Effect of the Oryungsan on the injured liver in CCl4-treated rats (오령산(五?散)이 CCl4 투여(投與)로 인한 흰쥐의 손상간(損傷肝)에 미치는 영향(影響))

  • This study was investigated to define effect of pretreatment of Oryungsan on hepatotoxicity induced by $CCl_4$ in rat. The experimental anmals were divided into normal group, control group, sample group, and sample group were divided sample1, sample2, sample3. Control group were injected diluted carbon tetrachloride and olive oil, $1m{\ell}/kg$ to intraperitoneal after 8th day. Sample group were injected Oryungsan water extracts, $0.33m{\ell}/kg/day$ to intraperitoneal of sample group1, $0.66m{\ell}/kg$ day to sample group2, $1.32m{\ell}/kg/day$ to sample group 3 each other for 7day, and injected the dosage of $CCl_4$ was $1.0m{\ell}/kg$ diluted 1 : 1 with olive oil after 8th day. The following results were made by observation GOT, GPT, Albumin, Globulin, Bile acid, hepatic tissue. 1. The serum GOT and GPT level of sample group were decreased. Sample group 1 were decreased significantly. 2. The serum A/G ratio of sample group was increased as compared with normaI group. 3. The serum bile acid of sample group was decreased significantly. 4. The destruction of hepatic tissue were repaired as compared with control group. These results demonstrated that oryungsan can be attributed to recovery from hepatotoxicity.

Hepatoprotective Activities of Daidzin, Daidzein, Genistein and Puerarin in Primary Cultured Rat Hepatocytes (흰쥐의 일차배양 간세포에서 Daidzin, Daidzein, Genistein 및 Puerarin의 간 보호 활성 평가)

  • The aim of this study was to investigate the protective activities of daidzin, daidzein, genistein or puerarin, active isoflavonoids of Puerariae Radix, on the hepatocyte injury induced by carbon tetrachloride (CCl$_4$, 10 mM), tert-butyl hydroperoxide (TBH, 0.5 mM) and D-galactosamine (GalN, 30 mM). Primary cultures of rat hepatocytes (18 hr cultured) were treated with CCl$_4$, TBH or GalN and various concentrations (0.1, 1, 10 and 100 ${\mu}$M) of daidzin, daidzein, genistein or puerarin. CCl$_4$ significantly increased the levels of lactate dehydrogenase (LDH), alanine aminotransferase (ALT) and aspartate aminotransferase (AST). The increase in LDH level was attenuated by daidzein, genistein and puerarin. Puerarin also inhibited the increase in AST level induced by CCl$_4$. The increases in LDH and ALT levels induced by TBH were significantly attenuated by daidzin and genistein treatments. GalN markedly increased the levels of LDH, ALT and AST These increases were significantly attenuated by daidzein. Daidzin also inhibited the increases in LDH and AST levels induced by GalN. The increases in LDH and ALT levels were attenuated by genistein and puerarin, respectively. These results suggest that daidzin and daidzein possess hepatoprotective activities.

Vitamin D Improves Intestinal Barrier Function in Cirrhosis Rats by Upregulating Heme Oxygenase-1 Expression

  • Intestinal barrier dysfunction always accompanies cirrhosis in patients with advanced liver disease and is an important contributor facilitating bacterial translocation (BT), which has been involved in the pathogenesis of cirrhosis and its complications. Several studies have demonstrated the protective effect of Vitamin D on intestinal barrier function. However, severe cholestasis leads to vitamin D depletion. This study was designed to test whether vitamin D therapy improves intestinal dysfunction in cirrhosis. Rats were subcutaneously injected with 50% sterile $CCl_4$ (a mixture of pure $CCl_4$ and olive oil, 0.3 mL/100 g) twice a week for 6 weeks. Next, $1,25(OH)_2D_3$ ($0.5{\mu}g/100g$) and the vehicle were administered simultaneously with $CCl_4$ to compare the extent of intestinal histologic damage, tight junction protein expression, intestinal barrier function, BT, intestinal proliferation, apoptosis, and enterocyte turnover. Intestinal heme oxygenase-1 (HO-1) expression and oxidative stress were also assessed. We found that vitamin D could maintain intestinal epithelial proliferation and turnover, inhibit intestinal epithelial apoptosis, alleviate structural damage, and prevent BT and intestinal barrier dysfunction. These were achieved partly through restoration of HO-1 and inhibition of oxidative stress. Taken together, our results suggest that vitamin D ameliorated intestinal epithelial turnover and improved the integrity and function of intestinal barrier in $CCl_4$-induced liver cirrhotic rats. HO-1 signaling activation was involved in these above beneficial effects.

