본 연구에서는 질소 시비수준이 생육시기별 식물체 및 종실에서 isoflavone함량에 미치는 영향을 구명하기 위하여 고속액체크로마토그래피(HPLC)를 이용하여 생육시기에 따른 식물체에서 isoflavone을 분석한 결과를 요약하면 다음과 같다. 1. 질소시비 수준에 따른 일반생육은 50%증비구가 다른 처리에 비하여 가장 좋았는데, 엽수 및 엽면적이 표준비의 72.7개, $619.7cm^{2}$에 비하여 81.9개와 $723cm^{2}$으로 증가하였다. 2. 수량 관련 형질 중에서 협수 및 종실수는 50%증비에서 개체당 31.9개와 72.3개로 가장 많았으며, 수량도 246 kg/10a로 다른 시비구에 비하여 가장 높았다. 무비와 N무비구에서는 협수가 24.9개/개체와 25.1개/개체, 립수는 60.3개/개체과 51.1개/개체, 수량은 181.0 kg/10a과 178.0 kg/10a으로 낮았다. 3. 생육시기에 따른 isoflavone 함량은 잎, 줄기, 뿌리에서 개화이후부터 립비대기(R5)까지는 감소하다가 이후 수확기(R7)까지 다시 증가하였다. 4. 식물체 부위별 isoflavone 함량은 뿌리에서 가장 높은 값을 나타내었다. 5. 시비수준에 따른 종실의 isoflavone 함량은 무비, N무비 및 50% 감비의 경우가 표준비나 증비보다 다소 높은 함량을 나타내었다.
The contents of lignan, protein, and oil of nine Sesamum indicum cultivars were analyzed. To determine the lignan contents, sesamin 1 and sesamolin 2 were isolated from the ethylacetate extracts of the Suwon cultivar, and the structures were fully characterized by spectral and physical methods. The seeds of nine cultivars were screened for two lignans, which were determined by HPLC using a $C_{18}$ reversed phase column coupled with a photodiode array detector. Suwon cultivar showed the highest concentration (1: $6.24{\pm}0.04$ and 2: $3.58{\pm}0.01\;mg/g$), whereas Soonheuk displayed the lowest (1: $0.91{\pm}0.01$ and 2: $0.73{\pm}0.01\;mg/g$). The average content ratio of sesamin 1 (3.64 mg/g) was significantly higher than that of sesamolin 2 (2.57 mg/g). The protein content ranged from $21.52{\pm}0.35$ to $31.22{\pm}0.25%$, Suwon containing the highest level and Soonheuk had the lowest. Kwangbaek showed the highest oil level ($49.84{\pm}0.40%$), while Soonheuk exhibited the lowest ($42.52{\pm}0.05%$). Sesamin 1 exhibited a stronger radical-scavenging activity in the ABTS ($IC_{50}:\;63.2{\pm}2.4\;{\mu}M$) than its DPPH radical-scavenging activity ($IC_{50}>200\;{\mu}M$). These results lead to the conclusion that lignan content is affected by protein and oil contents. Concentration of the chemical components in the cultivars could be a key factor in the selection process of a high quality species.
