Development of the method to extend shelf life of Backsulgie with enzyme treatment (효소처리에 의한 백설기의 저장성을 연장하기 위한 방법의 개발)

  • ${\alpha}$-amylase was investigated as an antistaling agent for Backsulgie, a traditional rice cake. Rice powder was mixed with ${\alpha}$-amylase, fermented for 2 hr at 37$^{\circ}C$, and steamed for 20 min. Rice cake was stored at room temperature or freezer for 4 days, and analyzed to determined the changes of chemical and sensory properties. When ${\alpha}$-amylase was added to rice cake, the content of reducing sugars and the yellow color of the cake were increased, and the water activity was decreased. Soft and moist textural properties were apparent in ${\alpha}$-amylase-added rice cakes by sensory evaluation. X-ray diffraction showed a V pattern after 4 days of storage which indicated the starch of rice cake was not retrograded. However, there was no significant difference in moisture content between enzyme-treated and non-treated rice cakes. Above results suggest that ${\alpha}$-amylase treatment produced dextrins which consequently bound with water and inhibited the retrogradation of rice cake.

Effect on Shear Strength of Ceramic Surface Treatment Materials and Three Resin Cements to IPS Empress 2 (표면 처리재와 레진 시멘트가 IPS Empress 2의 전단결합강도에 미치는 영향)

  • The purpose of this study is to analyze the shear bond strength according to kinds of surface treatment agents and resin cements after acid etching of IPS Empress 2. For this purpose, test groups were classified into silane-treated bonding groups, silica-coated group and Targis link applied group. Then, nine bonding groups in total, each three groups per kind, were prepared by using three kinds of resin cements-Panavia F, Variolink II and Rely-X ARC, and thirty test specimens per group were prepared. To examine any changes in the oral environment, the shear bond strength of each test specimen was measured after dipping test for 24 hours and for five weeks, respectively, in distilled water at $37^{\circ}C$ and performing heat cycle 10,000 times in total, each 2,000 times per week, during a five weeks of dipping, under the condition similar to the oral environment. The bond failure modes were also observed by means of a scanning electron microscope. The results are summarized as follows 1. Statically significant differences between the surface conditioning materials were observed. The shear strength of the silane treatment was the highest of all three types of surface treatments(p<0.001). 2. Shear strengths varied significantly for different types of resin luting agents. But bond strength of Targis link surface treatments were not significantly different(p<0.05). 3. No significant difference of bonding strengths was found between storage time conditions(24 hours and 5 weeks). Only group II, IV, VII, IX were significantly different(p<0.05). 4. After thermocycling, the shear bond strengths of all groups were significantly decreased (p<0.05). Group III, V, VI were no significantly different. 5. On the SEM observation of fractured surfaces, all groups were shown complex failure.

In vivo and In vitro Metabolism of Recombinant Human Epidermal Growth Factor (DWP401) in Rats (재조합 인간 상피세포성장인자(DWP401)의 흰쥐에서의 in vivo와 in vitro 대사)

  • Metabolism of DWP401, recombinant juman epidermal growth factor, was examined in vivo and in vitro in rats. When $^{125}I$-labeled DWP401 was administered at a dose of 50 ${\mu}g$/kg by i.v. injection. $^{125}I$-labeled DWP401 was rapidly degraded within 30 minytes above 93%. Thin layer chromatography analysis of urine collected for 24 hr after i.v. administration of $^{125}I$-labeled DWP401 showed ohly one spot on a X-ray film which was considered as diiodo-tyrosine. This result suggests tha $^{125}I$-labeled DWP401 was completely digested into free amino acids without any specific intermediate polypeptides. About 42.1% of the administered iodine was recovered in 24 hr. For in vitro degradation study, $^{125}I$-labeled DWP401 was added to plama and tissue homogenates of rats and incubated at $37^{\circ}C$. Almost 98% of the added radioactivity recovered from the protein fraction of the liver, kidey, small intestine, stomach and spleen decreased rapidly. For examplem the recovery rates of $^{125}I$-labeled DWP401 were 58.6, 63.2, 39.9, 52.9 and 66.8% after 4hrs of incubation in respective organ homogenates.

Nelumbo nucifera Leaves Inhibit HASMC Proliferation and Migration Activated by TNF-$\alpha$ (Human Aortic Smooth Muscle Cell에서 하엽(荷葉)의 항동맥경화 활성 연구)

  • Objectives : The proliferation and migration of human aortic smooth muscle cells (HASMC) in response to activation by various stimuli plays a critical role in the initiation and development of atherosclerosis. This study was conducted to examine the effects of Nelumbo nucifera leaves (NNL) on the proliferation and migration of HASMC. Additionally, the mechanisms involved in any observed effects were also evaluated. Methods : Apoptotic cells were measured by staining with FITC-labeled annexin V, followed by flow cytometric analysis. The expression level of apoptosis related proteins was confirmed by western blot. And MMP-9 activity was measured by gelatin zymography and MMP-9 expression was measured by ELISA Results : NNL completely inhibited the proliferation of HASMC via induction of the expression of apoptotic proteins including annexin V, cleaved poly ADP-ribose polymerase (PARP), and caspase-3 and -8. NNL treatment resulted in the release of cytochrome c into cytosol, a loss of mitochondrial membrane potential, a decrease in Bcl-2 and Bcl-xL and an increase in Bax expression. NNL also blocked HASMC migration via suppression of MMP-9. Conclusions : Taken together, these results indicate that NNL has the potential for use as an anti-artherosclerosis agent.

