Evaluation of an Automated ELISA (VIDAS(R)) and Real-time PCR by Comparing with a Conventional Culture Method for the Detection of Salmonella spp. in Steamed Pork and Raw Broccoli Sprouts (편육과 브로콜리싹에서 Salmonella spp. 검출을 위한 배지법과 Real-time PCR 및 신속 검사키트(VIDAS(R))의 비교검증)

  • Salmonellosis is an important worldwide foodborne infectious disease that is transmitted by many food vehicles including raw and processed animal products and fresh produce. In this study, the effectiveness of automated ELISA ($VIDAS^{(R)}$) and realtime PCR in the detection of Salmonella spp. in steamed pork and raw broccoli sprouts was evaluated by comparing their results with those of a conventional culture method. Bulk samples (500 g) of steamed pork and raw broccoli sprouts were inoculated with various levels of Salmonella and divided into 20 samples (25 g each). All the samples, including the controls, were analyzed using a conventional culture method, $VIDAS^{(R)}$, and real-time PCR to detect the presence of Salmonella. In addition, the levels of background flora in the steamed pork and the raw broccoli sprouts were determined. In the steamed pork that contained less than 100 CFU/g of aerobic bacteria, all three methods detected low levels of Salmonella without a statistical difference in their performance. In the broccoli sprouts with high quantities of background flora (ca. $6.7{\times}10^7$ CFU/g), however, all three methods were unable to detect low levels of Salmonella, and real-time PCR and $VIDAS^{(R)}$ more sensitively detected Salmonella than the culture method, with significant statistical differences. In conclusion, $VIDAS^{(R)}$ and real-time PCR could be superior to conventional culture methods in detecting Salmonella in food with high levels of background flora.

Three Dimensional Measurements of Pore Morphological and Hydraulic Properties (토양 공극 형태와 수문학적 특성에 대한 3 차원적 측정)

  • Pore network models are useful tools to investigate soil pore geometry. These models provide quantitative information of pore geometry from 3D images. This study presents a pore network model to quantify pore structure and hydraulic characteristics. The objectives of this work were to apply the pore network model to characterize pore structure from large images to quantify pore structure, calculate water retention and hydraulic conductivity properties from a three dimensional soil image, and to combine measured hydraulic properties from experiments with calculated hydraulic properties from image. Soil samples were taken from a site located at the Baltimore science center, which is located inside of the city. Undisturbed columns were taken from the site and scanned with a computer tomographer at resolutions of 22 ${\mu}m$. Pore networks were extracted by medial-axis transformation and were used to measure pore geometry from one of the scanned samples. Water retention and unsaturated hydraulic conductivity values were calculated from the soil image. Properties of soil bulk density, water retention and unsaturated hydraulic conductivity were measured from three replicates of scanned soil samples. 3D image analysis provided accurate detailed pore properties such as individual pore volumes, pore length, and tortuosity of all pores. These data made possible to calculate accurate estimations of water retention and hydraulic conductivity. Combination of the calculated and measured hydraulic properties gave more accurate information on pore sizes over wider range than measured or calculated data alone. We could conclude that the hydraulic property computed from soil images and laboratory measurements can describe a full structure of intra- and inter-aggregate pores in soil.

Effect of Soil Environment on Diversity and Population of Aerobic Soil Bacteria from Baekdudaegan Mountain Forests in Gyeongsangbuk-do, Korea (경상북도 산림지역의 토양 환경이 호기성 토양 세균의 다양성과 밀도에 미치는 영향)

