Concentration of Hazardous Substances of Before/after a Decoction- In Prescription of High Frequency - (전탕 전과 후의 한약재 및 처방에 포함된 위해물질의 농도변화 -다빈도 한약 처방을 중심으로-)

  • Objectives: To compare the contents of hazardous substances before/after a decoction. Methods : The heavy metal contents before/after a decoction were measured by Inductively Coupled Plasma Atomic Emission Spectrometer (ICP-AES) and mercury analyzer. In order to analyze pesticides in 6 samples we used simultaneous multi-residue analysis of pesticides by GC/ECD, which was followed by GC/MSD analysis to confirm the identity of the detected pesticide in each sample. In addition, the contents of sulfur dioxide (S02) were performed by Monier-Williams distillation method. Results : 1. The mean values of heavy metal contents (mg/kg) for the samples were as follows: Socheongryong-tang (before decoction - Pb; 1.115, Cd; 0.179, As; 0.069 and Hg; 0.028, after decoction - Pb; 0.110, Cd; 0.011, As; 0.005 and Hg; 0.002), Insampaedok-san (before decoction - Pb; 1.207, Cd; 0.148, As; 0.171 and Hg; 0.026, after decoction - Pb; 0.075, Cd; 0.006, As; not detected and Hg; O.OOD, Oryung-san (before decoction - Pb; 1.955, Cd; 0.430, As; 0.063 and Hg; 0.027, after decoction - Pb; 0.083, Cd; 0.013, As; 0.006 and Hg; 0.002), Hwangryunhaedok-tang (before decoction - Pb; 1.825, Cd; 0.210, As; 0.050 and Hg; 0.009, after decoction - Pb; 0.107, Cd; 0.010, As; 0.005 and Hg; O.OOD, Bangpungtongseong-san (before decoction - Pb; 1.740, Cd; 0.162, As; 0.585 and Hg; 0.018, after decoction - Pb; 0.041, Cd; 0.006, As; 0.022 and Hg; not detected) and Oyaksungi-san (before decoction - Pb; 1.199, Cd; 0.183, As; 0.321 and Hg; 0.031, after decoction - Pb; 0.096, Cd; 0.008, As; 0.021 and Hg; 0.0004). 2. Contents (mg/kg) of sulfur dioxide (S0$_2$) before a decoction in Socheongryong-tang, Insampaedok-san, Oryung-san, Hwangryunhaedok-tang, Bangpungtongseong-san and Oyaksungi-san exhibited 3.2, 5.7, 4.5, 49.8, 7.8 and 22.4, respectively. However, contents of sulfur dioxide after a decoction in all samples were not detected. 3. Contents (mg/kg) of residual pesticides before/after a decoction in all samples were not detected. Conclusions : These results will be used to establish a criterion of heavy metals, residual pesticides and sulfur dioxide.

Contents (mg/kg) of sulfur dioxide ($SO_2$) before a decoction in Banhabaikchulcheunma-tang, Pyungwi-san, Leejung-tang and Kwibi-tang exhibited 3.5, 3.4, 3.8 and 12.4, respectively. However, contents of sulfur dioxide after a decoction in all samples were not detected. 3. Contents (mg/kg) of residual pesticides before/after a decoction in all samples were not detected. Conclusions : These results will be used to establish a criterion of heavy metals, residual pesticides and sulfur dioxide.

  • Cadmium telluride (CdTe) films have been prepared on Corning 7059 glass, molybdenum (Mo), and polyimide (PI) substrates by r.f. magnetron sputtering technique. The influence of the sputter pressure on the structural and optical properties of these films was evaluated. In addition, a comparison of the properties of the films deposited on fferent substrates was performed.

Tissue Distribution, SNP Detection and Association Study with Immune Traits of Porcine LBP and CD14 Genes

  • Lipopolysaccharide binding protein (LBP) and CD14 protein play important roles in the defense against infection of Gram-negative bacteria. In the present study, tissue distribution and polymorphism of porcine LBP and CD14 genes were analyzed. Real-time PCR results showed that the porcine LBP gene was especially highly expressed in liver, while CD14 gene was highly expressed in liver and spleen tissues. A 1,732 bp cDNA fragment of porcine LBP gene and a 1,682 bp genomic DNA fragment of CD14 gene were isolated. Polymorphisms were identified in these two fragments and showed that there were 14 potential SNPs in the porcine LBP gene and 3 potential SNPs in the porcine CD14 gene. Three SNPs, 292G/A (Gly/Ser), 1168G/A (Ala/Thr) of the LBP gene and -61G/A of the CD14 gene, were genotyped using restriction fragment length polymorphism (RFLP) method. Association analyses indicated that polymorphism of the 292G/A locus was significantly associated with porcine immune traits hematocrit (HCT), IgG and delayed-type hypersensitivity (DTH) (p<0.01), and the 1168G/A locus was significantly associated with HCT and mean corpuscular volume (MCV) traits (p<0.05). No significant association was found between the -61G/A locus and immune traits of the pig. Our data indicated that the LBP gene was significantly associated with immune traits of pig. Also, we identified some SNPs which may be useful markers for disease-resistant breeding of pigs.

