Anti-Diabetic Effects of DA-11004, a Synthetic IDPc Inhibitor in High Fat High Sucrose Diet-Fed C57BL/6J Mice

  • DA-11004 is a synthetic, potent NADP-dependent isocitrate dehydrogenase (IDPc) inhibitor where $IC_{50}$ for IDPc is 1.49 $\mu$M. The purpose of this study was to evaluate the effects of DA-11004 on the high fat high sucrose (HF)-induced obesity in male C57BL/6J mice. After completing a 8-week period of experimentation, the mice were sacrificed 1hr after the last DA-11004 treatment and their blood, liver, and adipose tissues (epididymal and retroperitoneal fat)were collected. There was a significant difference in the pattern of increasing body weight between the HF control and the DA-11004 group. In the DA-11004 (100 mg/kg) treated group the increase in body weight significantly declined and a content of epididymal fat and retroperitoneal fat was also significantly decreased as opposed to the HF control. DA-11004 (100 mg/kg) inhibited the IDPc activity, and thus, NADPH levels in plasma and the levels of free fatty acid (FFA) or glucose in plasma were less than the levels of the HF control group. In conclusion, DA-11004 inhibited the fatty acid synthesis in adipose tissues via IDPc inhibition, and it decreased the plasma glucose levels and FFA in HF diet-induced obesity of C57BL/6J mice.

Deletion of the Lmna Gene Induces Growth Delay and Serum Biochemical Changes in C57BL/6 Mice

  • The A-type lamin deficient mouse line ($Lmna^{-/-}$) has become one of the most frequently used models for providing insights into many different aspects of A-type lamin function. To elucidate the function of Lmna in the growth and metabolism of mice, tissue growth and blood biochemistry were monitored in Lmna-deficient mice, heterozygous ($Lmna^{+/-}$) and wide-type ($Lmna^{+/+}$) backcrossed to C57BL/6 background. At 4 weeks after birth, the weight of various organs of the $Lmna^{-/-}$, $Lmna^{+/-}$ and $Lmna^{+/+}$ mice was measured. A panel of biochemical analyses consisting of 15 serological tests was examined. The results showed that Lmna deficient mice had significantly decreased body weight and increased the ratio of organ to body weight in most of tissues. Compared with $Lmna^{+/+}$ and $Lmna^{+/-}$ mice, $Lmna^{-/-}$ mice exhibited lower levels of ALP (alkaline phosphatase), Chol (cholesterol), CR (creatinine), GLU (glucose), HDL (high-density lipoprotein cholesterol) and higher levels of ALT (alanine aminotransferase) (p<0.05). $Lmna^{-/-}$ mice displayed higher AST (aspartate aminotransferase) values and lower LDL (lowdensity lipoprotein cholesterol), CK-MB (creatine kinase-MB) levels than $Lmna^{+/+}$ mice (p<0.05). There were no significant differences among the three groups of mice with respect to BUN (blood urea nitrogen), CK (creatine kinase), Cyc C (cystatin C), TP (total protein), TG (triacylglycerols) and UA (uric acid) levels (p>0.05). These changes of serological parameters may provide an experimental basis for the elucidation of Lmna gene functions.

Regulation of PPAR and SREBP-1C Through Exercise in White Adipose Tissue of Female C57BL/6J Mice

  • Previous study showed that swimming improved obesity but was not through $PPAR{\alpha}$ activation in liver and skeletal muscle in high fat diet-fed female mice with functioning ovaries as an animal model of obese premenopausal women. Thus, this study was aimed at investigation of the effects of swimming on the promotion of health and its molecular mechanism in adipose tissue of high fat diet-fed female mice. Eight-week-old female C57BL/6J mice were randomly divided into two groups (a non-swim control group and a swim group, n=8/group). Mice in the swim group swam for 2 h daily for 6 weeks in water bath with temperature of $35{\pm}1^{\circ}C$. All the animals received high fat diet (45% kcal fat) for 6 weeks. Reverse transcription-polymerase chain reaction was used to elucidate the molecular mechanism. Female mice subjected to swimming had significantly decreased body weight gain and white adipose tissue mass compared with the female control mice. Histological studies illustrated that swimming decreases the hepatic lipid accumulation. As expected, swimming did not affect the expression of mRNA levels of peroxisome proliferator-activated receptor (PPAR) ${\alpha}$ and $PPAR{\alpha}$ target genes responsible for mitochondrial fatty acid ${\beta}$-oxidation, such as carnitine palmitoyltransgerase-1 and medium chain acyl-CoA dehydrogenase in the white adipose tissue. However, mice that underwent 6-weeks of swimming exercise had decreased the mRNA expression of lipogenic genes, such as sterol regulatory element-binding proteins-1C and fatty acid synthase in comparison to sedentary control mice, with decreased $PPAR{\gamma}$ target genes involved in adipocyte-specific marker genes, such as adipocyte fatty acid binding protein and leptin in the white adipose tissue. These results suggest that swimming can effectively prevent obesity induced by high fat diet-fed, in part through down-regulation of adipogenesis and lipogenesis in white adipose tissue of female obese mice. Moreover, these results suggest that swimming maybe contributing the promotion of health through regulation of adipogenesis and lipogenesis in overweight premenopausal women.

