A simple screening method using lignoceullulose biodegradation for selecting effective breeding strains in Agaricus bisporus (리그노셀룰로오스 생물학적 분해를 이용한 간단한 양송이 육종효율 우수 균주 선발)

  • The white button mushroom, Agaricus bisporus, is commercially the fifth most important edible mushroom, accounting for the production of 9,732 tons of mushrooms in Korea in 2015. The genus Agaricus has been known for its potential to degrade lignocellulosic materials. Chemical analyses carried out during the cultivation of A. bisporus indicated that the cellulose, hemicellulose, and lignin fractions were changed preferentially for both vegetative growth and sexual reproduction. We screened A. bisporus strains for effective biodegradation through extracellular enzyme activity using cellulase, xylanase, and ligninolytic enzymes. The enzyme biodegradations were conducted as follows: mycelia of collected strains were incubated in 0.5% CMC-MMP (malt-mops-peptone), 0.5 Xylan-MMP, and 0.5% lignin-MMP media for 14 days. Incubated mycelia were stained with 0.2% trypan blue. Eighteen strains were divided into 8 groups based on different extracellular enzyme activity in MMP media. These strains were then incubated in sterilized compost and compost media for 20 days to identify correlations between mycelial growth in compost media and extracellular enzyme activity. In this study, the coefficient of determination was the highest between mycelial growth in compost media and ligninolytic enzyme activity. It is suggested that comparison with ligninolytic enzyme activity of the tested strains is a simple method of screening for rapid mycelial growth in compost to select good mother strains for the breeding of A. bisporus.

Studies on Breeding and Cultivation Characteristics of Lentinula edodes Strains for Sawdust Cultivation (표고 톱밥재배용 균주의 육종과 재배특성에 관한 연구)

  • For the breeding purpose, in this study we attempted to select a new Lentinula edodes strain by comparing mycelial growth rate, fruit-body quality and mushroom productivity per substrate weight of hybrid strains with parent strains. Nineteen hybrid strains were made by cross of di-karyotic and mono-karyotic to develop strains suitable for sawdust cultivation of L. edodes. Among the nineteen strains, ten were selected by comparing mycelial growth rate on PDA media and wood rot on sawdust-based substrates. The most mushroom harvesting was achieved by 100 days incubation from KFRI 960, KFRI 961, KFRI 962, KFRI 963, KFRI 964, KFRI 971, KFRI 973 and 125 days from KFRI 968, 970. Experiment of incubation periods with the ten strains showed that there were productivity differences between 100 and 150 days incubations. Through comparison of hybrid strains' mushroom productivity it was found that there was no difference in mushroom productivity through crossing high temperature types with high temperature types, crossing low temperature types with high temperature types and crossing unknown temperature types with high temperature types, but there was difference through crossing middle temperature types with high temperature types. Hybrid strains showed better productivity compared with parent strains. Fruiting temperature type analysis of hybrid strains confirmed that strains of high temperature types have a dominant character.

Cross-breeding of Neopyropia spp. (Bangiales, Rhodophyta) Using CAPS (Cleaved Amplified Polymorphic Sequence) Markers (CAPS (Cleaved Amplified Polymorphic Sequence) 마커를 적용한 김 교잡육종 기술 개발)

  • This study aimed to cross between Korean and Japanese pure lines of Neopyropia strains to establish cross breeding technology and identify a superior variety that harbors the strength of both parents. Four crossing combinations were tried using three methods, resulting in 1,476 single conchocelis colonies. The three co-dominant Cleaved Amplified Polymorphic Sequence (CAPS) markers (EF-1α/Mse I, TOP2/Mse I, car A/ApaL I) were used to distinguish heterozygotic sporophytes and their maternal lines obtained from the inter and intraspecific cross-fertilization within the wild type of Neopyropia strains. Of the 1,476 colonies, 26.9% (218) were heterozygotes obtained from the nuclear CAPS markers. Their maternal line was clearly confirmed using organelle CAPS marker and chimeric thallus was obtained from crossing experiment of Japanese N. yezoensis (♀) and Korean N. yezoensis (♂). The use of CAPS markers improved the efficiency of crossbreeding by quickly screening heterozygotes and maternal lines in the conchocelis phase, which otherwise required pigmentation mutants as genetic markers.

  • Korean ginseng has been widely used as medicine from ancient times in Asia. Current breeding efforts in Korea include the individual plant selection and the subsequent pure - line isolation, and considerable number of lines with desirable traits have thus been isolated. However, there were rare data on genetic maker and its analysis for selection of superior varieties. For taxonomic characterization and development of genetic markers for ginseng breeding, molecular biological methods including the RFLP and RAPD methods were applied. Cytoplasmic DNA of ginseng was analyzed for RFLP analysis. However. there is no different pattern among the chloroplast DNA or mitochondrial DNA of variants. In the case of RAPD analysis, the band patterns using 4 of 10 RAPD primers show the distinctive polymorphism among 9 ginseng variants, and lines, and Similarity Index(SI) on polymorphism was calculated for the extent and nature of these variabilities in ginseng. The sequences of 4 selected primers were TGCCGAGCTG, AATCGGGCTG. GAAACGGGTG, and GTGACGTAGG. By SI based on the polymorphic band patterns, Chungkyung - Chong and Hwangskoog - Chong, and JakyungChong 81783 and Jinjakyung of Russia showed the most close SI. The data of KG10l coincided with the fact that it was released from Hwangskoog - Chong. and Jakyung - Chong 81783 and Jinjakyung of Russia showed the most close SI. The data of KG101 coincided with the fact that it was released from Hwangskoog - Chong by breeding process. The data of Jakyung strains indicated the significant variation among the strains. From these results, RAPD analysis method could be succesively applied to the classification and genetic analysis for breeding of Korean ginseng.

