삼백초의 종근 저장온도, 용토 및 처리물질에 따른 생육특성 (Growth Effect by Storage Temperature, Soil Type and Treatment Chemical of Saururus chinensis (Lour.) Baill.)

  • Background: The objective of the present study was to investigate the effect of storage conditions on the growth of Saururus chinensis (Lour.) Baill. rootstock. Methods and Results: Rootstocks of S. chinensis were stored in either soil or vermiculite that had been treated with a control treatment, diluted wood vinegar (50 or 100-fold), DF-100 (50-fold), or 1-naphthylacetamide and at $5^{\circ}C$ or $15^{\circ}C$. After 8 weeks, the stored roots were planted in the field, and both plant height and leaf number were observed after transplantation. The greatest number of leaves ($5.60{\pm}0.80$) was produced by roots that had been stored in soil treated with 100 fold dilution of wood vinegar and at $5^{\circ}C$. Meanwhile the maximum plant height ($6.92{\pm}0.78cm$) at 30 d after transplanting was observed for rootstocks that had been stored in soil treated with the 100 fold dilution of wood vinegar and at $15^{\circ}C$, whereas the maximum plant height at 60 d after transplanting ($26.46{\pm}0.71cm$) was observed for rootstocks that had been stored in soil treated with the 100-fold dilution of wood vinegar and at $5^{\circ}C$. Therefore, the storage of rootstocks in soil treated with the 100-fold dilution of wood vinegar and at temperatures at or below $5^{\circ}C$ was most effective, and it can be used to prevent the decay of roots during the postharvest management of S. chinensis rootstocks. Conclusions: The results of the present study indicate that, among the parameters examined, the storage of roots in soil that had been treated with the 100-foil dilution of wood vinegar is the most effective method for improving the growth of S. chinensis.

Stock Characterization of the Fleshy Prawn (Penaeus chinensis) in the Yellow Sea by Intraspecific Sequence Variation of the Cytochrome c Oxidase Subunit I Gene

  • To determine the amount of genetic variation among populations of Penaeus chinensis (Osbeck) in the Yellow Sea, 342 bp region of the mitochondrial cytochrome c oxidase subunit I gene was amplified and sequenced. Six haplotypes, which differ by from one to four nucleotide sustitutions, were detected from 34 individuals of 4 populations examined. Mean sequence divergence between pairs of haplotypes was $0.68\%$. Most individuals from 4 populations were shared by the most common genotype. This genotype was distributed evenly in the Korean and Chinese populations. This result is in accordance with findings observed using RFLPs analysis of mtDNA (Hwang et al., 1997). Therefore, it is suggested that P. chinensis should be treated as one unit stock in the Yellow Sea.

오미자(Schizandra chinensis)의 병원성 미생물에 대한 항균효과 (Antimicrobial Effects of Schizandra chinensis Extract on Pathogenic Microorganism)

  • The growth of pathogens such as Staphylococcus aureus ATCC 29737, Bacillus subtilis KCTC 1021 and Escherichia coli ATCC 11775 was inhibited as much as 1 to 4 log cycles in tryptic soy broth containing 1% of Schizandra chinensis extract but Salmonella tuphimrium ATCC 14028 and Vibrio parahaemolyticus ATCC 17802 were not inhibited. The essential oil of Schizandra chinensis showed stronger inhibitory activity than ethanol extract against above menioned pathogens. The growth of Listeria monocytogenes was not changed in minced pork containing 1% of Schizandra chinensis extract for 12 days at 4$^{\circ}C$. The sensory quality of hamburger patty such as taste, flavor and overall accepability were not decreased by the addition of 1% Schizandra chinensis extract(p<0.05).