HFD 유도 C57BL/6J 비만 mice에서 AMPK/ACC/CPT-1 경로 촉진을 통한 산딸기 추출물의 비만 및 비알코올성 지방간 질환에 대한 보호 효과 (Protective Effect of Rubus crataegifolius Extracts Against Obesity and Non-alcoholic Fatty Liver Disease via Promotion of AMPK/ACC/CPT-1 Pathway in HFD-induced C57BL/6J Obese Mice)

  • Rubus crataegifolius (RC)는 장미과에 속하는 전통적인 아시아 약용 식물이다. RC 열매는 항산화 작용을 통해 성인병을 예방하는 것으로 알려져 있다. 본 연구에서는 RC 열매 추출물(RCex)이 비만과 비알코올성 지방간 질환(NAFLD)에 미치는 영향을 동물 모델을 통해 평가하였다. 28마리의 수컷 C57BL/6J 마우스에 8주간 비만을 유도한 후, 추출물을 8주간 경구 투여하였다. 그룹 1은 일반 대조군으로 표준사료를 섭취하였다. 그룹 2는 HFD 대조군으로, 그룹 3에는 심바스타틴(6.5 mg/kg/일)을, 그룹 4에는 RCex (200 mg/kg)을 투여하였다. RCex투여는 실험 마우스의 체중, 지방 조직, 간 무게를 감소시켰으며, 또한 지질 대사(ALT, AST, TC, TG, LDL, HDL)를 포함한 생화학적 바이오마커를 개선하였다. AMPK의 활성화는 지방생성 유전자(LXR, SREBP-1c, FAS, ACC1)의 발현을 감소시켰으며, RCex에 의한 CPT 활성 증진 효과를 검증하였다. RCex는 또한 에너지 소비 및 신진대사와 관련된 호르몬(adiponectin 및 leptin)의 혈장 수준에도 영향을 미쳤다. 또한, RCex가 HFD로 유도된 비만 mice의 포도당 불내성을 개선했음을 확인 하였다. RCex는 AMPK의 인산화를 통해 지방산 산화 및 지방산 합성을 조절함으로써 항비만 및 항NAFLD 효과를 가짐을 처음으로 입증하였다. 이는 R. crataegifolius가 비만 및 관련 NAFLD 예방에 좋은 보충제가 될 수 있음을 시사한다.

고려엉겅퀴 주정 추출물을 함유하는 임상시험제품의 항비만 활성 평가 (Anti-obesity Activities of Cirsium setidens Nakai Ethanolic Extract)

  • 본 연구는 고려엉겅퀴 주정추출물을 가지고 제작된 임상시험용제품(CNTM)이 3T3-L1 지방세포 및 고지방식이로 유도된 비만 쥐에 미치는 항비만 효능 관찰을 통하여 체지방 개선 기능성식품을 개발하기위하여 인체적용시험 시료를 제작한 후, 인체적용시험전에 그 효력이 유지됨을 확인하기위해 수행되었다. 본 연구에 사용된 시료 CNTM은 $80{\sim}320{\mu}g/mL$ 농도에서 세포독성이 관찰되지 않았으며, 지방 축적억제 효능 및 지방세포 분화, 지질대사 관련 유전인자들을 유의적으로 변화시키는 것으로 확인되었다. 또한 동물실험에서 CNTM 처리에 의하여 체중의 감소를 확인하였으며 혈중지질 성분 가운데 HDL-C/TC의 비율은 유의적으로 증가하며 LDL-cholesterol (LDL-C)과 non-esterified fatty acid (NEFA)의 비율은 유의적으로 감소하여 혈중 유리지방산 농도를 감소시키며 혈중 콜레스테롤 상승을 억제시키고 혈당을 감소시키는 효과가 있는 것으로 평가되었다. 따라서 고려엉겅퀴 주정추출물이 함유된 인체적용시험시료(CNTM)는 체지방 개선에 우수한 효과가 있는 것으로 확인되었다.

