Physical Properties of Cd2GeSe4 and Cd2GeSe4:Co2+ Thin Films Grown by Thermal Evaporation (진공증착법에 의해 제작된 Cd2GeSe4와 Cd2GeSe4:Co2+ 박막의 물리적 특성)

  • $Cd_2GeSe_4$ and $Cd_2GeSe_4:Co^{2+}$ films were prepared on indium-tin-oxide(ITO)-coated glass substrates by using thermal evaporation. The crystallization was achieved by annealing the as-deposited films in flowing nitrogen. X-ray diffraction spectra showed that the $Cd_2GeSe_4$ and the $Cd_2GeSe_4:Co^{2+}$ films were preferentially grown along the (113) orientation. The crystal structure was rhomohedral(hexagonal) with lattice constants of $a=7.405\;{\AA}$ and $c=36.240\;{\AA}$ for $Cd_2GeSe_4$ and $a=7.43\;{\AA}$ and $c=36.81\;{\AA}$ for $Cd_2GeSe_4:Co^{2+}$ films. From the scanning electron microscope images, the $Cd_2GeSe_4$ and $Cd_2GeSe_4:Co^{2+}$ films were plated, and the grain size increased with increasing annealing temperature. The optical energy band gap, measured at room temperature, of the as-deposited $Cd_2GeSe_4$ films was 1.70 eV and increased to about 1.74 eV and of the as-deposited $Cd_2GeSe_4:Co^{2+}$ films was 1.79 eV and decreased to about 1.74 eV upon annealing in flowing nitrogen at temperatures from $200^{\circ}C$ to $500^{\circ}C$. The dynamical behavior of the charge carriers in the $Cd_2GeSe_4$ and $Cd_2GeSe_4:Co^{2+}$ films were investigated by using the photoinduced discharge characteristics technique.

Variation of Cadmium and Zinc Content in Rice and Soil of the Mangyeong River Area (만경강 유역의 토양과 수도체중 Cd 및 Zn 함량의 변화)

  • To investigate differences in Cd and Zn contents of paddy soils and rice plants polluted by the municipal and industrial waste water in the Mangyeong River Area, soil and plant samples were collected at several distances from the main inlet and at different depths of the soil. Soil samples were extracted with $4M-HNO_3$ and plant samples were digested with a mixture of $HNO_3$and $HClO_4$for analyzing heavy metals by atomic absorption spectrophotometry. The contents of Cd and Zn in soils ranged from 0.38 to 1.17 and from 33.8 to 464.6mg kg^{-1}, respectively. The average Cd level in 1990 was less than that in 1982, but the Zn level in 1990 was higher than that in 1982 in general. No variation in Cd contents was observed in soils at the different distances from the source of waste water, but Zn contents in soils were lower with the increasing distances from the source of waste water. A significant correlation was observed among Cd content, OM, available silicate, CEC and $Ca^{++}$. Similar results existed among Zn content of 1982, OM and $Ca^{++}$. The Cd content in subsurface soils of 1992 was significantly correlated with Zn, Cu, and Pb in soils, and the Zn content in soils was significantly correlated with the Cu and Pb in soils, regardless of years. The Cd content in leaf blades of rice was more than seven times higher than that in brown rice. The Zn content in rice was higher than that in leaf blades and in panicle axis. The Cd content in panicle axis and the Zn content in all parts of rice were correlated with Zn, Cu and Pb contents in soils. The Cd and Zn contents in brown rice ranged from 0.10 to 0.90mg $kg^{-1}$ and from 4.2 to 95.9mg $kg^{-1}$ in the Mangyeong River Area, respectively.

Concurrent Innate Immunity Activation and Anti-inflammation effects of Dialyzed Coffee Extract in RAW 264.7 Cells, Murine Macrophage Lineage (RAW 264.7 세포에서 투석시킨 커피 추출액의 선천면역활성화와 항염증의 동시발생)

  • Coffee (Coffea spp.) is one of the most important agricultural commodities, being widely consumed in the world. Various beneficial health effects of coffee have been extensively investigated, but data on habitual coffee consumption and its bio-physiological effect have not been clearly explained as well as it is not proved the cause and effect between drinking coffee and its bio-physiological reactions. We made the dialyzed coffee extract (DCE), which is absorbable through gastrointestinal tract, in order to elucidate the cellular effect of whole small coffee molecules. RAW 264.7 cells, a murine macrophage lineage, were directly treated with DCE, i.e., DCE-2.5 (equivalent to 2.5 cups of coffee a day), DCE-5, and DCE-10, for 12 hours, and their protein extracts were examined by immunoprecipitation high performance liquid chromatography (IP-HPLC). RAW 264.7 cells differently expressed the inflammation-related proteins depending on the doses of DCE. RAW 264.7 cells treated with DCE showed marked increase of cathepsin C, cathepsin G, CD20, CD28, CD31, CD68, indicating the activation of innate immunity. Particularly, the macrophage biomarkers, cathepsin G, cathepsin C, CD31, and CD68 were markedly increased after DCE-5 and DCE-10 treatments, and the lymphocyte biomarkers, CD20 and CD28 were consistently increased and became marked after DCE-10 treatment. On the other hand, RAW 264.7 cells treated with DCE showed consistent increase of IL-10, an anti-inflammatory factor, but gradual decreases of different pro-inflammatory proteins including $TNF{\alpha}$, COX-2, lysozyme, MMP-2, and MMP-3. In particular, the cellular signaling of inflammation was gradually mitigated by the reduction of $TNF{\alpha}$, COX-2, IL-12, and M-CSF, and also the matrix inflammatory reaction was reduced by marked deceases of MMP-2, MMP-3, and lysozyme. These anti-inflammatory expressions were consistently found until DCE-10 treatment. Therefore, it is presumed that DCE may have dynamic effects of innate immunity activation and pro-inflammation suppression on RAW264.7 cells simultaneously. These effects were consistently found in the highest dose of coffee, DCE-10 (equivalent to 10 cups of coffee a day in man), that might imply the small coffee molecules were accumulated in RAW 264.7 cells after DCE-10 treatment and produce synergistic cytokine effects for innate immunity activation and anti-inflammatory reaction concurrently.

