Mungbean is used for not only seed but sprout, so, consumption of mungbean has been on the rise in Korea. Life cycle of mungbean tends to be short among Legume. For that reason, Mungbean can be harvested for various cropping system and season per regions and farmers regardless of sowing date and harvesting date. So, Prior research is needed about growth characters and life cycle of mungbean per sowing period. Mungebean cultivar 'Dahyun' and 'Sanpo' supplied by Korea Agriculture Technology Promotion Agency(KOAT) is cultivated in wagner pot. Sowing period is proper time of seeding in Jeollanam-do and Gyeongsangnam-do that is major cultivation region of mungbean in korea from early May to mid July every 2 weeks. Length at maturity stage tends to increase from early May(sowing date: 4th May) to early July(sowing date: 5th July), but after that, It tends to decrease from mid July(sowing date: 19th July). Number of branches and nods shows a similar trend. Length of pod has no tendency and no difference per sowing date. Number of pod per plants has also no tendency per sowing date. Test plots sowing in late May has the most number of pods.(Sanpo 22.9pods, Dahyun 16.8) Number of seeds per pod tends to increase to late May and Test plots sowing in mid July has the most number of seed per pod. In case of sowing at early May, Days of emergence is 7d. its summation of temperature is 132.2℃. After that, it tends to decrease to mid June. After mid June, Days of emergence is fixed to 3d. Average temperature growing up in this season, Summation of temperature from sowing to emergence takes the lowest point in test plots sowing in mid June.(Sanpo 88.6℃, Dahyun 88.6) Days of flowering tends to fasten from early May to mid July. Two cultivar shows same level. Days of maturity tends to fasten to mid June, after that tends to slow. In case that many research results about growth characters and life cycle mungbean per sowing period are drawn, it is expected that it result in increase of cultivation area and income of farmer.
A random regression model was applied for the first time for the analysis of test day records and to study the genetic persistency of first lactation milk yield of Indian Murrah buffaloes. Wilmink's Function was chosen to describe the shape of lactation curves. Heritabilities of test day milk yield varied from 0.33 to 0.58 in different test days. The highest heritability was found in the initial test day ($5^{th}$ day) milk yield. Genetic correlations among test day milk yields were higher in the initial test day milk yield and decreased when the test day interval was increased. The magnitude of genetic correlations between test day and 305 day milk yield varied from 0.25 to 0.99. The genetic persistencies of first lactation milk yield were estimated based on daily breeding values using two methods. $P_1$ is the genetic persistency estimated as a summation of the deviation of estimated daily breeding value on days to attain peak yield from each day after days to attain peak yield to different lactation days. $P_2$ is the genetic persistency estimated as the additional genetic yield (gained or lost) from days to attain peak yield to estimated breeding value on different lactation days relative to an average buffalo having the same yield on days to attain peak yield. The mean genetic persistency on 90, 120, 180, 240, 278 and 305 days in milk was estimated as -4.23, -21.67, -101.67, -229.57, -330.06 and -388.64, respectively by $P_1$, whereas by $P_2$ on same days in milk were estimated as -3.96 (-0.32 kg), -23.94 (-0.87 kg), -112.81 (-1.96 kg), -245.83 (-2.81 kg), -350.04 (-3.28 kg) and -407.58 (-3.40 kg) respectively. Higher magnitude of rank correlations indicated that the ranking of buffaloes based on their genetic persistency in both methods were similar for evaluation of genetic persistency of buffaloes. Based on the estimated range of genetic persistency three types of genetic persistency were identified. Genetic correlations among genetic persistency in different days in milk and between genetic persistencies on the same day in milk were very high. The genetic correlations between genetic persistency for different days in milk and estimated breeding value for 305 DIM was increased from 90 DIM to 180 DIM, and highest around 240 DIM which indicates a minimum of 240 days as an optimum first lactation length might be required for genetic evaluation of Indian Murrah buffaloes.
Mauremys reevesii is a Korean endemic turtle, and designed as an endangered species and national monument in South Korea. Recently, the population of the species has been dramatically declining because of habitat destruction, pollution and illegal capture. Moreover, small population size, difficulty of securing individuals, and lack of research are factors that impede the effective management of the species. In this study, we tested the effect of individual breeding and feeding on the seven juveniles of M. reevesii. Our results showed individual breeding and feeding were guaranteed the effective growth and development. Noticeable growth was confirmed in both body weight and carapace length. Moreover, the size difference among the individuals appeared at the start of this study decreased at the end of this study. Artificial breeding during the wither season was not caused disorders on the growth, behavior and morphology. This individual breeding may lead to effective growth and development, and it will be a way to increase the survival rate when the juveniles released into the wild.
