Analysis of miRNA expression in the trachea of Ri chicken infected with the highly pathogenic avian influenza H5N1 virus

  • Background: Highly pathogenic avian influenza virus (HPAIV) is considered a global threat to both human health and the poultry industry. MicroRNAs (miRNA) can modulate the immune system by affecting gene expression patterns in HPAIV-infected chickens. Objectives: To gain further insights into the role of miRNAs in immune responses against H5N1 infection, as well as the development of strategies for breeding disease-resistant chickens, we characterized miRNA expression patterns in tracheal tissues from H5N1-infected Ri chickens. Methods: miRNAs expression was analyzed from two H5N1-infected Ri chicken lines using small RNA sequencing. The target genes of differentially expressed (DE) miRNAs were predicted using miRDB. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis were then conducted. Furthermore, using quantitative real-time polymerase chain reaction, we validated the expression levels of DE miRNAs (miR-22-3p, miR-146b-3p, miR27b-3p, miR-128-3p, miR-2188-5p, miR-451, miR-205a, miR-203a, miR-21-3p, and miR-200a3p) from all comparisons and their immune-related target genes. Results: A total of 53 miRNAs were significantly expressed in the infection samples of the resistant compared to the susceptible line. Network analyses between the DE miRNAs and target genes revealed that DE miRNAs may regulate the expression of target genes involved in the transforming growth factor-beta, mitogen-activated protein kinase, and Toll-like receptor signaling pathways, all of which are related to influenza A virus progression. Conclusions: Collectively, our results provided novel insights into the miRNA expression patterns of tracheal tissues from H5N1-infected Ri chickens. More importantly, our findings offer insights into the relationship between miRNA and immune-related target genes and the role of miRNA in HPAIV infections in chickens.

Evaluation of Salt Tolerance in Sorghum (Sorghum bicolor L.) Mutant Population

  • Sorghum (Sorghum bicolor L.) is a promising biomass crop with a high lignocellulose content. This study aimed to select high salt-tolerance sorghum lines for cultivation on reclaimed land. Using 7-day seedlings of the sorghum population consisted of 71 radiation-derived mutants (M2 to M6) and 33 genetic resources, survival rate (SR), plant height (PH), root length (RL), fresh weight (FW), and chlorophyll content (CC) were measured for two weeks after 102 mM (0.6%) NaCl treatment. Furthermore, the characteristics of the sorghum population were confirmed using correlation analysis, PCA (principal component analysis), and the FCE (fuzzy comprehensive evaluation) method. Under 102 mM NaCl conditions, SR ranged from 4.9 (IS645-200-6) to 82.4% (KLSo79125-200-1), with an average of 49.9%. PH varied from 7.5 (Mesusu-100-2) to 33.2 cm (DINE-A-MITE-100-2-10), with an average of 20.4 cm. RL ranged from 1.0 (IS645-200-1) to 17.0 cm (30-100-2), with an average of 7.7 cm. FW varied from 0.1 (IS645-200-6) to 4.5 g/plant (DINE-A-MITE-100-2-10), with an average of 2.1 g/plant. CC ranged from 0.9 (DINE-A-MITE-100-2-2) to 3.1 mg/g (IS12937), with an average of 1.7 mg/g. An overall positive correlation, with SR and FW (r = 0.86, P < 0.01), and FW and CC (r = 0.79, P < 0.01), was shown by correlation analysis. Among the five traits, two principal components were extracted by PCA analysis. PC1 was significantly associated with FW, while PC2 was highly involved with RL. To evaluate the salt tolerance level of the sorghum population when an FCE based on trait data was performed, MFV (membership function value) was 0.68. As a result of compiling the MFV of each line, eight lines with MFV > 0.68 were selected. Ultimately, the radiation-derived mutant lines, DINE-A-MITE-100-2-10 and DINE-A-MITE-100-2-12 were selected as salt-tolerant sorghum lines. The results are expected to inform salt-tolerant sorghum breeding programs, and the high salt-tolerance sorghum lines might be advantageous for cultivation on reclaimed land.

