Synthetic MR을 이용한 정량적 평가에서 Coil channel 수 변화에 따른 재현성 평가 (Evaluation of Reproducibility According to Variation of Coil Channel in Quantitative Evaluation Using Synthetic MR)

  • 최신 영상 기법인 MAGiC sequence의 RF coil channel 수 변화에 따른 T1, T2 이완 시간 및 T1, T2 이완율을 측정하여 분석하였고 그에 따른 재현성을 평가하였다. Channel 수가 각각 (1, 8, 16, 32)channel 인 RF coil에 조영제 phantom(1.0, 0.6, 0.2, 0mM)을 위치시키고 MAGiC sequence를 이용하여 T1, T2, R1, R2 map을 획득하였다. 그리고 각각의 channel 수와 농도별로 T1, T2, R1, R2 value 그리고 relaxation rate를 측정하였고 이를 기반으로 각 농도에 따른 Relaxivity를 구하였다. 각각의 coil에서 T1, T2, R1, R2 value를 측정한 결과 T1, R1 value는 coil간의 유의한 차이가 없었다(p>0.05). 하지만 T2, R2 value는 유의한 차이가 있는 것으로 나타났다(p<0.05). 사후분석 결과에서는 T2 value의 경우 1 channel coil에서 측정한 결과가 8, 16, 32 channel coil에서 측정한 결과와 유의한 차이가 나타났고(p<0.05) 8, 16, 32 channel coil간의 차이는 나타나지 않았다(p>0.05). R2 value의 경우도 마찬가지로 1 channel coil에서 측정한 결과가 8, 16, 32 channel coil에서 측정한 결과와 유의한 차이가 나타났으며 또한 8 channel coil에서 측정한 결과와 16 channel coil에서 측정한 결과가 유의한 차이가 나타났다(p<0.05). 결론적으로 T1, R1 value는 coil의 channel 수 변화에 따른 차이가 크지 않지만 T2, R2 value는 큰 차이를 나타냈다. 따라서 MAGiC sequence를 이용하여 T2, R2 value를 정량적으로 측정할 경우 재현성을 위해 동일한 channel 수의 coil을 사용해야 할 것으로 사료된다.

개심수술 102례 의 임상적 고찰 (Clinical Analysis of 102 Cases of Open Heart Surgery)

  • A total of 102 patients who had an Open Heart Surgery from April 1976 to July 1981 were reviewed. 55 paeitnts were congenital heart disease and 47 patients were acquired heart disease. Among SS patients of congenital heart disease, 18 T 0 F, 18 V S D, 8 A S D, and each one case of l\ulcorner 0 R V, Truncus arteriosus, Ebstein anomaly, Single ventricle, P D A, P 5, A S D + P 5, E C D, V 5 D + P D A, A - P window, D C R V were noted respectively. In 47 patients of acquired heart disease and one Ebstein patient, 46 prosthetic values were implanted: 17 had M V R, 4 had A V R, 2 had M V R + A V R, and 4 had M V R + T V R and one T V R. The operative mortality was 8.S% in acquired heart disease and 17% in congenital heart disease. The follow up period was between 6 months and 6 years. There were 3 cases of late mortality in acquired heart disease and one case in congenital heart disease.

미생물을 이용한 트립신 과대 생산 연구 - Streptomyces용 숙주-벡터계를 이용한 트립신 유전자의 대량발현 최적화 - (Overproduction of Bacterial Trypsin in Streptomyces - Optimization for Streptomyces griseus Trypsin Production by Recombinant Streptomyces)

  • Streptomyces griseus trypsin (SGT)을 코드하는 sprT 유전자와 그 하류에 존재하는 두 개의 조절 유전자 rsgtR1 및 sgtR2를 동시에 갖고 있는 재조합 벡터 pWHM3-TR1R2를 S. lividans TK24 및 S. griseus IFO 13350에 도입하여, 트립신의 생산성을 더욱 증대시킬 수 있는 배지를 조사하였다. S. lividans TK24/pWHM3-TR1R2의 경우 배양 5일을 기준으로 R2YE에서 가장 높은 생산성(0.74 unit/mL)을 나타냈고, C5/L. (0.66 unit/mL), Livid (0.08 unit/mL), NDSK(0.06 unit/mL) 순으로 나타났다 S. griseus IFO 13350/pWHM3-TR1R2의 경우에는 전반적으로 배양 7일에 트립신 활성이 가장 높았으며, C5/L (1.518 unit/mL), R2YE(1.284 unit/mL), NDSK (0.932 unit/mL), Livid (0.295 unit/mL) 순으로 나타났다. S. griseus IFO 13350/pWHM3-TR1R2를 C5/L 배지에서 7일간 배양한 배양액으로부터 $25%{\sim}60%$ ammonium sulfate 침전, CM-sepharose 및 Sp-sepharose column chromatography를 통하여 트립신을 고순도로 정제할 수 있었다. 최종 purification fold는 6.5배, 순수 정제된 트립신의 specific activity는 69,252 unit/mg, 회수율은 1.4%이었다.

