Significance of Perianal Lesion in Pediatric Crohn Disease

  • Purpose: Despite the increasing incidence of pediatric Crohn disease (CD) in Korea, data on the characteristics of perianal lesions are scarce. Therefore, we aimed to investigate the characteristics of pediatric CD with accompanying perianal lesions in Korea. Methods: We retrospectively reviewed the medical records of children (age ${\leq}18$ years) with confirmed CD at Gachon University Gil Medical Center between 2000 and 2014. Patients were classified into two groups based on the presence or absence of any perianal lesions including skin tags. Additional analysis was performed according to the presence or absence of perianal perforating lesions. Results: Among the 69 CD children (mean age, 15.4 years) include in the analysis, 54 (78.3%) had a perianal lesion and 29 (42.0%) had a perianal perforating lesion. The median duration of chief complaints was longer in pediatric CD with any accompanying perianal lesions (5.40 months vs. 1.89 months, p=0.02), while there was no difference between pediatric CD with and without perianal perforating lesions (5.48 months vs. 4.02 months, p=0.18). Perianal symptoms preceded gastrointestinal symptoms in 13 of 29 (44.8%) patients with perianal perforating lesions. Conclusions: CD should be suspected in children with perianal lesions, even in circumstances when gastrointestinal symptoms are absent.

고려홍삼이 HIV-1 감염자에서 혈청 soluble CD8 항원 농도에 미치는 영향 (Effect of Korean Red Ginseng on Serum Soluble CD8 in HIV-1-Infected Patients)

  • 고려홍삼이 HIV-1 감염자에게서 바이러스의 활발한 증식으로 유발된 면역계의 과활성화를 장기간에 걸쳐 억제하는지 살펴보기 위해 면역 활성화 지표인 soluble CD8항원(sCD8)을 측정하였다. HIV-1 감염 대조군 49명, zidovudine (ZDV) 복용군 22명, KRG 복용군 48명, KRG와 ZDV 병용군(49명)을 대상으로 하였다. HIV-1 감염 대조군에서는 혈청 sCD8 농도가 31-48개월 동안 33% 증가하였으며(P < 0.05), sCD8/CD8+T 림프구 비도 $21{\pm}13$개월동안 54% 증가하였다(P < 0.001). ZDV 복용군에서는 혈청 sCD8농도가 초기 6개월 동안은 감소하였으나 그 이후로는 서서히 증가하였으며, sCD8/CD8+T 림프구 비도 증가하였다. KRG 복용군은 sCD8 농도가 31-48개월 동안 45% 감소하였으며(P < 0.01), sCD8/CD8+T 림프구 비도 $19\;{\pm}\;11$개월 동안 19% 감소하였다(P < 0.05). 병용군에서는 sCD8 농도가 31-48개월 동안 29%(P < 0.01) 감소하였다. 추적관찰 기간 동안 각 군간의 sCD8 농도 변화가 명확한 차이를 보였으며, KRG 복용군에서는 ZDV 복용군에서 관찰된 'rebound현상'(sCD8이 초기에 감소하다가 다시 증가하는 경향)이 없었다. 결론적으로 본 연구에서는 HIV-1 감염환자에서 KRG를 장기간 복용시켰을 경우 sCD8 농도가 지속적으로 감소하여, KRG가 면역계의 소모적인 과활성화 상태를 방지함을 알 수 있었다.

만성적인 저 농도 톨루엔 노출이 세포성면역 기능에 미치는 영향 (Influence of Chronic Low-Level Exposure to Toluene on Cell-Mediated Immunity)

  • Objectives: This study aimed to evaluate the effects of low-level exposure to toluene on T lymphocytes subpopulations.s. Methods: The study lasted from April to October 2010. The subjects were 390 male workers, among whom 137 were chronically exposed to toluene in video-tape manufacturing factories and 253 were controls had never been occupationally exposed to hazardous chemicals. The subpoupulations of CD4+, CD8+, CD16+ (natural killer cells) and total (CD3+) T lymphocytes were examined by two-color staining using monoclonal antibodies. The general and job characteristics of subjects were assessed through a self-administered questionnaire. Results: There was no significant difference in general and job characteristics between both groups. No significant difference in lipid peroxide level was observed between the control and exposed workers, but the concentration of hydrogen peroxide was significantly higher in the exposed workers. The numbers of CD16+ T lymphocytes in controls were significantly higher than those in exposed workers, but no significant differences were found in CD4+, CD8+ and CD3+ T lymphocytes. Hydrogen peroxide levels showed a significantly negative correlation with CD8+ (r = -0.29, p < 0.01), CD16+ (r = -0.56, p < 0.01) and CD3+(r = -0.22. p < 0.01), and toluene levels was significantly negative correlated with CD3+ (r = -0.29, p < 0.05). Conclusions: Our results suggest that chronic low-level exposure to toluene affects cell-mediated immunity and the effects might mediate through ROSs (Reactive Oxygen Species) such as hydrogen peroxide.