  • This study was carried out to compare species diversity of soil bacteria from Baekdudaegan mountain forests (Bonghwa-gun, Mungyeong-si and Sangju-si) in Gyeongsangbuk-do and to analyze the effects of soil environments on diversity and population of soil bacteria. Soil bacteria were isolated from soil samples by streak plate method, and identified by DNA extaction and 16S rDNA sequence analyses. The population of soil bacteria from the soil samples of Bonghwa-gun was the highest with $5.1{\times}10^5cfu/g$, and followed by those from Mungyeong-si and Sangju-si with $1.9{\times}10^5cfu/g$ and $1.1{\times}10^5cfu/g$, respectively. The population of soil bacteria from surface layer soil was the highest, and then gradually decreased according to soil depth. The increase in population of soil bacteria from soil samples of different sites was correlated with the increase of the altitude of soil sampling site, depth of A horizon, liquid phase among three phases of soil, water content and bulk density of soil. Two hundreds and sixty eight bacterial colonies from Bonghwa-gun were classified into 10 species, 8 genera. One hundred and thirty four bacterial colonies from Mungyeong-si were classified into 15 species, 9 genera. Forty four bacterial colonies from Sangju-si were classified into 5 species, 2 genera. The dominant species (occupancy rate) from Bonghwa-gun and Mungyeong-si were Bacillus weihenstephanensis (36% and 40%, respectively), and Sangju-si was Bacillus cereus (39%). The relationships between soil environment and community structure of soil bacteria were analyzed statistically by using ecological indices. The diversity, evenness and dominance indices of soil bacteria were 6.30, 2.04 and 0.59 in Bonghwa-gun, 9.09, 2.94 and 0.51 in Mungyeong-si, and 4.55, 2.34 and 0.71 in Sangju-si, respectively. The diversity and evenness indices were increased by the increase of water content, drainage condition and gravel content of soil, while the dominance index was decreased.

GPU Based Feature Profile Simulation for Deep Contact Hole Etching in Fluorocarbon Plasma

  • Recently, one of the critical issues in the etching processes of the nanoscale devices is to achieve ultra-high aspect ratio contact (UHARC) profile without anomalous behaviors such as sidewall bowing, and twisting profile. To achieve this goal, the fluorocarbon plasmas with major advantage of the sidewall passivation have been used commonly with numerous additives to obtain the ideal etch profiles. However, they still suffer from formidable challenges such as tight limits of sidewall bowing and controlling the randomly distorted features in nanoscale etching profile. Furthermore, the absence of the available plasma simulation tools has made it difficult to develop revolutionary technologies to overcome these process limitations, including novel plasma chemistries, and plasma sources. As an effort to address these issues, we performed a fluorocarbon surface kinetic modeling based on the experimental plasma diagnostic data for silicon dioxide etching process under inductively coupled C4F6/Ar/O2 plasmas. For this work, the SiO2 etch rates were investigated with bulk plasma diagnostics tools such as Langmuir probe, cutoff probe and Quadruple Mass Spectrometer (QMS). The surface chemistries of the etched samples were measured by X-ray Photoelectron Spectrometer. To measure plasma parameters, the self-cleaned RF Langmuir probe was used for polymer deposition environment on the probe tip and double-checked by the cutoff probe which was known to be a precise plasma diagnostic tool for the electron density measurement. In addition, neutral and ion fluxes from bulk plasma were monitored with appearance methods using QMS signal. Based on these experimental data, we proposed a phenomenological, and realistic two-layer surface reaction model of SiO2 etch process under the overlying polymer passivation layer, considering material balance of deposition and etching through steady-state fluorocarbon layer. The predicted surface reaction modeling results showed good agreement with the experimental data. With the above studies of plasma surface reaction, we have developed a 3D topography simulator using the multi-layer level set algorithm and new memory saving technique, which is suitable in 3D UHARC etch simulation. Ballistic transports of neutral and ion species inside feature profile was considered by deterministic and Monte Carlo methods, respectively. In case of ultra-high aspect ratio contact hole etching, it is already well-known that the huge computational burden is required for realistic consideration of these ballistic transports. To address this issue, the related computational codes were efficiently parallelized for GPU (Graphic Processing Unit) computing, so that the total computation time could be improved more than few hundred times compared to the serial version. Finally, the 3D topography simulator was integrated with ballistic transport module and etch reaction model. Realistic etch-profile simulations with consideration of the sidewall polymer passivation layer were demonstrated.