The Gene Pyramiding Effect of qBK1 and qFfR1 Genes of Major Resistance Genes in Bakanae Disease

  • Bakanae disease, caused by Gibberella fujikuroi, is one of the most devastating diseases threatening rice production in Korea. In recent years, the incidence of bakanae disease became alarming due to the mechanical transplanting practice where the spread of bakanae can be amplified during accelerating seeds growth, due to the use of seeding boxes. The development of resistant rice cultivars could be the primary and effective method for controlling bakanae disease. However, the effects of individual resistance genes are relatively small. Therefore, pyramiding of bakane R genes in rice breeding is a promising strategy having a high potential to mitigate the advert effects of bakanae disease. This study employed a gene pyramiding approach to develop bakanae disease resistant rice lines carrying qBK1, qFfR1 introduced from rice line MY299BK and cv. Nampyeong, respectively. The MY299BK carries qBK1 introduced from cv. Shingwang, which was found to have a high resistance compare to Nampyeong. In addition, the pyramiding effect of the qBK1 and qFfR1 resistance genes were investigated, and the presence or absence of these genes helped us investigate their interaction through bioassay method and MAS. Furthermore, the distribution of resistance in the population showed a biased distribution toward resistance in the F6:7 populutions. However, we could not confirm the accumulation effect of the resistance gene, but the difference between the two genes by the SN2 marker was confirmed. Therefore, the qBK1 gene harbored by MY299BK appears to be different from the qFfR1 carried by Nampyeong, suspected to possess a different bakanae disease resistant gene different from those found in MY299BK and Nampyeong.

Analyzing environmental factors influencing the gestation length and birth weight of Hanwoo cattle

  • Background: This study focused on reproductive traits in Hanwoo cattle, specifically the environmental factors affecting gestation length and birth weight. Methods: The records of 1,540 cows calved at the Hanwoo Research Institute from 2015 to 2023 were examined. This study analyzed two populations, line-breeding Hanwoo (LBH) and general Hanwoo (GH), with all cows undergoing estrus synchronization and artificial insemination. The R software was used to compare the differences between the two populations and analyze the environmental factors affecting each trait. Results: The results showed that the average gestation length for LBH was 283.28 ± 5.93 days, which was significantly shorter than that of the GH, which had an average of 285.63 ± 6.21 days (p < 0.001). The average birth weight of LBH calves was 25.10 ± 3.69 kg, significantly lighter than GH calves, which weighed 27.26 ± 4.11 kg on average (p < 0.001). Analysis of environmental factors revealed significant differences in the gestation length of LBH based on dam parity, year, and season of calving. However, no significant differences were observed based on calf sex. For LBH, birth weight showed significant differences based on dam parity, year of calving, and sex of the calf, but not the season of calving. In GH, gestation length varied with dam parity and calving season, but not with calving year or calf sex. The GH birth weight showed differences based on dam parity, year of calving, and calf sex, but not the season of calving. Conclusions: Reproductive traits in the Hanwoo cattle industry are economically vital but are heavily influenced by environmental factors due to their low heritability. An accurate evaluation of the genetic potential of these traits requires an analysis of the environmental factors affecting them. The results of this study serve as foundational data for predicting the potential for genetic improvement in the gestation length and birth weight of Hanwoo cattle.

New Classification of Plasmodiophora brassicae Races Using Differential Genotypes of Chinese Cabbage

  • Clubroot disease caused by Plasmodiophora brassicae induces severe losses of cruciferous vegetables worldwide. To control clubroot of Chinese cabbage, many CR (clubroot resistance) F1 hybrid cultivars have been bred and released in Korea, China and Japan. In this study, we determined the race of P. brassicae 12 field isolates, which collected from 10 regions in Korea, using Williams' differential varieties including two cabbage ('Jersey Queen', 'Badger Shipper') and two rutabaga ('Laurentian', 'Whilhelmsburger'). By Williams' differential varieties, 12 clubroot pathogens were assigned into one (GN2), two (HS and YC), two (HN1 and HN2), three (DJ, KS and SS) and four (GS, GN1, JS and PC) isolates for races 1, 2, 4, 5 and 9, respectively. In addition, the degree of resistance of 45 CR cultivars that were from Korea, China and Japan was tested with the 12 isolates. The 45 CR cultivars of Chinese cabbage were differentiated into three genotypes according to their resistance responses. Even though the 12 P. brassicae isolates were same race by Williams' differential varieties, three CR genotypes showed different resistance response to the isolates. These results indicate that races of P. brassicae by Williams' differentials were not related with resistance of CR cultivars, and three CR genotypes represented qualitative resistance to the P. brassicae isolates. CR genotype I including 'CR-Cheongrok' showed resistance to GN1, GN2, JS, GS, HS, DJ and KS isolates and susceptibility to YC, PC, HN1, HN2 and SS isolates. And CR genotype II such as 'Hangkunjongbyungdaebaekchae' was resistant to GN1, GN2, JS, GS, HS, YC, PC and HN1 and susceptible to DJ, KS, SS and HN2. CR genotype III including 'Chunhajangkun' and 'Akimeki' represented resistance to 10 isolates except for SS and HN2 isolates. Based on these results, we selected 'CR-Cheongrok', 'Hangkunjongbyungdaebaekchae', and 'Chunhajangkun' as a representative cultivar of three CR genotypes and 'Norangkimjang' as a susceptible cultivar. Furthermore, we investigated the resistance of 15 lines of Chinese cabbage, which were provided by seed companies, to 11 isolates except for HN1 of P. brassicae. The results showed that three lines were susceptible to all the tested isolates, whereas five, four, and three lines represented the similar responses corresponding to the CR genotypes I, II, and III, respectively; there is no line of Chinese cabbage showing different resistance patterns compared to three CR genotypes. In particular, line 'SS001' showing resistance responses of CR genotype II was a parent of 'Saerona' that have been commercialized as a CR $F_1$ cultivar of Chinese cabbage. Together, we divided 12 isolates of P. brassicae into 4 races, designated by wild type, mutant type 1, mutant type 2, and mutant type 3. Wild type including GN1, GN2, JS, GS, and HS isolates of P. brassicae was not able to infect all the cultivars of three CR genotypes, whereas, mutant type 3 such as SS and HN2 isolates developed severe clubroot disease on all the CR genotype cultivars. To mutant type 1 including DJ and KS isolates, CR genotypes I, II and III were resistant, susceptible and resistant, respectively. In contrast, to mutant type 2 including YC, PS, and HN1 isolates, CR genotypes I, II and III showed susceptibility, resistance and resistance, respectively. Taken together, our results provide the extended knowledge of classification of P. brassicae races, which is useful information for the breeding of resistant crops, with a suggestion that 'Norangkimjang', 'CR-Cheongrok', 'Saerona' and 'Chunhajangkun' cultivars of Chinese cabbage could be used as new race differentials of P. brassicae for clubroot disease assay.

