고온에서의 브로콜리 부위별 추출물의 항산화성 및 항균성 (Antioxidant Activity and Antibacterial Activities from Different Parts of Broccoli Extracts under High Temperature)

  • 브로콜리의 잎, 꽃, 꽃-줄기,줄기 그리고 껍질을 에탄올, 아세톤 및 증류수로 추출하여 그 추출물의 라디칼소거 활성을 조사하였다 각각의 시료는DPPH 라디칼에 대한 라디칼 소거 활성 (FRSA)을 나타내었다. 다섯 가지의 시료 중, 줄기 추출물, 꽃 추출물 및 잎 추출물에서 dot-blot 분석을 통해 라디칼소거 활성을 확인하였다. 브로콜리의 에탄을 추출물 에서는 $pH\;2\~6$의 산성영역 및 $60\~80^{\circ}C$의 온도 범위에서 강력한 라디칼소거 활성을 보였다. 녹차 추출물과 쑥 추출물에서는 Staphylococcus aureus에 대한 항균성을 보였고, 다섯 가지의 추출물중에서 단지 꽃 추출물 및 꽃-줄기 추출물이 고온 하에서 Bacillus arnyloliquefaciens에 대해 강력한 항균성을 나타내었다.

NF-κB 저해를 통한 브로콜리 잎 추출물의 PGE2 저해효과 (Inhibitory effect of broccoli leaf extract on PGE2 production by NF-κB inhibition)

  • Objective : Broccoli is edible green plant that has a wide variety of health benefits including cancer prevention and cholesterol reduction. However, leaves of broccoli are not eaten and are mostly left as waste. This study was conducted to evaluate the effects of the broccoli leaf extract (BLE) on prostaglandin E2 (PGE2) production related to nuclear factor kappa B (NF-κB) signaling in lipopolysaccharide (LPS)-activated macrophages. Methods : BLE was prepared by extracting dried leaf with ethanol. Cell viability was determined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. PGE2 and inflammatory cytokines were detected by enzyme-linked immunosorbent assay (ELISA). Expression level of each protein was monitored by Western blot analysis. Results : In LPS-activated Raw264.7 cells, PGE2 release into culture medium was dramatically enhanced compared to control cells. However, increased PGE2 was attenuated dose-dependently by treatment with BLE. Inhibition of PGE2 production by BLE was due to the suppression of cyclooxygenase-2 (COX-2) expression determined by Western blot analysis. BLE also inhibited the production of inflammatory cytokines such as interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α). Inhibition at PGE2 and cytokine was mediated from inhibition of nuclear translocation of NF-κB due to the repression of inhibitory kappa B alpha (IκBα) phosphorylation and degradation. Conclusion : This study showed that BLE exerted inhibitory activities against PGE2, which is critical for the initiation and resolution of inflammatory responses, and that inhibition of PGE2 was mediated by suppression of NF-κB signaling. These results suggest that the waste broccoli leaves could be used for controlling inflammation.

브로콜리 가루 첨가 머핀 제조 조건의 최적화 (Optimization of Formulation Condition for Muffins with Added Broccoli Powder)

  • The principal objective of this study was to develop the optimal recipe for muffins containing dried broccoli powder. In this study, broccoli powder was substituted for wheat flour in order to reduce its content. The study was conducted by determining the optimal sensory composite recipe, by preparing muffins with different levels of broccoli powder (A), sugar (B), and butter (C), by C.C.D (Central composite design) and performing sensory evaluation and analysis via RSM (Response surface methodology). The sensory measurements yielded significant values for appearance, flavor (p<0.01), texture (p<0.05), overall quality (p<0.05) and color (p<0.05), whereas instrumental measurements yielded significant values in lightness (p<0.01), redness (p<0.05), yellowness, baking loss rate (p<0.05), hardness (p<0.05), cohesiveness (p<0.01) and gumminess (p<0.05). The optimum formulations processed by numerical and graphical optimization were determined as 13.58g of broccoli powder, 92.02g of sugar, and 71.97g of butter.

