Stiudies on Enzyme of the Thermophilic Mold -(Part 4) Xylanase and Laminaranase from Thermophilic Mold- (고온성 사상균의 효소에 관한 연구 -(제4보) 고온성 사상균의 Xylanase 와 Laminaranase-)

  • In order to investigate the action of thermophilic fungi on the xylan and its related substances, xylanase and laminaranase were examined using Myriococcum albomyces. In the extract from bran culture of Myriococcum albomyces, xylanase and laminaranase activity were recognized. 1) The optimum pH for xylanase and laminaranase activity were found to be pH 5.0 and pH 6.0. 2) The optimum temperature for xylanase and laminaranase activity were found to be $55^{\cicr}C$. 3) Thermal stability of xylanase for 55 minutes at $65^{\cicr}C$ and laminaranase for 60 minutes at $65^{\cicr}C$ did not influence their stability.

  • A bark comprises about 10 to 20 percents of a typical log by volume, and is generally considered as an unwanted residue rather than a potentially valuable resources. As the world has been confronted with decreasing forest resources, natural resources pressure dictate that a bark should be a raw material instead of a waste. The utilization of the largely wasted bark of genus Pinus, Quercus and Populus grown in korea can be enhanced by Knowing its chemical Properties. Specially, uses of bark in mulching, growing media and soil amendment can be enhanced by knowing pH and C.E.C values. In this paper, an investigative study is carried out on the pH-and C.E.C-values. The results may be summarized as follows: 1. Bark is acid in nature, and pH values of bark varying from 3.3 to 4.7, is lower than that of wood.pH value of Pinus is the lowest. 2. Cation exchange capacity of bark is greater than that of wood, silt loam soil, and wheat straw.CEC of bark is 45.7 meq/100gm in Pinus, 41.8 meq/100gm in populus, 37.8 meq/100gm in Quercus.

Isolation and Characterization of Exopolysaccharide Producing Lactic Acid Bacteria from Kimchi (김치로부터 Exopolysaccharide 생성 유산균의 분리 및 특성 규명)

  • Three slime-forming lactic acid bacteria were isolated from Kimchi and shown to produce viscous exopolysaccharides (EPS) in sucrose media. The isolated strains, GJ2, C3 and C11, were identified as Leuconostoc kimchii, Leuconostoc citreum and Leuconostoc mesenteroides, respectively, by examining their metabolic characteristics and determining their 16S rDNA sequences. Leu. kimchii GJ2, Leu. citreum C3 and Leu. mesenteroides C11 exhibited high viability (maintained initial viable cell count of $10^8$ CFU/ml) in 0.05 M sodium phosphate buffer (pH 3.0) for 2 h, in artificial gastric juice for 2 h and in 0.3% oxgall for 24 h. When tested, Leu. kimchii GJ2, in particular, displayed antimicrobial activity against pathogenic microorganisms. Leu. kimchii GJ2, Leu. citreum C3 and Leu. mesenteroides C11 produced 21.49 g/l, 16.46 g/l and 22.98 g/l EPS, respectively, in sucrose (5%) medium. The amount of purified EPS extracted from Leu. kimchii GJ2, Leu. citreum C3 and Leu. mesenteroides C11 was 14.61 g/l, 7.73 g/l and 4.77 g/l, respectively. Although the EPS produced by Leu. kimchii GJ2, Leu. citreum C3 and Leu. mesenteroides C11 differed in viscosity, TLC and HPLC analysis revealed that each contained only one type of monosaccharide, glucose. The average molecular mass of EPS produced by Leu. kimchii GJ2 was 306,606 Da.

  • The molecular and crystal structure of Tricyclazole, C/sub9/H/sub7/N₃S, has been determined by single crystal x-ray diffraction study. Crystallographic data for title compound: Pca2₁, a=14.889(1) Å, b=7.444(1) Å, c=15.189(2) Å, V=1683.3(3) ų, Z= 8. The molecular structure model was solved by direct methods and refined by full-matrix least-squares. The final reliable factor, R, is 0.047 for 1533 independent reflections (F/sub o//sup 2/)). The asymmetry unit contains two molecules which are in plate conformation, parallel to each other and related by a pseudo four-fold screw on the b-direction.

  • The structure of $C_{12}H_{15}N_3O_3$ has been determined by X-ray diffraction methods. The crystal system is monoclinic, space group $P2_1/c$, unit cell constants, a = 12.9955(9) ${\AA}$, b = 7.7137(5) ${\AA}$, c = 13.4699(11) ${\AA}$, ${\beta}$ = 107.86(1)$^{\circ}$, V = 1285.2(1) ${\AA}^3$, T = 296 K, Z = 4, $D_c$ = 1.350 $Mgm^{-3}$. The intensity data were collected on an Enraf-Nonius CAD-4 Diffractometer with graphite monochromated Mo $K{\alpha}$ radiation (${\lambda}$ = 1.71073 ${\AA}$). The molecular structure was solved by direct methods and refined by full-matrix least squares to a final R = 4.19% for 1644 unique observed $F_0\;>\;4{\sigma}(F_0)$ reflections 193 parameters.

