Presence of Foxp3-expressing CD19(+)CD5(+) B Cells in Human Peripheral Blood Mononuclear Cells: Human CD19(+)CD5(+)Foxp3(+) Regulatory B Cell (Breg)

  • Foxp3 is a transcript factor for regulatory T cell development. Interestingly, Foxp3-expressing cells were identified in B cells, especially in CD19(+)CD5(+) B cells, while those were not examined in CD19(+)CD5(-) B cells. Foxp3-expressing CD5(+) B cells in this study were identified in human PBMCs and were found to consist of $8.5{\pm}3.5%$ of CD19(+)CD5(+) B cells. CD19(+)CD5(+)Foxp3(+) B cells showed spontaneous apoptosis. Rare CD19(+)CD5(+) Foxp3(+) regulatory B cell (Breg) population was unveiled in human peripheral blood mononuclear cells and suggested as possible regulatory B cells (Breg) as regulatory T cells (Treg). The immunologic and the clinical relevant of Breg needs to be further investigated.

Autocrine stimulation of IL-10 is critical to the enrichment of IL-10-producing CD40hiCD5+ regulatory B cells in vitro and in vivo

  • IL-10-producing B (Breg) cells regulate various immune responses. However, their phenotype remains unclear. CD40 expression was significantly increased in B cells by LPS, and the Breg cells were also enriched in $CD40^{hi}CD5^+$ B cells. Furthermore, CD40 expression on Breg cells was increased by IL-10, CD40 ligand, and B cell-activating factor, suggesting that $CD40^{hi}$ is a common phenotype of Breg cells. LPS-induced CD40 expression was largely suppressed by an anti-IL-10 receptor antibody and in IL-$10^{-/-}CD5^+CD19^+$ B cells. The autocrine effect of IL-10 on the CD40 expression was largely suppressed by an inhibitor of JAK/STAT3. In vivo, the LPS treatment increased the population of $CD40^{hi}CD5^+$ Breg cells in mice. However, the population of $CD40^{hi}CD5^+$ B cells was minimal in IL-$10^{-/-}$ mice by LPS. Altogether, our findings show that Breg cells are largely enriched in $CD40^{hi}CD5^+$ B cells and the autocrine effect of IL-10 is critical to the formation of $CD40^{hi}CD5^+$ Breg cells.

Entinostat, a histone deacetylase inhibitor, increases the population of IL-10+ regulatory B cells to suppress contact hypersensitivity

  • IL-10+ regulatory B (Breg) cells play a vital role in regulating the immune responses in experimental autoimmune encephalomyelitis, colitis, and contact hypersensitivity (CHS). Several stimulants such as lipopolysaccharide (LPS), CD40 ligand, and IL-21 spur the activation and maturation of IL-10+ Breg cells, while the epigenetic mechanism for the IL-10 expression remains largely unknown. It is well accepted that the histone acetylation/deacetylation is an important mechanism that regulates the expression of IL-10. We found that entinostat, an HDAC inhibitor, stimulated the induction of IL-10+ Breg cells by LPS in vitro and the formation of IL-10+ Breg cells to suppress CHS in vivo. We further demonstrated that entinostat inhibited HDAC1 from binding to the proximal region of the IL-10 expression promoter in splenic B cells, followed by an increase in the binding of NF-κB p65, eventually enhancing the expression of IL-10 in Breg cells.

불안정한 바닥위에서 발목각도가 기립균형에 미치는 영향 (The Influence of Foot Angle on Standing Balance Upon the Unstable Platform)

  • The purpose of this study was to compare the balance ability at different foot angle with KAT 2000 (Breg, Inc., Vista, CA. 1994). Forty-nine (male 24, female 25) normal subjects participated in this study. All subjects were assessed under two conditions. One was eye-opened condition and the other one was eye-closed one in 3 psi surface condition. All subjects were tested at different foot angle that were toe-in $25^{\circ}$, $45^{\circ}$, toe-out $25^{\circ}$ and $45^{\circ}$. The subject attempted to keep the platform as stable as possible with eyes closed and with eyes opened for every 20 seconds. The starting position was that subject crossed their arms across chest and flexed knees slightly. The results of each test were showed by a score on screen, which meant balance index. The se collected data were analyzed by using oneway ANOVA, Scheffe test, and t-test. The results of this study were as follows: 1. When the foot angle were changed, balance index was the lowest in toes-out $25^{\circ}$ condition and greatest with toes-in $45^{\circ}$ with eyes opened and with eyes closed. There were statistically significant difference with eyes opened and with eyes closed (p<0.05). 2. There was statistically significant difference in balance index according to visual condition and the balance index tested with eyes closed was higher than with eyes closed(p<0.05). 3. There was statistically significant difference in balance index by gender(p<0.05).