New generation of tall and complex buildings systems are now introduced that are reflective of the latest development in materials, design, sustainability, construction, and IT technologies. While the complexity in design is being overcome by the availability and advances in structural analysis tools and readily advanced software, the design of these buildings are still reliant on minimum code requirements that yet to be validated in full scale. The involvement of the author in the design and construction planning of Burj Khalifa since its inception until its completion prompted the author to conceptually develop an extensive survey and real-time structural health monitoring program to validate all the fundamental assumptions mad for the design and construction planning of the tower. The Burj Khalifa Project is the tallest structure ever built by man; the tower is 828 meters tall and comprises of 162 floors above grade and 3 basement levels. Early integration of aerodynamic shaping and wind engineering played a major role in the architectural massing and design of this multi-use tower, where mitigating and taming the dynamic wind effects was one of the most important design criteria established at the onset of the project design. Understanding the structural and foundation system behaviors of the tower are the key fundamental drivers for the development and execution of a state-of-the-art survey and structural health monitoring (SHM) programs. Therefore, the focus of this paper is to discuss the execution of the survey and real-time structural health monitoring programs to confirm the structural behavioral response of the tower during construction stage and during its service life; the monitoring programs included 1) monitoring the tower's foundation system, 2) monitoring the foundation settlement, 3) measuring the strains of the tower vertical elements, 4) measuring the wall and column vertical shortening due to elastic, shrinkage and creep effects, 5) measuring the lateral displacement of the tower under its own gravity loads (including asymmetrical effects) resulting from immediate elastic and long term creep effects, 6) measuring the building lateral movements and dynamic characteristic in real time during construction, 7) measuring the building displacements, accelerations, dynamic characteristics, and structural behavior in real time under building permanent conditions, 8) and monitoring the Pinnacle dynamic behavior and fatigue characteristics. This extensive SHM program has resulted in extensive insight into the structural response of the tower, allowed control the construction process, allowed for the evaluation of the structural response in effective and immediate manner and it allowed for immediate correlation between the measured and the predicted behavior. The survey and SHM programs developed for Burj Khalifa will with no doubt pioneer the use of new survey techniques and the execution of new SHM program concepts as part of the fundamental design of building structures. Moreover, this survey and SHM programs will be benchmarked as a model for the development of future generation of SHM programs for all critical and essential facilities, however, but with much improved devices and technologies, which are now being considered by the author for another tall and complex building development, that is presently under construction.
Helicobacter pylori에 대하여 항균효과가 가장 높은 Fomitopsis pinicola를 동정하고 저해물질의 성질에 관한 연구를 수행한 결과 다음과 같은 결론이 얻어졌다. H. pylori에 대한 항균력이 가장크게 나타난 네팔에서 채취한 F. pinicola의 동정결과는 F. pinicola AY854083로 밝혀졌다. F. pinicola의 균사체는 액상배양을 시작한 후 5일이 경과하면서 H. pylori에 대한 항균력이 나타났으며 8일 이후부터 가장 높았다. 그러나 균사체 추출물에서는 항균력이 나타나지 않았다. F. pinicola 균사체 8일 배양액의 H. pylori에 대한 최소억제 농도는 $0.25\;mg/m{\ell}$ 이었고, 배양액의 낮은 pH (pH 2.0)가 H. pylori의 생육억제에 영향을 주지만 중화를 하여도 H. pylori에 대한 항균력은 유지되었다. F. pinicola 균사체 8일 배양액을 DEAE-sephadex A-25 LC로 분리하여 Fp-P1, Fp-P2 및 Fp-P3를 얻었으며 H. pylori에 대한 항균력은 Fp-P1에서만 나타났다. Fp-P1을 thin layer chromatography로 분리하였으며 H. pylori에 대한 항균력은 $R_{f}$ 값이 0.67인 Fp-T3에서만 나타났다. F. pinicola 균사체 8일 배양액, Fp-P1, Fp-T3를 HPLC의 Waters symmetry $300\;C_{18}\;5\;{\mu}m$ column ($3.9{\times}150\;mm$)으로 분리한 결과 Fp-T3에서 단일 peak만 남았으며 이것이 H. pylori에 대한 항균력을 확인하였다. H. pylori에 대한 항균력을 나타내는 F. pinicola 균사체 배양액에서 분리 정제한 물질은 BioLC 분석과 TLC 발색결과로써 아미노기를 포함한 당으로 사료된다.
