  • In this study firstly we survey the calculation method and the characteristics of the way of estimating CBL(Customer BaseLine Load) that is important calculation tool for DRP internationally. Also we analyze the power consumption pattern using the 15 minutes load profiles of about 120,000 customers in domestic. Based on this pattern, we provide the CBL calculation method that can be utilized in DRP to save the cost, and analyze the accuracy of the CBL calculation proposed in this paper through the simulation.

Proteolysis of the Reverse Transcriptase of Hepatitis B Virus by Lon Protease in E. coli

  • Hepatitis B virus (HBV) polymerase, which possesses the activities of terminal binding, DNA polymerase, reverse transcriptase and RNaseH, has been shown to accomplish viral DNA replication through a pregenomic intermediate. Because the HBV polymerase has not been purified, the expression of HBV polymerase was examined in an E. coli expression system that is under the regulation of arabinose operon. The expressed individual domain containing terminal binding protein, polymerase, or RNaseH turned out to be insoluble. The activities of those domains were not able to be recovered by denaturation and renaturation using urea or guanidine-HCI. The expressed reverse transcriptase containing the polymerase and RNaseH domains became extensively degraded, whereas the proteolysis was reduced in a Ion- mutant. These results indicate that Lon protease proteolyzes the HBV reverse transcriptase expressed in E. coli.

  • This paper proposes the mechanism of cell buffering in the output buffer of ATM switch for the traffic with the different QOS in the ATM environment. The proposed mechanism, VQS(Variable Queue Sharing) can minimize the cell loss ratio(CLR) of bursty traffic through the sharing of CBR queue, VBR queue, ABR queue to maximize the utilization of queue resource. To evaluate VQS performance, we make simulator using Visual Slam 2.0. We compare the CLP and cell average delay of VQS and HOL, QLT_HOL using the bursty traffic patterns.

  • This paper propose the queue structure of cell buffering in the output buffer of ATM switch for the traffic with the different QoS. The proposed queue structure can minimize the cell loss ratio of bursty traffic, maximize the queue utilization through the sharing of real-time queue and non-real-time queue. To evaluate the proposed queue structure, we compare the CLP and cell average delay of the proposed queue and fixed queue using the bursty traffic patterns.

Complete Genome Sequence of Chryseobacterium mulctrae KACC 21234T : A Potential Proteolytic and Lipolytic Bacteria Isolated from Bovine Raw Milk

  • Chryseobacterium mulctrae KACC 21234T is a novel species isolated from raw bovine milk. Psychrotrophic bacteria are considered contaminants and are hypothesized to originate from the environment. In this investigation, the C. mulctrae KACC 21234T genome was determined to be 4,868,651 bp long and assembled into four contigs with a G+C ratio of 33.8%. In silico genomic analyses revealed the presence of genes encoding proteases (endopeptidase Clp, oligopeptidase b, carboxypeptidase) and lipases (phospholipase A(2), phospholipase C, acylglycerol lipase) that can catalyze the degradation of the proteins and lipids in milk, causing its quality to deteriorate. Additionally, antimicrobial resistance and putative bacteriocin genes were detected, potentially intensifying the pathogenicity of the strain. The genomic evidence presented highlights the need for improved screening protocols to minimize the potential contamination of milk by proteolytic and lipolytic psychrotrophic bacteria.

  • Severe acute respiratory syndrome (SARS) is a severe respiratory infectious disease caused by a novel human coronavirus, SARS-CoV. The 3CL protease is a key enzyme in the proteolytic processing of replicase polyprotein precursors, pp1a and pp1ab, which mediate all the functions required for viral genomic replication and transcription. Therefore, this enzyme is a target for the development of chemotherapeutic agents against SARS. A large quantity of active SARS-3CL protease is required for development of anti-SARS agents. Here we have constructed overexpression vector for the production of the SARS-3CL protease. The gene encoding SARS-3CL protease was amplified using polymerase chain reaction and cloned into the pET29a expression vector, resulting in pET29a/SARS-3CLP. Recombinant SARS-3CL protease was successfully synthesized by the dialysis mode of the cell-free protein expression system, and purified by three-step fast protein liquid chromatography using HighQ and MonoP column chromatographies and Sephacryl S-300 gel filtration. In addition, the produced SARS-3CL protease was found to be an active mature form. This study provides efficient methods not only for the development of anti-SARS materials from natural sources, but also for the study of basic properties of the SARS-3CL protease.