Genetic Clarification of Auricularia heimuer Strains Bred and Cultivated in Korea Using the ITS and IGS1 rDNA Region Sequences

  • Auricularia is one of the broadly cultivated edible mushrooms in Korea. Most of the Korean Auricularia strains used for cultivation and breeding are known as A. auricula-judae. Recently, this species has been reported to belong to a species complex. Therefore, this study was carried out to genetically clarify the bred and cultivated Korean A. auricula-judae strains. The internal transcribed spacer (ITS) and IGS1 rDNA region sequences were determined from 10 A. auricula-judae strains by PCR and sequencing. Variation in the nucleotide sequence and sequence length of the two rDNA regions were found among the seven A. auricula-judae strains. A maximum-likelihood (ML) phylogenetic tree based on the ITS sequences clearly placed all the 10 Korean A. auricula-judae strains in the A. heimuer clade of the A. auriculajudae complex. A. heimuer is diverged from A. auricula-judae. An ML phylogenetic tree based on the IGS1 sequences revealed the close relationship between Korean A. heimuer strains to Chinese A. heimuer strains. But each strain could be distinguishable by the IGS1 sequence. Furthermore, progeny strains in the seven Korean strains could be differentiated from their parental strains by the IGS1 sequence based phylogenetic tree. Our results are expected to be used to complement the distinction of domestic Auricularia cultivars.

YOLK CHOLESTEROL CONTENT IN EGGS FROM THE MAJOR DOMESTIC STRAINS OF BREEDING HEN

  • A survey of the cholesterol content of eggs obtained from commercial sources in Korea was conducted. Eggs from 17 different strain birds were obtained from the Poultry Random Sample Test Station at Ansung, Kyonggi province. Yolk cholesterol contents, determined by the colorimetric procedure, were obtained from three consecutively laid eggs from birds of about 63 weeks of age. The overall mean of cholesterol value was 17.25 mg/g of wet yolk and ranged from 16.40 to 18.18 mg/g yolk. Analysis of t-test between white shell eggs and brown shell eggs showed no difference in cholesterol content: however, the white shell eggs were somewhat high in cholesterol (17.41 vs. 17.08 mg/g yolk). Significant differences (p<0.05) were found in yolk cholesterol contents among strains within the breed.

Bigger females, more eggs: the impact of female body weight on egg-laying ability in Protaetia brevitarsis seulensis (Kolbe)

  • One of the necessary conditions for the mass production of the edible insect, the white-spotted flower chafer Protaetia brevitarsis seulensis (Kolbe), is to breed a strain with excellent egg-laying ability. To identify external morphological traits related to egg-laying ability, we investigated the effects of the weight, length, and width of female adults on egg production. Correlation and multiple regression analyses revealed a positive correlation between the weight of female adults and egg production. This study suggests that selection of heavier females is a good strategy for breeding strains with superior egg-laying characteristics. The results of this study will serve as important foundational data for future breeding of superior strains.

Pseudorecombination between Two Distinct Strains of Cucumber mosaic virus Results in Enhancement of Symptom Severity

  • Recently, a Cucumber mosaic virus (CMV) strain, named as CMV-209, was isolated from Glycine soja. In this study, symptom expression of CMV-209 was analyzed in detail in Nicotiana benthamiana by comparing with that of CMV-Fny, which is a representative strain of CMV. Using infectious cDNA clones of CMV strains 209 and Fny, symptom expression of various pseudorecombinants between these two strains were examined in the early and late infection stages. In the early infection stage, the pseudorecombinants containing Fny-RNA2 induced stunting and leaf distortion on the newly emerged leaves whereas the pseudorecombinants containing 209-RNA2 caused no obvious symptoms. In the late infection stage, the pseudorecombinants containing 209-RNA1 and Fny-RNA2 induced severe leaf distortion and stunting, while CMV-209 induced mild symptom and CMV-Fny caused typical mosaic, general stunting, and leaf distortion symptoms, indicating that RNA 2 encodes a symptom determinant(s) of CMV, which is capable of enhancing symptoms. Furthermore, our results support the possibility that natural recombination between compatible viruses can result in emergence of novel viruses causing severe damages in crop fields.

  • A number of Chinese, Japanese and European silkworm strains with coarse size filament were used for breeding the coarse size variety. Through cross breeding combined with pedigree selection within thirteen generations, a pair of coarse size silkworm variety, named ${CU_1}{\times}{CU_2}$, was obtained after five years. Laboratory trials showed that the filament size of its 4F_1$ hybrid was over 4.3 D, and the other economic characters were also good.
