Survival Rate of Lactic Acid Bacteria and the Change of ${\beta}-Galactosidase$ Activity in Commercial Yogurts Under the Acidic Conditions (산성조건하에서 시판요구르트의 유산균 생존률과 ${\beta}-galactosidase$의 활성도)

  • Four samples of commercially manufactured yogurts (plain, drinking type) were purchased and evaluated their physico-chemical properties, buffering capacity. And the survival rate of lactic acid bacteria and their ${\beta}-galactosidase$ activity under the acidic conditions (in vitro) were investigated. The values of pH, titratable acidity, viscosity and viable cell counts of yogurts were $3.71{\sim}4.08$, $0.990{\sim}1.045%$, $256{\sim}3164\;cps.$ and $10^8{\sim}10^9\;cfu/ml$, respectively. The volume of 1.0 M-HCl required to reduce the pH of yogurt (50 ml) to minus 2 value was $3.58{\sim}4.33\;ml$. When commercial yogurts were incubated at $37^{\circ}C$ for 120 minutes under the acidic conditions (pH 3.5, 2.5, 1.5), the survival rates of lactic acid bacteria in yogurt were $3.5{\times}10^{-2}{\sim}3.6{\times}10^{-1}%$ at pH 2.5, $8.3{\times}10^{-5}{\sim}4.2{\times}10^{-3}%$ at pH 1.5, respectively, but there was no significant difference at pH 3.5. The remaining activities of ${\beta}-galactosidase$ were $9.4{\sim}36.2%$ at pH 2.5, $4.2{\sim}19.0%$ at pH 1.5, respectively. These results suggested that a significant number of lactic acid bacteria in yogurt might be destroyed in the hostile environment of the stomach, but ${\beta}-galactosidase$ activity from yogurt might be somewhat maintained probably due to the protecting effect by its cell wall and membrane.

  • This study was conducted to investigate the shelf-life and quality of breads made with 0.5, 1 and 2% of Myagropsis myagroides fermented ethanol extracts (MOE). The total microbial count in breads made with 2% MOE decreased to about 1.6 log cycles as compared to that of breads not containing MOE. The protection index measured by rancimat increased with an increase in the quantity of MOE in the breads. During the storage period, the pH value was not different between breads containing MOE and breads not containing MOE. The lightness and redness of the breads decreased with an increase in the quantity of MOE, while the yellowness increased. In the sensory evaluation, the breads containing 0.5% MOE were more preferred than the breads not containing MOE. These results suggest that the addition of 0.5% MOE to breads has a good effect on improving the shelf-life and overall quality.

Response of Early-season Asian Pear 'Hanareum' Treated with GA4+7 to Postharvest Application of 1-methylcyclopropene (1-MCP) (조생종 배 '한아름'에 대한 GA4+7 및 1-methylcyclopropene(1-MCP) 처리 반응)

  • This study was conducted to investigate the effect of 1-methylcyclopropene (1-MCP, $1.0{\mu}L{\cdot}L^{-1}$), a known ethylene action inhibitor, on fruit quality and incidence of physiological disorders during a simulated marketing period at $25^{\circ}C$ for 20 days in early-season Asian pear (Pyrus pyrifolia Nakai) 'Hanareum' that had been treated with 0, 0.5, 1.2 or 2.4% $GA_{4+7}$. Weight loss of stored fruits increased with $GA_{4+7}$ concentration, and the 1-MCP treatment slightly reduced the weight loss rates during the marketing period. Flesh firmness decreased abruptly in all 1-MCP-untreated fruits as the storage period extended to 10 d, whereas the firmness of 1-MCP-treated fruits remained high (> 30 N) during 15 days shelf-life. The effect of 1-MCP was significantly reduced when fruits were subjected to increased GA concentration. Higher soluble solids content and acidity during extended shelf-life were also apparent in 1-MCP-treated 'Hanareum' pears. The L-values (lightness) and hue angles of 1-MCP treated samples were higher than those of controls during 20 days shelf-life, but the a-value (redness) was lower in 1-MCP treated fruits. 1-MCP treatment did not decrease the level of ethylene evolution regardless of $GA_{4+7}$ concentration during shelf-life in early-season Asian pear 'Hanareum'. By contrast, 1-MCP treatment decreased the respiration rate significantly during shelf-life. The efficacy of 1-MCP was greatest in the GA-untreated fruit and was reduced as the $GA_{4+7}$ concentration increased. 1-MCP treatment influenced the severity of physiological disorders including core browning and mealiness: 1-MCP treatment completely blocked the incidence of core browning of during 15 days shelf-life, and reduced the severity of mealiness during 20 days shelf-life regardless of $GA_{4+7}$ concentration. Based on our results, we conclude that the use of $1{\mu}L{\cdot}L^{-1}$ 1-MCP can be of great benefit for maintaining quality and preventing physiological disorders in early-season pear cultivar 'Hanareum' pear, whereas its efficacy decreases with the concentration of $GA_{4+7}$ whereas its efficacy gradually decreases when the concentration of $GA_{4+7}$ paste increased.