본 연구는 싸리나무의 기능성 식의약품 개발을 위해 부위별 최적의 에탄올 추출조건을 탐색하고 HPLC-DAD 분석방법에 대한 밸리데이션을 실시하였다. 싸리나무뿌리 추출물로부터 반복적인 크로마토그래피 기법으로 주요성분을 분리하여 NMR, MS와 같은 기기분석 data를 해석하여 xanthoangelol 화합물임을 확인하였다. Xanthoangelol를 표준물질로 선정하여 HPLC-DAD 분석법이 타당한지를 검토하기 위하여 직선성, 일간 일내 정확성, 정밀성 검출한계 및 정량한계를 확인하는 분석법 밸리데이션을 수행하였다. Xanthoangelol 분석은 ZORBAX Eclipse plus C18 (4.6×150 mm, 5 ㎛) 칼럼과 기울기 용출(gradient elution) 방법으로, 370 nm 파장에서 다른 물질의 간섭 없이 안정되게 분석되는 것을 확인하였다. 기능성 소재로 적용하기에 적합한 물과 에탄올을 이용해 농도별(water, 30, 50, 70, 95% EtOH)로 추출하고 xanthoangelol의 함량을 분석하였으며, 에탄올 비율에 따라 대상 성분의 추출율을 조절함으로써 기능성 바이오 식의약품 원료 개발을 위한 기초 자료로 활용할 수 있을 것으로 생각한다.

A 2,172-bp full-length gene (aga-F78), encoding a protease-resistant $\alpha$-galactosidase, was cloned from Rhizopus sp. F78 and expressed in Escherichia coli. The deduced amino acid sequence shared highest identity (45.0%) with an $\alpha$-galactosidase of glycoside hydrolase family 36 from Absidia corymbifera. After one-step purification with a Ni-NTA chelating column, the recombinant Aga-F78 migrated as a single band of ~82 and ~210 kDa on SDS-PAGE and nondenaturing gradient PAGE, respectively, indicating that the native structure of the recombinant Aga-F78 was a trimer. Exhibiting the similar properties as the authentic protein, purified recombinant Aga-F78 was optimally active at $50^{\circ}C$ and pH 4.8, highly pH stable over the pH range 5.0-10.0, more resistant to some cations and proteases, and had wide substrate specificity (pNPG, melidiose, raffinose, and stachyose). The recombinant enzyme also showed good hydrolytic ability to soybean meal, releasing galactose of $415.58\;{\mu}g/g$ soybean meal. When combined with trypsin, the enzyme retained over 90% degradability to soybean meal. These favorable properties make Aga-F78 a potential candidate for applications in the food and feed industries.
Residual solvents in foods are defined as organic volatile chemicals used or produced in manufacturing of extracts or additives, or functional foods. The solvents are not completely eliminated by practical manufacturing techniques and they also may become contaminated by solvents from packing, transportation or storage in warehouses. Because residual solvents have no nutritional value but may be hazardous to human health, there is a need to remove them from the final products or reduce their amounts to below acceptable levels. The purpose of this study was to develop and evaluate an analytical method for the screening of residual solvents in health functional foods. Furthermore, the aim of this study was to constitute a reasonable management system based on the current state of the market and case studies of foreign countries. Eleven volatile solvents such as MeOH, EtOH, trichloroethylene and hexane were separated depending on their column properties, temp. and time using Gas Chromatography (GC). After determining the GC conditions, a sample preparation method using HSS (Head Space Sampling) was developed. From the results, a method for analyzing residual solvents in health functional foods was developed considering matrix effect and interference from the sample obtained from the solution of solvents-free health functional foods spiked with 11 standards solutions. Validation test using the developed GC/HSS/MS (Mass Spectrometry) method was followed by tests for precision, accuracy, recovery, linearity and adequate sensitivity. Finally, examination of 104 samples grouped in suits was performed by the developed HSS/GC/MS for screening the solvents. The 11 solvents were isolated from health functional foods based on vapor pressure difference, and followed by separation within 15 minutes in a single run. The limt of detection (LOD), limit of quantification (LOQ), recovery and coefficient of variation (C.V.) of these compounds determined by the HSS/GC/MS were found to be 0.1 pg/mL, 0.1-125 pg/g, 51.0-104.6%, and less than 15%, respectively. Using the developed HSS/GC/MS method, residual solvent from 16 out of 104 health functional products were detected as a EtOH. This method therefore seems t o be a valuable extension ofanalytical method for the identification of residual solvents in health functional food.
