Development of CAPS marker for identifying a Formosan lily (Lilium formosanum) (흰나리(Lilium formosanum Wallace) 식별을 위한 CAPS 마커의 개발)

  • This study was conducted to identify lily species native to Korea from formosan lily (Lilium formosanum) belonging to Longiflorum section. Due to flowering time, flower color and orientation, long shelf life and resistant to diseases, the native lily species can be valuable genetic resources for interspecific hybrids. One of the chloroplast genes, matK, was used to clone and sequence to explore any base changes. The matK was successfully amplified into 1,539 bp (94% of the gene) and phylogenetic tree demonstrated 6 clades for those 11 lily species used in this study. There were one or two base substitutions among 10 lilies native to Korea, while formosan lily native to Taiwan exhibited 6 base substitutions in matK gene, rendering it genetically distant. A restriction enzyme NruI recognized one of the six base changes, and digested the matK gene of 10 native lily species only, but not in formosan lily. The confirmed cleavage characteristic of the target region in matK gene was designed into a CAPS (cleaved amplified polymorphic sequences) marker which will be available to estimate compatibility of interspecific hybridization and to trace the pedigree when those native lilies are crossed with the formosan lily.

A Study of the Types of Winter Caps Found in the Literature of the Joseon Dynasty (문헌을 통매 본 조선시대 방한모 명칭에 관한 연구)

  • The study aimed to classify and analyze about the winter cap in the literature of the Joseon Period, as the period of using, synonyms, user, shape and materials. The following are the result of the study. 1. The winter caps can be classified into two kinds of group by times. The first ones have been used continuously during the whole Joseon Dynasty, for example, Nanmo(煖帽, 暖帽), Ieom(耳掩) and Pigyeon(披肩). These names are winter caps for the bureaucracy. The second ones have been used a part of the Joseon Period. Some of the names used in the first half of the Joseon Period, for example, Mogwan(毛冠), Chopigwan(貂皮冠), Chogwan(貂冠) and Seobokpigwan(鼠腹皮冠). The other names used in the latter half of the Joseon Period, for example, Noepo(腦包), Nanja(暖子), Jeomgyeon, Chageompi(遮?皮), Pimaho(皮馬虎), Wibo, Chipo, pungryeong(風領), Nani(煖耳), Hohang(護項), Hwihang(揮項), Manseonduri, Pungcha(風遮), Samsangeon(三山巾), Iwi(耳衣), Aekeom(額掩), Yangjeongeon(凉轉巾), Bolkki, Ayam and Guile. 2. Synonyms with connoting relationship in which one name perfectly connotes meaning of another name are Nanmo, Nani and Pigyen, Nanmo connotes Ieom, Chomo, Homo, Iwi and Hwihang. Nani connotes Hwihang, Hoieom and Pungcha. Pigyen connotes Hwihang and Ieom. In these cases, the range of the meaning of clothing terms can be confirmed since names with broad meaning connote names with narrow meaning. 3. Some names related with the other names, as the user, shape and materials, for example, Ieom, Hwihang and Pungcha.

Phylogenetic relationships of medicinal mushroom Sparassis crispa strains using the rDNA-ITS and CAPS analysis (rDNA-ITS 및 CAPS 분석에 의한 꽃송이버섯 (Sparassis crispa) 수집균주의 계통분류학적 특성구분)

  • This study was carried out to analyze the genetic relationships among 22 strains of Sparassis crispa, which were collected from various regions of worldwide. The cleaved amplified polymorphic sequence were obtained from the ribosomal DNA ITS regions of each strain. Based on the sequence analysis, the presence of five different groups were observed. Most strains shared the high nucleotide sequence similarity (about 90%) to each other, except only one strain, KACC50866. Nucleotide sequence similarity of KACC50866 was below 10% to other strains, indicating the genetic relatedness of strain KACC50866 was low compared to other strains. More works such as mitochondria genome analysis should help to determine the precise genetic diversity of S. crispa strains.