Geochemical Variation of Authigenic Glauconite from Continental Shelf of the Yellow Sea, off the SW Korea (한반도 남서부, 황해 대륙붕에서 자생하는 해록석의 지구화학적 변화)

  • The massive, fractured and porous-type of glauconite, which is subdivided by surface morphology, occur in subtidal sand and semiconsolidated intertidal sand/mud from continental shelf of the southeastern Yellow Sea. This area is presumed to be a part of Holocene transgressive tidal systems tract. The glauconite, pellet-like grains with diameter of 0.1 to 1 mm, is scattered in surface sand sediments. Results of X-ray diffraction data of the minerals are monoclinic with $a=5.242{\AA}$, $b=9.059{\AA}$, $c=10.163{\AA}$, ${\beta}=100.5^{\circ}$, $V=474.53{\AA}^3$. Thermal treatments on the oriented glauconite increase the X-ray diffraction intensity near $10{\AA}$ (001), suggesting the presence of some expandable layers. Specific gravity of the glauconite is $2.60{\pm}0.45gm/cc$ on the basis of chemical composition and unit-cell dimensions. Based on $O_{10}(OH)_2$, chemical composition of glauconites, octahedral Fe content ranges from 1.19 to 2.06 atoms, corresponding octahedral AI is 0.18 to 0.76 atoms, which progressively substitute Fe for AI with increasing from porous to massive-type. The Mg content ranges from 0.35 to 0.54 atoms, and shows higher with increasing Al contents. A systematic increase of interlayer K from 0.34 to 0.71 is also observed with apparent increases from porous to massive-type, and related to a proportion of expandable layers. The clay preserved in glauconite, which is recognized as ordered/disordered (massive to fractured-type). The interstratified illite/smectite (porous-type), contains 7 to 27 % expandable layers. The glauconite seems to originate from post depositional authigenic growth in reducing environments promoted by the dissolution of clay minerals and biogenic debris.

Analysis of Signal Properties in accordance with electrode area of x-ray conversion material (X선 검출 물질의 전극 면적에 따른 신호특성 분석)

  • In recent, a digital x-ray detector attracted worldwide attention and there are many studies to commercialize. There are two methods in digital x-ray detector. This method is an Indirect method and Direct method. This study is to see the differences between the digital x-ray detector based on a-Se used in the existing indirect conversion method and an x-ray conversion material that has better SNR(Signal-to-noise ratio) and property than the a-Se. To solve the problem that is difficult to make a large area film using Screen-Print method, we used a Screen-Print method. In this study, we used a polyclystal $HgI_2$ as x-ray conversion material and a sample thickness is $150{\mu}m$ and an area is $3cm{\times}3cm$. ITO(Indium-Tin-Oxide) electrode was used as top electrode using a Magnetron Sputtering System and each area is $3cm{\times}3cm$, $2cm{\times}2cm$ and $1cm{\times}1cm$ and then we evaluated darkcurrent, sensitivity and SNR of the $HgI_2$ film are measured, then we evaluated the electrical properties. And we used a current integration mode when I-V test. This experiment shows that the sensitivity increases in accordance with the area of the electrode but the SNR is decreased because of the high darkcurrent. Through fabricating of various thicknesses and optimal electrodes, we will optimize SNR in the future work.

Saccharification of Foodwastes Using Cellulolytic and Amylolytic Enzymes from Trichoderma harzianum FJ1 and Its Kinetics

