First Report of Leaf Spot Caused by Alternaria tenuissima on Black Chokeberry (Aronia melanocarpa) in Korea

  • In July 2015, diseased leaves of black chokeberry (Aronia melanocarpa) were observed in Danyang and Gochang, Korea. The symptoms appeared as circular or irregular brown leaf spots, from which Alternaria tenuissima was isolated. The isolates were cultured on potato dextrose agar, and their morphological characteristics were observed under a light microscope. The colonies were whitish to ash colored. The pathogenicity test on healthy black chokeberry leaves produced circular brown spots, in line with the original symptoms. Molecular analyses of the ITS, GPD, RPB2, and TEF genes were conducted to confirm the identity of the pathogen. The phylogeny of the multi-gene sequences indicated that the causal agent was A. tenuissima. This study is the first report of A. tenuissima leaf spot on black chokeberry (A. melanocarpa).

Occurrence of Stem Rot of Petunia hybrida Caused by Sclerotium rolfsii (Sclerotium rolfsii에 의한 페튜니아흰비단병 발생)

  • From 2006 to 2008, the stem rot of Petunia hybrida Vilm. caused by Sclerotium rolfsii occurred sporadically at Jinju city in Gyeongsangnam-do. The typical symptom is water-soaking, brown on the stem and rotting, wilting and blighting. The infected plants were mostly died. White mycelial mats were spread over lesions, and then sclerotia were formed on stem and near soil line. The sclerotia were globoid in shape, $1{\sim}3\;mm$ in size and white to brown in color. The optimum temperature for mycelial growth and sclerotia formation was at $30^{\circ}C$ on PDA and the hyphal width ranged from 4 to $8{\mu}m$. The typical clamp connections were observed in the hyphae of the fungus. On the basis of mycological characteristics and pathogenicity to P. hybrida, this fungus was identified as Sclerotium rolfsii Saccardo. This is the first report on the stem rot of P. hybrida caused by S. rolfsii in Korea.

Incidence of Diseases in Codonopsis lanceolata with Different Cultivation Method (재배양식에 따른 더덕 병해 발생양상)

  • Disease incidence of Codonopsis lanceolata was surveyed at the major cultivating fields in Chonbuk province in 1996 to 1997. The main diseases of Codonopsis lanceolata were ovserved as leaf spot caused by Septoria codonopsis, anthracnose by Glomerella cingulata, brown leaf spot by Cercospora sp., rust by Coleosporium koreanum, powdery mildew by Erysiphe sp., Fusarium wilt caused by Fusarium oxyporum, and white root rot by Sclerotium rolfsii. Anthracnose, leaf spot and brown leaf spot occurred severely on leaves from early July to late August. They were caused early fallen leaves. Fusarium wilt and white root rot occurred severely on stem and below the soil line in late August. They resulted in withering to death or chlorosis and fallen of leaves. Disease incidence of Codonopsis lanceolata was also substantially different in occurrence with a method of cultivation in late growth stage. Fusarium wilt and white root rot were more severe with a method of no support cultivation than those with a method of support cultivation with a stick. Fusarium wilt occurred 48.8% in a method of no support cultivation but 3.1% in a method of support cultivation with a stick. And white root rot occurred 18.9% in a method of no support cultivation but 0.3% in a method of no support cultivation with a stick. Thus, it proved that soil-borne diseases could be controlled support cultivation with a stick.

The CO outflow survey toward the Very Low Luminosity Object candidates: a progress report

  • We present the preliminary results of CO outflow survey toward the 56 Very Low Luminosity Object (VeLLO) candidates at CO J=2-1 and J=3-2 transitions with two radio telescopes of the Caltech Submillimeter Observatory (CSO) and the Atacama Submillimeter Telescope Experiment (ASTE). The survey is aimed to understand the origin of the formation of low-mass stars or substellar objects. The VeLLO is a very faint (${\leq}0.1$ $L_{\odot}$) object deeply embedded in dense molecular clouds and believed to be a proto-brown dwarf which will be a brown dwarf or a faint protostar which has just formed with little mass accretion or which is in quiescent stage of episodic accretion. The candidates were searched for over all nearby ($d{\leq}450$ pc) Gould belt clouds and listed in a new catalogue of the VeLLO candidates by Kim et al. (2014 submitted). To diagnose present status and future fate of the VeLLOs, we conducted a systematic observation for the CO molecular outflows of the 56 VeLLOs to infer how accretion is being made around the VeLLOs. We found 17 VeLLO candidates either having a prominent wing in line profiles or showing bipolar intensity distribution of high velocity components. We will discuss the physical properties of these CO outflows and the identity of the VeLLO candidates.

First Record of the Oblique-banded Grouper, Epinephelus radiatus (Perciformes: Serranidae) from Korea (한국산 바리과(family Serranidae) 어류 1 미기록종, Epinephelus radiatus)

  • A single serranid specimen of Epinephelus radiatus was collected by a hook for the commercial longline fisheries occurred near Marado, Jeju Island, Korea. The present specimen was characterized by five irregular dark brown bands passing downward and forward from upper edge of body, scales in longitudinal row 107, and pored lateral line scales 55. This species is easily distinguishable from the morphologically similar Korean serranid species of E. poecilonotus based on band patterns on body. That is, the former has five irregular oblique dark-edged brown bands, and the latter has several long horizontal bands on lateral body. We propose a new Korean name, "Ma-ra-bari," for Epinephelus radiatus.