Allelic variation of melanocortin-1 receptor locus in Saudi indigenous sheep exhibiting different color coats

  • Objective: This study was designed to characterize the DNA polymorphisms of the melanocortin-1 receptor (MC1R) gene in indigenous Saudi Arabian sheep breeds exhibiting different color coats, along with individuals of the Sawaknee breed, an exotic sheep imported from Sudan. Methods: The complete coding region of MC1R gene including parts of 3' and 5' untranslated regions was amplified and sequenced from three the indigenous Saudi sheep; Najdi (generally black, n = 41), Naeimi (generally white with brown faces, n = 36) and Herri (generally white, n = 18), in addition to 13 Sawaknee sheep. Results: Five single nucleotide polymorphisms (SNPs) were detected in the MC1R gene: two led to nonsynonymous mutations (c.218 T>A, p.73 Met>Lys and c.361 G>A, p.121 Asp>Asn) and three led to synonymous mutations (c.429 C>T, p.143 Tyr>Tyr; c.600 T>G, p.200 Leu>Leu, and c.735 C>T, p.245 Ile>Ile). Based on these five SNPs, eight haplotypes representing MC1R $E^d$ and $E^+$ alleles were identified among the studied sheep breeds. The most common haplotype (H3) of the dominant $E^d$ allele was associated with either black or brown coat color in Najdi and Sawaknee sheep, respectively. Two other haplotypes (H6 and H7) of $E^d$ allele, with only the nonsynonymous mutation A218T, were detected for the first time in Saudi indigenous sheep. Conclusion: In addition to investigating the MC1R allelic variation in Saudi indigenous sheep populations, the present study supports the assumption that the two independent nonsynonymous Met73Lys and Asp121Asn mutations in MC1R gene are associated with black or red coat colors in sheep breeds.

ON CLEAN AND NIL CLEAN ELEMENTS IN SKEW T.U.P. MONOID RINGS

  • Let R be an associative ring with identity, M a t.u.p. monoid with only one unit and ${\omega}:M{\rightarrow}End(R)$ a monoid homomorphism. Let R be a reversible, M-compatible ring and ${\alpha}=a_1g_1+{\cdots}+a_ng_n$ a non-zero element in skew monoid ring $R{\ast}M$. It is proved that if there exists a non-zero element ${\beta}=b_1h_1+{\cdots}+b_mh_m$ in $R{\ast}M$ with ${\alpha}{\beta}=c$ is a constant, then there exist $1{\leq}i_0{\leq}n$, $1{\leq}j_0{\leq}m$ such that $g_{i_0}=e=h_{j_0}$ and $a_{i_0}b_{j_0}=c$ and there exist elements a, $0{\neq}r$ in R with ${\alpha}r=ca$. As a consequence, it is proved that ${\alpha}{\in}R*M$ is unit if and only if there exists $1{\leq}i_0{\leq}n$ such that $g_{i_0}=e$, $a_{i_0}$ is unit and aj is nilpotent for each $j{\neq}i_0$, where R is a reversible or right duo ring. Furthermore, we determine the relation between clean and nil clean elements of R and those elements in skew monoid ring $R{\ast}M$, where R is a reversible or right duo ring.