Sphagnum 피트모스에서의 카드뮴, 구리, 납의 흡착 (Adsorption of Cadmium, Copper, and Lead on Sphagnum Peat Moss)

  • Sphagnum 피트모스에서의 Cd(II), Cu(II) Pb(II)의 흡착에 대한 회분식 실험을 수행하였다. 실험결과 10-50 mg/L의 Cd(II), Cu(II), Pb(II)이 1.0 g/L의 sphagnum 피트모스에 1시간 내에 흡착되어 효과적으로 제거되었다. 초기농도에 따라 sphagnum 피트모스에 홉착된 Cd(II), Cu(II), Pb(II)의 반응속도는 유사이차 반응모델을 적용하여 설명하였다. 또한 Langmuir 등온흡착으로부터 Cd(II), Cu(II), Pb(II)에 대한 최대흡착량을 계산하여, 각각 33.90, 29.15, 91.74 mg/g가 sphagnum 피트모스에 흡착될 수 있음을 알아내었다. Sphagnum 피트모스가 Cd(II), Cu(II), Pb(II)를 흡착하는데 매우 효과적인 흡착제임을 실험결과를 통해 나타내었다.

FD-CD 트랜스코팅기법에서 CD에 의한 전파/누적 왜곡의 분석과 모델링 (An Analysis and Modeling of Propagation/Accumulation Errors Incurred by CD in the FD-CD Transcoding)

  • 최근에 FD(frame Dropping)-CD(Coefficient Dropping) 트랜스코딩기법은 계산량을 줄이고, 손쉽게 구현할 수 있다는 측면에서 주목을 받고 있다. 그러나, 기존의 기법에서는 CD로 인하여 발생되는 왜곡 특성이 GOP내의 부호화 의존성을 갖는 프레임의 복호화 영상 품질에 전파 및 누적되는 성질을 고려하고 있지 않다. 본 논문에서는 CD로 인하여 발생되는 왜곡을 수식적으로 분석하고, CD왜곡이 전파/누적됨을 모의실험을 통하여 분석한다. 또한, 실험직인 결과에 기초하여, CD에 의한 전파/누적 왜곡 특성을 지수 감소함수 형태로 모델링한다. 모의실험을 통하여, 제안된 모델은 CD로 인해 초래되는 전체 누적왜곡 크기를 잘 예측할 수 있음을 보인다.

20S-dihydroprotopanaxadiol, a ginsenoside derivative, boosts innate immune responses of monocytes and macrophages

  • 20S-dihydroprotopanaxadiol (2H-PPD) is a derivative of protopanaxadiol, a glycone of ginsenosides prepared from Panax ginseng. Although ginsenosides and acidic polysaccharides are known to be major active ingredients in ginseng, the immunopharmacological activities of their metabolites and derivatives have not been fully explored. In this study, we aimed to elucidate the regulatory action of 2H-PPD on the function of monocytes and macrophages in innate immune responses. 2H-PPD was able to boost the phagocytic uptake of fluorescein isothiocyanate-dextran in macrophages and enhance the generation of radicals (reactive oxygen species) in sodium nitroprusside-treated RAW264.7 cells. The surface levels of the costimulatory molecules such as CD80 and CD86 were also increased during 2H-PPD treatment. In addition, this compound boosted U937 cell-cell aggregation induced by CD29 and CD43 antibodies, but not by cell-extracellular matrix (fibronectin) adhesion. Similarly, the surface levels of CD29 and CD43 were increased by 2H-PPD exposure. Therefore, our results strongly suggest that 2H-PPD has the pharmacological capability to upregulate the functional role of macrophages/monocytes in innate immunity.

