수용성 치자(水溶性 梔子)(Gardenia jasminoides열매)색소(色素)를 첨가(添加)한 배지(培地)에 배양(培養)한 각종(各種) 효모균류(酵母菌類) 균집락(菌集落)의 형태(形態) (Colonial Morphology of Yeasts Grown on the Medium Containing Pigments Extracted from the Fruits of Gardenia jasminoides)

  • 수용성 치자(水溶性 梔子)(Ga. jasminoides의 열매)색소(色素)(등황색)(橙黃色)를 냉수(冷水)로 추출하여 함유시킨 yeast morphology agar(Difco)에 각종 효모균류(酵母菌類)를 배양하여 그들의 균집락(菌集落) 색깔변화를 관찰하여 다음과 같은 결과를 얻었다. 치자배지(梔子培地)에서 증식(增殖)한 Ca. guilliermondii 와 Ca. lusitaniae 의 균집락(菌集落)은 $28^{\circ}C$에서 3일이내에 군청색(群靑色)을 띠었고, Ca. tropicalis와 Ca. viswanathii 등은 옅은 회색(灰色)을 띠었는데 Ca. viswanathii는 7일후에 군청색(群靑色)으로 변했다. Ca. krusei와 To. glabrata 등은 $1{\sim}2$주후 회색(灰色)내지 옅은 회청색(灰靑色)을 띠었고 Sa. cerevi-siae는 연두색을 띠었다. 그러나 Ca. albicans와 Ca. parapsilosis 등은 색깔변화가 없었고, 적색(赤色) 균집락(菌集落)의 Rhodotorula sp.도 변화가 없었다. 그러나 Cr. neofor-mans는 갈색(褐色) 내지 자갈색(紫褐色) 균집락(菌集落)을 형성해 다른 모든 균종(菌種)과 뚜렷하게 구별되어 기존(旣存) 선별동정배지(選別同定培地)인 caffeic acid 또는 dopamine배지(培地)를 대체할 수 있을 뿐 아니라 몇몇 Candida spp.의 특정적 색깔을 관찰할 수 있어서 치자배지(梔子培地)가 임상검체(臨床檢體)로부터 효모균종(酵母菌種)의 분리배양(分離培養)에 매우 유용하다고 생각한다.

Detection of PIK3CA Gene Mutations with HRM Analysis and Association with IGFBP-5 Expression Levels in Breast Cancer

  • Breast cancer is the second most common cancer and second leading cause of cancer deaths in women. Phosphatidylinositol-3-kinase (PI3K)/AKT pathway mutations are associated with cancer and phosphatidylinositol-4, 5-bisphosphate 3-kinase catalytic subunit alpha (PIK3CA) gene mutations have been observed in 25-45% of breast cancer samples. Insulin growth factor binding protein-5 (IGFBP-5) can show different effects on apoptosis, cell motility and survival in breast cancer. We here aimed to determine the association between PIK3CA gene mutations and IGFBP-5 expressions for the first time in breast cancer patients. Frozen tumor samples from 101 Turkish breast cancer patients were analyzed with high resolution melting (HRM) for PIK3CA mutations (exon 9 and exon 20) and 37 HRM positive tumor samples were analyzed by DNA sequencing, mutations being found in 31. PIK3CA exon 9 mutations (Q546R, E542Q, E545K, E542K and E545D) were found in 10 tumor samples, exon 20 mutations (H1047L, H1047R, T1025T and G1049R) in 21, where only 1 tumor sample had two exon 20 mutations (T1025T and H1047R). Moreover, we detected one sample with both exon 9 (E542Q) and exon 20 (H1047R) mutations. 35% of the tumor samples with high IGFBP-5 mRNA expression and 29.4% of the tumor samples with low IGFBP-5 mRNA expression had PIK3CA mutations (p=0.9924). This is the first study of PIK3CA mutation screening results in Turkish breast cancer population using HRM analysis. This approach appears to be a very effective and reliable screening method for the PIK3CA exon 9 and 20 mutation detection. Further analysis with a greater number of samples is needed to clarify association between PIK3CA gene mutations and IGFBP-5 mRNA expression, and also clinical outcome in breast cancer patients.

  • 칼슘과 인은 동물체내에 존재하는 광물질로 칼슘이 대략 $1.6{\~}2.0{\%}$, 인이$0.9{\~}1.1{\%}$ 함유되어 있다. 칼슘과 인은 주로 뼈에 많이 함유되어 있는데 주로 $Ca_3(PO_4)_2$의 형태로 존재한다.