Effect of Aging on the Xylene Metabolism in $CCl_4$-Induced Liver Damaged Rats

  • To investigate an effect of aging on the xylene metabolism in liver damaged animals, a study was conducted. 50% carbon tetrachloride ($CCl_4$) in olive oil (0.1 ml/100 g body weight) was intraperitoneally given to 5-week and 12-week rats 12 times every other day and then one dose of 50% xylene in olive oil (0.25 ml/100 g body weight) was intraperitoneally given to the rats, and after 24 hr, the animals were sacrificed. On the basis of the functional findings in rat liver, ie, serum levels of alanine aminotransferase activity, liver protein and malonedialdehyde contents, 5-week rats showed less liver damage than 12-week rats. The increasing rate of urinary methylhippuric acid concentration to the control was significantly higher in 5-week rats than 12-week rats in case of xylene treatment after induction of liver damage. On the other hand, liver damaged 5-week rats showed significant rise of hepatic cytochrome P45O content compared with the liver damaged 12-week rats by the xylene treatment. And increasing rate of hepatic alcohol or aldehyde dehydrogenase activities to each liver damaged animals was higher tendency in 5-week rats than 12-week rats by the xylene treatment. In conclusion, 5-week rat showed greater metabolic rate of xylene than 10-week rats in case of liver injury because 5-week rats led to a slight liver damaged compared with 12-week rats.

Protective Effects of the Phenolic Compounds from the Leaves of Hedera rhombea on Hepatic Injury (송악의 페놀성 물질의 간 보호효과)

  • Hedera rhombea (HR) has been used for treatments of hemorrage, chronic catarrh, jaundice, lithisis and convulsion. This study was done to isolate active compounds that have protective effect on liver damage. BuOH and EtOAc fractions of HR recovered serum glutamic pyruvic transaminase (GPT), glutamic oxaloacetic transaminase (GOT) and ${\gamma}-glutamyltranspeptidase$ (${\gamma}-GTP$) activities in CCl4 treated rats. We isolated 7 phenolic compounds from BuOH and EtOAc fractions, which were identified as 3-caffeoyl quinic acid, 3,4-di-O-caffeoyl quinic acid, 3,5-di-O-caffeoyl quinic acid, 4,5-di-O-caffeoyl quinic acid, caffeic acid, methyl 3,4-di-O-caffeoyl quinic acid and methyl 3,5-di-O-caffeoyl quinic acid by chemical and spectral analysis. These compounds reduced significantly serum GOT and GPT elevated by CCl4 treatment in rats, and 3-caffeoyl quinic acid, 3,5-di-O-caffeoyl quinic acid and caffeic acid also showed mild inhibitory activity against human immunodeficiency virus.

Quercetin suppress CCL20 by reducing IκBα/STAT3 phosphorylation in TNF-α/IL-17A induced HaCaT cells (TNF-α/IL-17A 유도된 HaCaT 세포주에서 Quercetin의 IκBα/STAT3 인산화 조절에 의한 CCL20 발현 억제)