Background: Heat shock proteins play an important role in protection from stress stimuli and metabolic insults in almost all organisms. Methods: In this study, computational tools were used to deeply analyse the physicochemical characteristics and, using homology modelling, reliably predict the tertiary structure of the blunt snout bream (Ma-) Hsp70 and Hsc70 proteins. Derived three-dimensional models were then used to predict the function of the proteins. Results: Previously published predictions regarding the protein length, molecular weight, theoretical isoelectric point and total number of positive and negative residues were corroborated. Among the new findings are: the extinction coefficient (33725/33350 and 35090/34840 - Ma-Hsp70/ Ma-Hsc70, respectively), instability index (33.68/35.56 - both stable), aliphatic index (83.44/80.23 - both very stable), half-life estimates (both relatively stable), grand average of hydropathicity (-0.431/-0.473 - both hydrophilic) and amino acid composition (alanine-lysine-glycine/glycine-lysine-aspartic acid were the most abundant, no disulphide bonds, the N-terminal of both proteins was methionine). Homology modelling was performed by SWISS-MODEL program and the proposed model was evaluated as highly reliable based on PROCHECK's Ramachandran plot, ERRAT, PROVE, Verify 3D, ProQ and ProSA analyses. Conclusions: The research revealed a high structural similarity to Hsp70 and Hsc70 proteins from several taxonomically distant animal species, corroborating a remarkably high level of evolutionary conservation among the members of this protein family. Functional annotation based on structural similarity provides a reliable additional indirect evidence for a high level of functional conservation of these two genes/proteins in blunt snout bream, but it is not sensitive enough to functionally distinguish the two isoforms.
Genomes contain a large number of unique genes which have not been found in other species. Although the origin of such "orphan" genes remains unclear, they are thought to be involved in species-specific adaptive processes. Here, we analyzed seven orphan genes (MoSPC1 to MoSPC7) prioritized based on in planta expressed sequence tag data in the rice blast fungus, Magnaporthe oryzae. Expression analysis using qRT-PCR confirmed the expression of four genes (MoSPC1, MoSPC2, MoSPC3 and MoSPC7) during plant infection. However, individual deletion mutants of these four genes did not differ from the wild-type strain for all phenotypes examined, including pathogenicity. The length, GC contents, codon adaptation index and expression during mycelial growth of the four genes suggest that these genes formed during the evolutionary history of M. oryzae. Synteny analyses using closely related fungal species corroborated the notion that these genes evolved de novo in the M. oryzae genome. In this report, we discuss our inability to detect phenotypic changes in the four deletion mutants. Based on these results, the four orphan genes may be products of de novo gene birth processes, and their adaptive potential is in the course of being tested for retention or extinction through natural selection.
The purpose of this morphometric study was to obtain detailed quantitative information on all cell types in the testis interstitium of Korean ring-necked pheasants combined with data on changes in the steroidogenic function of the testis during the breeding and nonbreeding seasons. Animals collected during the breeding season, testis weights, sperm production, serum testosterone levels, leuteinizing hormone-stimulated testosterone secretion, and the length of the seminiferous tubules were significantly (p < 0.05) increased as compared to the nonbreeding season. Seminiferous tubules occupied 93.25% of testis volume in the breeding season. Leydig cells constituted 0.82% of the testicular volume. The mean volume of an Leydig cell was $1039{\mu}m^3$, and each testis contained about 24.53 million Leydig cells. Testes of the pheasants during the nonbreeding season displayed a 98% reduction in testis volume that was associated with a decrease in the absolute volume of seminiferous tubules (98% reduction), tubular lumen(100%), interstitium(90%), blood vessels(84%), lymphatic spaces(97%), Leydig cells(79%), mesenchymal cells(51%), and myoid cells(61%). The number of Leydig cells, mesenchymal cells, myoid cells per testis in the breeding season was higher (p < 0.05) than in the nonbreeding season. Although the average volume of a Leydig cell was 74% lower in the nonbreeding season, the average volume of a myoid and mesenchymal cell remained unchanged. These results demonstrate that there are a striking differences in the testicular structure of the Korean ring-necked pheasant in the breeding and nonbreeding seasons. Every structural parameter of the Leydig cell was pasitively correlated with both serum and LH-stimulated secretion concentrations of testosterone. Correlation of changes in hormonal status with morphometric alterations of all Leydig cell suggests that the Korean-ring necked pheasant may be used as a model to study structure-function relations in the avian testis.

