국내산 해조류 4종의 물과 에탄올 추출물이 3T3-L1에서 지방세포 분화에 미치는 영향 (Effects of Water and Ethanol Extracts from Four Types of Domestic Seaweeds on Cell Differentiation in 3T3-L1 Cell Line)

  • The aim of this study was to examine the cytotoxicity and potential inhibitory effects from four types of edible domestic brown seaweeds, Undaria pinnatifida (UP), Laminaria japonica (LJ), Sargassum fulvellum (SF), and Hizikia fusiforme (HF), on preadipocyte differentiation in 3T3-L1 cell line. Water and ethanol extracts from the four types of seaweeds were prepared and tested for cell viability in the 3T3-L1 cell line by using MTT assay. In addition, various doses of the water extract of seaweeds (WES) and ethanol extract of seaweeds (EES) were treated at the beginning of 3T3-L1 differentiation and continued until the cells were fully differentiated to adipocytes. Oil Red-O staining was performed to determine the potential cell differentiation inhibitory effects of the WES and EES by measuring the levels of lipid droplet accumulation in 3T3-L1 adipocytes. $PPAR{\gamma}$ mRNA expression levels were significantly reduced by WESs of UP, LJ, and HF as well as EESs of LJ and HF. As a result, we observed the superior cell differentiation inhibitory effects of WES compared to that of EES in a dose-dependent manner without any significant cytotoxicity in mouse adipocytes.

섬괴불나무(Lonicera insularis Nakai) 추출물의 면역자극 및 항비만 활성 (Immunostimulatory and Anti-Obesity Activity of Lonicera insularis Nakai Extracts in Mouse Macrophages RAW264.7 Cells and Mouse Adipocytes 3T3-L1 Cells)

  • 본 연구에서는 섬괴불나무 열매(LIF), 잎(LIL) 그리고 줄기(LIS) 추출물의 면역증진 활성과 섬괴불나무 열매(LIF) 추출물의 항비만 활성을 평가하였다. 섬괴불나무 열매(LIF), 잎(LIL) 그리고 줄기(LIS) 추출물은 RAW264.7 세포에서 NO, iNOS, COX-2, IL-1𝛽, TNF-𝛼와 같은 면역증진인자의 생성을 증가시켰으며, IL-1𝛽의 발현은 NO생성과 관련된 것으로 보여진다. 면역증진인자은 TLR2/4를 통해 MAPKs중 p38 그리고 JNK를 자극하여 발현이 유도되는 것으로 판단된다. 항비만 실험에서, 섬괴불나무 열매(LIF) 추출물은 AMPK, HSL, ATGL의 발현 증가와 perilipin-1 발현 억제를통해 지질분해를 유도하여 세포 내 지질축적을 억제하는 것으로 나타났으며, 갈색지방세포로의 분화유도와 에너지 대사에 관여하는 인자인 PRDM16, PGC-1𝛼의 발현유도를 통해서도 지질축적을 억제하는 것으로 판단된다. 향후 섬괴불나무 추출물은 건강 보조제 및 기능성 식품으로의 활용이 가능할 것으로 판단되지만, 섬괴물나무 추출물의 어떠한 성분이 면역과 항비만 활성에 영향을 미치는지에 대한 성분분석이 필요하다. 또한, 본 연구는 세포를 이용한 실험으로 정확한 분석을 위해서는 동물모델을 이용한 섬괴불나무 추출물의 면역증진 및 항비만 활성에 관한 추가적인 연구가 진행되어야 할 것이다.

Biocellulose Reduces Body Weight Gain of Rats Fed High-Fat Diet

  • Anti-obesity effect of biocellulose and diet formulation containing it was evaluated using obese rats. Thirty male Sprague-Dawlely (SD) rats were randomly assigned to high-fat diet group (CON), high-fat diet group containing 5% biocellulose (BIO), and high-fat diet group containing 5% dietary formulation (DF). After 10 weeks, body weight gains of BIO and DF groups were reduced by 15.1 and 6%, respectively, as compared with CON group. Epididymus, parirenal, and visceral fat pads of BIO and DF groups were lower than those of CON group. Weight of interscapular brown adipose tissues increased slightly in BIO group and significantly in DF group. Size of adipocyte in rats decreased in BIO and DF groups. Results indicate biocellulose-containing diet has anti-obesity effect by reducing body weight gain and white adipocytes depots in rats fed high-fat diet.