Laboratory Model Tests on the Load Transfer in Geosynthetic-Reinforced and Pile-Supported Embankment System (토목섬유보강 성토지지말뚝시스템에서의 하중전이 효과에 관한 모형실험)

  • A series of model tests were performed to investigate the load transfer by soil arching in geosynthetic-reinforced and pile-supported(GRPS) embankment systems. In the model tests, model piles with isolated cap were inserted in the model container and geosynthetics was laid on the pile caps below sand fills. The settlement of soft ground was simulated by rubber form. The loads acting on pile caps and the tensile strain of geosynthetics were monitored by data logging system. At the given interval ratio of pile caps, the efficiency in GRPS embankment systems increased with increasing the height of embankment fills, then gradually converged at constant value. Also, at the given height of embankment fills, the efficiency decreased with increasing the pile spacing. The embankment loads transferred on pile cap by soil arching increased when the geosynthetics installed with piles. This illustrated that reinforcing with the geosynthetics have a good effect to restraint the movement of surrounding soft grounds. The load transfer in GRPS embankment systems was affected by the interval ratio, height of fills, properties of grounds and tensile stiffness and so on.

Considerations on the Safety of Electric Caps Based on Measured Electrical Resistivity of Rock Samples (암석의 전기비저항 측정을 통한 전기뇌관의 사용 안전성 검토)

  • Much care should be taken when electric caps are used in blast site than when non-electric initiation systems are used. This is because electric caps can cause premature firing or misfires if stray currents of high magnitude flow into the blasting circuit. If the rock has higher electrical conductivity or lower electrical resistivity, such risks will be increased because the rock will provide more passages for the stray currents to flow into the blasting circuit. In this study, several rock samples obtained at a blast site were tested for electrical resistivity to decide whether electric caps could be used or not in the site. The measured electrical resistivity was $39{\sim}47{\Omega}{\cdot}m$ for the rock samples that had a higher content of metal sulfides. Contrary, the resistivity was $15000{\sim}21000{\Omega}{\cdot}m$ for ordinary rocks. Especially, in the case of the rock of electric resistivity of $39{\Omega}{\cdot}m$, only 2-V electric potential enables a stray current to flow through the rock of 1-m length, which can cause the premature firing of a detonator whose initiation current is 0.4 A. This result shows that electric initiation system should not be used in the site because rocks containing much amount of metal sulfides are widely distributed there.

Effect of Terazosin on Posttraumatic Nightmares : A Pilot Study (외상 후 스트레스장애 환자의 악몽치료에서 Terazosin의 효과 : 예비 연구)

  • Objective : Chronic posttraumatic stress disorder (PTSD) is defined by the coexistence of symptoms (reexperiencing, avoidance and hyperarousal), persisting for more than months or years that causes significant impairment in social occupational functioning. This study was conducted to evaluate the effects of terazosin on posttraumatic nightmares which is one of chronic PTSD symptoms. Methods : Twenty patients with chronic PTSD and severe trauma-related nightmares were treated with terazosin (3-7 mg/day) for 8 weeks. Recurrent distressing dreams item of the Clinician administered PTSD Scale (CAPS), Total CAPS score, Clinical Global Impressions-Severity Scale (CGI-S) and Clinical Global Impressions-Change Scale (CGI-C) were performed at baseline, 4-week and 8-week. Mississippi Scale for Combat-Related PTSD and Combat Exposure Scale (CES) were used to evaluate PTSD symptom pattern and degree of exposure. Results : Significant decrease in recurrent distressing dreams item, reexperiencing, avoidance and hyperarousal symptom score and total CAPS score were at 4-week and 8-week treatment compared to baseline (recurrent distressing dreams item : $2.70{\pm}1.88$ and $1.25{\pm}1.49$ ; reexperiencing, avoidance and hyperarousal symptom score : $2.30{\pm}2.49$, $1.20{\pm}1.05$, $3.10{\pm}1.68$ and $4.00{\pm}3.59$, $1.55{\pm}1.50$, $5.60{\pm}3.23$ ; total CAPS score : $8.20{\pm}6.26$ and $5.40{\pm}5.89$). There were significant correlation between dosage of terazosin and improvement of PTSD symptoms. Adverse effects such as fatigue and orthostatic hypotension were showed, which were mild and self-limited. Conclusion : These results suggest that terazosin may improve severe trauma-related nightmares and overall PTSD symptoms. Randomized controlled study with more subjects would be necessary in the future.

