Occurrence and Identification of the Fungus Causing Yellow Rot on Ganoderma lucidum (불로초버섯 노랑썩음병의 발생과 병원균의 분류 동정)

  • Since 1990, destructive yellow rots on Ganoderma lucidum caused by a soilborne fungus have been occurred in major cultivation areas of Korea. Incidences of the disease were 61% in Chulwon and 94% in Kanghwa area where the mushroom has been cultivated for 10 years, whereas the disease has not been found yet in new cultivation areas such as Moonkyung and Hongsung. when severely infected, inner tissues of bed-logs showed severe yellow and fruiting bodies of the mushroom was not produced. Infected tissues of bed-logs were readily distinguished from those of healthy ones by a distinctive brown border line. When the disease progressed, mycelia of Ganoderma lucidum were totally destroyed, and abundant ascocarps of the pathogen were formed inside the tissues of bed-logs showing yellowish green. The fungus derived from a single ascospore strongly lysed mycelia of Ganoderma lucidum growing on bottle media, and non-volatile components secreted by the pathogen were also highly inhibitory to mycelial growth of the mushroom fungus. The pathogen was identified as Arthrographis cuboidea based on its distinctive cultural and morphological characters. The fungus produced arthroconidia and unbrached conidiophores. The width of fungal conidia was distinctively wide as compared with the length. Colonies of the fungi were pale yellow to yellowish green on agar media. As a causal pathogen of yellow rot of Ganoderma lucidum., this fungus has not been reported yet in Korea.

  • This study was conducted to investigate the interaction of vitamins A and E on their transfer from diet to chicken eggs and the effect of vitamin A flood-dosing on its concentration in eggs and livers. In Experiment I, forty-two 45-wk-old brown layers (Bobeock) were divided into seven groups and fed one of seven diets: control, three vitamin A supplemented diets(8, OOO, 16, 000, and 64, 000 IU /kg diet) or three vitamin E supplemented diets (50, 100, and 200 IU/kg diet). In Experiment II, a total of thirty-two 35-wk-old white layers (Hy-ine) were divided into four groups and fed one of four diets :control, vitamin A 20, 00O+vitamin E 200 TU /kg, vitamin A 50, O00+vitamin E 200 IU /kg or vitamin A 100, OO0+vitamin E 200 lU/kg supplemented diets. In Experiment III, a total of fifty-six 35-wk-old white layers (Hy-line) was divided into four groups and fed one of four diets: control or three vitamin A supplemented diets (80, 000, 120, 000 and 160, 000 lU/kg diet). Breeding of a New Pink Gerbera hybrida 'Party Time' with Strong Peduncle for Cut Flower (단단한 화경의 핑크색 절화용 거베라 신품종 '파티타임' 육성)

  • A new pink gerbera (Gerbera hybrida Hort.) cultivar 'Party Time' was developed from a cross between 'Hope' and 'Sunnyeo', followed by seedling and line selections at the Flower Research Institute of Gyeongnam Agricultural Research and Extension Services (GARES) in 2007. Detailed characteristics of the new cultivar were evaluated from 2005 to 2007. 'Party Time' has semi-double type large flowers in a good harmony with pink (RHS, 65-C) ray floret and brown center. It has a good, stable flower shape and strong peduncles, and its vase life was 12.7 days. The average flower yield of 'Party Time' was about 47.3 stems per plant a year in greenhouse yield trails carried out 2005 to 2007. 'Party Time' (Grant No. 2691) was registered to the Korea Seed and Variety Service (KSVS) for commercialization in 2009.

A New Variety "Singang" with resistance to Soybean Mosaic Virus by Molecular Marker Assisted Selection (분자표지선발(MAS)을 이용한 콩모자이크병 저항성 신품종 "신강")

  • A new soybean variety, "Singang", was released by NICS, RDA in 2008. It is resistant to soybean mosic virus (SMV), which is one of the most serious diseases in soybean production. "Singang" had been developed for seven years by molecular marker-assisted selection (MAS) breeding method using recurrent parent, "Sowon", and donor parent, "L29" which has the SMV-resistance gene, Rsv3. Selected promising breeding line, Suwon 242, was evaluated in yield trials for 2 years from 2005 to 2006 at NICS. Regional yield trials were performed for 3 years from 2006 to 2008 in five locations including Jeju. Agronomic characteristics of "Singang" is a determinate growth habit, purple flower color, grey pubescence, yellow testa, light brown hilum and lanceolate leaflet shape. "Singang" showed the similar performance to "Sowon" cultivar. This variety showed 3 days shorter in days to maturity than the check variety (Pungsannamul). The average yield under regional yield trials in five location was 2.78t/ha. It is adaptable to the whole country except for Gangwon province.