Genetic and Agronomic Analysis of a Recombinant Inbred Line Population to Map Quantitative Trait Loci for Blast Resistance and Select Promising Lines in Rice (벼 RIL집단의 유전 분석과 농업형질 분석을 통한 도열병 저항성 QTL 탐색 및 유망계통 선발)

  • Koshihikari has been one of the most popular rice cultivars with good eating quality since the 1960s despite its susceptibility to blast disease and lodging. To map the genes controlling blast resistance and to develop promising blast-resistant breeding lines inheriting Koshihikari's high eating quality, a recombinant inbred line (RIL) population was developed from a cross between Koshihikari and a blast resistance donor with early maturity, Baegilmi. A total of 394 Koshihikari × Baegilmi RILs (KBRIL), and the two parents, were evaluated for blast resistance and major agronomic traits including heading date, culm length, panicle length, and tiller number. A linkage map encompassing 1,272.7 cM was constructed from a subset of the KBRIL (n = 142) using 130 single nucleotide polymorphisms. Two quantitative trait loci (QTL) for blast resistance, qBL1.1 harboring Pish/Pi35 and qBL2.1 harboring Pib, were mapped onto chromosomes 1 and 2, respectively. qBL1.1 was detected in both of the experimental sites, Namwon and Jeonju, while qBL2.1 was only detected in Namwon. qBL1.1 and qBL2.1 did not affect agronomic traits, including heading date, culm length, panicle length, and tiller number. From the 394 KBRILs, lines that were phenotypically similar to Koshihikari were selected according to heading date and culm length and were further divided into the following two groups based on blast resistance: Koshishikari-type blast resistant lines (KR, n = 15) and Koshishikari-type blast susceptible lines (KS, n = 15). Although no significant differences were observed in the major agronomic traits between the two groups, the KR group produced a greater mean head rice ratio than the KS group. The present study provides useful materials for developing blast-resistant cultivars that inherit both Koshihikari's high eating quality and Baegilmi's blast resistance.

Development and Characterization of EMS-induced Mutants with Enhanced Salt Tolerance in Silage Maize (EMS 유도 내염성 증진 사료용 옥수수 돌연변이체 선발 및 특성 분석)

  • Maize (Zea mays L.) is one of the most valuable agricultural crops and is grown under a wide spectrum of environmental conditions. However, maize is moderately sensitive to salt stress, and soil salinity is a serious threat to its production worldwide. In this study, we used ethyl methane sulfonate (EMS) to generate salt-tolerant silage maize mutants. We screened salt-tolerant lines from 203 M3 mutant populations by evaluating the morphological phenotype after salt stress treatment and selected the 140ES91 line. The 140ES91 mutant showed improved plant growth as well as higher proline content and leaf photosynthetic capacity compared with those of wild-type plants under salt stress conditions. Using whole-genome re-sequencing analysis, 1,103 single nucleotide polymorphisms and 71 insertions or deletions were identified as common variants between KS140 and 140ES91 in comparison with the reference genome B73. Furthermore, the expression patterns of three genes, which are involved in salt stress responses, were increased in the 140ES91 mutant under salt stress. Taken together, the mutant line identified in our study could be used as an improved breeding material for transferring salt tolerance traits in maize varieties.