Prediction of Dietary Protein-Energy Balance by Milk Urea Nitrogen and Protein Contents in Dairy Cow (젖소의 우유 중 단백질과 요소태질소 측정에 의한 사료의 에너지와 단백질 균형 상태 예측)

  • Milk urea nitrogen (MUN) and Milk protein (MP) are being used as indicators of the protein-energy balance and for actual farm feeding practices. The purpose of this study was to investigate the MUN and MP concentrations of individual cows and bulk tank milk to evaluate the dietary protein-energy balance from lactating Holstein cows. Mean MUN and MP concentrations in the milk samples obtained from 132,636 cows of 4,856 herd during Jan. 1999 to Dec. 2001 were 16.2 5.2mg/dl and 3.30 0.35%, respectively. The highest values were found during summer and lowest valued during winter in MUN. But, the average contents of MP were the highest during winter and the lowest during summer. In order to evaluate protein-energy balance for feeding, we set the level of recommended MP range of 2.90${\sim}$3.29% in early lactation considering a negative energy balance. The recommended level of MP in mid-lactation and late lactation were set as 3.10${\sim}$3.49%, and 3.30${\sim}$3.69%, respectively. Recommended MUN range of 12${\sim}$18 mg/dl was determined through the whole lactation period. Individual cows milk were analyzed by the 9 types based on this levels of MP and MUN in this study. Among the total cows investigated, 26.8%, 25.8%, and 22.2% have shown the recommended criteria of MP and MUN values, respectively. Also, of total herds surveyed, 11.6% had MUN values lower than 12.0 mg/dl and 32.9% had values higher than 18.0 mg/dl and 44.5% of total herd have not met with the recommended criteria of MP values in bulk tank milk. In case of MP, out of the total herd surveyed, 26.0% had MP values lower than 3.10% and 24.0% had values higher than 3.30% and 50.0% had MP values outside the recommended interval (3.1${\sim}$3.3%). This study has indicates that many dairy farms are under improper feeding management practice of the dietary protein-energy balance.

Physico-Chemical Properties of Organically Cultivated Upland Soils (유기농경지 밭 토양의 물리화학적 특성)

  • The upland soils (56 samples) from organic farms in Gyeonggi-do (12 sites), Gangwon-do (8 sites), Chungcheong-do (14 sites), Gyeongsang-do (4 sites), Jeollado (18 sites) in Korea were collected and their physical and chemical properties were analyzed by RDA's methods. In the results of physical property, the bulk density of soils averaged $1.14Mgm^{-3}$ (surface soil), $1.38Mgm^{-3}$ (subsoil), respectively. The porosity of them was 57%, 48%. Organically managed soil's (OS) bulk density was lower than conventional soil's but OS's porosity was a little higher than conventionally managed soil in surface soil. The depth of plough layer in organically managed soils was 21.2 cm indicating that the organic farming had good effect on soil physical property. In the results of chemical property, the surface soil pH was 6.9 and the contents of organic matter (OM) was $26gkg^{-1}$, available phosphate (Avail. $P_2O_5$) was $554mgkg^{-1}$, exchangeable calcium (Exch. Ca) was $8.9cmol_ckg^{-1}$, exchangeable potassium (Exch. K) was $0.89cmol_ckg^{-1}$, exchangeable magnesium (Exch. Mg) was $2.0cmol_ckg^{-1}$. The subsoil pH was 6.8 and the contents of OM was $21gkg^{-1}$, avail. $P_2O_5$ was $491mgkg^{-1}$, exch. Ca was $7.9cmol_ckg^{-1}$, exch. K was $0.68cmol_ckg^{-1}$, exch. Mg was $1.8cmol_ckg^{-1}$. The nutrient accumulation emerged in organic farming. Compared to the optimum nutrient range for the conventional upland soils, the exceed rate of pH, OM, available phosphate, and exchangeable Ca, K, and Mg was 79, 52, 64, 84, 66% and 55%, respectively, which mainly resulted from the over-application of lime materials or livestock manure compost. With these results it is suggested that organic farm need to reduce the use of inputs, which make soil alkalification or nutrient accumulation. More study on effects of inputs on lowering soil pH from alkalification could help organically managed soil to be improved.