Molecular mapping of plant genomes has progressed rapidly since Bostein et al.(1980) introduced the idea of constructing linkage maps of human genome based on restriction fragment length polymorphism (RFLP) markers. In recent years, the development of protein and DNA markers has stimulated interest for the new approaches to plant improvement. While classical maps based on morphological mutant markers have provided important insights into the plant genetics and cytology, the molecular maps based on molecular markers have a number of inherent advatages over classical genetic maps for the applications in genetic studies and/or breeding schemes. Isozymes and DNA markers are numerous, discrete, non-deleterious, codominant, and almost entirely free of environmental and epistatic interactions. For these reasons, they are widely used in constructing detailed linkage maps in a number of plant species. Plant breeders improve crops by selecting plants with desirable phenotypes. However a plant's phenotyes is often under genetic control, positioning at different "quantitative trait loci" (QTLs) together with environmental effects. Molecular maps provide a possible way to determine the effect of the individual gene that combines to produce a quantitative trait because the segregation of a large number of markers can be followed in a single genetic cross. Using market-assisted selection, plants that contain several favorable genes for the trait and do not contain unfavourable segments can be obtained during early breeding processes. Providing molecular maps are available, valuable data relevant to the taxonomic relationships and chromosome evolution can be accumulated by comparative mapping and also the structural relationships between linkage map and physical map can be identified by cDNA sequencing. After constructing high density maps, it will be possible to clone genes, whose products are unknown, such as semidwarf and disease resistance genes. However, much attention has to be paid to level-up the basic knowledge of genetics, physiology, biochemistry, plant pathology, entomology, microbiology, and so on. It must also be kept in mind that scientists in various fields will have to make another take off by intensive cooperation together for early integration and utilization of these newly emerging high-techs in practical breeding. breeding.

Trichomes are specialized epidermal structure having the functions of physical and chemical block against biotic and abiotic stresses. Several studies on $Capsicum$ species revealed that virus and herbivore resistance is associated with trichome-formation. However, there is no research on the structural characterization of trichomes developed on the epidermis of $Capsicum$ spp. Thus, this study attempts to charaterize the trichome morphologies in 5 species of $Capsicum$ using a Field Emission Scanning Electron Microscopy (FESEM). Six main trichome types were identified by their morphology under FESEM. Both glandular and non-glandular types of trichomes were developed on the epidermal tissues of $Capsicum$ spp. The glandular trichome were further classified into type I, IV and VII according to their base, stalk length, and stalk. Non-glandular trichomes were also classified into type II, III, and V based on stalk cell number and norphology. Almost all the species in $C.$$chinense$ and $C.$$pubescens$ had glandular trichomes. To our knowledge, this is the first study on classification of trichomes in the genus $Capsicum$ and, our results could provide basic informations for understanding the structure and function of trichomes on the epidermal differentiation and association with biotic stress tolerance.

As a consequence of tissue culture of rice, RAPD analysis was peformed to determine whether extended culture periods as undifferentiated calli affected the subsequent genetic constancy, and whether any resulting DNA rearrangements could be detected between sibling plants produced from the same callus. Somaclonal variation was induced at the initial stage of tissue culture and it increased with the length of culture maintenance. Of the 192 total bands, the number of polymorphic bands was 22 (11.5%), 33 (17.2%), and 49 (25.5%) in the callus of 1,3, and 6 months culture, respectively. A significantly higher level of genotypic polymorphisms between regenerants from two different somaclones was also detected, although all the regenerants were derived from a single genotype. In comparison of DNA polymorphisms between regenerants from non-irradiated and from irradiated calli, a scope of variation spectrum by gamma ray irradiation was larger than that by tissue culture. Consideration must be given to this genomic variation where attempts are to be made to use desirable somaclonal variants for plant breeding purpose and in genetic engineering program.
MyoD, expressed in skeletal muscle lineages of vertebrate embryo, is one of muscle-specific basic helix-loop-helix (bHLH) transcription factors, which plays a key role in the determination and differentiation of all skeletal muscle lineages. In this study, a cDNA of grass carp MyoD was cloned and characterized from total RNA of grass carp embryos by RT-PCR. The full-length cDNA of grass carp MyoD is 1597 bp. The cDNA sequence analysis reveals an open reading frame of 825 bp coding for a protein of 275 amino acids, which includes a bHLH domain composed of basic domain (1-84th amino acids) and HLH domain (98-142th amino acids), without signal peptide. Then the MyoD cDNA of grass carp was cloned to yeast expression vector pPICZ$\alpha$A and transformed into P. pastoris GS115 strain, the recombinant MyoD protein with a molecular weight of about 31KD was obtained after inducing for 2d with 0.5% methanol in pH 8.0 BMGY medium, and the maximum yield was about 250 mg/L in shaking-flask fermentation. The results were expected to benefit for further studies on the crystal structure and physiological function of fish MyoD.