  • Quercetin is a polyphenol compound with excellent antioxidant and anti-inflammatory activity. However, little has been reported about the efficacy of quercetin to control psoriasis. Thus, we aimed to investigate the effect of quercetin to regulate psoriatic dermatitis with HaCaT cell lines activated by TNF-α and IL-17A, which are in vitro psoriasis skin models. When quercetin was treated with TNF-α-activated HaCaT cell line, inflammatory cytokine expressions such as IL-1α, IL-1β and IL-6 were reduced by 49.1±7.14, 42.8±8.16, and 34.5±2.52%, respectively. In addition, mRNA expression levels of IL-8 and CCL20 the chemokines that attract immune cells such as Th17 cells and dendritic cells to the inflammatory reaction site, were also reduced by 38.4±5.83 and 52.9±4.59% compared to the TNF-α treatment group. The expression of proteins KRT6A and KRT16, which was nonspecifically increased in psoriatic skin was also significantly suppressed. Moreover, phosphorylation of IκBα and STAT3 proteins activated by TNF-α was also significantly inhibited. After stimulating the HaCaT with IL-17A, known as another psoriasis-inducing cytokine, it was observed that IκBα mRNA expression decreased by 55.8±5.28%, and STAT3 phosphorylation was downregulated by 36.3±6.81%. Finally, after co-activation by TNF-α/IL-17A, quercetin inhibited all of IL-1α, IL-1β, IL-6, TNF-α and CCL20 gene expression. The above results strongly suggest that quercetin is a material that has not only anti-oxidant and anti-inflammatory activities, but also has an activity in improving psoriasis.

Functional Analysis of Fibroblastic Reticular Cells Derived from Mouse Lymph Node via Bidirectional Crosstalk with T Cells (T세포와 양방향 작용을 통한 마우스 림프절로부터 분리된 fibroblastic reticular cell의 기능적 분석)

  • Fibroblastic reticular cells (FRCs) form the structural backbone of the T zone provide a guidance path for immigrating T cells in the lymph node (LN). FRCs may contribute directly to developing T-cell biology in the LN and allow analyses of fundamental aspects of FRC biology related to T cells. FRCs inhibited T-cell apoptosis, and FRC culture supernatants strongly induced the expression of Bcl-xL in T cells against doxorubicin. Coculture of FRC and T cells resulted in rearrangements of the actin cytoskeleton, as well as global changes in the morphology of the FRCs. In addition, when cocultured, the T cells adhered to the FRC monolayer, and the membrane intercellular adhesion molecule (ICAM)-1 was slightly increased by day-dependent manner. In contrast, the expression of soluble ICAM-1 was dramatically increased in a day-dependent manner. Several chemokines, such as CCL5, CXCL1, CXCL5, CXCL16, CCL8, CXCL13, and ICAM-1, and MMPs were expressed in FRCs sensed by tumor necrosis factor (TNF) families. Nuclear factor kappa B ($NF{\kappa}B$)-RelA of the $NF{\kappa}B$ canonical pathway was translocated into FRC nuclear by $TNF{\alpha}$. In contrast, p52 proteolyzed from p100, a counterpart of RelB of the noncanonical $NF{\kappa}B$ pathway, accumulated in the peripheral FRC nucleus by agonistic anti-$LT{\beta}R$ antibody. In summary, we propose a model in which FRCs engage in bidirectional crosstalk to increase the efficiency of T-cell biology. This cooperative feedback loop may help to maintain tissue integrity and function during immune responses.

Changes of Serum Gamma-Glutamyl Transpeptidase Values in Experimentally Induced Toxic Hepatitis, Obstructive Jaundice and Pancreatitis in Dogs (실험적으로 일으킨 개의 중독성(中毒性) 간염(肝炎), 폐쇄성(閉鎖性) 황달(黃疸) 및 췌장염(膵臟炎)에 있어서 혈청(血淸) Gamma-Glutamyl Transpeptidase치(値)의 변화)

  • In order to assess the diagnostic aid of serum gammaglutamyl transpeptidase values in hepatitis, obstructive jaundice and pancreatitis, four groups of 14 health dogs were subjected to the gastric intubatin of $CCl_4$(1.5ml/kg of body weight), the ligation of common bile duct, the ligation of pancreatic ducts and the injection of chloroform(0.2ml/kg of body weight) in the parenchyma of the pancreas. Some serum enzymes serum glutamic pyruvic transaminase(SGPT), serum glutamic oxalacetic transaminase(SGOT), total bilirubin, amylase and lipase known to be indicative of hepatic and pancreatic diseases were monitored. In comparision of these enzymes, gamma-glutamyl transpeptidase(GGTP) valuers were determined in these dogs before and after the experimental procedures. The results were summarized as follows: 1. In $CCl_4$ intoxication gorup, there were no significant changes in serum GGTP activities(mean: 6.0~14.6 IU/L). 2. In bile duct ligation group, serum GGTP activities shelved marked increases, beginning at postsurgical day 1 and rose the highest mean value(342.7 IU/L) on day 12. Then the activities never approached to the base-line values. 3. After the ligation of pancreatic ducts and the injection of chloroform in the pancreas, serum GGTP activities did not rise throughout the experiment. 4. SGPT:GGTP ratio did not increase in bile duct ligation group, but increase markedly in $ccl_4$ intoxication group. 5. The results indicated that serum GGTP values or SGPT:GGTP ratio could provide valuable indicators for differential diagnosis between hepatobiliary obstruction and hepatocellular disease.