Comparison of the Antioxidant Effects of Ethyl Alcohol Extracts of a Maillard-type and a Caramelization-type Browning Reaction Mixtures (Maillard 형(形) 및 Caramelization 형(形) 갈색화(褐色化) 반응물(反應物)에서 얻어진 알콜 추출물(抽出物)들의 항산화(抗酸化) 효과(效果)의 비교(比較))

  • The antioxidant effects of the alcohol extracts obtained from a Maillard-type and a caramelization-type browning reaction mixtures were determined and compared. The Maillard-type reaction mixtrue contained 0. 2 M glucose and 0. 2 M glycine while the caramelization-type reaction mixture contained only 0. 2 M glucose and both were heated at $100^{\circ}C$. The results obtained are as follows. 1. The color intensity of the Maillard-type reaction mixture appeared to increase in proportion to the length of reaction time. However, the antioxidant activity of the extracts did not seem to increase in proportion to the length of reaction time. The antioxidant activity of the extracts from the reaction mixture heated for 16 hours was not much greater than that of the extracts from reaction mixture heated for 2 hours. 2. The color intensity of the caramelization-type browning reaction appeared to increase in proportion to the length of reaction time. The antioxdant activity of the extracts did not seem to increase in proportion to the length of reaction time. 3. It appeared that the antioxdant effects of the alcohol extracts from the Maillard-type browning reaction mixture were far greater than those from the caramelization-type browning reaction mixture, compared on the basis of the same length of reaction time. Substrates, containing the alcohol extracts of the caramelization reaction mixture taken after 4 and 120 hours, developed peroxide values of 88. 9 and 33. 0 after a 20 day storage period (control, 135. 0) whereas substrates, containing the alcohol extracts of the Maillard-type reaction mixture taken after 1 and 16 hours, developed peroxide value of 9. 5 and 7. 5 after the same storage period.

COMPARISON OF MICROTENSILE BOND STRENGTH OF COMPOSITE RESTORATION TO ERODED ENAMEL BY SURFACE TREATMENT (접착제의 종류에 따른 침식치아에 대한 복합레진의 결합강도)

  • Composite resin has been widely used for eroded enamel. But, as there have been many reports about the differences in physicochemical characteristics of eroded enamel compared with sound enamel, an additional effort was thought necessary to obtain the optimal bond strength. As a possible answer, we came to think about the application of infiltrant resin which is known to have an excellent penetration capacity into enamel. This study was performed for the purpose of comparing the bond strength of composite restoration with or without infiltrant resin under adhesives on the artificially eroded enamel. 60 extracted sound maxillary primary incisors were selected and divided into group 1, 2, 3 according to the number of artificial erosion cycling for 5 minute duration in 1% citric acid of pH 3.2 at $37^{\circ}C$. And the labial surfaces were divided into 3 areas; group A, only resin adhesive was used, group I, only infiltrant resin, group IA, infiltrant resin followed by resin adhesive. Afterwards, every specimen was restored with composite resin. Microtensile bond strength was measured and failure modes were observed. The obtained results were as follows: 1. In comparing the bond strength by the degree of enamel erosion, it was revealed the highest bond strength in group 1, followed by group 2 and 3, showing the lowest bond strength in most eroded group(p<0.05). 2. In comparing the bond strength by surface treatment methods, group IA and I showed higher value than group A(p<0.05), with unsignificant difference between group I and IA(p>0.05). 3. In observation of failure mode, it was shown higher frequency of cohesive failure in order of 1-2-3 and IA-I-A. Conclusively, it was shown decreasing tendency of bond strength as the enamel is more eroded, and infiltrant resin was thought helpful to replace or add to the resin adhesive for optimal bonding with eroded enamel.