Molecular and functional characterization of the adiponectin (AdipoQ) gene in goat skeletal muscle satellite cells

  • Objective: It is commonly accepted that adiponectin binds to its two receptors to regulate fatty acid metabolism in adipocytes. To better understand their functions in the regulation of intramuscular adipogenesis in goats, we cloned the three genes (adiponectin [AdipoQ], adiponectin receptor 1 [AdipoR1], and AdipoR2) encoding these proteins and detected their mRNA distribution in different tissues. We also determined the role of AdipoQ in the adipogenic differentiation of goat skeletal muscle satellite cells (SMSCs). Methods: SMSCs were isolated using 1 mg/mL Pronase E from the longissimus dorsi muscles of 3-day-old female Nanjiang brown goats. Adipogenic differentiation was induced in satellite cells by transferring the cells to Dulbecco's modified Eagle's medium supplemented with an isobutylmethylxanthine, dexamethasone and insulin cocktail. The pEGFP-N1-AD plasmid was transfected into SMSCs using Lipofectamine 2000. Expression of adiponectin in tissues and SMSCs was detected by quantitative polymerase chain reaction and immunocytochemical staining. Results: The three genes were predominantly expressed in adipose and skeletal muscle tissues. According to fluorescence and immunocytochemical analyses, adiponectin protein expression was only observed in the cytoplasm, suggesting that adiponectin is localized to the cytoplasm of goat SMSCs. In SMSCs overexpressing the AdipoQ gene, adiponectin promoted SMSC differentiation into adipocytes and significantly (p<0.05) up-regulated expression of AdipoR2, acetyl-CoA carboxylase, fatty-acid synthase, and sterol regulatory element-binding protein-1, though expression of CCAAT/enhancer-binding $protein-{\alpha}$, peroxisome proliferator-activated receptor ${\gamma}$, and AdipoR1 did not change significantly. Conclusion: Adiponectin induced SMSC differentiation into adipocytes, indicating that adiponectin may promote intramuscular adipogenesis in goat SMSC.

3종 분말식초의 항산화, 항혈전 및 항비만 활성 (Antioxidant, Anti-thrombosis, and Lipid Accumulation Inhibition Activities of Different Dried Vinegars)

  • 식초는 당과 전분을 함유하는 다양한 재료를 초산균으로 발효시켜 제조되며, 다양한 유기산, 당류, 아미노산, 에스터 및 기타 화합물을 포함하여 독특한 맛과 향미를 나타낸다. 식초는 소화를 원활하게 하고 혈당 수준을 낮추는 데 도움을 주며, 항비만 및 항산화 활성이 알려져 있으며, 체액의 알칼리화에 기여한다고 알려져 있다. 본 연구에서는 천연 발효식초로부터 기능성 분말식초(dried vinegar, DV)를 제조하기 위해, 풋사과, 현미 및 아로니아를 각각 Gluconacetobacter xylinus로 90-180일 발효시킨 발효식초를 말토덱스트린(37.46%), 포도당(5%), 구연산(1%) 및 비타민 C (0.04%) 용액에 현탁 후 분무건조하여 3종의 DV를 제조하였다. 제조된 DVs의 산도, 색상, 수분 및 용해성 고형 내용물 및 열 안정성 분석 결과, 분무건조가 분말제조에 적합함을 확인하였으며, DVs는 우수한 관능과 용해성을 나타내었다. 제조된 DVs중 아로니아-DV가 0.5 mg/ml 농도에서 가장 높은 1,1-diphenyl-2-picryl hydrazyl 및 2,2-azobis (3-ethylbenzothiazoline-6-sulfonate) 자유 라디칼 소거 활성(각각 36.7% 및 75.3%) 및 환원력 (0.334)을 보였으며, nitrite 소거 활성은 풋사과-DV에서 가장 높았으며, 아로니아-DV 및 현미-DV순으로 나타났다. 항혈전 활성 평가결과 아로니아-DV가 prothrombin 억제 활성이 가장 높게 나타난 반면, 현미-DV는 3T3-L1 지방세포에서 세포 독성 없이 지질 축적 억제 활성을 나타내었다. 상기 결과는 분말식초의 산업화 가능성을 제시하며, 식품가공 소재로 응용 가능함을 제시하고 있다.