Comparison of Heavy Metal Contents in Women's Hair Collected from Residental and Factory Areas in Pusan (부산시 주거지역과 공업지역 여성모발에서 검출된 중금속 농도에 대한 비교 연구)

  • The concentrations in women's hair of essential and toxic elements such as Zn, Ni, Fe, Mn, Cu, Cr, Pb and Cd in residential and factory areas in Pusan were determined, and all the elements were detected from all the samples. It was apparent that employees working in factory areas in Pusan were exposed directly to the essential and toxic heavy matals. This is based on the fact that average concentrations of Ni, Fe, Mn, Cu, Cr, Pb, and Cd in hair samples from women aged 10 to 29 in factory area, who were considered to be main members among women employees, were much higher than those of residential areas. That is, the highest concentrations found in the hair samples taken from women 10 to 29 years old living in factory areas were 110 times higher in Cr (in the women in their 20's), 92 times higher in Pb (in the women in their teens) and 23 times higher in Cd (in the women in their 20's) than those living in residential areas. Furthermore, it was suggested that the heavy metals may be accumulated gradually in human bodies as humans get older, based on the fact that the concentrations of Cr, Pb, and Cd in women living in residential areas were higher in older age groups, although the average contents of these elements in residents from residential areas were similar with those of several foreign countries.

Characterization of Two Novel mAbs Recognizing Different Epitopes on CD43

  • JL1, a specific epitope on CD43, is a potential biomarker for the diagnosis of acute leukemia. Although qualitative assays for detecting leukemia-specific CD43 exist, there is a need to develop quantitative assays for the same. Here, we developed two novel monoclonal antibodies (mAbs), 2C8 and 8E10, recognizing different epitopes on CD43. These clones are capable of pairing with YG5, another mAb against JL1 epitope, because they were selectively obtained using sandwich ELISA. Antigens recognized by 2C8 and 8E10 were confirmed as CD43 by western blotting using the CD43-hFC recombinant protein. When expression on various leukemic cell lines was investigated, 2C8 and 8E10 displayed a disparity in the distribution of the epitope. Enzyme assays revealed that these mAbs recognized a sialic acid-dependent epitope on CD43. Using normal thymus and lymph node paraffin-embedded tissues, we confirmed a difference in the epitopes recognized by the two mAbs that was predicted based on the maturity of the cells in the tissue. In summary, we developed and characterized two mAbs, 2C8 and 8E10, which can be used with YG5 in a sandwich ELISA for detecting leukemia-specific CD43.

Bactericidal Effects of Food-borne Bacteria using Chlorine Dioxide and Electrolyzed Water (이산화염소수와 전해수를 이용한 식중독균의 살균효과)

  • The present study investigated the bactericidal effects of chlorine dioxide (CD) and electrolyzed water (EW) on pathogenic bacteria, such as Bacillus cereus, Staphylococcus aureus, Salmonella Typhimurium, and Escherichia coli O157:H7, by treatment them with CD and EW, respectively, for 0, 2, 4, 6, 8, and 10 min. Additionally, the sensitivities of Gram-positive (B. cereus and S. aureus) and Gram-negative (S. Typhimurium and E. coli O157:H7) to CD and EW were compared, respectively. In CD, the D-values for B. cereus, S. aureus, S. Typhimurium, and E. coli O157:H7 were 1.85±0.64, 2.06±0.85, 2.26±0.89, and 2.59±0.40 min, respectively. In EW, the D-values for B. cereus, S. aureus, S. Typhimurium, and E. coli O157:H7 were 2.13±0.32, 1.64±0.64, 1.71±0.32, and 1.86±0.36 min, respectively. All strains decreased consistently for 10 min in both CD and EW. However, the D-values of each bacterial species did not differ significantly between CD and EW (P>0.05). When comparing the bactericidal effect of CD and EW, no difference in D-value was observed, even though the pH and available chlorine concentration of CD were significantly lower than those of EW. These data could be used for the application of CD and EW in the food industry, considering characteristics such as the selection of optimal disinfectants, determination of optimal concentrations, and sensitivity to disinfection targets.