Two Crystal Structures of Fully Dehydrated $Ag_{12-2x}Co_x-A (x = 3 and 4.5)$ (완전히 탈수한 $Ag_{12-2x}Co_x-A$ (x = 3 및 4.5)의 결정구조)

  • The crystal structures of $Co^{2+}\;and\;Ag^+\;exchanged\;zeolite\; A,\; Ag_6Co_3$-A(a = 12.131(5)$\AA$) and $Ag_3Co_{4.5}$-A(a = 12.145(1)$\AA$), have been determined by single crystal X-ray diffraction techniques. Both structures were solved and refined in the cubic space group Pm3m at 21(1)$^{\circ}C$. Full-matrix leastsquares refinement converged to the final error indices of R1 = 0.045 and R2 = 0.041 for $Ag_3Co_{4.5}-A,\; and\; R1 = 0.066\; and\; R2 = 0.076\; for\; Ag_6Co_3$-A using the 258 and 189 reflections, respectively, for which I > 3$\sigma$(I). Both structures indicate that CO(Ⅱ)ions are coordinated by three framework oxygens; the Co(II) to O(3) distances are 2.118(4)$\AA$ for $Ag_3Co_{4.5}$-A and 2.106(1)$\AA$ for $Ag_6Co_3-A$, respectively. In each structure, the angle substended at Co(II), O(3)-Co(II)-O(3) is ca 120°, close to the idealized trigonalplanar value. $Co^{2+}$ ions prefer to 6-ring sites and $Ag^+$ ions prefer to 8-ring site when total number of cations is more than 8. The crystals of hydrated and dehydrated $Ag_{12-2x}Co_x-A (x > 4.5)$ had no crystalline diffraction pattern, indicating the apparent exchange limit of $Co^{2+}\; into\; Ag_{12}-A\; is\; 4.5 Co^{2+}$ ions per unit cell. $Co^{2+}$ ions hydrolyze $H_2O$ molecules and $H_3O^+$ concentraction is accumulating. These $H_3O^+$ ions destroy the zeolite structures.

Quality characteristics and biological activity of Yanggaeng with Glechoma hederacea var. longituba Nakai powder (긴병꽃풀의 생리활성과 이를 첨가한 양갱의 품질특성)

  • The objective of this study is to investigate the quality characteristics of Yanggaeng by using the functional properties of Glechoma hederacea (GH). Sample was dried at $50^{\circ}C$ dry oven. The results of the study were as follows : The Phenolic compounds of GH was $12.99{\pm}0.3mg/g$ in water extract (GHWE), $3.14{\pm}0.07mg/g$ in 70% ethanol extract (GHEE). The antioxidant activity of GH was determined in various phenolic concentrations at $50-200{\mu}g/mL$. DPPH activities of GHWE and GHEE were 77.16-78.24% and 73.04-77.00%, respectively. The ABTS were 84.35-99.75% and 83.74-99.55%. The anti-oxidant protection factor (PF) were 1.54-1.62 PF and 1.62-2.09 PF and TBARS were 42.93-94.09% and 91.05-95.19%, respectively. Tyrosinase inhibitory activity of GHEE increased concentration dependently. Hyaluronidase inhibition activity of GHEE and GHWE, showing that there were increasing pattern depending on the increases in the phenolics concentration of GH. In texture, Hardness and springiness were significantly different in the control and 2% groups, but cohesiveness and chewiness did not show any significant difference. In color, L value decreased in proportion to concentration, and a and b values did not change. Sensory characteristics showed that the 1% group had the highest score and the 2% group had the lowest score. Thus, when the GHP Yanggaeng was prepared, in consideration of its sensory characteristics, and at appropriate concentration on below 1%.