Breeding of a New Yellow Gerbera hybrida "Golden Time" with High Yield for Cut Flower (황색 대륜 절화용 다수성 거베라 신품종 "골든타임" 육성)

  • A new yellow gerbera (Gerbera hybrida Hort.) cultivar "Golden Time" was developed from a cross between '99-ga-2' and '99-na-7-6', followed by seedling and line selections at the Flower Research Institute, Gyeongnam Agricultural Research and Extension Services (ARES) in 2007. Detailed characteristics of the new cultivar were evaluated from 2004 to 2007. "Golden Time" has semi-double type large flowers in a good harmony with yellow (RHS, 17-B) ray floret and brown center. It has good, stable flower shape and strong peduncles, and its vase life was 12.3 days. The average flower yield of "Golden Time" was about 49.5 stems per plant/year in greenhouse trails during 2004 and 2007. This cultivar was registered to the Korea Seed and Variety Service (KSVS) for commercialization in 2009. Year-round production of this cultivar is possible in the greenhouse condition throughout the Korean climate.

Effect of different sources and inclusion levels of dietary fat on productive performance and egg quality in laying hens raised under hot environmental conditions

  • Objective: This experiment aimed to investigate the effect of different sources and inclusion levels of dietary fat on productive performance and egg quality in laying hens raised under hot environmental conditions. Methods: A total of 480 Hy-Line Brown laying hens at 31 wk of age were randomly allotted to 1 of 5 experimental diets. The control diet contained 2,800 kcal/kg nitrogen-corrected apparent metabolizable energy with no fat addition. Four additional diets were prepared by adding 2.0% or 4.0% of animal fat (AF) or soybean oil (SO). Energy and nutrient concentrations were consistent among all diets. Diets were fed to hens for 4 weeks. Average daily room temperature and humidity were $26.7^{\circ}C{\pm}1.52^{\circ}C$ and $77.4%{\pm}4.50%$. The heat stress index was approximately 76, indicating that hens were raised under heat stress conditions. Results: Final body weight (BW) was greater (p<0.05) for hens fed diets containing 2.0% or 4.0% AF than for those fed the control diet or diets containing 2.0% or 4.0% SO. The BW gain and feed intake were greater (p<0.05) for hens fed diets containing additional AF or SO than those fed the control diet. Eggshell thickness was the greatest (p<0.05) for hens fed the control diet, but the least (p<0.05) for hens fed diets containing 4.0% SO. Egg yolk color was the greatest (p<0.05) for hens fed the control diet, but the least (p<0.05) for hens fed diets containing 4.0% SO. Conclusion: Inclusion of supplemental fat (AF and SO) in diets exhibits preventative effects on BW loss for hens raised under hot environmental conditions when energy and nutrient concentrations in diets were maintained. The effects were greater for AF than for SO. However, inclusion of supplemental fat in diets decreases eggshell thickness and egg yolk yellowness, possibly due to a reduction in Ca absorption and intake of egg yolk colorants.

'Hyowon6', a Mid-Late Maturing Rice Variety with High Quality (고품질 중만생 벼 품종 '효원6호')

  • Hyowon6, a mid-late maturing, good eating quality rice variety with multi-resistance, was developed by the rice breeding team of Pusan National University in 2013. This variety was derived from a cross between Hwayeong (as a multi-resistant parent) and Koshihikari (as a good eating quality parent). Selection by the pedigree breeding method was carried out until the $F_8$ generation. A promising line, JS14-12-36-8-5-3-1-1-1, was developed and designated as HY103 in 2013. This variety headed on August 22, which was 11 days later than that of Hwayeong. The culm length and panicle length of Hyowon6 was 85.7 cm and 21.1 cm, respectively. The number of panicles per hill was 14.5 and the number of grains per panicle was 101.7. The ratio of ripened grain was approximately 92% and the 1,000-grain weight was approximately 21.36 g for brown rice, which were similar to those of Hwayeong. Hyowon6 was moderately resistant to lodging and also to neck blast, leaf blight, and stripe virus. The glossiness value of Hyowon6 was 83, which was considerably higher than that of Hwayeong.