Growth and Morphological Characteristics of Introduced Sorghum Germplasm (도입 수수 유전자원의 생육 및 형태적 특성)

  • This study was conducted to obtain fundamental information on forage sorghum breeding in forage crop field of Livestock Experiment Station at Suwon from 1986 to 1991. The charcterization of sorghum germplasm was performed through 1986 to 1987, and after parental lines were selected from diverse sorghum germplasm on the basis of flowering date, plant height and several morphological characters for forage sorghum Fl hybrids. The range of variation of 50% flowering date and plant height were greater in order of forage sorghum sudangrass and male sterile line of grain sorghum. The average flowering date was earlier in sudangrass and male sterile line of grain sorghum than forage sorghum lines from the tested sorghum germplasms. And the average plant height was tall in order of forage sorghum, sudangrass and male sterile lines of grain sorghum. There were remarkable morphological variations between sudangrass lines and male sterile lines of grain sorghum such as plant color, leaf midrib color, glume color, seed coat color, head compactness and shape, awns, grain covering and 100 seed weight.

Characteristics of Early Growth in Inbred Line of Dangyang Waxy Maize ("단양찰" 자식계통의 초기생육 특성)

  • The objective of this research was to know useful genetic characteristics for breeding program of corn. These materials were inbreds after selfing of six or eight generations and had useful genetic informations. The emergence of Danyang Waxy Corn was faster than those of other inbred lines at 2~3 days in field conditions. The plant height of Dangyang Waxy Corn was the highest at 8 days after emergence. However the plant height of New Dangjin was the shortest with 6.25cm at 8 days after emergence, the fresh weight of New Dangjin was 0.046g at 2 days after emergence but that of Dangyang was the heaviest with 0.180g. The fresh weight of 9 inbred lines had more increments in 2 days after emergence. The mean values of dry weight also showed similar trends in 9 inbred lines. The shoot dry weight of inbred lines, Dangyang and New Dangjin was 0.045g and 0.018g at 8 days after emergence, respectively. The root length of inbred line, Dangyang, was the longest with 64.4cm at 8 days after emergence. But the root length of New Dangjin was the shortest with 20.4cm at 8 days after emergence. The fresh weight of endosperm was 0.35g at 2 days after emergence and 0.26g at 8 days after emergence in Dangyang Waxy Corn, because of reduced nutrition of endosperm.

Influence of the Hoping Number of Children, the Policy of Childbirth, and the Causes of Reduced Childbirth to the Perception of Childbirth in Health-Line College Students (일 지역 보건계열 대학생을 대상으로 한 희망자녀수, 출산정책, 출산저하 원인이 출산인식에 미치는 영향)

  • Purpose: The influence of the hoping number of children, the policy of childbirth, and the causes of reduced childbirth to the perception of childbirth, which will be the subject of future childbirth, was surveyed by health-line college students. Results: In terms of the attitude to the perception of childbirth, the quality of personal life, childcare, insufficient support of childbirth, and breeding of descendants had the largest influence on childbirth, and in terms of the attitude to the hoping number of children, economic reasons, childcare expenses, and education expenses had the largest influences. Second, when the causes of social demographic properties influencing childbirth was analyzed, sex showed significance to all the factors on the hoping number of children in terms of growing areas, to all the factors excluding childbirth policy in terms of the economic level, and hoping number of children in terms of the parents' jobs. Third, when the correlation was analyzed, a correlation was found in every area excluding the hoping number of children in terms of the causes of childbirth reduction and economy, childbirth policy in terms of the personal causes, personal factor in terms of the causes of childbirth policy, and family factor in terms of the causes of childbirth reduction and economy. Fourth, when the influences of the causes of hoping number of children, childbirth policy, and childbirth reduction to perception of childbirth were analyzed, the case hoping no or one child, the case hoping 2 or more children, childbirth policy, and childbirth reduction had a positive significant influence on the perception of childbirth. Conclusion: The childbirth rate will be improved if the following occur: childbirth policy and laws are established at the level of government, education programs are developed and operated for the formation of the value of perception of childbirth, and a new support policy is established after analyzing the current childbirth support policies. To change the perception of childbirth in college students who are future subjects of childbirth, successive studies will be needed to verify the effect of aforesaid measures.
