Effects of Jeju Citrus unshiu Peel Extracts Before and After Bioconversion with Cytolase on Anti-Inflammatory Activity in RAW264.7 Cells (면역세포에서 Bioconversion 전후 제주 감귤 과피 추출물의 항염증 효과)

  • Citrus and its peels, which are by-products from juice and/or jam processing, have long been used in Asian folk medicine. Citrus peels show an abundant variety of flavanones, and these flavanones have glycone and aglycone forms. Aglycones are more potent than glycones with a variety of physiological functions since aglycone absorption is more efficient than glycones. Bioconversion with cytolase converted narirutin and naringin into naringenin and hesperidin into hesperetin. Therefore, this study aimed to investigate the anti-oxidant and anti-inflammatory effects of bioconversion of Citrus unshiu (CU) peel extracts with cytolase (CU-C) in RAW264.7 cells. HPLC chromatograms showed that CU and CU-C had 23.42% and 29.39% total flavonoids, respectively. There was substantial bioconversion of narirutin to naringenin and of hesperidin to hesperetin. All citrus peel extracts showed DPPH scavenging activities in a dose-dependent manner, and CU-C was more potent than intact CU. RAW264.7 cells were pre-treated with $0{\sim}500{\mu}g/mL$ of citrus peel extracts for 4 h and then stimulated by $1{\mu}g/mL$ of lipopolysaccharide (LPS) for 8 h. All citrus peel extracts showed decreased mRNA levels and protein expression of LPS-induced inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in a dose-dependent manner. Especially, CU-C markedly inhibited mRNA and protein expression of iNOS and COX-2 compared to intact citrus peel extracts. All citrus peel extracts showed decreased NO production by iNOS activity. This result suggests that bioconversion of citrus peel extracts with cytolase may provide potent functional food materials for prevention of chronic diseases attributable to oxidation and inflammation by boosting the anti-inflammatory effects of citrus peels.

  • To further our understanding of sugar accumulation in 'Shiranuhi' mandarin [(C. unshiu ${\times}$ C. sinensis) ${\times}$ C. reticulate], we investigated the patterns of sugar uptake in juice sacs exposed to different concentrations of sucrose, fructose and glucose in vitro. Data was also collected on the change in weight and shape of the in vitro juice sacs over time. Soluble solids content, sugar content and acidity content were highest at 20% sucrose, fructose and glucose solution content; while fructose content was highest at 5% sucrose concentration. Furthermore, the juice sac's fresh weight was highest at 5% sucrose and lowest at 20% fructose content. The shape of the juice sacs also differed in different sugar concentration and type. Overall, sucrose, fructose and glucose content in juice sacs increased with the sugar concentration. These results suggest that sugar translocation into juice sacs is actively induced by high sugar concentration in the medium. Thus, it can be concluded that sugar and acid accumulation in juice sacs increased with sugar concentration in vitro culture.

Development of a SCAR Marker Linked to Male Fertility Traits in 'Jinkyool' (Citrus sunki) ('진귤' (Citrus sunki) 의 웅성가임 연관 SCAR 마커 개발)

  • In Citrus, an $F_1$ segregation population of 150 plants was constructed from a cross between 'Kiyomi' (C. unshiu ${\times}$ C. sinensis) carrying the male sterility trait and 'Jinkyool' (C. sunki). Sequence-related amplification polymorphism (SRAP) combined with bulked segregant analysis was used to develop markers linked to male fertility. In the $F_1$ population, 66 out of 150 seedlings had aborted anthers and the ratio of male sterile plants to fertile plants in the progenies matched the expected Mendelian segregation ratio of 1:1 ($x^2$ =2.16 at p=0.05). From the profiling of the 197 SRAP primer sets, three SRAP primer sets (F4/R27, F39/R60, and F15/R37) that were closely linked to the target trait were identified and successfully converted into a sequence characterized amplified region (SCAR) marker for selection of male fertility in citrus. The SCAR marker, using the pMS 33U/pMS 1462L primer set specifically, produced a single 1.4-Kb fragment that was linked to male fertility. Our results suggested that this SCAR marker can be useful for marker-assisted selection of male sterile individuals in breeding $F_1$ progenies in Citrus.

RT-PCR Detection of Citrus Tristeza Virus form Early Satsuma Nandarin and Yuzu in Cheju Island

  • Citrus tristeza virus (CTV) was identified form CTV-infected early satsuma mandarin (Citus unshiu) and yuzu (C.junos) by RT-PCR. The total RNAs were isolated from citrus bark and seaf tissues infected with CTV and reverse transcription was followed with primers designed for amplifying CTV coat protein gene. DNA fragments 738 bp were amplified by RT-PCR and these products were colned for sequence analysis. Based on the sequence analysis, this PCR product has 97% sequence homology to CTV (T-385) CP gene isolated from USA. RT-PCR assay for CTV detection was more sensitivity than ELISA assay which was done with anti-CTV CP antibody. This is the frist report about CTV identification in Cheju island Korea.