브로콜리 추출물의 화장품 안정성 평가 연구 (A Study on the Evaluation of Cosmetic Stability of Broccoli Extracts)

  • 본 연구는 브로콜리 추출물의 DPPH radical 소거활성을 통한 항산화 활성과 저온(5℃이하), 고온(50℃이상)의 보관 조건을 통해 화장품 제조 시 나타날 수 있는 온도 변화에 따른 항산화력 변화를 비교해보고, 브로콜리 추출물 5%가 함유된 크림을 제조하여 화장품의 제형 안정성, 변색 및 변취, pH, 1차 첩포 테스트를 통해 물리적, 화학적 안정성에 대해 확인하고 하였다. 실험 결과, 브로콜리 추출물 1%의 농도에서 95.5%의 높은 DPPH radical 소거 활성이 확인되었으며, 온도 변화에 따른 보관 조건에도 항산화력은 높에 유지되는 것을 확인되었다. 브로콜리 추출물 5% 함유된 크림에서도 제형의 안정성, 변색 및 변취, pH, 1차 첩포 테스트 모두 안전성과 안정성이 유지되는 것이 확인되었다. 본 연구들을 통해 브로콜리 추출물의 높은 항산화 활성과 온도 변화에 따른 항산화력 유지력을 통해 피부에 안전하고 효과적으로 사용가능한 화장품 소재로써의 가능성이 확인되었다.

소포자 배양 유래 계통을 활용한 브로콜리 신품종 조기 육성 (Development of new broccoli varieties from elite lines obtained by microspore cultivation method)

  • Since the year 2000, the production and consumption of broccoli have rapidly increased in Korea. And, the average production area and amount were about 1,700 ha and 29,000 ton for the past 5 years. Even with the increase of these cultivation and consumption, more than 95% of the broccoli seeds are currently imported from foreign countries such as Japan and Netherlands. Therefore, development of domestic broccoli varieties is needed to relieve Korean farmers' production cost for broccoli. In this situation, National Institute of Horticultural and Herbal Science (NIHHS) of Korea has tried to develop F1 hybrid varieties from elite lines that were obtained by microspore cultivation method from 2008. As the results, about 850 lines of broccoli were obtained and self-pollinated. Then their ploidy levels of the genome were confirmed to select double haploid (D.H.) lines. And the D.H. lines' horticultural traits were evaluated in open field. After the selection of 17 elite D.H. lines, they were cross-pollinated with a male sterile (MS) line to produce F1 hybrid seeds. After 2 to 3 years field trials of these F1 hybrid varieties at the area of Suwon, Gangneung, and Jeju respectively, two hybrid varieties such as 'Wongyo8011' and 'Wongyo8012' are selected for the application of variety protection. With these 4 years of research, we found that the microspore cultivation method is a powerful tool for the conventional breeding program, especially for the development of various inbred lines and even F1 hybrid varieties in short time.

Bioactivities of Sulfur Compounds in Cruciferous Vegetables

  • Cruciferous vegetables are rich in organosulfur compounds such as isothiocyanates and sulfides. While the isothiocyanates, corresponding to pungent principle, are generated from myrosinase-catalyzed hydrolysis of glucosinolates, the sulfides can be generated non-enzymatically. Recent studies provide evidences that some sulfur compounds in these vegetables show a chemopreventive action against carcinogenesis; while isothiocyanates such as sulforaphane induce phase 2 enzymes (glutathione S-transferase/quinone reductase), disulfides tends to elevate the level of phase 1 and 2 enzymes. Especially, sulforaphane rich in Cruciferae vegetables has been reported to express anticarcinogenic effect in some organs such as liver, kidney or intestine. When the level of sulfur compounds in Cruciferous and Alliaceous vegetables was determined by GC/MS (SIM), the richest in sulforaphane is broccoli followed by turnip, cabbage, radish, kale, cauliflower and Chinese cabbage. Meanwhile, the sulfides are predominant in Alliaceous vegetables such as onion. In related study, the administration of vegetable extract elevated the GST level by 1.5 fold for broccoli, 1.4 fold for radish, and 1.3 for onion. Thus, the vegetables frequently used in Korean dish contain relatively high amount of anticarcinogenic sulfur compounds. Moreover, the combination of broccoli and radish extracts elevated the GST induction up to 1.84 folds of control. In addition, the Kakdugi, fermented radish Kimchi was observed to show a comparable GST induction despite the decomposition of methylthio-3-butenylisothiocyanate (MTBI). Therefore, the combination of vegetables including broccoli, and fermented radish Kimchi would be useful as a functional food for chemoprevention.