Soil Properties of Chestnut (Castanea crenata) Stands by Regions in Gyeongnam Province (경상남도 밤나무임지의 지역별 토양특성)

  • This study was carried out to evaluate soil properties by regions from chestnut (Castanea crenata Sieb. et Zucc) stands in Gyeongnam province. Soil physical and chemical properties were measured from soil samples of top 20 em collected from three hillslopes (upper, middle, lower) of the chestnut stands in six regions (Jinjusi, Sacheonsi, Sancheonggun, Hadonggun, Goseonggun, Hapcheongun) where are major chestnut cultivation areas throughout the province. Soil properties were significantly different among regions (p<0.05), while were not significantly different among hillslopes (p>0.05). Soil bulk density, soil pore space, soil pH, organic matter content, total nitrogen, available phosphorus, and CEC were significantly different among regions (p<0.05). Soil bulk density was significantly lower (p<0.05) in Hadonggun ($0.96g/cm^3$ than in other regions ($1.12{\sim}1.22g/cm^3$). Soil pH was below pH 5.03 in most regions and Sancheonggun showed the lowest soil pH value (pH 4.62), followed by Jinjusi, Hadonggun, Hapcheongtm, Goseonggun, and Sacheonsi. Organic matter content was highest in Hadonggun (6.46%), while other regions ranged between 2.93% and 3.47%. Total nitrogen content showed a similar trend like the organic matter content. Available phosphorus was above 100 ppm in Jinjusi, Hadonggun and Sancheonggun, but Sacheonsi showed the lowest concentration (15 ppm) among the regions. Cation Exchange Capacity (CEC) was above 10 cmolc/kg in Goseonggun and Hadonggun, but below 8.6 cmolc/kg in Jinjusi and Hapcheongtm. Potassium content ranged between 0.07 and 0.14 cmolc/kg, and magnesium was above 0.66 cmolc/kg in all regions. The results indicate that soil property in chestnut stands was different among regions in Gyeongnam province. This suggested that the chestnut stands should be managed by the fertilization application reflected in the variability of regional soil property in chestnut stands.

Compare Physicochemical Properties of Topsoil from Forest Ecosystems Damage patterns (산림생태계 훼손 유형별 표토의 이화학적 특성 비교)

  • This study was carried out to evaluate the physicochemical properties of different types of topsoil in forest ecosystems by damage pattern and analyse the possibility of using the topsoil as a planting ground construction material. There were 72 samples from 36 sites of 12 damaged areas and 36 sites of 12 non-damaged areas. The results showed that the physicochemical properties of topsoil from non-damaged areas of forest ecosystems were on an average clay loam~sandy loam in soil texture, showing $0.95{\sim}1.10Mg/m^3$ in soil bulk density, $35.7{\sim}44.0m^3/m^3$ in solid phase, 56.0~64.3 in soil porosity, 8.4~35.8% in aggregate stability, 5~13 mm in soil hardness, 5.3~6.1 in pH, 0.14~0.65 dS/m in EC, 0.28~0.42% in T-N, $14{\sim}22cmol^+/kg$ in CEC, $0.15{\sim}0.31cmol^+/kg$ in Ex. $K^+$, $2.07{\sim}2.84cmol^+/kg$ in Ex. $Ca^{2+}$, $0.45{\sim}1.97cmol^+/kg$ in Ex. $Mg^{2+}$, 17~96 mg/kg in Av. $P_2O_5$ and 3.2~5.6% in OM. On the other hand, damaged areas were on an average clay loam~loamy sand in soil texture, showing $1.54{\sim}1.75Mg/m^3$ in soil bulk density, $52.8{\sim}58.0m^3/m^3$ in solid phase, 42.0~47.2 in soil porosity, 4.2~22.5% in aggregate stability, 13~25 mm in soil hardness, 4.8~5.5 in pH, 0.13~0.62 dS/m in EC, 0.02~0.12% in T-N, $5{\sim}15cmol^+/kg$ in CEC, $0.11{\sim}0.18cmol^+/kg$ in Ex. $K^+$, $0.45{\sim}2.36cmol^+/kg$ in Ex. $Ca^{2+}$, $0.39{\sim}0.96cmol^+/kg$ in Ex. $Mg^{2+}$, 15~257 mg/kg in Av. $P_2O_5$ and 0.4~2.2% in OM. After conducting a comparison of physicochemical characteristics of non-damaged forest area and damaged areas, it was found that the physicochemical characteristics of damaged areas were more deteriorated compared to that of non-damaged areas. Therefore, it is judged that it is necessary to establish countermeasures for the conservation and management of the damaged areas for topsoil recycling in the future.