녹농균(Pseudomonas aeruginosa)의 쿼럼 센싱 수용체인 QscR의 활성에 영향을 미치는 아미노산 잔기 분석 (Analysis of Amino Acid Residues Affecting the Activity of QscR, a Quorum Sensing Receptor of Pseudomonas aeruginosa)

  • 그람 음성균인 녹농균(Pseudomonas aeruginosa)은 다양한 환경에 존재하는 기회감염성 병원균으로, 병원성의 발현에 쿼럼센싱(QS) 기전이 중요한 역할을 담당한다. 녹농균의 여러 QS 신호물질 수용체들 중 하나인 QscR은 다른 QS 수용체들과는 구분되는 특별한 특성들을 가진다. 본 연구에서는 이러한 특성들 중 특히 넓은 신호물질 특이성을 QscR에 부여해 주는 아미노산 잔기가 무엇인지 알아보기 위해, QscR의 72번째 threonine, 132번째 arginine, 140번째 threonine 잔기가 각각 isoleucine, methionine, isoleucine 잔기로 치환된 돌연변이 QscR들($QscR_{T72I}$, $QscR_{R132M}$, $QscR_{T140I}$)을 제조하였다. 이들의 활성을 측정해 보았을 때 $QscR_{R132M}$은 N-3-oxododecanoyl homoserine lactone (3OC12-HSL)에 대한 반응성이 사라졌고, $QscR_{T72I}$$QscR_{T140I}$는 민감성이 많이 감소하기는 하였으나 여전히 3OC12-HSL에 대한 반응성을 가지고 있었다. 이들 돌연변이 QscR들에 다양한 구조의 acyl-HSL을 처리해 보았을 때, $QscR_{T72I}$$QscR_{T140I}$는 야생형 QscR처럼 자기 자신의 신호물질인 3OC12-HSL 보다 N-decanoyl HSL (C10-HSL)이나 N-dodecanoyl HSL (C12-HSL)처럼 10개 혹은 12개의 탄소 사슬을 가지면서 3번째 탄소에 oxo-moiety가 없는 acyl-HSL에 대해 더 높은 반응성을 보였으며, $QscR_{R132M}$은 3OC12-HSL 뿐만 아니라 본 연구에서 사용된 어떤 acyl-HSL에도 반응성을 보이지 않았다. 또한 $QscR_{T72I}$$QscR_{T140I}$는 QscR 억제제인 5f에 의해 야생형 QscR과 비슷한 수준으로 활성이 억제되었다. 이러한 결과들은 130번째 arginine의 경우 QscR의 활성과 acyl-HSL들과의 결합에 중요한 역할을 하는 반면, 72번째와 140번째 threonine들의 경우 QscR의 활성에는 중요하지만, 다른 구조의 acyl-HSL들에 대한 선택적 결합이나, 경쟁적 억제자들의 결합 간섭에는 영향을 주지 않음을 시사하는 것이다.

Effects of Temperature on the Pharmacokinetics of Ciprofloxacin in the Cultured Black Rockfish (Sebastes schlegeli) and Olive Flounders (Paralichthys olivaceus)

  • Temperature-dependent pharmacokinetics of ciprofloxacin (CIP) was studied in the cultured olive flounders, Paralichthys olivaceus, and black rockfish, Sebastes schlegeli using high performance liquid chromatography (HPLC) originally developed for quinolone determination from livestock. Pharmacokinetics of CIP was apparently affected by ambient water temperature. In a two-compartment model for flounders after oral dosage of 20 mg/kg, $K_{01},\;at\;13^{\circ}C$ and $23^{\circ}C$ were 4.18 and 1.20/hr, respectively. The $K_{10},\;T_{max}\;and\;C_{max}\;at\;13^{\circ}C$ were 5.574/hr, l4.37${\mu}g/mL\;and\;3.15{\mu}g/mL,$ respectively. The corresponding values at $23^{\circ}C$ were l2.84/hr, 15.39${\mu}g/mL\;and\;6.38{\mu}g/mL$, respectively. The AUC, $T_{1/2} (\alpha)\;and\;T_{1/2}\;(\beta)$ were 278.23 ${\mu}g \cdot hr/mL$, 0.24hr and 47.02hr at $13^{\circ}C$ and 3l7.8l${\mu}g \cdot hr/mL$, 0.30 hrs and 60.78hrs at $23^{\circ}C$ for the flounder, respectively. Similar CIP pharmacokinetics were revealed in the black rockfish after oral dosage of 20 mg/kg under the two water temperature regimes. These pharmacokinetical results have some implication in the optimal usage of recently introduced antibacterials in the farmed fish, which were primarily adapted for poultry and mammalian species.

For large problems H and the constraint matrix A are assumed to be sparse.