꽈배기모자반 추출물이 3T3-L1 지방세포 분화에 미치는 영향 (Inhibitory Effect of Crude Extracts from a Brown Alga Sargassum siliquanstrum on 3T3-L1 Adipocyte Differentiation)

  • In this study, the potential capacity of the crude extract and its solvent fractions from S. siliquanstrum against adipocyte differentiation were evaluated in 3T3-L1 adipocytes. The anti-adipogenic effect of S. siliquanstrum was evidenced by the fact that its crude extract decreases the lipid accumulation of differentiating cells and the expression levels of crucial adipogenesis markers, peroxisome proliferator-activated receptor $(PPAR){\gamma}$ and CCAAT/enhancer-binding protein $(C/EBP){\alpha}$. All solvent fractions except the water fraction showed an observable decrease in lipid accumulation and $PPAR{\gamma}$ and $C/EBP{\alpha}$ expressions. In conclusion, these results suggest that S. siliquanstrum possesses obesity inhibiting components, which may possibly be used as a valuable anti-obesity agent for reducing the risk of obesity.

Wild Ginseng Prevents the Onset of High-Fat Diet Induced Hyperglycemia and Obesity in ICR Mice

  • Ginseng is a shade-loving perennial herb that is cultivated mainly in Korea, Japan, and China. The ginseng root has been used as a tonic remedy, and its antidiabetic activity has been demonstrated as early as 1920s. Although wild ginseng was anecdotally thought to be superior to cultivated ginseng as far as pharmacological properties were concerned, there have been no prior reports on the antidiabetic effect of wild ginseng. In this study, we investigated the preventative anti-diabetic and anti-obese effects of wild ginseng ethanol extract (WGEE). In the preventive experiment, WGEE co-administered with a high fat diet significantly inhibited body weight gain, fasting blood glucose, triglyceride, and free fatty acid levels in a dose dependent manner. WGEE-treated mice at doses of 250 and 500 mg/kg improved the insulin resistance index by 55% and 61% compared to the high fat diet (HFD) control, respectively. Diameters of white and brown adipocytes were also decreased by 62% and 46% in the WG500-treated group compared to those in HFD fed control mice. Taken together, WGEE has potential as a preventive agent for type 2 diabetes mellitus (and possibly obesity) and deserves clinical trial in the near future.

Effects of Dyglomera® on leptin expression, pro-inflammatory cytokines, and adipocyte browning in 3T3-L1 cells

  • Dyglomera® is an aqueous ethanol extract derived from the fruit and pods of Dichrostachys glomerata. A previous study has revealed that Dyglomera regulates adipogenesis and lipolysis by modulating AMP-activated protein kinase (AMPK) phosphorylation and increased expression levels of lipolysis-related proteins in white adipose tissue of high fat diet-induced mice and 3T3-L1 adipocyte cells. To further investigate mechanisms of Dyglomera, additional studies were performed using 3T3-L1 cells. Results revealed that Dyglomera downregulated adipogenesis by inhibiting the protein kinase B/mammalian target of rapamycin signaling pathway and reconfirmed that it downregulated gene expression levels of proliferator-activated receptor (PPAR)-γ, CCAAT enhancer binding protein α, sterol-regulation element-binding protein-1c. Dyglomera also reduced adipokines such as tumor necrosis factor alpha, interleukin-1β, and interleukin 6 by regulating leptin expression. Moreover, Dyglomera promoted beige-and-brown adipocyte-related phenotypes and regulated metabolism by increasing mitochondrial number and expression levels of genes such as T-box protein 1, transmembrane protein 26, PR domain 16, and cluster of differentiation 40 as well as thermogenic factors such as uncoupling protein 1, proliferator-activated receptor-gamma co-activator-1α, Sirtuin 1, and PPARα through AMPK activation. Thus, Dyglomera not only can inhibit adipogenesis, but also can promote lipolysis and thermogenesis and regulate metabolism by affecting adipokine secretion from 3T3-L1 adipocytes.