일반적으로 많이 사용하고 있는 한약재 중 항산화효과가 있는 대상을 선발하였고 그 중 효과가 우수한 건칠을 대상으로 항산화물질을 분리, 효과를 측정하였다. 건칠(Rhus verniciflua STOKES)은 옻나무과(Anacardiaceae)에 속하는 옻나무(Rhus verniciflua STOKES)의 수액으로 한방에서는 구충, 복통, 통경, 변비의 목적으로 사용되어왔다. 옻나무 에탄올추출물은 돈지와 팜유를 기질로 Rancimat test에서 항산화효과가 인정되었으며, 용매의 극성을 각각 달리하여 분획하였을때 chloroform 층에서 항산화 활성이 가장 높았다. 이 분획물을 다시 phenolic compound를 중심으로 용매 분획한 후, 활성이 높은 free phenolic acid부분을 silica gelcolumn chromatography하여 RCF-11 분획하였으며, 이 분획물을 돈지, 팜유 및 대두유에 각각 200 ppm 수준으로 첨가 하였을 때 AI (antioxidant index: 추출물을 넣은 유지의 유도기간을 기질유지의 유도기간으로 나눈 값)는 각각 5.57, 1.53 및 1.54를 나타내었다. 이것은 같은 농도에서의 ${\delta}-tocopherol$, BHT, BHA 보다 동물성유지인 돈지에서는 4배, 식물성유인 팜유와 콩기름에서는 1.5배 정도 효과가 우수하였다. RCF-11에 항산화작용이 알려진 ascorbic acid, citric acid, ${\delta}-tocopherol$ 200 ppm을 첨가한 후 분획물의 농도를 달리해가며 동물성유지와 식물성유지에 적용해보았다. 식물성유에서는 ascorbic acid를 첨가했을 때 단독으로 사용했을 때보다 AI가 1.5배 정도 상승효과가 있었으며, 동물성유에서는 ${\delta}-tocopherol$를 첨가했을 때 단독으로 사용했을 때보다 AI가 2배 정도 상승하였다. Rancimat을 통해 항산화 활성이 확인 된 옻나무 75% ethanol 추출물의 free phenolic acid 성분은 TLC, UV spectra를 통해 본 결과 flavanone이나 flavone계 화합물로 추정되며, 앞으로 그 원인물질에 대한 구조동정이 요구된다.
산양유 $\beta-casein$의 효소에 의한 가수분해 특성과 가수분해물의 ACE 저해 효과를 측정하고자 산양유의 $\beta-casein$을 양이온 교환 컬럼인 Mono S HR 5/5를 이용하여 분리하였으며 분리된 $\beta-casein$을 동물성 분해효소인 trypsin으로 처리하여 가수분해 특성을 확인하였고 가수분해물의 ACE 저해활성을 측정하였다. Mono S HR 5/5 양이온 교환 컬럼을 이용한 산양유 산 케이신으로 부터 순수한 $\beta-casein$의 분리는 SDS-PAGE를 이용하여 확인한 결과 순수한 $\beta-casein$의 분리가 이루어졌음을 확인할 수 있었다. $\beta-casein$을 $37^{\circ}C4$에서 trypsin으로 처리하여 전기영동으로 확인한 결과 가수분해 직후부터 $\beta-casein$ 위치의 band가 희미해지기 시작하고 저분자량의 band가 나타나기 시작하였으나 120분이 지난 후에는 모든 band가 가수분해되어 사라졌고 산양유에서 분리된 $\beta-casein$을 trypsin으로 처리하여 120분 경과 후 그 가수분해물을 이용하여 ACE 저해효과를 측정한 결과 가수분해하지 않은 $\beta-casein$은 $1.80\pm1.21\%$의 ACE 저해활성을 보였으나 trypsin으로 가수분해하여 ACE 저해 활성을 측정하였을 때 $25.36\pm0.79\%$의 저해 활성을 나타내었으며, trypsin에 의한 $\beta-casein$가수분해물의 $IC_{50}$을 측정한 결과 $308.7\pm2.77({\mu}g/mL)$로 나타났다.