Differential characterization of myogenic satellite cells with linolenic and retinoic acid in the presence of thiazolidinediones from prepubertal Korean black goats

  • Objective: Myogenic satellite cells were isolated from semitendinosus muscle of prepubertal Korean black goat to observe the differential effect of linolenic and retinoic acid in thepresence of thiazolidinediones (TZD) and also to observe the production insulin sensitive preadipocyte. Methods: Cells were characterized for their stemness with cluster of differentiation 34 (CD34), CD13, CD106, CD44, Vimentin surface markers using flow cytometry. Cells characterized themselves as possessing significant (p<0.05) levels of CD13, CD34, CD106, Vimentin revealing their stemness potential. Goat myogenic satellite cells also exhibited CD44, indicating that they possessed a % of stemness factors of adipose lineage apart from their inherent stemness of paxillin factors 3/7. Results: Cells during proliferation stayed absolutely and firmly within the myogenic fate without any external cues and continued to show a significant (p<0.05) fusion index % to express myogenic differentiation, myosin heavy chain, and smooth muscle actin in 2% horse serum. However, confluent myogenic satellite cells were the ones easily turning into adipogenic lineage. Intriguingly, upregulation in adipose specific genetic markers such as peroxisome proliferation-activated receptor ${\gamma}$, adiponectin, lipoprotein lipase, and CCAAT/enhancer binding protein ${\alpha}$ were observed and confirmed in all given treatments. However, the amount of adipogenesis was found to be statistically significant (p<0.01) with linolenic acid as compared to retinoic acid in combination with TZD's. Conclusion: Retinoic acid was found to produce smaller preadipocytes which have been assumed to have insulin sensitization and hence retinoic acid could be used as a potential agent to sensitize tissues to insulin in combination with TZD's to treat diabetic conditions in humans and animals in future.

Study on Immunological Response on Salmonella gallinarum in Immunosuppressed Chickens (면역억압계군에서 Salmonella gallinarum 감염에 대한 면역반응)

  • The immune response against Salmonella gallinarum infection was investigated in immunosuppresed chickens. Newly hatched chickens were treated with cyclophosphamide at the first and second day after birth and were challenged intraperitoneally with S gallinarum ($1{\times}10^7CFU/m{\ell}$) on day 6. Group 1, Immunosuppresed and Challenged group, was treated with cyclophaiphamide and challenged with S gallinarum; group 2, Immunosuppressed group, was only treated with cyclophsphamide; group 3, Challenged group, was only challenged with S gallinarum; group 4, Control group. In each group, the localization of lymphocytes of the lymphoid organs and intestine was immunohistochemically compared using a variety of monoclonal antiboies ($CD4^+$, $CD8^+$, and B lymphocyte). Also, S gallinarum were assessed by Maccallum-Goodpasture stain and immunohistochemical analysis in the paraffin-embedded intestinal tissues. In S gallinarum challenged chickens, $CD4^+$ and $CD8^+$ lymphocytes of the intestinal organs such as duodenum, jejunum, ileum and colon were increased. However, in cyclophophamide treated chickens, $CD4^+$ and $CD8^+$ lymphocytes and especially B lymphocytes of the lymphoid organs such as thymus, spleen, and bursa of Fabricius were dramatically decreased. These results suggest that cyclophsophamide is an immunosuppressive agent that especially causes depletion of B lymphocytes, suppress humoral immunity and eventually suppresses avian immune responses. Its protection against S gallinarum infection is mainly dependent on both cell-mediated mechanism and the humoral immune response.

  • This paper focuses on the after synthesis of CdTe quantum dots(QDs) in aqueous solution. CdTe nanoparticles were prepared in aqueous solution using mercaptocarboxylic acid or thioglycolic acid(TGA) as stabilizing agents. QDs emit light smaller than the nano size. The contents of the mercaptocarboxylic acid, and a kind of raw material, were revealed for a period of time. We succeeded in synthesizing a very high quality QDs solution; we discussed how to make QDs better and to keep them stabilized. TGA is known as one of the best stabilizing agents. Many papers have mentioned that TGA is a good stabilizing agent. We dramatically confirmed the state of QDs after the experiments. The QDs solution can be influenced by several factors. Different content of TGA can influence the stability of the CdTe solution. Most papers deal with the synthesis of CdTe, so we decided to discuss the after synthesis process for the stability of the CdTe solution.