The antioxidant activity of steamed ginger and its protective effects on obesity induced by high-fat diet in C57BL/6J mice

  • BACKGROUND/OBJECTIVES: Ginger, a root vegetable, is known to have antioxidant and antiobesity effects. Preparation, such as by steaming, can affect the chemical composition of prepared root vegetables or herbs and can change their functional activities. In the present study, we investigated the protective effects of steamed ginger against oxidative stress and steatosis in C57BL/6J mice fed a high-fat diet. MATERIAL/METHODS: The levels of polyphenols and flavonoids in two different extracts of steamed ginger, i.e., water extract (SGW) and ethanolic extract (SGE); as well, their antioxidant activities were examined. Forty male C57BL/6J mice were fed a normal diet (ND, n = 10), high-fat diet (HFD, 60% fat, w/w, n = 10), HFD supplemented with 200 mg/kg of SGE or garcinia (GAR) by weight (SGED or GARD, respectively, n = 10) for 12 weeks. Serum chemistry was examined, and the expressions of genes involved in lipid metabolism were determined in the liver. Histological analysis was performed to identify lipid accumulations in epididymal fat pads and liver. RESULTS: The SGE had higher contents of polyphenols and flavonoids and higher DPPH and $ABTS^+$ free radical scavenging activities compared to those of SGW. Treatment with SGE or GAR significantly decreased the HFD-induced weight gain. Both SGE and GAR significantly reduced the high serum total cholesterol (TC), triglyceride (TG) and low-density lipoprotein levels induced by HFD. Compared to ND, HFD significantly increased hepatic TC and TG levels. SGE or GAR supplementation significantly decreased the increase of hepatic lipids by HFD. Interestingly, SGE had a more significant effect in reducing hepatic TC and TG levels than GAR. Furthermore, hepatic genes involved in lipogenesis and lipolysis were altered in both the SGED and GARD groups. CONCLUSIONS: The present study indicates that steamed ginger supplementation can decrease plasma TC and TG and can inhibit liver steatosis by regulating the expressions of hepatic genes.

Grape seed extract (Vitis vinifera) partially reverses high fat diet-induced obesity in C57BL/6J mice

  • The aim of the present study was to assess the anti-obesity effects of grape seed extract (GSE) supplement in C57BL/6J mice. Thirty mice were divided into three groups; normal diet control group (ND), high fat diet control group (HD) and high fat diet plus grape seed extract supplemented group (HD+GSE). Results were as follows: 1. GSE supplement reduced the weight gain in mice fed high fat diets; epididymal and back fat weights, were lower compared to non-supplemented HD group. 2. Blood lipid concentrations were lower in the HD+GSE group than in the HD group. Serum HDL-C concentrations were higher in the HD+GSE group compared with the other groups. 3. The concentrations of acid-insoluble acylcarnitines, (AIAC) in serum and liver were higher in the HD+GSE group than in the HD group. 4. GSE supplementation increased mRNA levels of lipolytic genes such as carnitine palmitoyltransferase-l (CPT-1) and decreased mRNA levels of lipogenic genes such as acetyl CoA carboxylase (ACC). These findings suggest that grape seed extract supplements in high fat diet might normalize body weight, epididymal and back fat weights, lipid concentrations, and carnitine levels through controlling lipid metabolism.

