CD Gene Microarray Profiles of Bambusae Caulis in Liquamen in Human Mast Cell

  • Bambusae Caulis in Liquamen(BCL) has been used to relieve the cough and asthma, and remove the phlegm in traditional Oriental medicine. In recent years, it was studied for its antiinflammatory, antiallergenic, immune-modulating, and anticarcinogenic capabilities. This experiment was performed to evaluate the microarray profiles of CD genes in human mast cells before and after BCL treatment. The results are as follows: The expression of 51 of the genes studied was up-regulated in the Bel-treated group; they include the genes coding L apoferritin, beta-2-microglobulin, ferritin light polypeptide, CD63, monocyte chemotactic and activating fact, heme oxygenase 1, CD140a, integrin alpha M, colony stimulating factor 2 receptor, eukaryotic translation elongation factor, CD37, interleukin 18, NADH dehydrogenase 1 beta, CD48, 5-lipoxygenase activating protein, interleukin 4, ribosomal protein L5, GABA(A) receptor-associated protein, beta-tubulin, integrin beta 1, CD162, CD32, lymphotoxin beta, alpha-tublin, integrin alpha L, CD2, CD151, CD331, 90 kDa heat shock protein, CD59, CD3Z, microsomal glutathione S-transferase 2, CD33, CD162R, cyclophilinA, CD84, interleukin 9 receptor, interleukin 11, CD117, CD39-Like 2, and so forth. The expression of 7 of the genes studied was down-regulated in the BCL-treated group; they include the genes coding con, CD238, SCF, CD160, CD231, CD24, and CD130. Consequently, the treatment of BCL on the human mast cells increased the expression of 51 genes and decreased the expression of 7 genes. These data would provide a fundamental basis to the traditional applications of Bambusae Caulis in Liquamen.

Purification and Characterization of Cyclodextrin Glycosyltransferase from Bacillus brevis CD162 (Bacillus brevis CD162 Cyclodextrin Glycosyltransferase의 정제 및 특성)

  • The cyclodextrin glycosyltransferase (CGTase, EC 3.2.1.19) from Bacillus brevis CD162 was purified by precipitating with ammonium sulfate, DEAE-Sepharose CL-6B column chromatography and Sephadex G-150 column chromatography. The molecular mass and pI of the purified enzyme were estimated to be 74,000 and 6.3 by SDS-PAGE and isoelectric focusing, respectively. The purified enzyme was clearly identified as the CGTase by zymogram after SDS-PAGE. The optimum pH and temperature for the enzyme activity were 8.0 and $55^{\circ}C$, respectively. The enzyme was stable at the range of pH $5.5{\sim}9.0$, and up to $50^{\circ}C$. The amino acid sequence from the $NH_2-terminal$ of the purified CGTase was Ser-Val-Thr-Asn-Lys-Val-Asn-Tyr-Ser-Lys-Asp-Val-Ile-Tyr-Gln. The yields of the products from starch as the substrate were 1.3% for ${\alpha}-$, 33.9% for ${\beta}-$, and 9.7% for ${\gamma}-cyclodextrin$.

Factors Controlling the Deposition of Airborne Metals on Plant Leaves in a Subtropical Industrial Environment

  • This study was conducted in an industrial city (Bilaspur) representative of subtropical area in central India. In order to assess the metal deposition on plant, concentrations of six target metals (i.e., Fe, Mn, Pb, Cu, Cd, and Cr) in both plant leaf and dust (deposited on its surface) samples were measured from six different sites. Metal concentrations in dust samples were found on the order of Fe>Mn> Cr>Pb>Cu>Cd. In contrast, the concentration of metals in plant leaves were seen on the order of Fe>Mn>Cr>Cd>Cu>Pb. As such, Cd showed significantly high concentration in leaves relative to their corresponding dust samples. A high accumulation potential for Fe and Cd was seen from Butea monosperma, while Mn and Pb were accumulated noticeably in Pongamia pinnata and Butea monosperma. Likewise, Cr and Cu were enriched in Calotropis procera, Alstonia scholaris, and Butea monosperma. The overall results of our study suggest that the foliar uptake pattern should vary considerably by an interactive role between plant and metal types.