Emission Characteristics of OLEDs Using LiF/Al/LiF Structure (LiF/Al/LiF 구조를 적용한 OLED 소자의 발광 특성)

  • We fabricated red and blue organic light emitting display (OLEDs) which had the two kinds of multi-structure of ITO/HIL/HTL/EML/ETL/LiF/Al and ITO/HIL/HTL/EML/ETL/LiF/Al/LiF. In the case of red OLED that had LiF/Al/LiF structure compared to LiF/Al structure, the current density increased from 4.3 mA/$cm^2$ to 7.3 mA/$cm^2$, and the brightness increased from 488 cd/$m^2$ to 1,023 cd/$m^2$ at 7.0 V, and as a result the current efficiency was improved from 11.28 cd/A to 13.95 cd/A. Also in the case of blue OLED that had LiF on Al cathode layer, the current density increased from 1.2 mA/$cm^2$ to 1.8 mA/$cm^2$, and the brightness increased from 45 cd/$m^2$ to 85 cd/$m^2$ at 7.0 V, and as a result the current efficiency was improved from 3.69 cd/A to 4.82 cd/A. Through these experimental results it could be suggested that the LiF layer formed on Al prevents the oxidation of Al surface, and the electrode resistance become low with increase of supplied electrons, therefore the brightness and the efficiency are improved from the influence to the well-balanced bonding of electron and hole at emitting layer.

Evaluation of a Rapid Immunochromatography Technique for Determination of Cd in Soils and Rice Grains (면역크로마토그래피를 이용한 토양 및 쌀의 Cd 간이진단법 평가)

  • Cadmium is known to be very toxic to human health and can be relative easily translocated from soils in plants. Therefore, a rapid method for screening Cd in soils and crops has become more and more important. For this reason, we examined a rapid immunochromatograpy (ICG) test kit which uses antigen-antibody reaction based on immunoassay and chromatography. Soils and rice grains collected from mine waste-contaminated sites were determined for their Cd contents using this kit. For comparison purposes, 0.1 M HCl and ICP-OES were employed as a conventional extraction and determination method. Cadmium contents in rice grains determined using ICG technique were $0.46{\sim}2.39mg\;kg^{-1}$ and Cd contents determined using 0.1 M HCl and ICP-OES were $0.52{\sim}1.97mg\;kg^{-1}$. The correlation between these two Cd contents were statistically significant ($r^2$=0.930). The results of Cd contents in soils also showed a statistically significant relationship between these two methods ($r^2$=0.975). On the basis of these results, ICG technique can be applied to rapidly quantify Cd in crops and soils. However, further research is necessary to apply ICG technique for the field screening.

The Expression of CD 18 on Ischemia- Reperfusion Injury of TRAM Flap of Rats (흰쥐의 복직근피부피판에 일으킨 허혈-재관류 손상에서 CD18의 발현)

  • Purpose: This study was to evaluate the expression pattern of CD 18(leukocyte adhesion glycoprotein) in ischemia-reperfusion injury of TRAM flap of rats. Through this study, we can obtain more information about ischemia-reperfusion injury. We want to develop specific medicine to improve the survival rate of TRAM flap in the future. Methods: A TRAM flap supplied by a single pedicle superior epigastric artery and vein was elevated on 60 Sprauge-Dawley rats. The rats were divide into 6 groups (each group n=10); Group O: sham, no ischemia-reperfusion injury, Group I: 2 hour reperfusion after 4 hour ischemia, Group II: 4 hour reperfusion after 4 hour ischemia, Group III: 8 hour reperfusion after 4 hour ischemia, Group IV: 12 hour reperfusion after 4 hour ischemia, and Group V: 24 hour reperfusion after 4 hour ischemia. This study consisted of gross examination for flap survival and flow cytometry study of CD18 on neutrophils. Results: The gross measurement of the flap showed different survival rate in group I(71%), II(68%), III(37%), IV(34%) and V(34%). All experimental groups showed an increase in the expression of CD18 compared to group O. The expression of CD18 was rapidly increased in ascending order in group I, II and III. But, the expression of CD18 was maintained in group IV and V. Conclusion: The results can be implemented in the study to develop drugs which are capable of reducing ischemia-reperfusion injury in microsurgical breast reconstruction.