Aqueous Solubility Enhancement of Some Flavones by Complexation with Cyclodextrins

  • The inclusion complexes of cyclodextrins (CDs) with flavones in aqueous solution were investigated by phase solubility measurements. The effect of b -cyclodextrin (b -CD), heptakis (2,6-di-O-methyl) b -cyclodextrin (DM-b -CD) and 2-hydroxypropyl-b -cyclodextrin (HP-b -CD) on the aqueous solubility of three flavones, namely, chrysin, apigenin and luteolin was investigated, respectively. Solubility enhancements of all flavones obtained with three CDs followed the rank order: HP-b -CD > DM-b -CD > b -CD, and besides, CDs show higher stability constant on luteolin than that on others flavones. 1H-nuclear magnetic resonance (NMR) spectroscopy and molecular modeling was used to help establish the model of interaction of the CDs with luteolin. NMR spectroscopic analysis suggested that A-C ring, and part of the B ring of luteolin display favorable interaction with the CDs, which was also confirmed by docking studies based on the molecular simulation. The observed augmentation of solubility of luteolin by three CDs was explained by the difference of electrostatic interaction of each complex, especially hydrogen bonding.

TiO2/CdS 복합광촉매의 밴드갭 에너지 특성과 광촉매 효율 (Photocatalytic Efficiency and Bandgap Property of the CdS Deposited TiO2 Photocatalysts)

  • To improve photocatalytic performance, CdS nanoparticle deposited TiO2 nanotubular photocatalysts are synthesized. The TiO2 nanotube is fabricated by electrochemical anodization at a constant voltage of 60 V, and annealed at 500 for crystallization. The CdS nanoparticles on TiO2 nanotubes are synthesized by successive ionic layer adsorption and reaction method. The surface characteristics and photocurrent responses of TNT/CdS photocatalysts are investigated by scanning electron microscopy (SEM), X-ray diffraction (XRD), UV-Vis spectrometer and LED light source installed potentiostat. The bandgaps of the CdS deposited TiO2 photocatalysts are gradually narrowed with increasing of amounts of deposited CdS nanoparticles, which enhances visible light absorption ability of composite photocatalysts. Enhanced photoelectrochemical performance is observed in the nanocomposite TiO2 photocatalyst. However, the maximum photocurrent response and dye degradation efficiency are observed for TNT/CdS30 photocatalyst. The excellent photocatalytic performance of TNT/CdS30 catalyst can be ascribed to the synergistic effects of its better absorption ability of visible light region and efficient charge transport process.

Preparation and Bioevaluation of 177Lu-labelled Anti-CD44 for Radioimmunotherapy of Colon Cancer

  • CD44 is a particular adhesion molecule and facilitates both cell-cell and cell-matrix interactions. In particular, splice variants of CD44 are particularly overexpressed in a large number of malignancies and carcinomas. In this study, the $^{177}Lu$-labelled CD44 targeting antibody was prepared and bioevaluated in vitro and in vivo. Anti-CD44 was immunoconjugated with the equivalent molar ratio of cysteine-based DTPA-NCS and radioimmunoconjugated with $^{177}Lu$ at room temperature within 15 minutes. The stability was tested in human serum. An in vitro study was carried out in HT-29 human colon cancer cell lines. For the biodistribution study $^{177}Lu$-labelled anti-CD44 was injected in xenograft mice. Anti-CD44 was immunoconjugated with cysteine-based DTPA-NCS and purified by a centricon filter system having a molecular cut-off of 50 kDa. Radioimmunoconjugation with $^{177}Lu$ was reacted for 15 min at room temperature. The radiolabeling yield was >99%, and it was stable in human serum without any fragmentation or degradation. The radioimmunoconjugate showed a high binding affinity on HT-29 colon cancer cell surfaces. In a biodistribution study, the tumor-to-blood ratio of the radioimmunoconjugate was 43 : 1 at 1 day post injection (p.i) in human colon cancer bearing mice. The anti-CD44 monoclonal antibody for the targeting of colon cancer was effectively radioimmunoconjugated with $^{177}Lu$. The in vitro high immunoactivity of this radioimmunoconjugate was determined by a cell binding assay. In addition, the antibody's tumor targeting ability was demonstrated with very high uptake in tumors. This radioimmunoconjugate is applicable to therapy in human colon cancer with highly expressed CD44.

자동차용 CD/DVD 데크의 진동 저감 및 성능 향상에 관한 연구 (A Study on the Vibration Reduction and Performance Improvement of the CD/DVD for the Vehicles)

  • Dynamic characteristics of CD/DVD deck fur vehicle are analyzed in this paper. Generally CD/DVD deck for vehicle is used from inferior environment with the vibration of the vehicle, shock of outside and so on. Therefore it must have the structure which is stabilized from the vibration to prevent read error. For this purpose, vibration characteristic of the deck for vehicle should be identified. To analyze characteristic of the deck system, we perform the signal analysis and modal testing using the EFT analyzer. Also we change the design factor degrading the performance of the deck system and verify the efficiency improvement using the acceleration measurement occurring to the sound discontinuation.
