Mixture of dimethylaminobenzaldehyde and cyanoacrylate to develop fingerprints with fluorescence: a preliminary test

  • This study suggests a new one-step fluorescent cyanoacylate-fuming method for developing fingerprints by using a CAB mixed with dimethylaminobenzalde (DMAB) and cyanoacylate (CA) in a specific ratio. CAB is prepared by mixing 2.5 % (w/w) DMAB with CA and fumigated at 180 ℃. Under these conditions, developing fingerprints showed the best results. The fuming method using CAB develops latent fingerprints into fluorescence and has a higher sensitivity than CA, and it showed comparable or better contrast to existing fluorescence enhancement methods. It was also applicable on a variety of non-porous surfaces that can be encountered at ordinary times. This method is more useful than conventional fluorescent dyeing methods in that it minimizes damage to fingerprints or samples, makes it easy to manufacture, saves time, and can use existing current equipment as it is.

Characterization of Bacillus thuringiensis subsp. tohokuensis CAB167 Isolate against Mosquito Larva (모기유충에 활성 있는 Bacillus thuringiensis subsp. tohokuensis CAB167 균주의 특성)

  • Eight Bacillus thuringiensis strains activated against mosquito larva were compared their characterization. Spherical-shaped parasporal inclusion of B. thuringiensis subsp. tohokuensis CAB167 was observed by phase-contrast microscopy and scanning electron microscopy. $LC_{50}$ values of B. thuringiensis subsp. tohokuensis CAB167 against Culex pipiens molestus, Culex pipiens pallens, and Aedes aegyti were 173, 190 and 580 ng/ml, respectively. B. thuringiensis subsp. tohokuensis CAB167 had a parasporal inclusion containing 4 major protein components, for example, 135, 80, 49 and 28-kDa by SDS-PAGE. Otherwise, after trypsin digestion of parasporal inclusion, SDS-PAGE was showed new protease-resistant peptides at 72 and 63-kDa. Activated toxins of isolated CAB167 were different from other reference strains on a serological by immuno-diffusion test.

Biological Control of White Rot in Garlic Using Burkholderia pyrrocinia CAB08106-4 (Burkholderia pyrrocinia CAB08106-4 균주를 이용한 마늘 흑색썩음균핵병의 생물학적 방제)

  • White rot caused by Sclerotium cepivorum was reported to be severe soil-born disease on garlic. Disease progress of white rot of garlic (Allium sativum L.) was investigated during the growing season of 2009 to 2011 at Taean and Seosan areas. The white rot disease on bulb began to occur from late April and peaked in late May. The antifungal bacteria, Burkholderia pyrrocinia CAB08106-4 was tested in field bioassay for suppression of white rot disease. As a result of the nucleotide sequence of the gene 16S rRNA, CAB008106-4 strain used in this study has been identified as B. pyrrocinia. B. pyrrocinia CAB080106-4 isolate suppressed the white rot with 69.6% control efficacy in field test. These results suggested that B. pyrrocinia CAB08106-4 isolate could be an effective biological control agent against white rot of garlic.

  • Display Unit(DU) of TCMS is very useful device display many data for operation and maintenance graphically. At This Time, rolling stock is reducing the wiring and size of desk for cutting down on their cost and making light weight. Dual Du(Display Unit) of TCMS could make the desk of cab simply by integrating cab's switches and gauges. Dual DU is not only can reduce the wiring and simplify desk of cab but also can making system of TCMS reliable by including backup function each other. For this reason. In this paper, advantages, application and example analysis of Dual DU of TCMS for rolling stock are described through the ongoing projects.

  • Samsung Ltd. has developed a new-type cab mount for specific use on construct ion machinery subjected to strong vibration and multi-directional impact force. These all make it possible to achieve an excellent damping effect over a wide frequency range against large amplitude vibration as well as excellent insulation against small-amplitude vibration. This new mount make lower vibration and noise levels while increasing riding comfort at the same time. Characteristics of Cab mount were optimized through computer simulation, advanced bench testing, ODS testing, and a real equipment offroad testing.

Mechanisms of thermally induced deflection of a long-span cable-stayed bridge

  • Variation of temperature is a primary environmental factor that affects the behavior of structures. Therefore, understanding the mechanisms of normal temperature-induced variations of structural behavior would help in distinguishing them from anomalies. In this study, we used the structural health monitoring data of the Shanghai Yangtze River Bridge, a steel girder cable-stayed bridge, to investigate the mechanisms of thermally induced vertical deflection ($D_T$) at mid-span of such bridges. The $D_T$ results from a multisource combination of thermal expansion effects of the cable temperature ($T_{Cab}$), girder temperature ($T_{Gir}$), girder differential temperature ($T_{Dif}$), and tower temperature ($T_{Tow}$). It could be approximated by multiple linear superpositions under operational conditions. The sensitivities of $D_T$ of the Shanghai Yangtze River Bridge to the above temperatures were in the following order: $T_{Cab}$ > $T_{Gir}$ > $T_{Tow}$ > $T_{Dif}$. However, the direction of the effect of $T_{Cab}$ was observed to be opposite to that of the other three temperatures, and the magnitudes of the effects of $T_{Cab}$ and $T_{Gir}$ were found to be almost one order greater than those of $T_{Dif}$ and $T_{Tow}$. The mechanisms of the thermally induced vertical deflection variation at mid-span of a cable-stayed bridge as well as the analytical methodology adopted in this study could be applicable for other long-span cable-stayed bridges.

