The Distribution of $^{14}C-chitosan$ by Different Molecular Weight in Mice (마우스에서 $^{14}C-chitosan$ 분자량별 체내 분포에 관한 연구)

  • Chitosan is a nontoxic natural chealtor which was made by chitin, and reduced a contamination of radiostrontium in animals. In this experiment, A different molecular weight of C-14 chitosan was intravenously administered to mice, and then the distribution of C-14 chitosan in the body was observed. Male mice (8 to 10 weeks, body weight of 30 to 35g) of ICR strain were used. C-14 chitosan was diluted with saline and then given intravenously in mice. After the administration of C-14 chitosan, mice was sacrificed at the 6th hour, 1st, 3rd, 5th, and 7th day. Beta radioactivities in the blood, liver, kidney, liver, muscle, testis, and urine was measured using a liquid scintillation analyzer. Most of the C-14 chitosan was excreted through urine within 6 hours. Biodistribution of C-14 chitosan was similar despite the difference of moleclar weight. Higher distributions of radioactivities were found in the liver, kidney, spleen. The relative concentration in tissue increased for the 6 hours and then decreased. In conclusion, most of C-14 chitosan was excreted through urine despite the difference of molecular weight. and, low molecular weight of C-14 chitosan showed higher distribution than high molecular weight of C-14 chitosan in tissues.

Enhancing the Efficacy of Burkholderia cepacia B23 with Calcium Chloride and Chitosan to Control Anthracnose of Papaya During Storage

  • The efficacy of the combination of Burkholderia cepacia B23 with 0.75% chitosan and 3% calcium chloride ($CaCl_2$) as a biocontrol treatment of anthracnose disease of papaya caused by Colletotrichum gloeosporioides, was evaluated during storage. The growth of B. cepacia B23 in papaya wounds and on fruit surfaces was not affected in presence of chitosan and $CaCl_2$ or combination throughout the storage period. The combination of B. cepacia B23 with chitosan-$CaCl_2$ was more effective in controlling the disease than either B. cepacia B23 or chitosan or other combination treatments both in inoculated and naturally infected fruits. Combining B. cepacia B23 with chitosan-$CaCl_2$ gave the complete control of anthracnose infection in artificially inoculated fruits stored at $14^{\circ}C$ and 95% RH for 18 days, which was similar to that obtained with fungicide $benocide^{(R)}$. Moreover, this combination offered a greater control by reducing 99% disease severity in naturally infected fruits at the end of 14 days storage at $14^{\circ}C$ and 95% RH and six days post ripening at $28\pm2^{\circ}C$, which was superior to that found with $benocide^{(R)}$ or other treatments tested. Thus, postharvest application of B. cepacia B23 with chitosan-$CaCl_2$ as enhancers represents a promising alternative to synthetic fungicides for the control of anthracnose in papaya during storage.

Effects of Chitosan on Anti-tumor Activity in Mice (키토산이 암세포성장에 미치는 효과)

  • Cytotoxic anticancer chemotherapeutic agents generally produce severe side effects, while reducing host resistance to cancer and infections, especially through the destruction of lymphoid and bone marrow cells. In this study, we have investigated the effect of chitosan on cytotoxic activity against cancer cells and life span in mice. The direct cytotoxicity of chitosan or chitosan-combinated chemotherapeutic agents for tumor cells was observed. In addition, the effect of life span extention was counted on sarcoma 180 mice injected with chitosan-combinated mitomycin C. The effect of growth inhibion for cancer cells, K562 and Yac-1 was shown in the cytotoxicity test of chitosan or chitosan-combinated chemotherapeutic agents. Also, the effect of life span extension was observed on sarcoma 180 mice injected with chitosan-combinated mitomycin C. Our results suggest that life span extension in sarcom 180 mice exposed with chitosan-combinated chemotherapeutic agents showed the probability of its usefulness for cancer therapy if more research results were accumulated.

  • Dyeing properties and crosslinking effect of bifunctional reactive dyes on chitosan fiber were investigated to improve the stability of chitosan fiber against the mild acidic solution and the wet processing. Chitosan fibers were crosslinked with epichlorohydrin for comparing purpose, and dyed with C. I. Reactive Red 194, C. I. Reactive Blue 50, and the reactive dye having two $\alpha$-bromoacrylamide groups at various concentrations. The initial dyeing rates of reactive dyes are rapid and chitosan fibers absorb the relatively large amount of dyes. The chitosan fibers dyed with these dyes show the low degree of swelling and the low solubility in 1 % acetic acid solution and also represent the higher thermal stability The reactive dye with two $\alpha$-bromoacrylamide groups shows higher crosslinking effect than other dyes.

  • The effects of dietary chitosan on lipid oxidation, fatty acid composition and blood profile of porks were investigated. A total 24 pigs(55$\pm$5kg) were fed a control diet (a commercial diet) or chitosan-supplemented diets (T1; 0.2% chitosan, T2; 0.4% chitosan, T3; 0.6% chitosan) for 6 weeks. After six weeks, pigs were slaughtered and bellies were collected from each treatment group. Samples were stored at 0$\pm$1$^{\circ}C$ for 14 days. The thiobarbituric acid reaction substance(TBARS) values of all the treatments increased until 7 days of storage. The fatty acid and crude fat composition of all the treatments were not changed during storage significantly. The total cholesterol and triglyceride in blood chemistry tend to lower chitosan supplemented groups then control group.