Soil Physical and Chemical Properties of Kaolinite Opencast Mines and Adjacent Red Pine Forests in Sancheong-gun (산청군 고령토(백토) 노천 광산 채굴지와 인접 소나무 임분의 토양 물리·화학적 성질)

  • Soil properties in opencast mines are a key factor in reclamation (revegetation) of mining areas. In this study we determined the soil physical and chemical properties of kaolinite tailings, reclaimed areas, and adjacent natural red pine (Pinus densiflora S. et Z.) forests in kaolinite opencast mines in Sancheong-gun, Gyeongsangnam-do. Six sites were chosen for collection of soil samples to determine soil physical and chemical properties at a soil depth of 10 cm. Soil bulk density was significantly higher (P < 0.05) in the kaolinite tailings (1.51 g·cm-3) than in the reclaimed areas (1.19 g·cm-3) and red pine forests (0.93 g·cm-3), whereas air phase in the kaolinite tailings (14.2%) was significantly lower than in the red pine forests (32.6%). Clay content in the red pine forests was significantly higher than in the reclaimed areas (18.7%) or kaolinite tailings (14.8%), whereas soil structural stability index was significantly lower in the reclaimed areas (1.61%) and kaolinite tailings (0.87%) than in the red pine forests (7.75%). Soil pH was significantly higher in the kaolinite tailings (pH 6.68) and reclaimed areas (pH 6.27) than in the red pine forests (pH 5.31). Soil organic carbon and total nitrogen were significantly higher in the red pine forests (C: 36.03 mg·g-1; N: 2.08 mg·g-1) than in the reclaimed areas (C: 5.00 mg·g-1; N: 0.31 mg·g-1) than in the kaolinite tailings (C: 2.12 mg·g-1; N: 0.07 mg g-1). The amount of available phosphorus was not significantly different among the three treatments. The concentration of exchangeable potassium was significantly lower in the kaolinite tailings (0.08 cmolc·kg-1) than in the reclaimed areas (0.21 cmolc·kg-1) and red pine forests (0.30 cmolc·kg-1). These results indicate that, because of high soil bulk density and low soil organic carbon, total nitrogen, available phosphorus, and exchangeable potassium in kaolinite tailings and reclaimed mining areas, soil nutrient management is needed in order to reclaim the vegetation in these type of areas.

Development of Correction Formulas for KMA AAOS Soil Moisture Observation Data (기상청 농업기상관측망 토양수분 관측자료 보정식 개발)

  • Soil moisture data have been collected at 11 agrometeorological stations operated by The Korea Meteorological Administration (KMA). This study aimed to verify the accuracy of soil moisture data of KMA and develop a correction formula to be applied to improve their quality. The soil of the observation field was sampled to analyze its physical properties that affect soil water content. Soil texture was classified to be sandy loam and loamy sand at most sites. The bulk density of the soil samples was about 1.5 g/cm3 on average. The content of silt and clay was also closely related to bulk density and water holding capacity. The EnviroSCAN model, which was used as a reference sensor, was calibrated using the self-manufactured "reference soil moisture observation system". Comparison between the calibrated reference sensor and the field sensor of KMA was conducted at least three times at each of the 11 sites. Overall, the trend of fluctuations over time in the measured values of the two sensors appeared similar. Still, there were sites where the latter had relatively lower soil moisture values than the former. A linear correction formula was derived for each site and depth using the range and average of the observed data for the given period. This correction formula resulted in an improvement in agreement between sensor values at the Suwon site. In addition, the detailed approach was developed to estimate the correction value for the period in which a correction formula was not calculated. In summary, the correction of soil moisture data at a regular time interval, e.g., twice a year, would be recommended for all observation sites to improve the quality of soil moisture observation data.
