2-Chloroethylethyl Sulfide Induces Apoptosis and Necrosis in Thymocytes

  • 2-chloroethylethyl sulfide (CEES) is an alkylating agent that readily reacts with a wide variety of biological molecules causing metabolic abnormality. The mechanism of cell death during CEES injury is poorly understood. We have examined the effect of exposure of thymocytes with various concentrations of CEES to determine the pattern of cell death in thymocytes injury induced by CEES. In the present study, we show that two patterns of cell death occurred by either one of two mechanisms: apoptosis and necrosis. Exposure to low level of CEES (100 ${\mu}M$) for 5 h caused an induction of apoptosis on thymocytes, as identified by the following criteria: DNA fragmentation visualized by the characteristic "ladder" pattern was observed upon agarose gel electrophoresis and morphological features were revealed by microscopical observations. In contrast, exposure to high levels of CEES (500 ${\mu}M$) induce necrotic features such as cell lysis. Thus, depending on the concentrations, CEES can result in either apoptotic or necrotic cell damage. Our findings suggest that thymocytes which are not killed directly, but merely injured by low levels of CEES, are able to activate an internally-programmed cell death mechanism, whereas thymocytes receiving severe damages apparently can not.

Protective Effect of Aminoglycosides and Their Combinations Against 2-Chloroethylethyl Sulfide Exposure

  • Exposure of splenocytes to 2-chloroethylethyl sulfide (CEES) resulted in the cell death, and the cytotoxicity of CEES was prevented by inhibitors of lysosomal hydrolases. Therefore, it has been postulated that the cytotoxicity of CEES may be partially due to the lysosomal labilization. This study, based on this mechanism, was undertaken to determine whether aminoglycoside antibiotics as inhibitors of lysosomal phospholipases and their combinations with other lysosome stabilizers can be useful as a treatment to reduce the CEES toxicity in mice. 2-Chloroethylethyl sulfide (20 mg/kg body weight) was injected ip into female ICR mice, and candidate compounds were administered ip before or after the CEES challenge. Kanamycin (40 mg/kg body weight) as effective as deferoxamine (100 mg/kg body weight) enhanced the survival rate after 5 days of intoxication from 10% of control to 50 - 60%. The most effective was found to be the combination of kanamycin, cycloheximide, deferoxamine and dextrose showing an almost full protection against 2LD50 of CEES. Consistent with the protection of the CEES toxicity, the decrease of body weight in mice intoxicated with CEES was effectively prevented by kanamycin or its combinations. It is suggested that kanamycin or its combination (kanamycin, cycloheximide, deferoxamine and dextrose) would be one of effective antidotes against the CEES poisoning in mice.

An Additional Mechanism for the Cytotoxicity of 2-Chloroethylethyl Sulfide in Spleen Lymphocytes; Lysosomal Labilization

  • Exposure of spleen lymphocytes to 2-chloroethylethyl sulfide (CEES) leads to a reduction of the intracellular ATP level, followed by a decrease in cell viability. Addition of nicotinamide, an inhibitor of poly(ADP-ribose) polymerase (PADPRP), restores both ATP level and viability, indicating that an activation of PADPRP is responsible for the cytotoxicity of CEES. The involvement of a $Ca^{2+}$-mediated process in cytotoxicity is suggested. Verapamil, EGTA, trifluoperazine, and butacaine exhibit a partial protection (20 to 58%) against the cytotoxicity of CEES. Investigation of the causative role of proteolytic degradation in cell death indicate that pepstatin and leupeptin exert a substantial protective effect (60 to 70%), suggesting the involvement of lysosomal destabilization in CEES-induced cytotoxicity. Also, lysosomotropic agents markedly decrease the cytotoxicity. Lysosomal labilization may be a mechanism for the cytotoxicity of CEES.