  • The flavor constituents of Korean Milgam were extracted with a nitrogen gas stream under partial vacuum and identified by gas liquid chromatography. By employing the extraction coefficient, it was possible to determine the concentration of components in Milgam as well as in the extracts. Among 53 GLC peaks, 26 components were identified. Ethanol was the most abundant component (140ppm), followed by limonene (120ppm). These two were the most important flavor constituents.

Composition and Antimicrobial Activities of Essential Oils in the Peel of Citrus Fruits

  • Volatile components of the essential oils of Satsuma mandarin (C. unshiu), Dangyuza (C. grandis), Yuza (C. junos), Byungkyul (C. playtymamma), Jinkyul (C. sunki), and Hakyul (C. natsudaidai) grown in Jeju Island were isolated from the fruit peels by hydro distillation and determined by GC-MS. GC-MS analysis identified 58 compounds, with main components being d-limonene $(64.01{\sim}79.34%),\;{\beta}-myrcene\;(3.01{\sim}26.53%),\;{\gamma}-terpinene\;(0.11{\sim}12.88%),\;{\beta}-pinene\;(0.78{\sim}4.74%),\;and\;{\alpha}-pinene\;(1.01{\sim}2.55%)$. Differences in compositions and contents of the essential oils were observed among citrus varieties. Effects of citrus oils on growth inhibitions of Escherchia coli, Staphyllococcus epidermidis, and Candida albicans were investigated using disc diffusion assay and minimal inhibitory concentration (MIC) assay. The essential oils inhibited growths of the test organisms, exhibiting higher levels of activity against Gram-positive S. epidermidis (MIC values $0.04{\sim}0.17mg/mL$), whereas Gram-negative E. coli was moderately resistant (MIC values $1.66{\sim}20.30mg/mL$). MIC of citrus essential oils ranged from $0.82{\sim}23.69mg/mL$ against C. albicans. The essential oils obtained from C. sunki, C. grandis, and C. playtymamma showed the highest antimicrobial activities against S. epidermidis and C. albicans, indicating their potential as natural antimicrobial agents.

Effect of Foliar Application of GA3 on the Flower Bud Formation and Fruit Quality of Satsuma Mandarine (C. unshiu Marc. cv. Miyagawa) (지베렐린 엽면살포가 '궁천조생' 감귤의 착화와 과실품질에 미치는 영향)

  • BACKGROUND: Control of alternate bearing satsuma mandarin in Jeju is very important to maintain the optimum price of fruit and get the sustainable income of farmers. Unlike orange, Satsuma mandarin is well known to sensitive on alternate bearing. We carried out the experiment to know the effect of foliar application of $GA_3$ on the flowering and fruit quality of satsuma mandarin (C. unshiu Marc. cv. Miyagawa). METHODS AND RESULTS: To experiment, the treatments consist of control, different concentration of $GA_3$ (25, 50 and 100 mg/L), machine oil emulsion 100 times and mixture of various concentration of $GA_3$ (25 and 50 mg/L) with machine oil emulsion 100 times which it was applied on 15 year-old Miyagawa satsuma mandarin at December 29, 2011. Foliar application of $GA_3$ in winter reduced the flowering of satsuma mandarin. Flower-leaf ratio was significantly reduced at 100 mg/L $GA_3$, while no differences observed in low concentration of $GA_3$ (25 and 50 mg/L). However, it was significantly decreased to 0.19 in application of $GA_3$ 25 and 50 mg/L with machine oil emulsion 100 times mixture. Number of leaves per fruit was significantly increased as foliar application of $GA_3$ also it reduced the fruits remarkably. Soluble solid contents and Hunter's a of peel color ratio showed no difference among $GA_3$ single treatments, but it was reduced in $GA_3$ 25 and 50 mg/L with machine oil emulsion 100 times mixtures significantly. From the results, it has been found that higher $GA_3$ concentration can reduce the number of flowers on the alternate bearing of satsuma mandarin. However, it was found that lower concentration of $GA_3$ with machine oil emulsion mixture 100 times can reduce flowering. CONCLUSION(S): The foliar application of $GA_3$ (100 mg/L) can alleviate alternate beraring. Also, mixture of lower concentration of $GA_3$ with machine oil emulsion 100 times can retard flowering more significantly while it needs further confirmation.