Development of Fiber-end-cap Fabrication Equipment (대구경 광섬유 엔드캡 제작장비 개발)

  • In this paper, we design and construct the equipment to manufacture large-diameter optical fiber end caps, which are the core parts of high-power fiber lasers, and we fabricate large-diameter optical fiber end caps using the home-made equipment. This equipment consists of a CO2 laser as a fusion-splice heat source, a precision stage assembly for transferring the position of a large-diameter optical fiber and an end cap, and a vision system used for alignment when the fusion splice is interlocked with the stage assembly. The output of the laser source is interlocked with the stage assembly to control the output, and the equipment is manufactured to align the polarization axis of the large-diameter polarization-maintaining optical fiber with the vision system. Optical fiber end caps were manufactured by laser fusion splicing of a large-diameter polarization-maintaining optical fiber with a clad diameter of 400 ㎛ and an end cap of 10×5×2 ㎣ (W×D×H) using home-made equipment. Signal-light insertion loss, polarization extinction ratio, and beam quality M2 of the fabricated large-diameter optical fiber end caps were measured to be 0.6%, 16.7 dB, and 1.21, respectively.

The First Report on Clinical Manifestation of Cryopyrin-Associated Periodic Syndrome in Korean Children

  • Purpose: The aim of this study was to evaluate the clinical characteristics of children diagnosed as cryopyrin-associated periodic syndrome (CAPS) in Korea. Methods: Diagnosis was made based on clinical features and confirmed by a mutation in the cold-induced autoinflammatory syndrome 1 (CIAS1) gene. Especially, osteocartilaginous overgrowth in the patella or distal femur was so characteristic that its presence warranted a diagnosis of chronic infantile neurologic cutaneous and articular/NOMID. Results: We observed the clinical features of 9 Korean CAPS patients. All the patients suffered from an urticarial rash with recurrent fever. Among the 9 patients, 6 presented with rash and 4 with fever on the 1st or 2nd days of birth. Eight patients showed myalgia, and 7 patients showed arthralgia in the joints, and 6 patients showed radiologic findings of arthropathy including cupping of the metaphysis, excessive growth of the epiphysis, osteopenia or overgrowth of the cartilage. Four patients showed brain atrophy, enlarged ventricles or leptomeningeal enhancement on magnetic resonance imaging. Intellectual disability was observed in 1 patient. Five patients had eye involvement as conjunctivitis, uveitis, chorioretinitis, avascular area or papillary edema, and 3 patients showed progressive hearing loss. All 9 patients showed increased C-reactive protein (CRP) and erythrocyte sedimentation rate (ESR). Conclusions: All the patients carried a mutation on exon 3 of the CIAS1 gene. After the anakinra (interleukin-1 receptor antagonist) therapy, the fever and rash immediately disappeared, and CRP and ESR were improved.

Development and Application of a Cleaved Amplified Polymorphic Sequence Marker for Discriminating A Mating Type Alleles of Lentinula edodes (표고 A 교배형 구분을 위한 CAPS 마커의 개발 및 적용)

  • Lentinula edodes is one of the most widely consumed edible mushrooms in Korea. Mating in L. edodes is regulated by a tetrapolar system, and two unlinked genetic loci, A and B, are known to be major determinants of the mating types, as reported in other heterothallic basidiomycetes. The A locus of L. edodes encodes a pair of homeodomain (HD) transcription factors. The highly variable N-termini of these HD transcription factors contribute to the diversity among the A mating types. In this study, we developed a cleaved amplified polymorphic sequence (CAPS) marker to discriminate 11 different A mating type alleles predominant among both cultivated and wild strains. Amplification of the variable region of the A locus followed by digestion with HaeIII and EcoRI restriction enzymes enabled successful discrimination among the 11 A mating type alleles. We also evaluated the applicability of this method in the identification of two A mating types of a dikaryotic strain.
