Legumin Accumulation in Endoplasmic Reticulum Cisternae at Early Stage of Seed Development and Protein Body Transformation in Pea Cotyledon Cells (완두의 종자 발달과정에서 소포체 내강에 대한 저장 단백질 legumin의 축적과 단백과립 변환)

  • Immunoelectron microscopy of storage protein at early stage of seed development showed legumin was firstly accumulated protein in between endoplasmic reticulum (ER) cisternae, and these accumulates were differentiated into protein body (PB) by transformation at later stage. Thin sections of pea cotyledons during the later stages of seed maturation showed three morphologically different types of protein bodies. One of these, presented as rough-surfaced cisternae with terminal dilations, which contained protein deposits and were often found interdigitated between stacks of rough endoplasmic reticulum. Conventional electron microscopy at earlier stages of cotyledon development showed this protein body type initially developed from the rough ER. This transformation of endoplasmic reticulum into a protein body is believed to represent a new pathway of protein body development.

Vegetation Change after A Forest Fire in a Rural Japanese Red Pine Forest and Applications of Effective Microorganism (농촌 소나무림에서 산불에 의한 식생변화와 유용미생물의 적용)

  • This study examined the natural restoration of vegetation through monitoring of the development of a vegetation community from 2006 through 2007 after a forest fire. Approximately 5,000 $m^2$ in a forest near Topyeon-ri, Kangnae-myeon, Chungcheongbuk-do with Japanese red pine (Pinus densiflora) forest and its floor vegetation had been completely burned by a fire in April 2005. This area and another nearby Japanese red pine forest were selected as the experiment site and the control site, respectively. Vegetation survey was conducted at the experiment site and the control site. A seed bank experiment was carried out in the greenhouse to examine underground vegetation. Effective microorganism(EM) was applied to the seed bank experiment to estimate its effects on the direction of ecological succession. According to the results, a total of 36 plant species including shrub and herbaceous species were discovered in the experiment site. Quercus serrata, Lespedeza cyrtobotrya, and Castanea crenata, Rubus crataegifolius, Oplismenus undulatifolius, and Carex lanceolata were among the most abundant species. Biomass in the experiment site reached 2.4 times biomass than those in the control site, indicating the productivities of shrub and herbaceous layers are better in the experiment site. According to the result of the soil seed bank experiment of the experiment site, a total of 182 plants of 14 species were recorded. In addition, a total of 13 plants of 2 species were found from soil seed bank of the experiment site applied by EM. If EM is applied to the burned site, it will control the budding of herbaceous plants, creating the gap between herbaceous plants. This loss of competition is expected to help the restoration of trees in the burned area.

As a result, must idealize initial deflection that occurrence is possible to endow stability and accuracy in the hull structure or marine structure and reflect in early structure design considering secondary buckling. Longi direction of compressive load interacts and analyzed finite element series analysis that apply various kinds initial deflection shape measured actually on occasion that is arranged simply supported condition in this research. Applied ANSYS (elasto-plasticity large deformation finite element method) to be mediocrity finite element program for analysis method and analysis control used in Newton-Raphson method & Arc-length method.

Effect of Environmental Stress on Morphological Change of an Extremely Cadmium-Tolerant Yeast, Hansenula anomala B-7

  • An extremely cadmium-tolerant budding yeast, Hansenula anomala B-7 underwent a morphological switch in response to either heat shock treatment or cadmium stress, respectively. It exhibited a morphological transition from a unicellular yeast form to a pseudohyphae-like coagulation when subjected to prolonged heat shock treatment. In contrast, the yeast cells showed an irregularity in surface morphology when given thermal stress for a short time. Patterns of proteins expressed in the pseudohyphae-like cells demonstrated that several proteins were overexpressed while others were underexpressed in comparison with those prepared from the cells in the yeast form. It was a striking feature, however, that nearly 40% of the proteins extracted from the cells in the pseudohyphae form appeared to be composed of a single polypeptide. This polypeptide was apparently overexpressed during the pseudohyphae phase and its molecular weight was estimated to be 58 kDa according to SDS-PAGE analysis. However, a significant level of the protein was not observed in the cells before transition to pseudohyphae. The architecture of the cell shape was also damaged when incubated in a medium containing more than 1,000 ppm (8.9mM) of cadmium ions, although able to proliferate at a slow rate. However, the irregularity in the cell morphology exerted either by the brief heat shock treatment or by the cadmium stress with the high concentrations of the metal ions was not repaired, even though the damaged cells were allowed to grow for sufficient time in fresh, cadmium-free medium.

  • Saccharomyces cerevisiae Hsp30 is a plasma membrane heat shock protein that is induced by various environmental stress conditions. However, the functional role of Hsp30 during diverse environmental stressors is not presently known. To gain insight into its function during thermal stress, we have constructed and characterized a ${\Delta}hsp30$ strain during heat stress. $BY4741{\Delta}hsp30$ cells were found to be more sensitive compared with BY4741 cells, when exposed to a lethal heat stress at $50^{\circ}C$. When budding yeast is exposed to either heat shock or weak organic acid, it inhibits Pma1p activity. In this study, we measured the levels of Pma1p in mutant and Wt cells both during optimal temperature and heat shock temperature. We observed that $BY4741{\Delta}hsp30$ cells showed constitutive reduction of Pma1p. To gain further insights into the role of Hsp30 during heat stress, we compared the total protein profile by 2D gel electrophoresis followed by identification of differentially expressed spots by LC-MS. We observed that contrary to that expected from thermal-stress-induced changes in gene expression, the ${\Delta}hsp30$ mutant maintained elevated levels of Pdc1p, Trx1p, and Nbp35p and reduced levels of Atp2p and Sod1p during heat shock. In conclusion, Hsp30 is necessary during lethal heat stress, for the maintenance of Pma1p and a set of thermal stress response functions.