Salmonella 실험계의 십자화과 채소류의 항돌연변이효과 (Antimutagenic Effect of Various Cruciferous Vegetables in Salmonella Assaying System)

  • The antimutagenic effects of juices and methanol extracts from cruciferous vegetables (cabbage, red cabbage, Korean cabbage, kale, cauliflower, broccoli, radish root, leafy radish, rape leaves and shepherd's purse) on the mutagenicity induced by aflatoxin B1(AFB1) and N-methyl-N'-nitrosoguanidine(MNNG) were studied using Salmonella assay system. In the case of juices from the cruciferous vegetables, the juices of cabbage, kale, cauliflower and radish root in the concentrations of 50, 200 and 500 ${mu}ell$/plate considerably decreased the mutagenicity induced by AFB1, and the juices of cabbage and broccoli in the concentrations of 200 and 500${mu}ell$/plate significantly reduced the mutagenicity induced by MNNG. The antimutagenic activities of the juices against AFB1 were stronger than those against MNNG. In the case of methanol extracts from the cruciferous vegetables, the methanol extracts of kale, broccoli and shepherd'purse appeared to inhibit the mutagenicity induced by AFB1 and MNNG in Salmonella typhimurium test strains. The effects of the juices against mutagens quite different from ones of the methanol extracts. The juice of cabbage showed antimutagenicity, but its methanol extract was less effective. However, both juices and methanol extracts from kale and broccoli exhibited strong antimutagenic activities against the mutagens.

Microbial Quality and Safety of Fresh-Cut Broccoli with Different Sanitizers and Contact Times

  • This study was conducted to investigate the effects of different sanitizers and contact times on storage quality and microbial growth in fresh-cut broccoli. Fresh broccoli samples were cut into small pieces, washed each for 90 s and 180 s in normal tap water (TW), $100\;{\mu}/l$ chlorinated water (CL, pH 7), electrolyzed water (EW, pH 7.2) containing $100\;{\mu}/l$ free chlorine, or $2\;{\mu}/l$ ozonated water ($O_3$). Then, samples were packaged in 30-${\mu}m$ polyethylene bags and stored at $5^{\circ}C$ for 9 days. No significant differences were observed in gas composition and color parameters ($L^*$, $a^*$, $b^*$, and hue angle) among different sanitizers with contact times. No off-odor was detected during the storage. A longer contact time was not effective in reducing microbial population, except with $O_3$ washing. $O_3$ with 90 s was not much effective in reducing microbial population compared with Cl or EW. However, samples washed with $O_3$ for 180 s observed the lowest numbers of total aerobic and coliform plate counts. The result suggested that, a longer contact time of ozone can be used as a potential sanitizer to maintain the microbial quality and safety of fresh-cut broccoli.

Antioxidant and Anti-inflammatory Activities of Broccoli Florets in LPS-stimulated RAW 264.7 Cells

  • Broccoli (Brassica oleracea var. italia) florets were extracted with 80% methanol and the extract was sequentially fractionated with n-hexane, ethyl acetate, n-butanol, and distilled water. The extract and the fractions were evaluated for total phenolic content, sulforaphane content, antioxidant activity, and anti-inflammatory activity in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. The total phenolic content and sulforaphane content of the ethyl acetate fraction (EF) were 35.5 mg gallic acid equivalents/g and $620.2{\mu}g/g$, respectively. These values were higher than those of the 80% methanol extract and organic solvent fractions. The oxygen radical absorbance capacity of the EF [$1,588.7{\mu}M$ Trolox equivalents (TE)/mg] was 11-fold higher than that of the distilled water fraction ($143.7{\mu}M\;TE/mg$). The EF inhibited nitric oxide release from LPS-stimulated RAW 264.7 cells in a dose-dependent manner and inhibited $I{\kappa}B-{\alpha}$ degradation and nuclear factor-${\kappa}B$ activation in LPS-stimulated RAW 264.7 cells. In conclusion, the EF of broccoli florets exerted potent antioxidant and anti-inflammatory effects.

Induction of Hepatic Glutathione S-transferase Activity in Mice Administered with Various Vegetable Extracts

  • The effect of various vegetables commonly consumed by Koreans on the induction of glutathione S-trasferase(GST) activity in mice was assessd. The extract of vegetable dissolved in propylene glycol (5ml/kg body wt.) was administered to ICR female mice 6 to 8 weeks old via gavage during 5 days. The changes of body weight and liver weight of all treated groups were not significantly different compared with control group. Hepatic protein contents of trated groups compared with control group were not significantly different except BHT treated group. The induction of GST activity in liver cytosol of mice was the greatest with broccoli, followed by radish, wild green onion, turnip, and green onion. The induction of GST activity in liver cytosol increased up to 1.5 to 1.8-folds at a dose of 24 g fresh vegetable/mouse. The induction of combination between vegetables was the highest with the combination of broccoli and radish (1.83-fold), followed by that of broccoli and green onion (1.72-fold), and that of broccoli and turnip (1.50-fold).