C57BL/6J 마우스에서 울금(Curcuma longa L.) 추출물이 첨가된 인조쌀의 체내 지질 수준 개선 효과 (Improvement Effect of Artificial Rice Containing Curcuma longa L. Extract on Lipid Parameters in C57BL/6J Mice)

  • 본 연구에서는 울금의 메탄올 추출물을 제조하고 3T3-L1 지방세포 모델을 이용하여 세포 수준에서 울금 인조쌀의 지방구 형성 조절을 통한 지질축적 억제능을 평가하였다. 더 나아가 C57BL/6J 마우스에 고지방식이와 함께 인조쌀, 울금 추출물이 첨가된 인조쌀을 공급하였을 때 체중과 지질대사의 변화를 관찰하였다. 세포 실험의 결과로부터 울금 메탄올 추출물을 처리한 후 $20{\mu}g/mL$의 농도에서 유의적인 지방구형성 억제를 관찰하였다. 동물 실험에서는 식이효율을 측정한 결과 인조쌀을 섭취한 군에서 그 효율이 낮아졌으며, 특히 울금 인조쌀을 섭취한 군에서 유의적으로 감소하였다. 부고환 지방 무게는 인조쌀 섭취군에서는 고지방섭취군과 차이를 보이지 않았지만, 울금 인조쌀을 섭취한 군에서는 유의적으로 낮아졌다. 혈청 지질 수준은 인조쌀군과 울금 인조쌀군 모두 고지방식이군에 비하여 개선되는 결과를 보였다. 혈청 중성지방은 인조쌀군과 울금 인조쌀군 모두유의적으로 낮아졌으며, 혈청 콜레스테롤 함량은 고지방식이군에 비해 인조쌀군에서 감소하는 경향을 보였고 울금 인조쌀군에서는 유의적으로 감소되었다. 이러한 결과를 통해 울금 이 첨가된 인조쌀에 다량 함유된 식이섬유소와 여러 생리활성물질이 지질대사를 개선시키고 지방 축적을 억제하여 일상식을 통한 체내 지질 수준 개선 효과를 기대해 볼 수 있을 것이라 사료된다.

Combined Effects of Cell Cultured Acanthopanax Senticosus Supplementation and Exercise on Lipid Profiles, Carnitine and Leptin Levels in Mice

  • The purpose of this study was to determine the independent and the combined effects of cell cultured Acanthopanax senticosus extracts (ASE) supplementation and swimming exercise on body weight, lipid profile, carnitine and leptin levels in C57 BL/6J mice. Forty C57BL/6J mice were divided into four groups: non-supplement and non-exercise (NSNE); non-supplement and exercise (NSE); supplement and non-exercise (SNE); supplement and exercise (SE) mice. They were allowed free access to food and water. The exercised groups were forced to swim (1hr, 6 days a week) in a water bath for 12 weeks. The supplemented groups were fed Cell cultured ASE (0.5 g/kg body weight/day) for 12 weeks. In this study, we found that the combination of Cell cultured ASE supplementation and exercise significantly decreased liver triglyceride (TG) level and serum leptin level but significantly increased serum HDL-cholesterol level compare to control (NSNE) group. These improved lipid profiles and decreased serum leptin would have positive effects on obesity and cardiovascular disease.

파벽(破壁) 영지포자가 고지방식이 섭취 마우스의 지방축적 및 체중감량에 미치는 영향 (Effect of Cell-Wall Broken Spores of Ganoderma lucidum(Leyss. ex.Fr.) Karst on the Lipid Accumulation and Body Weight Reduction in C57BL/6J Mice fed High-Fat Diet)

  • This study was performed to investigate the effects of cell-wall broken spores of Ganoderma lucidum on the lipid accumulation and body weight reduction in C57BL/6J mice. Six-week-old C57BL/6J mice were randomly divided into 5 groups and assigned to one of these groups; normal chew diet(Nor) group, high-fat diet(HFD) group, HFD plus spores of Ganoderma lucidum 800 mg/kg/day (HFD + GS/B) group, HFD plus cell-wall broken spores of Ganoderma lucidum 400 mg/kg/day (HFD + BGS/A) group and finally HFD plus cell-wall broken spores of Ganoderma lucidum 800mg/kg/day (HFD + BGS/B). The experimental groups which were treated oral co-administration with cell-wall broken(or original) spores of Ganoderma lucidum and HFD significantly attenuated accumulative body weight gain, compared with HFD group. Administration of these experimental materials also resulted in significant reduction not only the serum levels of total cholesterol, homocysteine but also the lipid accumulation in liver tissue. But in the almost of results the cell-wall broken spores of Ganoderma lucidum were evaluated superior than the original one. These results indicate that cell-wall broken spores of Ganoderma lucidum may inhibit the lipid accumulation in blood as well as liver tissue. Therefore it may be a valuable candidate for the therapy preventing obese induced hyperlipidemic symptoms.