오미자(Schizandra chinensis)추출물이 김치 숙성에 미치는 영향

  • Shizandra chinensis(SC) and Pinus regida(PR) showed antimicrobial activity against 3 strains(B-5, D-1, A-1) of lactic acid bacteria(LAB) isolated from kimchi among eight kinds of plant extracts such as Shizandra chinensis, Phellodendron amurense, ornus officinalis, Pinus regida, Allium tuberosum, Machilus thunbergii, Cyperus rotundus and Schizonepeta tenuifloia. The growth of LAB was inhibited apparently in modified MRS broth containing 1% Schizandra chinensis at $35^{\circ}C$. Pinus regida showed weaker inhibitory effect on the growth of isolated LAB than Shizandra chinensis. pH of SC added kimchi did not change greatly compare with control during 25 days of fermentation. Degree of titratable acidity change and ratio of reducing sugar utilization in control were more higher than in SC added kimchi during fermentation. Growth of total bacteria and lactic acid bacteria was inhibited about 1 to 2 $log_10$ cycle by addition of SC extracts during kimchi fermentation for 10 days at $10^{\circ}C$. Fermentation of kimchi was delaved about 5 to 7 days by addition of 1 or 2% of SC. extract, but sensory quality (falvor, taste and overall acceptability) of SC added kimchi was lower than that of control (p>0.05).

사육환경과 먹이종류에 따른 개량조개, Mactra chinensis 유생의 성장과 생존 (Effects of Rearing Condition and Species of Microalgae on Growth and Survival of Larvae of the Sunray Surf Clam, Mactra chinensis)

  • 개량조개의 인공종묘생산을 위한 유생사육의 최적 사육환경과 먹이생물의 종류에 따른 유생의 성장 및 생존을 조사하였다. 수온에 따른 유생의 성장은 $18^{\circ}C$에서 $33^{\circ}C$까지 수온이 높을수록 빨랐으며, $23^{\circ}C$ 이상에서는 사육 14일째 $230{\mu}m$ 이상 성장하였으나 $18^{\circ}C$에서는 $151.1{\mu}m$로 성장이 부진하였다. 염분별 유생사육 시 염분 30 psu에서 일간 성장은 $11.3{\mu}m$로 가장 빨랐고, 생존율도 65.8%로 가장 높았다. 개량조개 유생은 사육수 1 ml 당 10개체 이하의 밀도에서는 성장과 생존율이 양호하였으나 20개체 이상에서는 다소 성장이 늦어 유생의 사육밀도는 1 ml 당 10개체 이하가 가장 효과적이었다. 개량조개 유생의 빠른 성장과 생존율을 높일 수 있는 유생사육 최적 조건은 수온 $23^{\circ}C$, 염분 30-35 psu, 유생 수용밀도 1-10 개체/ml이었다. 유생사육 시 먹이생물은 미세조류 4종 [Isochrysis galbana, Pavlova lutheri, Isochrysis sp. (green), Chlorella ellipsoidea] 을 혼합 공급한 실험구에서 빠른 성장과 생존율을 높일 수 있으며, 먹이생물을 단일종만을 공급시 I. galbana와 Isochrysis sp. (green) 실험구에서 빠른 성장과 생존율을 높일 수 있었다. 따라서 개량조개 유생의 적정먹이는 I. galbana와 Isochrysis sp. (green)를 포함한 2종 이상을 혼합 공급하는 것이 적합할 것으로 판단된다.

Rapid Origin Determination of the Northern Mauxia Shrimp (Acetes chinensis) Based on Allele Specific Polymerase Chain Reaction of Partial Mitochondrial 16S rRNA Gene

  • Acetes chinensis is an economically important shrimp that belongs to the Sergestidae family; following fermentation, A. chinensis' economic value, however, is low in China, and much of the catch in China is exported to Korea at a low price, thus leading to potential false labeling. For this reason, we developed a simple method to identify A. chinensis' origin using allele-specific polymerase chain reaction (PCR). Ten single nucleotide polymorphisms (SNPs) were identified from partial (i.e., 570 bp) DNA sequence analysis of the mitochondrial 16s rRNA gene in 96 Korean and 96 Chinese individual shrimp. Among 10 SNP sites, four sites were observed in populations from both countries, and two sites located in the middle with SNP sites at their 3'-ends were used to design allele-specific primers. Among the eight internal primers, the C220F primer specific to the Chinese A. chinensis population amplified a DNA fragment of 364 bp only from that population. We were able to identify the A. chinensis population origin with 100% accuracy using multiplex PCR performed with two external primers and C220F primers. These results show that the 16S rRNA gene that is generally used for the identification of species can be used for the identification of the origin within species of A. chinensis, which is an important finding for the fair trade of the species between Korea and China.