Investigation of Regulatory Mechanism of Flux of Acetyl-CoA in Alcaligenes eutrophus Using PHB-negative Mutant and Transformants Harboring Cloned phbCAB Genes

  • The regulatory mechanism of the flux of acetyl-CoA in Alcaligenes eutrophus in unbalanced growth conditions was investigated using a PHB-negative mutant and transformants reintroduced PHB-biosynthesis enzymes through the transformation of cloned phbCAB genes. The PHB-negative mutant was defected absolutly in PHB synthase but partially in ${\beta}$-ketothiolase and acetoacetyl-CoA reductase, and excreted substantial amount of pyruvate to culture broth at late growth phase. The excretion was due to the inhibitory effect of acetyl-CoA on the activity of pyruvate dehydrogenase. The cloned phbC and phbCAB genes were transformed to the PHB-negative mutant strain to reintroduce PHB biosythesis enzymes. Pyruvate excretion could be decreased substantially but not completely by transformation of PHB synthase alone, while pyruvate excretion was ceased by transformation of all three PHB biosynthesis enzymes. To identify the most critical PHB biosynthesis enzyme influencing on the flux of acetyl-CoA, the effect of the variation of PHB biosynthesis enzymes on pyruvate dehydrogenase was investigated. ${\beta}$-Ketothiolase influenced the activity of pyruvate dehydrogenase more sensitively than PHB synthase. ${\beta}$-Ketothiolase, the first step enzyme of PHB biosynthesis that condense acetyl-CoA to acetoacetyl-CoA, seems to be the major enzyme determining the flux of acetyl-CoA to PHB biosynthesis or TCA cycle, and the rate of PHB biosynthesis in A. eutrophus.

Light Regulated Plant Gene Expression (빛에 의한 식물 유전자의 발현)

  • Light regulates a variety of genes in higher plants. The expression of light-induced plant genes is regulated at the level of transcription via red- light photomorphogenic receptor, phytochrome, as well as unknown blue light photoreceptor(s). Ribulose-5-phosphate carboxylase/oxygenase (Rubisco) small subunit (SSB) and light harvesting chlorophyll a/b (Cab) protein are those of the best understood genes regulated by light. 5'-upstream flanking sequence (- -400) of Rubisco SSB and Cab genes sis known as a light responsive, enhance-like element. It responses to red and blue light in transgenic plant system as a tissue specific manner. Phytochrome gene is also regulated by light. In contrast to most of the light regulated plant genes, it is negatively controlled by red light. Search for the cis- and trans-acting factors responsible for the light signal is in progress to understant photomorphogenesis and development in higher plants.

Comparison of Vibration Characteristics of a Multi-leaf Spring and a Tapered Leaf Spring of a Heavy Truck (대형트럭 다판 스프링과 테이퍼 판스프링의 진동특성 비교)

  • This paper develops the flexible computational model of a heavy truck by interfacing the frame modeled as a flexible body to the heavy truck's computational model composed of rigid bodies. The frame is modeled by the finite element method. Three torsional modes and three bending modes of the frame are considered for the interface of the heavy truck's computational model. The actual vehicle test is conducted off road with a velocity of 20km/h. The vertical accelerations at the cab and front axle are measured in the test. For the verification of the developed computational model, the measured vertical acceleration profiles are compared with the simulation results of the heavy truck's flexible computational model. E grade irregular road profile of ISO is used as an excitation input in the simulation. The verified flexible computational model is used to compare the vibration characteristics of a front suspension system having a multi-leaf spring and that having a tapered leaf spring. The comparison results show that the front suspension having a tapered leaf spring has a higher vertical acceleration at the front axle but a lower vertical acceleration at the cab than the suspension system having a multi-leaf spring.

  • Seoul Metro has been continuously growing after its foundation on August 15, 1974 and is currently acting the leading role in the city transportation which is now used by 4,130 thousand people (as of early 2011) on a daily average. The number of the electric rail cars also increased significantly from 60 cars at the time of foundation to 1,954. And the types of the cars also started and operated with the rheostatic control cars in the 70s to the chopper control cars in the 80s and VVVF control cars in the 90s. But the control devices of the driver's cabs haven't been accustomed to the drivers because of it's diversity. Therefore, This study carried out the survey for the preferences of control devices from the incumbent drivers at Seoul Metro. And we analyzed it and suggest a method for standardization of it's locating and naming the major control devices in the driver's cab as the consideration of their preference for it's efficiency, safety, and convenience.