Effects of Different Dietary Cadmium Levels on Growth and Tissue Cadmium Content in Juvenile Parrotfish, Oplegnathus fasciatus

  • This feeding trial was carried out to evaluate the effects of different dietary cadmium levels on growth and tissue cadmium content in juvenile parrotfish, Oplegnathus fasciatus, using cadmium chloride ($CdCl_2$) as the cadmium source. Fifteen fish averaging $5.5{\pm}0.06$ g (mean${\pm}$SD) were randomly distributed into each of twenty one rectangular fiber tanks of 30 L capacity. Each tank was then randomly assigned to one of three replicates of seven diets containing 0.30 ($C_0$), 21.0 ($C_{21}$), 40.7 ($C_{41}$), 83.5 ($C_{83}$), 162 ($C_{162}$), 1,387 ($C_{1,387}$) and 2,743 ($C_{2,743}$) mg cadmium/kg diet. At the end of sixteen weeks of feeding trial, weight gain (WG), specific growth rate (SGR) and feed efficiency (FE) of fish fed $C_{21}$ were significantly higher than those of fish fed $C_{83}$, $C_{162}$, $C_{1,387}$ and $C_{2,743}$ (p<0.05). Weight gain, SGR and FE of fish fed $C_0$, $C_{21}$ and $C_{41}$ were significantly higher than those of fish fed $C_{162}$, $C_{1,387}$ and $C_{2,743}$. Protein efficiency ratio of fish fed $C_0$, $C_{21}$ and $C_{41}$ were significantly higher than those of fish fed $C_{1,387}$ and $C_{2,743}$. Average survival of fish fed $C_0$, $C_{21}$, $C_{41}$ and $C_{162}$ were significantly higher than that of fish fed $C_{2,743}$. Tissue cadmium concentrations increased with cadmium content of diets. Cadmium accumulated the most in liver, followed by gill and then muscle. Muscle, gill and liver cadmium concentrations of fish fed $C_0$, $C_{21}$, $C_{41}$ and $C_{83}$ were significantly lower than those of fish fed $C_{162}$, $C_{1,387}$ and $C_{2,743}$. Based on the ANOVA results of growth performance and tissue cadmium concentrations the safe dietary cadmium level could be lower than 40.7 mg Cd/kg diet while the toxic level could be higher than 162 mg Cd/kg diet.

Epidemiological Observation on Recent Outbreaks of Canine Distemper in Korea (최근 국내발병 개 디스템퍼에 대한 역학적 조사)

  • To characterize the recent outbreaks of canine distemper (CD) in Korea, we carried out epidemiological investigations by clinical observations, serum neutralizing (SN) antibody titer determination and RT-PCR on the 315 dogs which were clinically suspected as canine distemper virus (CDV) infection. One hundred and sixty two of 315 dogs were infected with CDV. Breed or gender did not seem to have effects on the prevalence of CD. The major part of dogs were in young age from 6 weeks to 18 weeks of age, and were not vaccinated or incompletely vaccinated. Clinical signs of dogs with CD were multi systemic and extremely variable. Dogs died from CD had significantly more ocular signs and neurologic signs than those of dogs survived (p<0.05). The SN titers against CDV of 157 (96.9%) dogs were under 1:16, which is less than protective level. One possible explanation for recent outbreaks of CD in Korea might be low antibody titers against CDV because of vaccination failure. Therefore, to reduce the impact of virulent infection in the dog population, dogs should be vaccinated adequately and prophylactic measures should include isolation of young dogs from the dog population until vaccination can be expected to provide protection.

