Antioxidative Effect of Chelidonium majus Extract on Cultured NIH3T3 Fibroblasts Injured by Cadmium Chloride of Toxicant (독성물질인 염화카드뮴으로 손상된 배양 NIH3T3 섬유모세포에 대한 애기똥풀 추출물의 항산화 효과)

  • The aim of this study was to evaluate the cytotoxicity of cadmium chloride ($CdCl_2$), toxicant, and the protective effect of Chelidonium majus (CM) extract on $CdCl_2$-induced cytotoxicity in cultured NIH3T3 fibroblasts. Cell viability, the effect of butylated hydroxytoluene (BHT) against $CdCl_2$, and the antioxidative effects including DPPH-free radical scavenging activity, superoxide anion-radical scavenging activity (SSA), and lactate dehydrogenase (LDH) activity were assessed. $CdCl_2$ caused a significant dose-dependent decrease in cell viability, and $XTT_{50}$ value was determined at 38.7uM of $CdCl_2$. It was determined as highly-toxic by Borenfreund and Puerner' toxic criteria. BHT of antioxidant significantly increased cell viability severely damaged by $CdCl_2$-induced cytotoxicity in these cultures. In the protective effect of CM extract on $CdCl_2$-induced cytotoxicity, CM extract significantly increased cell viability, DPPH-free radical scavenging activity, SSA and inhibitory activity of LDH. From these results, it is suggested that oxidative stress is involved in the cytotoxicity of $CdCl_2$, and CM extract showed protective efficacy on $CdCl_2$-induced cytotoxicity via antioxidative effects. Conclusively, natural resources like CM extract may be a putative antioxidative agent for the detoxification or diminution of toxicity correlated with oxidative stress.

Optical properties of $Ag_2CdSnSe_4$ and $Ag_2CdSnSe_4:CO^{2+}$ single crystals ($Ag_2CdSnSe_4$$Ag_2CdSnSe_4:Co^{+2}$단결정의 광학적 특성)

  • Optical properties of $Ag_2CdSnSe_4$ and $Ag_2CdSnSe_4:Co^{+2}$ quaternary semiconductor single crystals grown by the chemical transport reaction method were investigated. The analysis of the X - ray powder diffraction measurements showed that these crystals have a wurtzite structure with lattice constants a = 4.357 $\AA$, c = 7.380 $\AA$, for $Ag_2CdSnSe_4$ and a = 4.885 $\AA$, c = 7.374 $\AA$, for $Ag_2CdSnSe_4:CO^{2+}$. The direct band gap at 298K, obtained from the optical absorption measurement, is found to be 1.21 eV for $Ag_2CdSnSe_4$ and 1.02 eV for $Ag_2CdSnSe_4:CO^{2+}$. The shrinkage of the band gap due to Co-doping is observed and is about 190 meV, We observed four absorption bands of $Co^{2+}$ ions in two near infrared regions of optical absorption spectra of $Ag_2CdSnSe_4$:$Co^{+2}$. These absorption bands were assigned as due to electronic transitions between the split energy levels of $Co^{2+}$ ions in $T_d$ crystal field under spin-orbit interactions.

Celiac disease in children: increasing prevalence and changing clinical presentations

  • Background: Celiac disease (CD) is a chronic autoimmune enteropathy. It results from genetic predisposition and exposure to gluten-containing food. The prevalence and presentation of CD vary among populations. Purpose: This study aimed to describe the prevalence and clinical characteristics of CD in children in Bahrain. Methods: We retrospectively reviewed the medical records of children diagnosed with CD in the pediatric department, Salmaniya Medical Complex, Bahrain, in 1988-2018. Their clinical, biochemical, serological, and histopathological findings were documented. Adherence to the recommended gluten-free diet (GFD) was assessed. Results: Of 86 patients with CD, 67 were included. The CD prevalence was 0.02%. A significant increase in prevalence in the last decade was observed (P<0.0001). Thirty-eight patients (56.7%) were males. The median (interquartile range) age at presentation was 4.45 (1.5-7.3) years. A family history of CD was positive in 13 out of 43 patients (30.2%). Pallor and failure to thrive were the most common presentations. The most frequent associated disease was iron-deficiency anemia in 23 patients (69.7%). Positive serology was found in 32 of 45 patients (71.1%). Marsh-Oberhuber type III was found in 16 of 35 patients (45.7%). Seropositive patients were significantly older (P=0.025) and had more severe duodenal histology (P=0.002). Adherence to GFD was poor in 27 patients (64.3%). Conclusion: This study revealed a significant increase in CD prevalence over the last decade. Atypical presentations were frequent. Most patients had poor adherence to GFD.