Optimization of a Medium for the Production of Cellulase by Bacillus subtilis NC1 Using Response Surface Methodology (반응 표면 분석법을 사용한 Bacillus subtilis NC1 유래 cellulase 생산 배지 최적화)

  • Previously, cellulase and xylanase producing microorganism, Bacillus subtilis NC1, was isolated from soil. Based on the 16S rRNA gene sequence and API 50 CHL test the strain was identified as Bacillus subtilis, and named as B. subtilis NC1. We cloned and sequenced the genes for cellulase and xylanase. Plus, the deduced amino acid sequences from the genes of cellulase and xylanase were determined and were also identified as glycosyl hydrolases family (GH) 5 and 30, respectively. In this study to optimize the medium parameters for cellulase production by B. subtilis NC1 the RSM (response surface methodology) based on CCD (central composite design) model was performed. Three factors, tryptone, yeast extract, and NaCl, for N or C source were investigated. The cellulase activity was measured with a carboxylmethyl cellulose (CMC) plate and the 3,5-dinitrosalicylic acid (DNS) methods. The coefficient of determination (R2) for the model was 0.960, and the probability value (p=0.0001) of the regression model was highly significant. Based on the RSM, the optimum conditions for cellulase production by B. subtilis NC1 were predicted to be tryptone of 2.5%, yeast extract of 0.5%, and NaCl of 1.0%. Through the model verification, cellulase activity of Bacillus subtilis NC1 increased from 0.5 to 0.62 U/ml (24%) compared to the original medium.

A Study on the Standardization of Sugar-preserved Prunus mume Manufactured in Ha-Dong (하동지역 매실 당절임의 표준화 방안 연구)

  • Prunus mume is rich in organic acid, and excels in antioxidant activity. However, development of processed foods using Prunus mume is very limited, and characteristics of its products are different according to the region they are processed in. In this study, we collected 13 kinds of sugar-preserved Prunus mume home-made products in Hadong, and investigated their physical and manufacturing characteristics to select a superior product. 5 kinds of sugar-preserved Prunus mume products were selected according to results of sensory evaluation, and the overall acceptability of product No.13 was superior. Moisture contents of the 5 samples ranged from 45.2~53.3%, and pH and acidity showed similar values at about 2.8 and 0.4%, respectively. Sugar content of product No.13 were slightly lower than other products. Crude protein and lipid contents were similar at about 0.4 and 0.5%, respectively. Hardness value of product No.13, selected by its texture in sensory evaluation, was higher than others. The main components of organic acids were oxalic, malic, citric and fumaric acid, and citric acid content occupied more than 60% of total organic acid content. Also, the main components of free sugar were glucose and fructose.

Early Changes after Death of Plaice, Paralichthys olivaceus Muscle -4. Effect of Killing Methods on Rigor Index and Breaking Strength of Muscle- (넙치(Paralichthys olivaceus)육의 사후 조기 변화 -4. 치사 방법이 육의 사후 경직도와 파괴 강도에 미치는 영향-)

  • This study was undertaken to clarify the effect of killing methods on physical and rheological changes of plaice, Paralichthys olivaceus muscle at early period after death. Plaices killed by the four different methods(1. spiking at the brain instantly. 2. drowning in air. 3. dipping in 1,000ppm ethylaminobenzoate dissolved sea water as an anesthetic. 4. electrifying in sea water.) were stored at $5^{\circ}C$, and the rigor-index and breaking strength through storage were monitored. The longest onset time of rigor-mortis and full rigor was in the samples killed by dipping in sea water with dissolved anesthetic among all samples, where rigor-mortis began at 20hrs after killing and maximum tension was attained after 56hrs. However, in the cases of plaice electrified in sea water or drowned in air, the onset of rigor-mortis began just after killing and maximum tensions were attained after 9hrs and 13hrs, respectively. The level of breaking strength in the muscle of fish killed by spiking the brain instantly was $950.30{\pm}50.23g$, immediately after killing. The value and time reached around the maximum breaking strength for each of the samples were $1,230.60{\pm}30.32g$ and Ohr (immediately after killing) for samples killed by electrifying in sea water, $1,235.83{\pm}35.37g$ and 2.5hrs for drowning samples, $1,186.29{\pm}55.90g$ and 10hrs for spiking samples, and $1,189.67{\pm}50.32g$ and 15hrs for samples dipped in anesthetic, respectively. From the results above, it could be concluded that electrification in sea water is the most effective method in accelerating rigor-mortis and shortening times of reaching the maximum breaking strength of fresh plaice flesh of all the killing methods at early periods after death, whereas dipping in sea water treated with anesthetic was the most effective way in delaying those changes.