'Jungmo2509', the First Rye Cultivar of Self-Fertility in the Korea (국내 최초의 자식성 호밀 품종 '중모2509')

  • Rye (Secale cereal L.) is the most tolerant to abiotic stress including low temperature, drought, and unfavorable soil conditions among the winter cereals. Rye is the rapid growth of early spring results from increasing areas for the use of the forage and green manure in the middle part of Korea. "Jungmo2509", a rye cultivar was developed by the National Institute of Crop Science (NICS), RDA in 2014. It was developed from a cross between "Olhomil", a self-compatible cultivar, and "Synthetic II", a self-incompatible line. "Jungmo2509" is an erect plant type and of a middle size, with a green leaf color, a yellowish-white colored culm, and a yellowish brown-colored, small-size grain. The heading date of "Jungmo2509" was April 23, which was 5 days later than that of "Gogu", respectively. But "Jungmo2509" showed greater resistance to lodging compared to that of the check cultivar, with similar to winter hardiness, wet injury, and disease resistance. "Jungmo2509" was a higher to than "Gogu" in terms of protein content (9.4% and 8.0%, respectively), total digestible nutrients (TDN) (55.7% and 55%, respectively). The seed productivity of "Jungmo2509" was approximately 2.08 ton 10a-1, which was 11% lower than that of the check. Almost all rye cultivars are out-crossing due to genes controlling incompatibility, but "Jungmo2509" is higher seed fertility (56%) than that of Gogu (0%). it has self-compatible genes. "Jungmo2509" is erect in plant type and resistance for lodging. Therefore, "Jungmo2509" can produce uniform seeds for processed grains of human consumption and utilize them as parents for breeding the rye hybrids with high forage yields.

Anti-cancer effects of kelp extract in mouse melanoma B16-F0 cell line through apoptosis (마우스 흑색종 세포주 B16-F0에서 다시마 추출물의 세포사멸을 통한 항암 효과)

  • Kelp belongs to the brown algae family and has been reported to exert anti-cancer effects on some cancer types, however studies have not been reported on the anti-cancer effects of kelp extracts on melanoma. In this study, the anti-cancer effects of kelp extract in B16-F0 cells were investigated, and the underlying molecular mechanisms were assessed. Kelp extract was found to inhibit the proliferation of B16-F0 cells, induce cytotoxicity, inhibit cell colony formation, and induce DNA fragmentation and apoptosis. The molecular mechanism was found to involve kelp extract increasing the expression of cytochrome-c and activated caspase-9 in the intrinsic apoptotic pathway. In addition, kelp extract upregulated the expression of Fas-associated protein with death domain and activated caspase-8 in the extrinsic apoptosis pathway. Activation of caspase-9 and caspase-8 by kelp extract induced activation of caspase-3 and cleaved poly adenosine diphosphate-ribose polymerase, consequently inducing apoptosis. These data suggest that kelp extract represents a potential therapeutic agent for melanoma.

A Scientific Analysis of Decorative Metal Foil Used in Pouch for the Sutra Embroidered with a Sun and Moon Design Designated as National Folklore Cultural Heritage (국가민속문화재 일월수 다라니 주머니 금속 장식지의 과학적 분석)

  • Through scientific analysis, this study identified the material characteristics of metal foil decorating the border line and knotting of the National Folklore Cultural Heritage 'Pouch for the Sutra Embroidered with a Sun and Moon Design'. Through Scanning Electron Microscope-Energy Dispersive Spectroscopy results, it was estimated that silver (Ag) and sulfur (S) were present in the metal foil, and silver leaf was also attached to the medium. S may discolor Ag from yellow to black depending on its concsentration and contact time. Yellow color could not be identified in metal foil at present. But there existed an example of the preparation of a gold-colored flat silver thread; therefore, further research is needed to estimate the original color. The lamella was reddish brown on the back. Aluminum, silicon, and iron were also detected and were the main components found in red soil. This is believed to be the red adhesive in traditional flat gold thread and is considered to be an adhesive-related component of the metal foil. From the gas chromatography mass spectrometry results, the adhesive component was confirmed to be animal glue.