Effects of dietary Cadmium and Protein Levels on the Body Protein Metabolism and Cadmium Toxicity in Growing Rats (식이내 Cadmium과 단백질 수준이 흰쥐의 체내 단백질 대사 및 Cadmium 중독에 미치는 영향)

  • This study were performed to investigate effect of dietary cadmium(Cd) and protein levels on growth, body protein metabolism and Cd toxicity in growing rats. Forty eight male rats of Sprague-Dawley weighing 113$\pm$2g were blocked into 6 groups accoridng to body weight. Dietary protein were given at the levels of 7, 15 and 40% of diet and Cd (200ppm)were either added or not. The result obtained were summerized as follow; 1) Food intake, weight gain, FER PER, liver and kidney weight, weight and length of bones, hematocrit, and hemoglobin content in Cd-added groups were low than those in Cd-free groups. 2) Serum total protein showed no significant difference with Cd addition, but it was significantly lower in low protein diet groups. Liver protein in Cd-added groups was lower than Cd-free groups, and was tend to be increased with increasing dietary protein level. 3) Daily urinary and fecal nitrogen excretions in Cd-added groups were lower than Cd-free groups, and were increased with increasing dietary protein level. 4) Cadmium contents in blood, liver, kidney, and femur were tend to be decreased with increasing dietary protein level. Especially, Cd content in kidney of Cd-added groups was significantly decreased with increasing dietary protein level. 5) Daily urinary and fecal Cd excretions were tend to be increased with increasing dietary protein level, and Cd-added-high protein diet group showed the highest Cd excretion among the Cd-added groups, Cd absorption ration and Cd retention ratio were tend to be decreased with increasing dietary protein level.

Adsorption Characteristics of the Heavy Metals, Cd(II) and Pb(II) Ions, on the Si-Immobilized Ulva pertusa (실리카고정 구멍갈파래에 대한 중금속 Cd(II), Pb(II)이온의 흡착 특성)

  • Adsorption characteristics of heavy metal ions, Cd(II) and Pb(II), on eastcoast-living algae, ulva pertusa, has been studied in our experiment. The Maximum adsorption amount of Cd(II) and Pb(II) ions on 1 g of the ulva pertusa were 2.3 mg, 3.1 mg in alkaline and 2.0 mg, 2.8 mg in acidic solution. However 3.4 mg, 7.3 mg in alkaline and 3.1 mg, 6.5 mg in acidic solution were shown on the Si-immobilized ulva pertusa in the same condition. Thus, Si-immobilized ulva pertusa adsorbs more amount of heavy metals, Cd(II), Pb(II), than the ulva pertusa, and more effective absorbent in alkaline. Furthermore, more amounts of Pb(II) ion were absorbed compare to Cd(II) ion in our work. Recovery ratio of Cd(II) and Pb(II) ions on the ulva pertusa were 55.0~61.0%, 59.7~66.8% respectively and 87.6~97.5%, 83.5~99.3% on the Si-immobilized ulva pertusa.

The Study on Absorption of $Cd^{2+}$ and $Pb^{2+}$ by Persicaria thunbergii and Rumex crispus, and Their Phytochelatin (고마리와 소리쟁이에 의한 $Cd^{2+}$$Pb^{2+}$의 흡수 및 phytochelatin에 관한 연구)