  • Pectin, hesperidin and naringin were extracted from hot air-dried peel ($60^{\circ}C$, 1 hr and air velocity 160 fpm) of citrus produced in Korea in order to see the amount of each component contained in the peel. Pectin was extracted by three different methods and the quality and contents of the pectins were determined respectively. 1. The pectin yield by the total pectic substance method was the highest (26.0% for unshiu (U) and 28.5% for natsudaidai (N) expectedly and the soluble pectic substance method the least (13.5%(U) and 15.6% (N)) The yield by method III (extraction by water at pH 1.5 followed by isopropanol precipitation) was intermediate (18.1% (U) and 20.8%(N)). Anhydrouronic acid (AUA) content was the highest (92.0% (U) and 90.3%(N)) in those by method III. The AUA contents of the other pectins were 80.0% for soluble pectin (for both U and N), 71.6% for the commercial pectin (Sunkist Groups Inc., U.S.A.), 58.0%(N) and 63.4%(U) for total pectic substance. 2. The methoxyl content of total pectic substance was the lowest (4.81%(U) and 4.88%(N)). However, there was no significant difference in methoxyl content among the rest which were found to have low levels(5.27-7.20%). 3. The pectin by method III gave the highest jelly strength. The commercial pectin, soluble pectice substance and total pectic substance were next in order. 4. The hesperidin content of unshiu was 5.07% (dry basis) and the naringin content of natsudaidai 3.03% (dry basis).

Effect of Foliar Application of Gibberellic Acid and Machine Oil Emulsion Mixture on Flowering and Fruit Quality of Satsuma Mandarin (C. unshiu Marc. cv. Miyagawa) Cultivated on Open Fields

  • In satsuma mandarin (C. unshiu Marc. cv. Miyagawa), alternate bearing is frequently occurred. It is very difficult to control and maintain the optimum production because satsuma mandarin is a very sensitive citrus cultivar. We carried out the experiment to find out the effects of foliar application of gibberellic acid ($GA_3$) and machine oil emulsion mixture on flowering and fruit quality of satsuma mandarin grown on open field. The treatments were composed of control (no application), $GA_3$ (25, 50 and $100mg{\cdot}L^{-1}$), machine oil emulsion 100 times and mixture of $GA_3$ (25 and $50mg{\cdot}L^{-1}$) with machine oil emulsion 100 times on 16 year-old 'Miyagawa' satsuma mandarin on December 30, 2012. Foliar application of $GA_3$ during winter season led to a reduction of the flowering in the following year. $GA_3$ decreased flowering in dose dependant manner. However, application of $GA_3$ and machine oil emulsion (100 times-diluted mixture) inhibited effectively the flowering compared to $GA_3$ only. Also, flower-leaf ratio was significantly decreased by the application of $GA_3$ 25 (0.54) and $50(0.41)mg{\cdot}L^{-1}$ with machine oil emulsion mixture in comparison to $GA_3$ 25 (0.86) and $50(0.60)mg{\cdot}L^{-1}$ only. The number of leaves per fruit increased in application of $GA_3$ and machine oil emulsion mixture as concentration of $GA_3$ increased. Fruit weight and sugar-acid ratio also increased in comparison to the control whereas fruit size, soluble solid content and acidity remained unchanged. From the results, it was suggested that $GA_3$ application can reduce the number of flowers on the alternate bearing of satsuma mandarin. Moreover, application of $GA_3$ (25 and $50mg{\cdot}L^{-1}$) with machine oil emulsion 100 times mixture can reduce the flowering effectively without altering fruit maturity and soluble solid contents. Therefore, it can be concluded that the application of $GA_3$ 25 and $50mg{\cdot}L^{-1}$ with machine oil emulsion 100 times might be useful in controlling alternate bearing in satsuma mandarin.

Physicochemical Properties of Hallabong Tangor(Citrus Kiyomi ${\times}$ ponkan) Cultivated with Heating (가온재배한 한라봉 감귤의 성분분석)

  • Physicochemical properties of Hallabong Tangor(Citrus Kiyomi ${\times}$ ponkan) cultivated in Heated greenhouse were investigated. Moisture contents of Hallabong and M16A (a variant species of Hallabong) were 87.42% and 88.12% total sugar were 8.01% and 7.81% and acid content were 1.09% and 0.99% respectively. Vitamin C content of Hallabong was 72.01 mg/100 g that was higher than Citrus unshiu. Potassium content of M16A was 938.33 mg/kg, while Hallabong was 1369.33 mg/kg. The contents of inorganic element in a decreasing order were K > Ca > P > Mg > Na in Hallabong, and K > P > Ca > Mg > Na in Ml6A. Sucrose in Hallabong and M16A were 3.60% and 4.36%, respectively, which is half of total free sugars. Fructose and glucose Hallabong and M16A were 2.22% and 1.90%, 1.94% and 1.65% respectively. Citric acid in Hallabong and M16A was 82.32% and 69.88%, respectively among total organic acids. The content of malic acid was higher in M16A, compared to Citrus unshiu. Hesperidin and narirutin were identified main flavonoids.
