairs (30 in normal term delivery group, NT; 30 in preform delivery group, PT). We also investigated the relationship between vitamin levels of maternal-umbilical cord plasma and pregnancy outcome. Mean energy intakes of NT and PT pregnant women were 93.2% and 85.4%, and their protein intakes were 113.3% and 110.9% of the recommended dietary allowance (RDA), respectively. While vitamin A intakes were only 51.2% and 39.6% of the RDA in NT and PT pregnant women. The vitamin E intake was about 50% of the RDA (NT 6.27 mg, PT 7.78 mg). The levels of retinol in maternal plasma of NT and PT were $1.51\mumol/\ell\;and\;1.43\mumol/\ell$, respectively. The retinol levels in umbilical cord plasma in NT and PT were $0.72\mumol/\ell\;and\;0.61\mumol/\ell$, respectively. The level of $\beta-carotene$in maternal plasma of NT was 0.49 $\mu$mol/$\ell$, significantly (p < 0.01) higher than that of PT ($0.31\mumol/\ell$).The $\beta-carotene$ of umbilical cord plasma of NT and PT were $0.702\mumol/\ell\;and\;0.01\mumol/\ell$, respectively. The plasma $\alpha$-tocopherol of maternal of NT and PT were $0.72\mumol/\ell\;and\;0.01\mumol/\ell\;29.51 /mumol/\ell\; and 27.17\mumol/\ell,\;respectively.\; The $\alpha$-tocopherol of umbilical cord plasma of NT and PT were $4.16\mumol/\ell\;and\;3.80\mumol/\ell$, respectively. The antioxidant vitamin levels (retinol, $\beta-carotene,\;and\;\alpha$-tocopherol) in maternal plasma were significantly higher (p<0.0001) than those in umbilical cord plasma. However, there was no correlation between the vitamin levels in maternal plasma and those in umbilical cord plasma. The maternal plasma $\beta$-carotene level showed a positive correlation to gestational age. Also Apgar score at 1 min produced a positive correlation to maternal plasma $\beta$-carotene level.

Pregnancy Outcomes in Relation to Plasma TRAP Concentration of Maternal and Umbilical Cord (임신말 모체와 제대혈의 혈장 총유리기포집 항산화능과 임신결과)

The vitamin A intakes of NT and W pregnant women were 559.7 RE and 497.8 RE, which were less than RDA. While the vitamin E and C intakes of both NT and PT pregnant women were more than RDA. The maternal plasma TRAP level of PT was 1.41 mmol/l and that of was 1.50 mmol/l, which was significantly higher than TRAP level of PT (p < 0.05) . The umblical cord plasma TRAP levels of NT and PT were 1.44 mmol/l and 1.23 mmol/l, which indicates the significant difference between those two groups (p < 0.001) . In case of comparing the TRAP level of maternal and umbilical cord blood, there was no significant difference in NT pregnant women, however, in PT group maternal the TRAP level significant higher than that of umbilical cord (p < 0.001). The length of gestation and plasma TRAP level of maternal and umbilical cord showed a positive correlation. However, other parameters of pregnancy outcomes such as birth weight, weight gain, and Apgar score were not affected by the plasma TRAP levels. Based on these results, preform infants could have a risk of oxidative stress because of low plasma TRAP level.