  • When Persicaria thunbergii and Rumex crispus were treated with Cd($NO_3$)$_2$ and Pb($NO_3$)$_2$ of 5 or 10 mM for 5 days, the amount of bioaccumulation of $Pb^{2+}$ in the leaf of P. thunbergii was 2.87-8.08$\mu\textrm{g}$/g and that of $Cd^{2+}$ was 0.82-2.79$\mu\textrm{g}$/g. In the case of P. thunbergii, the concentration of $Pb^{2+}$ in the leaf was higher than that of $Cd^{2+}$. On the other hand, in R. crispus, the concentration of $Cd^{2+}$ and $Pb^{2+}$ were similar as follows ; 1.49$\mu\textrm{g}$/g in $Cd^{2+}$ 5mM, 2.90$\mu\textrm{g}$/g in Cd2+ 10mM, 1.83$\mu\textrm{g}$/g in $Pb^{2+}$ 5mM and 2.73$\mu\textrm{g}$/g in $Pb^{2+}$ 10mM. The remaining rate of heavy metals and the variation of pH in the cultured soil decreased as compared with control (100 % and pH 6.48) after 5 days as follows; to 77.l% and pH 6.39 in $Cd^{2+}$ 5mM, 90.2% and pH 5.79 in $Cd^{2+}$ 10 mM, 81.1% and pH 6.00 in $Pb^{2+}$ 5mM, and 85.7% and pH 5.80 in $Pb^{2+}$ 10 mM. The result of size exclusion chromatography, several phytochelatins were seperated from the extract of the leaf of both plants treated with heavy metals. The molecular mass of these phytochelatins were estimated as follows; in the case of P. thunbergii, about 4,300-8,600 da by $Cd^{2+}$ and about 3,200-9,700 da by $Pb^{2+}$, and in R. crispus, about 4,300 da by $Cd^{2+}$ and about 3,200-7,500 da by $Pb^{2+}$.   • Pot and field experiments were conducted to find out the effects of lime application on growth and Cd uptake of paddy rice in Cd added and contaminated soils. Lime application to soil-applied Cd could reduce Cd content in brown rice as well as increasing rice yields compared to that of no lime. The optimum requirement of lime to maintain Cd content in brown rice below 0.4 ppm (precaution concentration) were 56,381 and 512kg/10a at 3, 6 and 12 ppm of Cd in soil by pot experiment and 142.7 and 623.9kg/10a at 1.5 and 5.4 ppm of Cd in soil by field trial, respectively. The recommendable soil pH before transplanting to keep the Cd content in brown rice below 0.4 ppm were 5.2, 7.2 and 7.5 at 3, 6 and 12 ppm of Cd in soil, respectively.

Adsorption Kinetics of metals (Cu, Cd, Pb) in Tidal Flat Sediments and Yellow Loesses (갯벌과 황토에 의한 중금속 (Cu, Cd, Pb)의 흡착 kinetics)

  • The purpose of this study was to investigate the adsorption kinetics of heavy metals (Cu, Cd and Pb) using three tidal flat sediments and two yellow loesses. The relationship between adsorption rate calculated by non-linear regression model and chemical parameters was estimated. The contents of ignitiot loss (I.L.) am Fe, Mn and Al oxides of yellow loess were higher $1.5{\~}6 times$ than those of tidal flat sediments. But the contents of silt and clay of tidal flat sediment in Eueunri was higher than others. Heavy metals adsorption were occured rapidly in the intial 30 min and the concentration of adsorbed heavy metals were $4.1{\~}14.7\;{\mu}g/g\;for\;Cu,\;2.8{\~}16.7\;{\mu}g/g\;for\;Cd\;and\;43.4{\~}101.7\;{\mu}g/g$ for Pb, showing a high cumulative adsorption of $8{\~}70{\%}\;for\;Cu,\;18{\~}31{\%}\;for\;Cd and\;19{\~}52{\%}$ for Pb after 3hr. In initial concentration of $0.5{\times}10^(-5)M$, adsorption rate of heavy metals by the tidal flat sediments and yellow loesses was the sequence Pb>Cu^gt;Cd. The adsorption kinetics of Cu, Cd and Pb was found to be one-site kinetic model. Especially, in the case of Cu, there was a high negative ($R^2= -0.88{\~}-0.99$) linear correlation between chemical parameter such as I.L., Al oxide, silt and clay, and adsorption rate coefficients ($K_a$) calculated by non-linear model.

The CD28-B7 Family in Anti-Tumor Immunity: Emerging Concepts in Cancer Immunotherapy

  • The interactions between B7 molecules and CD28-family receptors are crucial in the regulation of adaptive cellular immunity. In cancer, the aberrant expression of co-inhibitory B7 molecules has been attributed to reduced anti-tumor immunity and cancer immune evasion, prompting the development of cancer therapeutics that can restore T cell function. Murine tumor models have provided significant support for the targeting of multiple immune checkpoints involving CTLA-4, PD-1, ICOS, B7-H3 and B7-H4 during tumor growth, and clinical studies investigating the therapeutic effects of CTLA-4 and PD-1 blockade have shown exceptionally promising results in patients with advanced melanoma and other cancers. The expression pattern of co-inhibitory B7 ligands in the tumor microenvironment has also been largely correlated with poor patient prognosis, and recent evidence suggests that the presence of several B7 molecules may predict the responsiveness of immunotherapies that rely on pre-existing tumor-associated immune responses. While monotherapies blocking T cell co-inhibition have beneficial effects in reducing tumor burden, combinatorial immunotherapy targeting multiple immune checkpoints involved in various stages of the anti-tumor response has led to the most substantial impact on tumor reduction. In this review, we will examine the contributions of B7- and CD28-family members in the context of cancer development, and discuss the implications of current human findings in cancer immunotherapy.