세라믹소지에 생성되는 석회에 의한 백화현상분석 (Analysis of Ca-rich efflorescence in ceramic bodies)

  • 세라믹소지에 발생하는 온도별 백화생성기구를 백화를 형성하는 양이온과 음이온을 분석하여 규명하였다. 백화가 발생된 세라믹 소지의 표면을 SEM과 EDS 분석을 하여 백화의 주 성분은 Ca와 S임을 확인하였다. 세라믹소지를 pH 7과 pH 10에서 각기 습식혼합 하였으며, 이때 여과된 액을 ICP와 IC 분석을 하여, 양이온 및 음이온 농도분석을 하였다. pH 7에서 제조된 세라믹 소지의 Ca 이온농도는 pH 10에서 제조된 세라믹소지의 Ca 이온농도의 8배 값을 보였으며, pH 10에서 제조된 세라믹소지는 $1100^{\circ}C$ 이상으로 소결된 소지에서도 백화가 발견되지 않았다.

  • $CaF_2-CaO-P_2O_5$계에서 congruent melting하는 apatite$(Ca_{10}(PO_4)_6F_2)$ 단결정을 FZ법으로 길이 50~60mm의 크기로 육성하였따. 분위기는 산소 분위기로 하였으며 회전수는 상하 상호 역방향으로 20~30rpm로, 성장속도와 분위기 등을 조절하면서 최적의 성장조건을 찾고자 하였다. 성분 원소중 Ca를 Co로 부분 치환시켜 적외선 흡수도 증가 및 발색효과를 얻었다. 결정성 및 성장 방위를 고찰하기 위해 laue back reflection pattern을 조사하였으며, XRD를 이용하여 apatite 합성을 확인하였고 광투과로 FTIR분석으로 발색 및 성분원소를 동정하였다.

복막의 섬유화 기전으로서의 상피중간엽전이 (Epithelial to Mesenchymal Transition in CAPD Patients)

  • Epithelial to mesenchymal transition (EMT) is an important etiologic factor for the development of peritoneal fibrosis in CAPD patients. Mesothelial cells are main source of trans-differentiated fibroblasts under stress from the bioincompatible peritoneal dialysate. In our study there was no difference in dialysate TGF-${\beta}$ and VEGF between the low and high GDP groups during an initial 12 months. However, after adjusting with D-CA125, the low GDPs group showed a significantly lower D-TGF-${\beta}$/D-CA125 and D-VEGF/CA125 during the initial 12 months. Among the adjusted peritoneal growth factors for CA125, VEGF/CA125 and TGF-b/CA125 were factors significantly associated with greater EMT in this study. Adjustment of the peritoneal growth factor for effluent CA125 (surrogate for mass of HPMCs) revealed significant association with EMT suggesting that the fibroblastoid transition from HPMCs could be affected by the amount of intraperitoneal growth factors (TGF-b, VEGF) per unit mass of HPMCs. There was significant improvement in both cell score and D-CA125 at the sixth and 12th months after switching from a high GDPs solution to a low GDPs solution. Use of icodextrin solution in patients who had average peritoneal transport showed not only better systemic effects such as decreased glucose absorption via dialysate but also preservation of the peritoneum, including less EMT and high mesothelial bulk mass. In conclusion, Therapy with low GDP solution including icodextrin may positively impact preservation of the peritoneal membrane integrity and prevention of peritoneal fibrosis with time on PD.

Synthesis of scheelite-type nanocolloidal particles by pulsed laser ablation in liquid and their size distribution analysis

  • A novel pulsed laser ablation process in liquid was investigated to prepare scheelite-type ceramic [calcium tungstate ($CaWO_4$) and calcium molybdate ($CaMoO_4$)] nanocolloidal particles. The crystalline phase, particle morphology, particle size distribution, absorbance and optical band-gap were investigated. Stable colloidal suspensions consisting of well-dispersed $CaWO_4$ and $CaMoO_4$ nanoparticles with narrow size distribution could be obtained without any surfactant. Particle tracking analysis using optical microscope combined with image analysis was applied for a fast determination of particle size distribution in the prepared nanocolloidal suspensions. The mean nanoparticle size of $CaWO_4$ and $CaMoO_4$ colloidal nanoparticles were 16 nm and 30 nm, with the standard deviations of 2.1 and 5.2 nm, respectively. The optical absorption edges showed blue-shifted values about 60~70 nm than those of reported in bulk crystals. And also, the estimated optical energy band-gaps of $CaWO_4$ and $CaMoO_4$ colloidal particles were 5.2 and 4.7 eV. The observed band-gap widening and blue-shift of the optical absorbance could be ascribed to the quantum confinement effect due to the very small size of the $CaWO_4$ and $CaMoO_4$ nanocolloidal particles prepared by pulsed laser ablation in liquid.
