의치환자에서 Candida albicans분리와 amphotericin B 및 miconazole에 시험관내 감수성 (Isolation of Candida albicans from Denture Patients and in Vitro Activities of Amphotericin B and Miconzole)

  • Candida albicans is now well recognized among the denture stomatitis patients. The broth macrodilution test is the most widely used technique for antifungal susceptibility testing. The purpose of this study was to determine the C. albicans carrier rate of the denture patients in Iksan, chonbuk. To determine the C. albicans carrier rate of denture patients, culture were made from 227 sample taken in Iksan, Chonbuk during July 1997 to August 1997. Also activities of amphotericin B and miconzole against isolates of denture patients of C. albicans were tested by broth macrodilution test using RPMI medium 1640. The results were as follows : First C. ablicans was isolated from 6.6% of denture patients samples and the frequency of isolation fo C. albicans was highest(50%) in the age group of 71-year-old to 80-year-old denture patients. Second, against C. albicans, the MIC range of amphotericn B was $0.06{\sim}0.25{\mu}g/ml$. MIC50 and MIC90 were $0.13{\mu}g/ml$ and $0.25{\mu}g/ml$, respectively. Third, the MIC range of miconazole was $10-{\ge}20{\mu}g/ml$ and MIC50 and MIC90 were $20{\mu}g/ml$ and ${\ge}20{\mu}g/ml$, respectively. It was concluded from this study that C. albicans acrriages from healthy denture individuals only over 60-year-old ages were isolated, they remain susceptible to amphotericin B and not rarely resistant to miconzole.

Candid albicans 바이오필름에 대한 전기분해 수소수의 항진균 효과 (Antifungal effect of electrolyzed hydrogen water on Candida albicans biofilm)

  • 목적: Candida albicans는 면역력이 약한 환자에게서 점액질환을 야기하며, 의치성 구내염과 밀접하게 관련되어 있다. 전기 분해 수소수는 금속 전극을 통해서 전기를 흘려 전기분해 한 물이며, 구강세균에 대해서 항균효과를 보였다. 본 연구에서는 칸디다 바이오필름에 대한 전기 분해 수소수의 항진균효과를 조사하였다. 연구 재료 및 방법: C. albicans는 발아세포 및 균사형태를 형성시키기 위해서 사부로포도당 액체배지 및 F-12 영양 배지를 이용하여 호기상태 및 5% 이산화탄소 환경에서 각각 배양하였다. 여러 금속 전극을 이용하여 수돗물을 전기분해 한후에 발아세포 형태와 균사형태의 칸디다를 전기분해 수소수로 처리하였다. 결과: 사부로포도당배지와 F-12 영양배지를 이용하여 칸디다를 배양하였을 때, 각각 발아세포 형태와 균사 형태를 보였다. 전기 분해 수소수는 발아세포 형태의 C. albicans에 대해서 항진균 효과를 보였다. 또한 칸디다 바이오필름에 대해서도 항진균 효과를 보였다. 여러 종류의 금속 전극을 이용한 전기분해 수소수중에 백금전극을 이용한 전기분해 수소수만 항진균 활성이 있는 것으로 나타났다. 결론: 백금 전기분해 수소수는 구강 칸디다증과 의치관련 구내염을 예방하기 위한 구강 청결제로 사용 가능 할 것이다.

In Vitro Antifungal Activity of Equol against Candida albicans

  • In this study, we demonstrate that equol has fungicidal activities against Candida albicans. The minimum inhibitory and minimum fungicidal concentrations of equol against C. albicans were 516 and $1,032{\mu}M$, respectively. Two separate viability assays found that equol changed the integrity of the C. albicans cell membrane, possibly by formation of membrane lesions. Scanning electron microscopy demonstrated ultrastructural changes.