Correlation between Magnifying Narrow-band Imaging Endoscopy Results and Organoid Differentiation Indicated by Cancer Cell Differentiation and its Distribution in Depressed-Type Early Gastric Carcinoma

  • Background: A close association between patterns identified by magnifying narrow-band imaging (M-NBI) and histological type has been described. M-NBI patterns were also recently reported to be related to the mucin phenotype; however, detials remain unclear. Materials and Methods: We investigated the cellular differentiation of gastric cancer lesions, along with their mucosal distribution observed by M-NBI. Ninety-seven depressed-type early gastric cancer lesions (74 differentiated and 23 undifferentiated adenocarcinomas) were visualized by M-NBI. Findings were divided into 4 patterns based on abnormal microvascular architecture: a chain loop pattern (CLP), a fine network pattern (FNP), a corkscrew pattern (CSP), and an unclassified pattern. Mucin phenotypes were judged as gastric (G-type), intestinal (I-type), mixed gastric and intestinal (M-type), and null (N-type) based on 4 markers (MAC5AC, MUC6, MUC2, and CD10). The relationship of each pattern of microvascular architecture with organoid differentiation indicated by cancer cell differentiation and its distribution in each histological type of early gastric cancer was investigated. Results: All CLP and FNP lesions were differentiated. The cancer cell distribution showed organoid differentiation in 84.2% (16/19) and 61.1% (22/36) of the two types of lesions, respectively, and there was a significant difference from the unclassified pattern with organoid differentiation (p<0.001). Almost all (94.7%; 18/19) CSP lesions were undifferentiated, and organoid differentiation was observed in 72.2% (13/18). There was a significant difference from the unclassified pattern with organoid differentiation (p<0.05). Conclusions: Cellular differentiation and distribution are associated with microvascular architecture observed by M-NBI.

  • The frequency of abnormality at birth is average 1-1.5%, and of these, cleft lip & palate is known to be the most frequent congenital abnormality, Cleft is considered to be due to multi-factorial heredity correlated with genetic and environmental factors, Cleft patients require the collaborative treatment with several medical departments, Clinics for Maxillo-Oral Disorders of Tohoku University Dental Hospital performs the total managements related to such as occlusion and language for the patients with congenital maxilla-facial abnormality, This study examined the patients with cleft lip and/or palate who came to the Clinics for Maxilla-Oral Disorders of Tohoku University Dental Hospital for the past 17 years from Jan. 1987 to Dec, 2002, and had the results as follows, 1. Annual mean number of patients The annual mean number of the patients for 17 years from Jan, 1987 to Dec, 2002 was 91 patients, ranging from 63 minimum to 116 maximum, 2, Gender and types of cleft There were 747(51%) males and 709(49%) females, with a male to female ratio 1,05:1. CLP was the most frequent cleft type as shown in 616 patients, and other patients manifested different complaints such as CL, CP, SMCP and MC in order. 3. The laterality in cleft type The lip cleft was frequently expressed orderly on left, right and both sides of CL patients while orderly being shown on left, both and right sides of CLP patients. Accordingly, lip cleft was most commonly found on the left side. 4. Address at first visit Of 1,456 subjects, 850(58.4%) patients were residing in Miyagi Prefecture, where this hospital is located. 5. Age at first visit 615(42.2%) patients came to the hospital at their age younger than 1 year old, comprising 282(19.4%) patients age younger than 2 months old and 333(22.9%) patients age between 2 month old and 1 year old. 6. Mother's age at birth For the mother's age at birth, 526(39.9%) patients were at the age of 25 to 30 years old, and 17(1.3%) patients were over 40 years old. 7. Birth weight 34.3%(443 patients) had a birth weight of 2500-3000gm and 56.0%(724 patients) had a 3000-4000gm. It was also found that 7.9%(102 patients) had a birth weight of less than 2500gm. 8. Familial expression The frequency of familial expression was 6.5%(94 patients).

  • Recent investigation into the integrity of nuclear containment buildings has highlighted the importance of developing an elaborate diagnostic method to evaluate the distribution and size of cavities inside concrete walls. As part of developing such a method, this paper presents a finite element approach to modeling elastic waves propagating in the containment building walls of a nuclear power plant. We introduce a perfectly matched layer (PML) wave-absorbing boundary to limit the large-scale nuclear containment wall to the region of interest. The formulation results in a semi-discrete form with symmetric damping and stiffness matrices. The transient elastic wave equations for a mixed unsplit-field PML were solved for displacement and stresses in the time domain. Numerical results show that the sensitivity of displacement, velocity, acceleration, and stresses is large depending on the size and location of the cavity. The dynamic response of the wall slightly differs depending on the existence of the containment liner plate. The results of this study can be applied to a full-waveform inversion approach for characterizing cavities inside a containment wall.

Flutter performance of central-slotted plate at large angles of attack

  • The flutter instability is one of the most important themes need to be carefully investigated in the design of long-span bridges. This study takes the central-slotted ideal thin flat plate as an object, and examines the characteristics of unsteady surface pressures of stationary and vibrating cross sections based on computational fluid dynamics (CFD) simulations. The flutter derivatives are extracted from the surface pressure distribution and the critical flutter wind speed of a long span suspension bridge is then calculated. The influences of angle of attack and the slot ratio on the flutter performance of central-slotted plate are investigated. The results show that the critical flutter wind speed reduces with increase in angle of attack. At lower angles of attack where the plate shows the characteristics of a streamlined cross-section, the existence of central slot can improve the critical flutter wind speed. On the other hand, at larger angles of attack, where the plate becomes a bluff body, the existence of central slot further reduces the flutter performance.