Inhibitory activity of Terminalia chebula extract against TNF-α/IFN-γ-induced chemokine increase on human keratinocyte, HaCaT cells (TNF-α/IFN-γ 유도된 각질형성세포 염증에서 가자 추출물의 케모카인 저해 효과)

  • Objectives : Terminalia chebula (TC) has been used as a traditional remedy to treat gastrointestinal infectious and inflammatory diseases. However, its protective effects and mechanisms against skin inflammation have not been well-elucidated. Thus, the aim of this study is to evaluate the protective effects of the TC water extract and also to suggest a putative mechanism of TC against skin injury on human keratinocytes, HaCaT cells. Methods : HaCaT cells were pre-treated with TC for 1 h and then stimulated with tumor necrosis factor-alpha (TNF-α) and interferon-gamma (IFN-γ) (10 ng/mL each) to induce skin inflammation and injury. After 24 h, the cells were harvested to evaluate the expression of Th2 chemokines, such as C-C motif chemokine ligand 5 (CCL5, also known as RANTES), C-C chemokine ligand 17 (CCL17, also known as TARC) and C-C chemokine ligand 22 (CCL22, also known as MDC). To investigate the regulatory mechanisms of TC, we also assessed the phosphorylation of signal transducer and activator of transcription 1 (STAT1) signaling pathways in HaCaT cells. Results : Treatment of TC decreased the mRNA levels of RANTES, TARC and MDC with a concentration dependent manner against co-stimulation of TNF-α and IFN-γ. In addition, TC significantly reduced TNF-α and IFN-γ induced phosphorylation of STAT1. Conclusions : In summary, we propose that TC may be a promising candidate for anti-inflammatory skin protector through the inhibition of chemokines via STAT1 deactivation.

Gene Expression Profiles in Genetically Different Mice Infected with $Toxoplasma$ $gondii$: ALDH1A2, BEX2, EGR2, CCL3 and PLAU

  • $Toxoplasma$ $gondii$ can modulate host cell gene expression; however, determining gene expression levels in intermediate hosts after $T.$ $gondii$ infection is not known much. We selected 5 genes ($ALDH1A2$, $BEX2$, $CCL3$, $EGR2$ and $PLAU$) and compared the mRNA expression levels in the spleen, liver, lung and small intestine of genetically different mice infected with $T.$ $gondii$. ALDH1A2 mRNA expressions of both mouse strains were markedly increased at day 1-4 postinfection (PI) and then decreased, and its expressions in the spleen and lung were significantly higher in C57BL/6 mice than those of BALB/c mice. BEX2 and CCR3 mRNA expressions of both mouse strains were significantly increased from day 7 PI and peaked at day 15-30 PI ($P$<0.05), especially high in the spleen liver or small intestine of C57BL/6 mice. EGR2 and PLAU mRNA expressions of both mouse strains were significantly increased after infection, especially high in the spleen and liver. However, their expression patterns were varied depending on the tissue and mouse strain. Taken together, $T.$ $gondii$-susceptible C57BL/6 mice expressed higher levels of these 5 genes than did $T.$ $gondii$-resistant BALB/c mice, particularly in the spleen and liver. And ALDH1A2 and PLAU expressions were increased acutely, whereas BEX2, CCL3 and EGR2 expressions were increased lately. Thus, these demonstrate that host genetic factors exert a strong impact on the expression of these 5 genes and their expression patterns were varied depending on the gene or tissue.