Computational Evaluation on the Interactions of an Opaque-Phase ABC Transporter Associated with Fluconazole Resistance in Candida albicans, by the Psidium guajava Bio-Active Compounds

  • Objectives: Candida albicans is an opportunistic pathogen that occurs as harmless commensals in the intestine, urogenital tract, and skin. It has been influenced by a variety of host conditions and has now evolved as a resistant strain. The aim of this study was thus detect the fluconazole resistant C. albicans from the root caries specimens and to computationally evaluate the interactions of an opaque-phase ABC transporter protein with the Psidium guajava bio-active compounds. Methods: 20 carious scrapings were collected from patients with root caries and processed for the isolation of C. albicans and was screened for fluconazole resistance. Genomic DNA was extracted and molecular characterization of Cdrp1 and Cdrp2 was done by PCR amplification. P. guajava methanolic extract was checked for the antifungal efficacy against the resistant strain of C. albicans. Further in-silico docking involves retrieval of ABC transporter protein and ligand optimization, molinspiration assessment on drug likeness, docking simulations and visualizations. Results: 65% of the samples showed the presence of C.albicans and 2 strains were fluconazole resistant. Crude methanolic extract of P. guajava was found to be promising against the fluconazole resistant strains of C. albicans. In-silico docking analysis showed that Myricetin was a promising candidate with a high docking score and other drug ligand interaction scores. Conclusion: The current study emphasizes that bioactive compounds from Psidium guajava to be a promising candidate for treating candidiasis in fluconazole resistant strains of C. albicans However, further in-vivo studies have to be implemented for the experimental validation of the same in improving the oral health and hygiene.

SeO2의 병원성 진균 Candida albicans에 대한 항균 활성 (Antifungal Activity of SeO2 against Pathogenic Fungus Candida albicans)

  • This study was carried out to determine the antifungal activity of $SeO_2$ against pathogenic fungus, Candida albicans. Concentration-dependent Inhibitory activity of $SeO_2$ against C. albicans was shown. When $400{\mu}g/disc$ of $SeO_2$ was tested, the inhibition size ranged from 20.7 to 26.8 mm (avg. 23.4 mm). MIC values of $SeO_2$ against cell growth of C. albicans ranged from 50 to $100{\mu}g/mL$ (avg. $70{\mu}g/mL$). These results could be applied to antibiotics-resistant C. albicans and other pathogenic fungi.

Resveratrol Impaired the Morphological Transition of Candida albicans Under Various Hyphae-Inducing Conditions

  • The ability of the human fungal pathogen Candida albicans to undergo the morphological transition from a single yeast form to pseudohyphal and hyphal forms in response to various conditions is known to be important for its virulence. Many studies have shown the pharmacological effects of resveratrol, a phytoalexin polyphenolic compound. In this study, we investigated the antifungal activity of resveratrol against C. albicans. Both yeast-form and mycelial growth of C. albicans were inhibited by resveratrol. In addition, normal filamentation of C. albicans was affected and yeast-to-hypha transition under serum-, pH-, and nutrient-induced hyphal growth conditions was impaired by resveratrol.

Utilization of Ferroproteins by Candida albicans during Candidastasis by Apotransferrin

  • Many reports have stated that some of the pathogenic bacteria can obtain iron from ferroproteins, such as cytochrome C, ferritin, hemin, hemoglobin, and myoglobin. These reports prompted us to determine if an opportunistic pathogenic fungus, Candida albicans, can utilize ferroproteins to circumvent the iron-regulatory effect of transferrin. The following assays were carried out to measure in vitro growth stimulation by the ferroproteins: as an initial step, C. albicans was cultured in iron-free (pretreated with apotransferrin for 24h) culture medium. Once Candida albicans yeast cell growth reached stasis from iron starvation, individual ferroproteins were added to the culture media. Results showed that hemin, hemoglobin, and myoglobin supported a partial growth recovery. Additional studies with haptoglobin, a serum protein that interacts with the globin moiety of certain ferroproteins, established that C. albicans could obtain iron from the haptoglobin-ferroprotein complexes. These data indicate that the heme part of the ferroproteins is the source of iron. This implies that heme oxygenase, CaHMX1 might be involved in bringing about dissociation of heme-containing protein for iron-acquisition. In addition, anticandidal activity of transferrin takes place not only by the process of iron regulation, but also by direct interaction with the yeast cells.