  • The industry has rapidly growth and energy supply technology advanced are become main factor which to contribute of the harmonic losses. This problem is one aspect that may affect the capability of the transmission line and also to the efficiency of electricity. This paper proposes a new scheme to allocate the cost pertaining to transmission loss due to harmonics. The proposed method, called as Generalized Harmonic Distribution Factor, uses the principle of proportional sharing method to allocate the losses among the transmission users especially for industry consumers. The IEEE 14- and 30 bus test system is used to compare the proposed method with existing method. The results showed that the proposed method provided a scheme better in allocating the cost of transmission loss, which could encourage the users to minimize the losses.

  • This paper analyses the power flow of a three-feeder/multi-bus distribution system by a custom Generalized Power Quality Conditioner (GUPQC). The GUPQC has been realized by three voltage source converters (VSCs) coupled back-to-back through a common DC-link capacitor on the DC-side. One feeder was controlled by the shunt compensator, whereas each of the other two feeders was controlled by the proposed novel series compensator. The GUPQC has the capability to simultaneously compensate voltage and current quality problems of a multi-bus/three-feeder distribution system. Besides that, the power can be transferred from one feeder to other feeders to compensate for poor power quality problems. Extensive simulation studies were carried out by using MATLAB/SIMULINK software to establish the ability of the GUPQC to improve power quality of the distribution systems under distorted supply voltage conditions.

  • 4차 산업혁명시대가 현대사회에 도래함에 따라 시대변화에 능동적으로 대처하고, 현재의 삶의 위기를 극복하기 위하여 직업전환을 꾀하거나 자기개발 및 노후 준비를 위한 성인학습자들의 창업에 대한 관심이 계속해서 증대되고 있다. 정부와 대학 등 여러 기관에서는 창업에 관심이 많아진 성인학습자들을 위하여 창업을 적극적으로 장려하는 정책을 추진하고 있다. 그러나, 제2차 대학 창업교육 5개년 계획(안)에 따르면 실습형 강좌보다 이론 중심의 창업교육이 대부분이며, 창업 이후의 고도화 교육지원 역량이 부족한 상황(교육부, 2018)이다. 성인학습자에게 창업교육 및 창업 환경 조성이 미흡하다는 문제점이 대두되어 창업교육이 성인학습자의 창업의지와 창업행동에 미치는 영향 관계에서 자기효능감의 효과를 실증적으로 규명하는 것이 본 연구의 목적이다. 연구대상은 성인학습자를 대상으로 2022년 9월부터 10월까지 온라인 설문조사를 실시였고, 총 207부가 수집되었다. 척도의 신뢰성을 검증하기 위하여 크론바흐 알파계수(Cronbach's α)를 산출하여 분석하고 측정하였다. 가설검증은 다중회귀분석 통계분석 방법을 사용하였고 SPSS 22.0 통계처리 프로그램을 이용하였다. 연구 결과, 첫째, 자기효능감은 창업교육에 유의한 영향을 미치는 것으로 나타났다. 둘째, 창업교육은 성인학습자의 창업의지에 유의한 영향을 미치는 것으로 나타났다. 셋째, 창업교육은 성인학습자의 창업행동에 유의한 영향을 미치는 것으로 나타났다. 넷째, 자기효능감은 성인학습자의 창업의지에 유의한 영향을 미치는 것으로 나타났다. 다섯째, 자기효능감은 성인학습자의 창업행동에 유의한 영향을 미치는 것으로 나타났다. 여섯째, 창업교육과 성인학습자의 창업의지 간의 관계에서 자기효능감은 매개 효과를 갖는 것으로 나타났다. 일곱째, 창업교육과 성인학습자의 창업행동 간의 관계에서 자기효능감은 완전매개 효과를 갖는 것으로 나타났다. 성인학습자들의 특성을 고려하여 다양한 교육방식 등을 적용하는 것이 필요함을 확인하였고 따라서 성인학습자들에게 본인들의 현장경험을 기반한 국내·외 창업 사례나 벤처기업 등의 경영관리의 전반적인 내용에 대해 실무적으로 학습하는 프로그램 또는 성인학습자들이 직접 창업을 구성하고 구상한 내용에 맞는 사업계획서를 작성하여 그 내용을 발표하고 토의하는 방식 등의 전문적인 창업교육을 위하여 지속적 개발이 필요하다는 시사점을 도출하였다. 또한, 창업교육을 통해 성인학습자들이 실제로 창업행동을 할 수 있도록 창업의지를 제고시키고 자기효능감을 심어주어 그들의 자기개발 또는 제2의 삶을 설계 할 수 있도록 지원체계를 만들어야 할 것이다.