  • Two specimens of Upeneus tragula were firstly collected from Jeju Island, Korea. This species is characterized by having pectoral fin rays 13, lateral line 29, first gill rackers 5+14, both lobes of caudal fin with dark bands, ventral part of body with numerous blackish-brown dots, and upper part of first and second dorsal fin dark brown. It is easily distinguished from three other Korean Upeneus species in having both lobes of caudal fin with dark bands (vs. no band in U. sulphureus; only upper lobe with dark bands in both U. moluccensis and U. japonicus). We suggest the new Korean name "Keom-eun-jul-chok-su" for this species.

Identification and genetic characterization of bacterial isolates causing brown blotch on cultivated mushrooms in Korea

  • Fluorescent bacteria were isolated from sporocarps that browned into various mushrooms during survey at places of the production in Korea. We examined the pathogenicity, biodiversity, and genetic characteristics of the 19 strains identified as Pseudomonas tolaasii by sequence analysis of 16S rRNA and White Line Assay. The results emphasize the importance of rpoB gene system, fatty acid profiles, specific and sensitive PCR assays, and lipopeptide detection for the identification of P. tolaasii. As a result of these various analyses, 17 strains (CHM03~CHM19) were identified as P. tolaasii. The phylogenetic analysis based on the 16S rRNA gene showed that all strains were clustered closest to P. tolaasii lineage, two strains (CHM01, CHM02) were not identified as P. tolaasii and have completely different genetic characteristics as a result of fatty acids profile, specific and sensitive PCR, lipopetide detection, rpoB sequence and REP-PCR analysis. Pathogenicity tests showed 17 strains produce severe brown discolouration symptoms to button mushrooms and watersoaking of sporophore tissue within three days after inoculation. But two strains did not produce discolouration symptoms. Therefore, these two strains will be further investigated for correct species identification by different biological and molecular characteristics.

Comparison of pH, Water Holding Capacity and Color among Meats from Korean Native Chickens

  • This study investigated the initial pH measured at 15 min post-mortem, ultimate pH, water holding capacity (WHC), and color of meats obtained from five lines of Korean native chicken (KNC) for the development of breed for high-quality meat. In addition, the effect of sex was examined. In total, 595 $F_1$ progeny (Black [B], Grey-Brown [G], Red-Brown [R], White [W] and Yellow-Brown [Y]) from 70 full-sib families were used. Chickens were slaughtered at 20 wk of age and the measurement traits of all breast and thigh meats from 595 chickens were analyzed. The initial pH at 15 min post-slaughter of the breast and thigh meats was affected by the line and sex of Korean native chicken (P<0.05). However, there was no line and sex effect on ultimate pH and WHC of the breast and thigh meat except that the ultimate pH of thigh meat had line effect. Except for the $L^*$ value of breast meat, the meat color was significantly affected by line and sex (P<0.05). The ultimate pH showed consistently negative correlation with the $L^*$ value and positive correlation with the $a^*$ value in breast and thigh meats. Based on the results, we concluded that the line W in male chickens and the line G and W in female chickens may be good candidates for the selection to develop breed for high-quality meat because these lines showed property of high initial pH or/and ultimate pH.

Expression of Steroidogenesis-related Genes in Rat Adipose Tissues

  • Adipose tissue is one of the major endocrine gland. More recently, local production of steroids in adipocytes differentiated from mouse 3T3-L1 cell-line was reported. We hypothesized that rat adipocytes have steroidogenic machinery and the expression patterns of the components might be differentially regulated, depending on the distribution and sex. To verify this hypothesis, we collected the adipose tissues depot-and sex-specifically at postnatal day (PND) 30, and performed quantitative RT-PCRs. In overall aspects, the abundances of the transcripts were lower in the brown adipose of both sexes. $3{\beta}-HSD$ transcript levels in female abdominal and reproductive adipose, CYP17 transcript levels in female reproductive adipose, $17{\beta}-HSD$ transcript levels in female abdominal and reproductive adipose, and CYP19 transcript levels in female abdominal adipose were significantly lower than those of male counterparts. Similar to steroidogenic factors, the abundance of the $ER-{\alpha}$ transcripts were generally lower in the brown adipose of both sexes. $ER-{\beta}$ transcripts were more abundant in male white adipose depots than their female counterparts. The levels of LHR transcripts in female reproductive adipose were significantly higher than those of male counterpart. In conclusion, our study demonstrated that the expressions of steroidogenesis-related genes were depot- and sex-specifically occurred in the immature male and female rat adipose tissues. Our study suggested that the adipose tissues are not only targets but de novo synthesizing sites of sex steroid(s), though the synthesizing activities could be much less than in gonads. Further researches in this field will be helpful for understanding the adipose physiology and for medical application such as sex-specific steroid supplement therapies for older populations.

Molecular Mapping of Resistant Genes to Brown Planthopper, Bphl and bph2, in Rice

  • This study was carried out to map Bphl and bph2 gene in Mudgo and Sangju13 (Oryza sativa L.) respectively conferring resistance to brown plan-thopper (BPH) and to establish the marker-assisted selection (MAS) system. Bulked seedling (grown for 20 days) test was conducted with the 73 F4 lines derived from a cross between Nagdongbyeo and Mudgo for Bphl and with 53 BC3F5 lines derived from the Milyang95/Sangju13 cross for bph2. Bph1 was mapped between RG413 and RG901 on chromo-some 12 at a distance of 7.5 cM from RG413 and 8.4 cM from RG90l. A recessive gene bph2 was located near RZ76 on chromosome 12 at a distance of 14.4 cM. Bphl and bph2 were linked to each other with a distance of about 30 cM. An RFLP marker, RG413 linked to Bphl, was converted to an STS marker to facilitate the marker-assisted selection. BPH resistant genotypes could be selected with 92% accuracy in a population derived from a line of NM47-B-B.