Cholesterol Removal from Whole Egg by Crosslinked β-Cyclodextrin

  • This study was carried out to optimize cholesterol removal in whole egg using crosslinked ${\beta}$-cyclodextrin (${\beta}$-CD) and to recycle the ${\beta}$-CD. Various factors for optimizing conditions were concentration of the ${\beta}$-CD, mixing temperature, mixing time, mixing speed and centrifugal speed. In the result of this study, the optimum conditions of cholesterol removal were 25% crosslinked ${\beta}$-CD, $40^{\circ}C$ mixing temperature, 30 min mixing time, 1,200 rpm mixing speed and $2,810{\times}g$ centrifugal speed. The recycling was repeated five times. The cholesterol removal was 92.76% when treated with the optimum conditions. After determining the optimum conditions, the recyclable yields of the crosslinked ${\beta}$-CD ranged from 86.66% to 87.60% in the recycling and the percentage of cholesterol removal was over 80% until third recycling. However, the cholesterol removal efficiency was decreased when the number of repeated recycling was increased. Based on the result of this study, it was concluded that the crosslinked ${\beta}$-CD was efficient for cholesterol removal in whole egg, and recycling is possible for only limited repeating times due to the interaction of the ${\beta}$-CD and egg protein.

Removal of Cd(II) and Pb(II) Ions in water by the Ulva pertusa and Sargassum horneri (Ulva pertusa 및 Sargassum horneri를 이용한 수중 Cd(II) 및 Pb(II) 이온의 제거)

  • Heavy metal ions in water were removed using algal biomass as adsorbents. Absorbents were dried for 3 days, ground them by 40~60 mesh and then were swelled in a buffer solution for 1hr. After being packed in the column, commercially available standard solution of Cd(II) and Pb(II) ions were diluted to get the suitable concentration and then it was eluted with the rate of 1mι/min. Heavy metals on the adsorbents were recovered with nitric acid. More amounts of Cd(II) or Pb(II) ions in green algae, Ulva pertusa, than in brown algae, Sargassum horneri, were adsorbed. Pb(II) ion was adsorbed more than Cd(II) ion in both algae. The pH effect of adsorbed amounts of Cd(II), Pb(II) ions on the biomass was shown the following order ; pH 10.5 > 8.5 > 7.0 > 5.5 > 3.5. Recovery ratio of metal ions front algae is shown higher in acidic or neutral conditions than it in alkalic ones. Pb(II) ion is recovered relatively more than Cd(II) ion in our system.

Bamboo Culm Extract Downregulated Activation of NKT- and B- cells and Production of IL-6 in Pristane-Induced Lupus Mice

  • Lupus is characterized by immunoregulatory abnormalities between T- and B-cells leading to autoantibody production and multiorgan injuries. We investigated whether bamboo culm extract (BC) ameliorates aberrant activation of T cells and B cells and attenuate production of IL-6 in pristane-induced lupus mice. Lupus was induced by i.p. a single injection of 0.5 ml of pristane in female BALB/c mice, which, later about 4 months, were used as a lupus model. The pristane-induced lupus mice and healthy mice were injected i.p. with BC 5 ${\mu}l$/kg or PBS once a day for 3 weeks. These results demonstrated that BC significantly decreased levels of serum and BAL IL-6 and production of IL-6 by macrophages with/without LPS, and downregulated expression of NKT cell and CD86+ CD45R/B220+, but not CD80+CD45R/B220+ and CD69+CD4+ in the splenocytes in pristaneinduced lupus mice. Moreover, BC greatly increased Con A-stimulated production of IL-6, IL-10 and IFN-${\gamma}$ by splenocytes obtained from pristane-induced lupus mice. Therefore, our findings suggest that BC may ameliorate lupus pathogenesis in pristane-induced lupus mice via downregulation of aberrant activation of NKT cells and B cells and inhibition of production of IL-6.

Determination of Optimal Toxic Concentration and Accumulation of Cadmium in Broiler Chicks

  • Cadmium is considered one of the most toxic, non biodegradable heavy metal for the human and animals. The purpose of the present study was to investigate the changes in biochemical parameters of blood and accumulation of cadmium in various tissue caused by various levels of dietary cadmium chloride ($CdCl_2$) in broiler chicks. $CdCl_2$ was administered through drinking water to broiler chicks. In spectral analysis, $CdCl_2$ treatment caused a significant increase in Glutamate pyruvate transaminase (GPT), creatinine and uric acid levels in all treated groups. Intriguingly, the GPT, creatinine, and uric acid levels were significantly higher at 75 mg/kg as compared to the groups treated with high doses (100, 125 and 150 mg/kg) of $CdCl_2$. Atomic Absorption Spectrophotometer (AAS) was used for the determination of Cd accumulation in kidney, liver and Breast muscles. AAS analysis revealed that Cd accumulation is increased in breast muscles as compared to liver and kidney at higher doses of Cd than 75 mg/kg.