Functional Analysis of the Putative BUB2 Homologues of C. elegans in the Spindle Position Checkpoint

  • Spindle position checkpoint monitors the orientation of mitotic spindle for proper segregation of replicated chromosomes into mother cell and the daughter, and prohibits mitotic exit when mitotic spindle is misaligned. BUB2 forms one of the key upstream element of spindle position checkpoint in budding yeast, but its functional homologues have not been identified in higher eukaryotes. Here, we analyzed the functions of two putative BUB2 homologues of C. elegans in the spindle orientation checkpoint. From the C. elegans genome database, we found that two open reading frames (ORFs), F35H12_2 and C33F10_2, showed high sequence homology with BUB2. We obtained the expressed sequence tag (EST) clones for F35H12_2 (yk221d4) and C33F10_2 (yk14e10) and verified the full cDNA for each ORF by sequencing and 5' RACE with SL1 primer. The functional complementation assays of yk221d4 and yk14e10 in ${\Delta}bub2$ of S. cerevisiae revealed that these putative BUB2 homologues of C. elegans could not replace the function of BUB2 in spindle position checkpoint and mitotic exit. Our attempt to document the component of spindle position checkpoint in metazoans using sequence homology was not successful. This suggests that structural information about its components might be required to identify functional homologues of the spindle position checkpoint in higher eukaryotes.

Depletion of ${\varepsilon}$-COP in the COPI Vesicular Coat Reduces Cleistothecium Production in Aspergillus nidulans

  • We have previously isolated ${\varepsilon}$-COP, the ${\alpha}$-COP interactor in COPI of Aspergillus nidulans, by yeast two-hybrid screening. To understand the function of ${\varepsilon}$-COP, the $aneA^+$ gene for ${\varepsilon}$-COP/AneA was deleted by homologous recombination using a gene-specific disruption cassette. Deletion of the ${\varepsilon}$-COP gene showed no detectable changes in vegetative growth or asexual development, but resulted in decrease in the production of the fruiting body, cleistothecium, under conditions favorable for sexual development. Unlike in the budding yeast Saccharomyces cerevisiae, in A. nidulans, over-expression of ${\varepsilon}$-COP did not rescue the thermo-sensitive growth defect of the ${\alpha}$-COP mutant at $42^{\circ}C$. Together, these data show that ${\varepsilon}$-COP is not essential for viability, but it plays a role in fruiting body formation in A. nidulans.

Granulation Characteristics of Mono-granular NPK(10-0-30) Fertilizer Incorporated with Rock-Phosphate Powder and its Effects on Tobacco Plant (인광석분말을 증량제로 사용한 연초(煙草)재배용 복합비료(10-0-30)의 조립(造粒)특성 및 비효)

  • Fertilizer granulation test was carried out by a small pan granulator. A premixture composed of SOP 60%, urea 22% and RP powder 18% was rolled in the pan granulator while 10% phosphoric acid solution(binder) was sprayed on the rolling powder bed. Granules were developed very fast along with a little amount of binder. Hardness, brittle ratio in water and hygroscopicity of granules were improved enough to evaluate physical properties of the fertilizer. Growth responses of tobacco plant to the fertilizer were investigated at seedling and flowering stage by pot experiment under plastic film roof. Seedlings showed poor growth at nursery pot cell. In virgin soil with deficient available phosphate tobacco plant showed poor growth until budding and flowering stage but good growth in tillage soil with high cumulative phosphate.

Expression pattern of floral scent genes in different flowering stages of Chrysanthemum cultivars

  • Among the various volatile organic compounds (VOCs) emitted by the plant, floral scent plays a key role in attracting pollinators for reproduction and mediates ecological interactions. Floral scent is an important trait and industry drives the competition for flowers with novel scents. Chrysanthemum is one of the well-known ornamental plants and is a popular cut flower across the world. Floral scent and the genes responsible for the floral scent emission are poorly studied in chrysanthemum. In the present study, floral scent and the expression pattern of floral scent genes were analyzed in two chrysanthemum cultivars 'Golden Egg' and 'Gaya Glory'. Initially, intensity of the floral scent in five developing stages of flower including 'budding (B), bud developing (BD), initial blooming (IB), almost open (AO) and open flower (OF)' was analyzed using electronic nose (E-nose) with six metal oxide sensors. Based on the distance analysis, different stages of flower showed different relative intensity of scent according to the sensory evaluation. Although the scent pattern differed by stage, scent intensity was strongest in the OF stage in the completely opened flower in both the cultivars. Further, expression pattern of six genes in the floral scent pathway including FDS, IDI, ISPH, TPS2, TPS5 and TPS6 was observed in all the five stages of the flower in both the cultivars. The expression pattern of all the six genes differed by stage and the terpene synthase genes TPS2, TPS5 and TPS6 showed good expression levels in the $5^{th}$ flower stage compared to other stages. This study provides a preliminary data for understanding the regulation of floral scent in chrysanthemum.