  • Anaplasma phagocytophilum major surface protein-2 (Msp2 or p44) is the immunodominant outer membrane protein of the bacterium. Recently, we disclosed that Msp2 was an A. phagocytophilum adhesin for binding to host neutrophils and HL-60 cells, probably mediated by attachment to platelet selectin glycoprotein ligand-1 (PSGL-1). In this study, we further elucidated that Msp2 bound to PSGL-1/FucT IV-transfected BJAB but not nontransfected BJAB cells. Binding of recombinant Msp2 or cell (lee bacteria to the surface of PSGL-1/FucT IV-transfected BJAB cells was significantly higher than to nontransfected BJAB cells (p<0.01 and p<0.01). Also, Msp2 monoclonal antibody and soluble recombinant Msp2 as antagonist led to concentration-dependent reductions in A. phagocytophilum adhesln (p<0.05 and p<0.01) to transfected BJAB cells. Thus, we conclude that Msp2 of. A. phagocytophilum acts as an adhesin by which the bacterium binds to PSGL-1 on host neutrophils and myeloid cells.

Relationships in Cd, Hg, Pb and Zn Concentrations Between Hair and Internal Organs of Korean (한국인 모발과 내부장기조직간 Cd, Hg, Pb 및 Zn함량의 상호관련성)

  • In order to study the relationships between trace element concentrations of hair and internal organs, autopsy samples of hair, liver, kidney cortex, kidney medulla, heart, lung, spleen, cerebrum, testis and bone from 162 Korean subjects were analyzed for Cd, Hg, Pb and Zn by inductively coupled plasma atomic emission spectrometry. High significant positive correlations were found between the Cd concentration in hair and bone and Pb in hair and lung. Hair Hg levels were highly significantly correlated with organ Hg levels in the liver, kidney cortex, kidney medulla, heart, lung, spleen, cerebrum, testis and bone. And hair Zn levels were highly significantly correlated with organ Zn levels in the kidney cortex, kidney medulla, spleen, testis and bone.

Effect of Korean Red Ginseng intake on the survival duration of human immunodeficiency virus type 1 patients

  • Background: Long-term ginseng intake can increase longevity in healthy individuals. Here, we examined if long-term treatment with Panax ginseng Meyer (Korean Red Ginseng, KRG) can also enhance survival duration (SD) in patients with human immunodeficiency virus type 1 (HIV-1) infection. Methods: We retrospectively analyzed 252 HIV-1 patients diagnosed from 1986 to 2013 prior to the initiation of antiretroviral therapy. Overall, 162 patients were treated with KRG ($3,947{\pm}4,943g$) for $86{\pm}63$ mo. The effects of KRG on SD were analyzed according to the KRG intake level and the length of the follow-up period. Results: There were significant correlations between the total amount of KRG and SD in the KRG intake group (r = 0.64, p < 0.0001) as well as between total amount of KRG and mean annual decrease in $CD4^+$ T-cell count in all 252 patients (r = -0.17, p < 0.01). The annual decrease in $CD4^+$ T-cell count (change in $cells/{\mu}L$) was significantly slower in KRG-treated patients than in patients receiving no KRG ($48{\pm}40$ vs. $106{\pm}162$; p < 0.001). The SD (in months) was also significantly longer in the KRG group than in the no-KRG group ($101{\pm}64$ vs. $59{\pm}40$, p < 0.01). Conclusion: KRG prolongs survival in HIV-1 patients, possibly by slowing the decrease in $CD4^+$ T-cell count.