미나리아재비과 한약재의 항진균성 검증 및 비교 연구 (A Study on the Antifungal Properties of Ranunculaceae Herbal Medicines)

  • 본 연구는 항진균성 천연물 의약품을 개발하기 위한 생물자원의 연구로서 미나리아재비과에 속한 여러 한약재의 항진균활성을 새롭게 검증하고 비교하는 것을 목표로 한다. 한약재에서 진균류 생장을 저해하는 물질을 효과적으로 추출하는 방법을 찾고 추출물들에 포함된 항진균성 유용 물질을 탐색하였다. 위령선, 황련, 백두옹 한약재의 열수 추출 및 유기용매 추출을 진행하여 액체배지에서 항진균활성을 측정하였을 때 클로로포름 분획물에서 효과적인 결과를 얻었다. 또한 단세포성 진균인 Candida albicans, Candida tropi- calis, Candida lusitaniae 그리고 사상성 곰팡이인 Pythium ultimum KACC 40705, Aspergillus fumigatus, Fusarium oxysporum을 대상으로 고체배지에서 disc diffusion 실험으로 진균 성장 저해 실험을 수행하였다. 백두옹 추출물은 Candida spp.에 대해서, 위령선은 사상성 곰팡에 대해서 우수한 항진균활성을 가지고 있다는 것으로 확인했다. 마지막으로 추출물에 존재하는 항진균성 유용물질을 탐색하기 위해 GC-MS 분석을 진행하였다. 연구 결과 미나리아재비과의 한약재에서 항진균활성을 확인할 수 있었으며 위령선에서 thur- berogenone, 황련에서 16-HCDO를 항진균성 후보물질로서 확인하였다. 결론적으로 백두옹, 위령선 및 황련은 다양한 진균류에 대하여 항진균활성을 가지고 있음을 확인하였으며, GC-MS 분석에서 세 한약재 모두 서로 다른 항진균성 후보물질을 가지고 있는 것으로 확인되었다. 이러한 결과는 미나리아재비과 생물자원이 진균류 저항성에 대해 여러 방향으로 진화되었음을 나타낸다.

오미자 리그난 성분의 정량분석 (Quantitative Analysis of Lignans from Fruits of Schizandra chinensis)

  • This study was carried out to establish the quantitative analysis of lignans for the quality evaluation of fruit from Schizandra chinensis. Five lignans, gomisin N, schizandrin, gomisin C, schisantherin C, and gomisin A were isolated from the fruits of S. chinensis and identified by the spectroscopic methods. Quantitative determination of three lignans, schizandrin, gomisin A, gomisin N, was conducted using HPLC. Average contents of three lignans in collected lines were 0.70% of schizandrin, 0.20% of gomisin A, 0.57% of gomisin N, and 1.47% of total lignans.

  • A reversed phase HPLC method was established for quantitative determination of lignans including schizandrin (1), gomisin A (2), deoxyschizandrin (3), gomisin N (4), and wuweizisu C (5) from Schizandra chinensis using a binary gradient of acetonitrile and water as a mobile phase with UV detection at 254nm. In result, contents of schizandrin (1), gomisin A (2), deoxyschizandrin (3), gomisin N (4), and wuweizisu C (5) in Schizandra chinensis from Moonkyung were 8.41${\pm}$0.30, 3.01${\pm}$0.13, 1.06${\pm}$0.05, 7.69${\pm}$0.30, and 1.68${\pm}$0.06 mg/g, respectively.

Morphology and distribution of antennal, maxillary palp and labial palp sensilla of the adult bruchid beetles, Callosobruchus chinensis (L.) (Coleoptera: Bruchidae)

  • Callosobruchus chinensis (Coleoptera: Bruchidae) is the main pest of mungbean (Vigna radiata) especially during seed storage. To provide background information for chemical ecology studies, we examined the externalmorphology of the antennal, maxillary palp and labial palp sensilla of this insect with scanning electron microscopy. The antennae of adult C. chinensis exhibited sexual dimorphism. The female have serrated antenna and the male have pectinate morphology. Eight sensillum types have been observed on the antennae of both sexes: $B\ddot{o}hm$ bristles, sensilla chaetica, sensilla trichodea, sensilla basiconica, sensilla styloconicum, sensilla coeloconica, sensillum campaniformia, sensilla cavity. There are no differences on the morphology of maxillary palp and labial palp between female and male. Four different types of sensilla are present on the labial and maxillary palps of both sexes of C. chinensis: sensilla chaetica, sensilla twig basiconica, sensilla basiconica, sensilla digitiformia.