Performance Analysis of Two-Way Relay NOMA Systems with Hardware Impairments and Channel Estimation Errors

  • In this paper, we consider a two-way relay non-orthogonal multiple access (TWR-NOMA) system with residual hardware impairments (RHIs) and channel estimation errors (CEEs), where two group users exchange their information via the decode-and-forward (DF) relay by using NOMA protocol. To evaluate the performance of the considered system, exact analytical expressions for the outage probability of the two groups users are derived in closed-form. Moreover, the asymptotic outage behavior in the high signal-to-noise ratio (SNR) regime is examined and the diversity order is derived and discussed. Numerical simulation results verify the accuracy of theoretical analyses, and show that: i) RHIs and CEEs have a deleterious effects on the outage probabilities; ii) CEEs have significant effects on the performance of the near user; iii) Due to the RHIs, CEEs, inter-group interference and intra-group interference, there exists error floors for the outage probability.

Bioconversion of Cyanidin-3-Rutinoside to Cyanidin-3-Glucoside in Black Raspberry by Crude α-ʟ-Rhamnosidase from Aspergillus Species

  • Cyanidin-3-glucoside (C3G) has been known to be more bioavailable than cyanidin-3-rutinoside (C3R), the most abundant anthocyanin in black raspberry (Rubus occidentalis). The aim of this study was to enhance the bioavailability of anthocyanins in black raspberry by cleaving ʟ-rhamnose in C3R using crude enzyme extracts (CEEs) from Aspergillus usamii KCTC 6956, A. awamori KCTC 60380, A. niger KCCM 11724, A. oryzae KCCM 12698, and A. kawachii KCCM 32819. The enzyme activities of the CEEs were determined by a spectrophotometric method using ρ-nitrophenyl-rhamnopyranoside and ρ-nitrophenyl-glucopyranoside. The CEE from A. usamii had the highest α-ʟ-rhamnosidase activity with 2.73 U/ml at 60℃, followed by those from A. awamori and A. niger. When bioconversion of C3R to C3G in black raspberry was analyzed by HPLC-DAD, the CEEs from A. usamii and A. awamori hydrolyzed 95.7% and 95.6% of C3R to C3G, respectively, after 2 h incubation. The CEEs from A. kawachii and A. oryzae did not convert C3R to C3G in black raspberry.

Chemiresistive Gas Sensors for Detection of Chemical Warfare Agent Simulants

  • Precautionary detection of chemical warfare agents (CWAs) has been an important global issue mainly owing to their toxicity. To achieve proper detection, many studies have been conducted to develop sensitive gas sensors for CWAs. In particular, metal-oxide semi-conductors (MOS) have been investigated as promising sensing materials owing to their abundance in nature and excellent sensitivity. In this review, we mainly focus on various MOS-based gas sensors that have been fabricated for the detection of two specific CWA simulants, 2-chloroethyl ethyl sulfide (2-CEES) and dimethyl methyl phosphonate (DMMP), which are simulants of sulfur mustard and sarin, respectively. In the case of 2-CEES, we mainly discuss $CdSnO_3-$ and ZnO-based sensors and their reaction mechanisms. In addition, a method to improve the selectivity of ZnO-based sensors is mentioned. Various sensors and their sensing mechanisms have been introduced for the detection of DMMP. As the reaction with DMMP may directly affect the sensing properties of MOS, this paper includes previous studies on its poisoning effect. Finally, promising sensing materials for both gases are proposed.