  • The study was targeted to saccharify foodwastes with the cellulolytic and amylolytic enzymes obtained from culture supernatant of Trichoderma harzianum FJ1 and analyze the kinetics of the saccharification in order to enlarge the utilization in industrial application. T. harzianum FJ1 highly produced various cellulolytic (filter paperase 0.9, carboxymethyl cellulase 22.0, ${\beta}$-glucosidase 1.2, Avicelase 0.4, xylanase 30.8, as U/mL-supernatant) and amylolytic (${alpha}$-amylase 5.6, ${\beta}$-amylase 3.1, glucoamylase 2.6, as U/mL-supernatant) enzymes. The $23{\sim}98\;g/L$ of reducing sugars were obtained under various experimental conditions by changing FPase to between $0.2{\sim}0.6\;U/mL$ and foodwastes between $5{\sim}20\%$ (w/v), with fixed conditions at $50^{\circ}C$, pH 5.0, and 100 rpm for 24 h. As the enzymatic hydrolysis of foodwastes were performed in a heterogeneous solid-liquid reaction system, it was significantly influenced by enzyme and substrate concentrations used, where the pH and temperature were fixed at their experimental optima of 5.0 and $50^{\circ}C$, respectively. An empirical model was employed to simplify the kinetics of the saccharification reaction. The reducing sugars concentration (X, g/L) in the saccharification reaction was expressed by a power curve ($X=K{\cdot}t^n$) for the reaction time (t), where the coefficient, K and n. were related to functions of the enzymes concentrations (E) and foodwastes concentrations (S), as follow: $K=10.894{\cdot}Ln(E{\cdot}S^2)-56.768,\;n=0.0608{\cdot}(E/S)^{-0.2130}$. The kinetic developed to analyze the effective saccharification of foodwastes composed of complex organic compounds could adequately explain the cases under various saccharification conditions. The kinetics results would be available for reducing sugars production processes, with the reducing sugars obtained at a lower cost can be used as carbon and energy sources in various fermentation industries.

  • We have measured the spatiotemporal behavior for the density of excited Xe atoms in the $1s_5$ metastable states by laser absorption spectroscopy in accordance with various shapes of ITO electrode. The maximum density of excited Xe atoms in the Is5 state in a discharge cell for fish-boned, T-shaped and squared ITO electrodes has been measured to be $3.01{\times}10^{13}\;cm^{-3}$, $2.66{\times}10^{13}\;cm^{-3}$ and $2.06{\times}10^{13}\;cm^{-3}$, respectively. Throughout this experiment, we could understand the influence of the shapes of ITO electrode of micro discharge cell on the high efficiency of AC-PDPs.

Evaluation of Biocompatibility of Anodized and Hydrothermally Treated Pure Niobium Metal (양극산화와 열수처리한 순수 니오비움 금속의 생체활성 평가)

  • This study was performed to investigate the surface properties of electrochemically oxidized pure niobium by anodic oxide and hydrothermal treatment technique. Niobium specimens of $10\times10mm$ in dimension were polished sequentially from #600, #800, #1000 emery paper. The surface pure niobium specimens were anodized in an electrolytic solution that was dissolved calcium and phosphate in water. The electrolytic voltage was set in the range of 250 V and the current density was 10 $mA/cm^2$. The specimen was hydrothermal treated in high-pressure steam at 300$^{\circ}C$ for 2 hours using an autoclave. Then, specimens were immersed in the Hanks' solution with pH 7.4 at 37$^{\circ}C$ for 30 days. The surface of specimen was characterized by scanning electron microscope(SEM), energy dispersive X-ray microanalysis(EDX), potentiostat/galvanostat test, and cytotoxicity test. The results obtained was summarized as follows; According to the result of measuring corrosion behavior at 0.9% NaCl, corrosion resistance was improved more specimens treated with anodic oxide than in hydrothermal treated ones. The multi-porous oxide layer on surface treated through anodic oxidation showed a structure that fine pores overlap one another, and the early precipitation of apatite was observed on the surface of hydrothermal treated samples. According to the result of EDX after 30 days deposition in Hanks' solution, Ca/P was 1.69 in hydrothermal treated specimens. In MTT test, specimens treated through anodic oxidation and hydrothermal treated ones showed spectrophotometer similar to that of the control group. Thus no significant difference in cytotoxicity was observed (P>0.05).

Mutation Detection of E6 and LCR Genes from HPV 16 Associated with Carcinogenesis

  • Human papillomavirus (HPV) is responsible for one of the most frequent sexually transmitted infections. The first phylogenetic analysis was based on a LCR region fragment. Nowadays, 4 variants are known: African (Af-1, Af-2), Asian-American (AA) and European (E). However the existence of sub-lineages of the European variant havs been proposed, specific mutations in the E6 and LCR sequences being possibly related to persistent viral infections. The aim of this study was a phylogenetic study of HPV16 sequences of endocervical samples from C${\acute{o}}$rdoba, in order to detect the circulating lineages and analyze the presence of mutations that could be correlated with malignant disease. The phylogenetic analysis determined that 86% of the samples belonged to the E variant, 7% to AF-1 and the remaining 7% to AF-2. The most frequent mutation in LCR sequences was G7521A, in 80% of the analyzed samples; it affects the binding site of a transcription factor that could contribute to carcinogenesis. In the E6 sequences, the most common mutation was T350G (L83V), detected in 67% of the samples, associated with increased risk of persistent infection. The high detection rate of the European lineage correlated with patterns of human migration. This study emphasizes the importance of recognizing circulating lineages, as well as the detection of mutations associated with high-grade neoplastic lesions that could be correlated to the development of carcinogenic lesions.