Interleukin-10-Producing B Cells Help Suppress Ovariectomy-Mediated Osteoporosis

  • Osteoporosis is prevalent in elderly women and it may cause dental implant failure. In particular, estrogen deficiency in postmenopausal women leads to higher rates of osteoporosis prevalence. Immune cell-mediated effects involving the development of osteoporosis have been studied previously; however, the role of IL-10-producing regulatory B (B10) cells in osteoporosis is largely unclear. Here, we examined the role of B10 cells in osteoporosis. C57BL/6 mice were subjected to ovariectomy (OVX). Fifteen weeks after OVX surgery, the first molar of the right maxillary was extracted, and twenty-four weeks after OVX surgery, serous progression of osteoporosis was observed in the alveolar bone. Moreover, the proportion of CD19+CD5+CD1dhigh regulatory B cells, B10, and CD4+CD25+FoxP3+ regulatory T cells from the spleen of OVX mice decreased during the progression of osteoporosis, compared to controls. In contrast to regulatory cells, IL-17-producing Th (Th17) cell levels were increased in OVX mice. Adoptive transfer of B10 cells to OVX mice led to a decrease in Th17 cell abundance and inhibited the development of osteoporosis in the alveolar bone from OVX mice. Thus, our results suggest that B10 cells may help suppress osteoporosis development.

불안정 발판(Unstable Platform)에서 20대 연령의 균형능력 평가 (Evaluation of the Balance Ability for 20 to 29 Years Old on the Unstable Platform)

  • The purpose of this study were to evaluate and compare the balance ability at different conditions in normal 20 to 29 years old on unstable platform, KAT 2000(Breg, Inc., Vista, CA. 1994). Static and dynamic BI(balance index) were measured 3psi and 5psi surface conditions. Static tests were done on right and left leg separately, then both legs together with the feet apart 20cm with the eyes opened and closed. Dynamic tests were done on both legs together with apart 20cm with the eyes opened. A dynamic test was performed in which the subject moved platform in a circular manner to chase a moving object on a computer screen. Seventy healthy students(average 21.6 years, male, female) were tested. In this study applied the paired t-test and correlation to determine the statistical significance of result. The results were as follow: 1) The mean static balance index of the Rt leg was $119.9{\pm}75.72$ on 3psi surface condition with the eyes opened, and that of the Lt leg was $224.3{\pm}121.16$. 2) The mean static balance index of the Rt leg was $93.1{\pm}24.16$ on 5psi surface condition with the eyes opened, and that of the Lt leg was $180.5{\pm}61.76$. 3) The mean static and dynamic balance index of both legs were $76.4{\pm}31.86$, $2187.6{\pm}696.99$ on 3psi with the eyes opened, and $68.3{\pm}14.82$, $1938.7{\pm}525.41$ on 5psi respectively. 4) The mean static balance index of the Rt leg was $517.8{\pm}220.87$ on 3psi surface condition with the eyes closed, and that of the Lt leg was $588.6{\pm}204.81$. 5) The mean static balance index of the Rt leg was $271.9{\pm}192.151$ on 5psi surface condition with the eyes closed, and that of the Lt leg was $363.4{\pm}98.97$. 6) The mean static balance index of both legs was $332.6{\pm}137.31$ on 3psi surface condition with the eyes closed, and that of the 5psi was $288.5{\pm}133.07$. 7) The balance index on 3psi surface condition was significantly higher than that of 5psi (p<0.05, p<0.01). 8) The balance index with the eyes closed was significantly higher than that of the eyes opened (p<0.05). 9) The balance index on the left leg was significantly higher than that of the right leg (p<0.05, p<0.01). 10) There was no correlation between static balance index and dynamic balance index. 11) Therewas no correlation between weight or height and balance index.