Aucklandia lappa Causes Cell Wall Damage in Candida albicans by Reducing Chitin and (1,3)-β-D-Glucan

  • The fungal cell wall is a major target of antifungals. In this study, we report the antifungal activity of an ethanol extract from Aucklandia lappa against Candida albicans. We found that the extract caused cell wall injury by decreasing chitin synthesis or assembly and (1,3)-β-D-glucan synthesis. A sorbitol protection assay demonstrated that the minimum inhibitory concentration (MIC) of the A. lappa extract against C. albicans cells increased eight-fold from 0.78 to 6.24 mg/ml in 72 h. Cell aggregates, which indicate damage to the cell wall or membrane, were commonly observed in the A. lappatreated C. albicans cells through microscopic analysis. In addition, the relative fluorescence intensities of the C. albicans cells incubated with the A. lappa extract for 3, 5, and 6 h were 92.1, 84.6, and 79.8%, respectively, compared to the controls, estimated by Calcofluor White binding assay. This result indicates that chitin content was reduced by the A. lappa treatment. Furthermore, synthesis of (1,3)-β-D-glucan polymers was inhibited to 84.3, 79.7, and 70.2% of that of the control treatment following incubation of C. albicans microsomes with the A. lappa extract at a final concentration equal to its MIC, 2× MIC, and 4× MIC, respectively. These findings suggest that the A. lappa ethanol extract may aid the development of a new antifungal to successfully control Candidaassociated disease.

Synergistic Antifungal Activity of Magnoliae Cortex and Syzyii Flos against Candida albicans

  • Candida albicans is a dermal fungus of the human body that is known to cause oral candidiasis, vaginal candidiasis, and bloodstream infections in immunocompromised people or in certain environmental conditions. As cases of strains resistant to antifungal agents in C. albicans have been reported, studies using plant materials as safe antifungal agents are being actively conducted. In this study, a total of 17 edible plant extracts showed antifungal activity against C. albicans as a result of evaluating a 280-plant extract library using paper disk diffusion method. Among them, the four extracts with the strongest antifungal activity (Cinnamomi Cortex, Cinnamomi Ramulus, Magnoliae Cortex, and Syzygii Flos) were selected and evaluated for synergistic antifungal activity against C. albicans. The combination of Magnoliae Cortex and Syzygii Flos showed a synergistic activity. The antifungal activity was evaluated based on the concentrations of magnolol and eugenol, the respective components of Magnoliae Cortex and Syzygii Flos. Magnolol and eugenol showed synergistic antifungal activities at the concentration ratio of 1:25 - 1:61. The antifungal activity of these two compounds contributes 28 to 48% to the synergistic antifungal activity of the combination of Magnoliae Cortex and Syzygii Flos extract. In this study, we propose that a combination of Magnoliae Cortex and Syzygii Flos can effectively inhibit the growth of C. albicans and that magnolol and eugenol are the responsible inhibitory compounds.

수종의 점막조정제간에 Candida albicans의 성장 양상 비교 (COMPARISON OF THE GROWTH OF CANDIDA ALBICANS AMONG THE TISSUE CONDITIONERS)

  • The use of tissue conditioners has come into favor for preprosthetic treatment and the treatment of Denture stomatitis, but the major problem in the use of the tissue conditioner is the growth of C.albicans. To compare the growth of C.albicans according to the type and the wearing period of the tissue conditioner, three commercial tissue conditioners were relined to intraoral plates which were delivered to 14 smoking, 12 nonsmoking healthy men. Cultures were made from the conditioner surface at 2, 7, 14, 21 days after after intraoral placement. The frequency of positive culture and colony counts of C.albicans were measured by imprint culture technique. The following results were achieved : 1. The frequency of positive cluture had increased significantly for all materials used. 2. The frequency of positive culture had incresed significantly around 2,14 days for day for COE-Comfort, SR-Ivoseal. 3. There were no significant difference in the colony counts of C.albicans among the mate rials used. 4. There were an over-all increase with the wearing period of tissue conditioners and significant increase around 2, 7, 14 days for COE-Comfort, and 2, 14 days for SR-Ivoseal, viscogel in colony counts. 5. Smoking had no effect on the growth of C.albicans. That is, there were no difference among the materials used in the growth of C.albicans. In the clinical application of tissue conditioners, we should avoid a long term use of it, but in inevitable cases, special disinfection procedures should be considered.