  • A novel ${\beta}$-cyclodextrin derivative (CCD-C) was synthesized with chitosan and carboxymethyl-${\beta}$-cyclodextrin. Its structure was characterized by elemental analysis, infrared spectra analysis, and X-ray diffraction analysis. The adsorption properties for guanosine 5'-monophosphate, cytidine 5'-monophosphate and uridine 5'-monophosphate were studied. Experimental results demonstrated that CCD-C had higher adsorption capability for guanosine 5'-monophosphate, and that the adsorption capacity for guanosine 5'-monophosphate was 74.20mg/g. The adsorption capacity was greatly influenced by pH, time and temperature. The introduction of chitosan enhanced the adsorption ability and adsorption selectivity of ${\beta}$-cyclodextrin for guanosine 5'-monophosphate. This novel derivative of chitosan is expected to have wide applications in the separation, concentration and analysis of nucleotides in biological samples.

Dyeing and Functional Properties of Cotton-Modal-Chitosan Blended Towel Fabric Dyed with Mugwort Colorants (면-모달-키토산 혼방타월의 쑥에 대한 염색성과 기능성)

  • The objective of this study is to develop eco-friendly, functional towel material utilizing cotton-Modal-chitosan blended(C-M-CH) yarn and natural dyeing with mugwort colorants. Dyeing properties of towels with mugwort colorants were studied by investigating the effect of dyeing conditions including concentration of mugwort colorants, dyeing temperature, and dyeing time, and the effects of mordants on dye uptakes were investigated. The C-M-CH towel showed better dye uptake than 100% cotton towel with mugwort colorants. The shade of towels got darker and red-yellowish tint increased by mordanting. Comparing with 100% cotton towel, the colorfastness of dyed C-M-CH towel was satisfactory showing above 3 grade which is the lowest grade to washing fastness. The antibacterial activity against Staphylococcus aureus and deodorization performance of towels were excellent and improved by dyeing with mugwort colorants. From the results obtained, it is concluded that the cotton-Modal-chitosan blended towel dyed with mugwort colorants can be used practically for an eco-friendly and multi-functional towel materials with excellent absorbance and drying properties.

  • The preservative effect of chitosan film packing on quality of lightly-salted and dried horse mackerel was studied. In preparation of chitosan film, blue crab shell chitosan was dissolved in dilute acetic acid$(1.0\%,\;v/v)$, filtered, and spreaded on plastic plate and dried at $50\pm2^{\circ}C$. The chitosan film thus obtained was neutralized with 1.0N NaOH for 2 hrs and dried at room temperature after washing several times with distilled water. The lightly-salted and dried horse mackerel product was prepared by drying for 4 hrs at $40\pm2^{\circ}C$ in hot air dryer after packing with the chitosan film. During storage at $5.0\pm0.5^{\circ}C$, moisture content of the product was higher than that of the reference, but contents of VBN(volatile basic nitrogen) , amino nitrogen, and TMA of the product on dry basis were lower than those of the reference. Viable cell count, TBA value, and peroxide value of the product were also lower than those of the reference. Judging from the result of sensory evaluation, the chitosan film packing in the storage of lightly-salted and dried horse mackerel was remarkably elongated shelf-life of the product. From the results of chemical and sensory evaluation, it was concluded that chitosan film packing was an effective method for retaining the quality of lightly-salted and dried horse mackerel.

  • Tyrosinase (monophenol. 3.4-${\beta}$-dihydroxyphenylalanin oxygen oxidoreductase. EC 1.14.18.1 J. which plays a pivotal role in melanogenesis. It is single chain glycoprotein catalyzing the hydroxylation of tyrosine to ${\beta$\mid$$-3.4-dihydroxyphenylalanin (DOPA) and the oxidation of DOPA to DOPA quinone. To investigate whitening effect of chitosan oligosaccharide. we obtained chitosan oligosaccharide [(glucosamine)2-6] by NaNO2 oxidation and measured the effect of chitosan oligosaccharide on tyrosinase activity. Chitosan oligosaccharide dose-dependently inhibited tyrosinase (2 unit) activity and inhibited by 18.8% at dose of 100${\mu}$g/ml. Vitamin C. arbutin and kojic acid that are well known to be inhibitor of melanin production dose-dependently inhibited tyrosinase (2unit) activity. These results suggest that chitosan oligosaccharide may be used as inhibitor of melanin production in melanocyte. which will be further studied.

Extension of Tofu Shelf-Life with Water Soluble Degraded Chitosan as a Coagulant (수용성 키토산 분해물질을 응고제로 이용한 두부의 저장성 증대)

  • CaCl2 and water soluble degraded chitosan mixtures were used as coagulants for extending the shelf life of tofu. Microbial counts, pH, and turbidity in tofu immersion solution were analyzed during the periods of storage at 4oC and physico chemical properties of tofu were determined by rheological properties, color and yield. There was no significant difference in moisture content and yield between tofu made with and without chitosan. Failure stress and stress relaxation values were reduced when chitosan was added to CaCl2, but there was no significant change among contents of chitosan added to tofu. The pH values were decreased at the beginning and then slowly increased after 14 days storage. The changes in turbidity were increased during storage, but the change in 2.0g degraded chitosan mixture with 7.0g CaCl2 coagulant(2.0:7.0 ratio) was lower than others and initial turbidity was lower as much as chitosan added to coagulant. These data showed that the water soluble degraded chitosan had a good anti microbial effect and increases the shelf life of tofu.