Anti-hyperlipidemic Effect of Polyphenol Extract (SeapolynolTM) and Dieckol Isolated from Ecklonia cava in in vivo and in vitro Models

  • The inhibitory effect of polyphenol extracts (Seapolynol$^{TM}$, SPN) of the marine brown algae Ecklonia cava and dieckol, a major component of SPN, on hyperlipidemia was investigated in ICR mice fed a high-fat diet (HFD) for five weeks. For analysis of the anti-hyperlipidemic effects of SPN and dieckol, these two agents were given orally on a daily basis to HFD-fed mice for four weeks, starting one week after the beginning of HFD feeding. Groups administered with SPN as well as dieckol showed lower body weight gains than the HFD only group. Administration of SPN and dieckol also resulted in a significant reduction of the level of total cholesterol (TCHO), triglyceride (TG), and low-density lipoprotein (LDL) cholesterol in the serum of HFD-fed mice. In Oil Red O staining using 3T3-L1 preadipocytes, it was shown that both SPN and dieckol markedly inhibited lipid accumulation of 3T3-L1 cells. Furthermore, SPN and dieckol (50 ${\mu}g$/mL) significantly inhibited 3-hydroxyl-methyl glutaryl coenzyme A (HMGCoA) reductase activity in vitro. Taken together, these results suggest that polyphenols of Ecklonia cava (SPN) and dieckol reduce body weight gain and fat accumulation in HFD-induced obese mice, and that their hypolipidemic effect is related to the inhibition of adipogenesis of adipocytes and HMGCoA reductase activity.

The difference of castration timing of Korean Hanwoo bulls does not significantly affect the carcass characteristics

  • It is already well known that castration improves marbling quality but exact timing of castration is still highly debated in beef cattle production industry. After castration, blood hormonal changes occur in steer and objective of this study was to investigate the effects of growth hormone (GH) levels on adipocyte differentiation in stromal vascular cells (SVCs) and transdifferentiation into adipocytes in C2C12 myoblasts. Total GH concentrations were measured via enzyme-linked immunosorbent assay (ELISA) in 24 male calves and 4 female calves. Cell proliferation, cellular triglyceride (TG) accumulation, and the cell's lipolytic capability were measured in C2C12 myoblasts and SVCs. Myogenic, adipogenic, and brown adipocyte-specific gene expression was measured via real-time polymerase chain reaction (PCR) using SYBR green. Serum GH levels were the highest in late-castrated calves. Treatment with 5 ng/mL GH resulted in greater TG accumulation as well as increased CCAAT-enhancer-binding protein (C/EBP)α and peroxisome proliferator-activated receptor (PPAR)γ expression compared to that after treatment with 15 ng/mL GH. Treatment with 5 ng/mL GH also resulted in lower myogenin (myo)G and myoD expression compared to that after treatment with 15 ng/mL GH. The expression of bone morphogenetic protein (BMP) 7 after treatment with 5 ng/mL GH was higher than that after treatment with 15 ng/mL GH. But carcass characteristics data showed no significant difference between early and late castrated steers. Therefore, our results indicate that castration timing does not seem to be inevitable determinate of carcass qualities, particularly carcass weight and marbling score in Hanwoo beef cattle.