Sporulation of Cercospora capsici causing Cercospora leaf spot of Pepper (고추 갈색점무늬병원균 (Cercospora capsici)의 배양특성과 다량 포자형성법)

  • High sporulation method and cultural characteristics of Cercospora capsici causing Cercospora leaf spot of pepper were examined. Optimum temperature for mycelial growth of Cercospora capsici was $25^{\circ}C$. The fungus did not grow below $5^{\circ}C$ and over $35^{\circ}C$. Optimum pH for mycelial growth was pH 4.0~pH 8.0. Mycelial growth was not influenced by light. C. capsici sporulated well on pepper leaf agar (5g/l). A standard method of sporulation established was as follows. The mycelial plugs were ground with some water in motar with pestle. The mycelial suspension was smeared on the surface of medium and incubated for 2~3 days at $20^{\circ}C$. The culture surface was lightly scraped with a brush after adding 1 ml of sterile water to stimulate sporulation and further incubated for 2~3 days.

  • Block copolymers of the general formula $[(-CO-R'-CO-HN-Ar-NH-CO-R'-CO)_xNH(CH_2)_3-(Me_2SiO)_y(CH_2)_3NH_2]_n$, [n=18.00 to 1175.0] where $R'=CH_2CH(CH_2GeCl_3)$;$CH_2CHGeCl_3CH_2$; and $Ar=-C_6H_4$;$-(o.CH_3C_6H_4)_2$;$-o.CH_3OC_6H_4)_2$;$-(o.CH_3C_6H_4)$ were prepared by a polycondensation reaction of polyamide containing a pendant trichlorogermyl group and terminal acid chloride $Cl(-CO-R'-CO-NH-Ar-NH-CO-R'-CO-)_xCl$ with aminopropyl-terminated polydimethylsiloxane $H_2N(CH_2)_3(Me_2SiO)_y-(CH_2)_3NH_2]$, (PDMS). These polymers were characterized by elemental analysis, $T_g$, FT-IR, $^1H$-NMR, solid state $^{13}C$-NMR, and molecular weight determination. The thermal stability of these copolymers was examined using thermal analysis techniques, such as TGA and DSC. Their molecular weights as determined by laser light scattering technique ranged $5.13{\times}10^5$ to $331{\times}10^5\;g/mol$. These polymers display their $T_g$ in the range of 337 to $393^{\circ}C$ with an average decomposition temperature at $582^{\circ}C$.

  • The geometrical parameters, total and relative energies, vibrational frequencies, the HOMO-LUMO energy gap, and reorganization energies for the neutral molecule, anion, and cation of $C_{16}H_{16}O_3$ have been determined using density functional method (DFT). The highest level of theory employed in this study is $B3LYP/6-311G^{**}$. Harmonic vibrational frequencies were determined at the $B3LYP/6-311G^{**}$ level of theory. All positive vibrational frequencies were obtained to confirm minimum structures. The HOMO-LUMO energy gap and reorganization energies were calculated to predict the charge transport property of liquid-crystal.

  • In this letter. we report on the investigation of Ti. Pt/Si/Ti Ohmic contacts to p-type 4H-SiC. The contacts were formed by a 2-step vacuum annealing at $500^{\circ}C$ for 1h. $950^{\circ}C$ for 10 min respectively. The contact resistances were measured using the transmission line model method. which resulted in specific contact resistivities in the $3.5{\times}10^{-3}$ and $6.2{\times}10^{-4}ohm/cm^{2}$, and the physical properties of the contacts were examined using x-ray diffraction. microscopy. AES(auger electron spectroscopy). AES analysis has shown that, at this anneal temperature, there was a intermixing of the Ti and Si. migration of into SiC. Overlayer of Pt had the effect of decreasing the specific contact resistivity and improving the surface morphology of the annealed contact.

Effects of Temperature, Soil Moisture, Soil pH and Light on Root Gall Development of Chinese Cabbage by Plasmodiophora brassicae (배추무사마귀병 뿌리혹의 형성에 미치는 온도, 토양수분, 토양 pH, 광의 영향)

  • Development of root galls of clubroot disease on Chinese cabbage seedlings was first observed 17days after inoculation of Plasmodiophora brassicae at $25^{\circ}C$ 4-11days earlier than at 5, 20, 3$0^{\circ}C$ and 35$^{\circ}C$. Subsequent enlargement of root galls was also fastest at $25^{\circ}C$ and 2$0^{\circ}C$ but delayed at 15$^{\circ}C$ and 3$0^{\circ}C$ or above. Chinese cabbage seedlings with root gall formation showed reduction in number of leaves above ground fresh weight and amount of root hairs but increase in root weight, Root galls development was highest at soil moisture level of 80% of maximum soil moisture capacity than at 60% and 100%. Optimum soil pH for root gall development was pH 6 although root galls were formed at a range of pH 5 to 8. Period of light illumination also affected root gall development with the greatest gall development at 12hr/12hr in light/dark period and the least at 8hr/16hr. Site of root gall formation and gall shape did not differ greatly among treatments of temperature soil moisture pH and light experiments.