A Low-Dose High-Resolution SPECT System with CdTe for Small-Animal Imaging Applications: A GATE Simulation Study (GATE 시뮬레이션을 통한 고해상도 저선량용 소동물 영상화를 위한 CdTe 검출기 기반의 SPECT 기기 연구)

  • Dedicated single-photon emission computed tomography (SPECT) systems based on pixelated semiconductors are being developed for studying small animal models of human disease. To clarify the possibility of using a SPECT system with CdTe for a high resolution low-dose small animal imaging, we compared the quality of reconstructed images from pixelated CdTe detector to those from a small SPECT system with NaI(Tl). The CdTe detector was $44.8{\times}44.8$ mm and the pixels were $0.35{\times}0.35{\times}5$ mm. The intrinsic resolution of the detector was 0.35 mm, which is equal to the pixel size. GATE simulations were performed to assess the image quality of both SPECT systems. The spatial resolutions and sensitivities for both systems were evaluated using a 10 MBq $^{99m}Tc$ point source. The quantitative comparison with different injected dose was performed using a voxelized MOBY phantom, and the absorbed doses for each organ were evaluated. The spatial resolution of the SPECT with NaI(Tl) was about 1.54 mm FWHM, while that of the SPECT with a CdTe detector was about 1.32 mm FWHM at 30 mm. The sensitivity of NaI(Tl) based SPECT was 83 cps/MBq, while that of the CdTe detector based SPECT was 116 cps/MBq at 30 mm. The image statistics were evaluated by calculating the CNR of the image from both systems. When the injected activity for the striatum in the mouse brain was 160 Bq/voxel, the CNR of CdTe based SPECT was 2.30 while that of NaI(Tl) based SPECT was 1.85. The CNR of SPECT with CdTe was overall higher than that of the NaI(Tl) based SPECT. In addition, the absorbed dose was higher from SPECT with CdTe than those from NaI(Tl) based SPECT to acquire the same quantitative values. Our simulation results indicated that the SPECT with CdTe detector showed overall high performance compared to the SPECT with NaI(Tl). Even though the validation study is needed, the SPECT system with CdTe detector appeared to be feasible for high resolution low-dose small animal imaging.

Alteration of $CD4^+CD25^+Foxp3^+$ T cell level In Kawasaki disease

  • Purpose: Exaggerated pro-inflammatory reactions during the acute phase of Kawasaki disease (KD) suggest the role of immune dysregulation in the pathogenesis of KD. We investigated the profiles of T regulatory cells and their correlation with the clinical course of KD. Methods: Peripheral blood mononuclear cells were collected from 17 KD patients during acute febrile and subacute afebrile phases. T cells expressing CD4, CD25, and Foxp3 were analyzed using flow cytometry, and the results were correlated with the clinical course of KD. Results: The percentage of circulating $CD4^+CD25^{high}Foxp3^+$ T cells among $CD4^+$ T cells was Significantly higher during the subacute afebrile phase than during the acute febrile phase ($1.10%{\pm}1.22%$ vs. $0.55%{\pm}0.53%$, P=0.049). Although levels of $CD4^+CD25^{low}Foxp3^+$ T cells and $CD4^+CD25^-Foxp3^+$ T cells were only slightly altered, the percentage of $CD4^+CD25^+Foxp3^-$ T cells among $CD4^+$ T cells was significantly lower during the subacute afebrile phase than during the acute febrile phase ($2.96%{\pm}1.95%$ vs. $5.64%{\pm}5.69%$, P=0.036). Consequently, the ratio of $CD25^{high}Foxp3^+$ T cells to $CD25^+Foxp3^-$ T cells was higher during the subacute afebrile phase than during the acute febrile phase ($0.45%{\pm}0.57%$ vs. $0.13%{\pm}0.13%$, P=0.038). Conclusion: Decreased $CD4^+CD25^{high}Foxp3^+$ T cells and/or an imbalanced ratio of $CD4^+CD25^{high}Foxp3^+$ T cells to $CD4^+CD25^+Foxp3^-$ T cells might playa role in KD development. Considering that all KD patients were treated with intravenous immunoglobulin (IVIG), recovery of $CD4^+CD25^{high}Foxp3^+$ T cells during the subacute afebrile phase could be a mechanism of IVIG.
