Plant Regeneration from Cotyledon and Hypocotyl Tissues of Chinese Cabbage (배추의 자엽과 배축 절편체로부터의 식물체 재분화)

  • The study was carried out to develop a simple and efficient system to regenerate plants from cotyledon and hypocotyl tissues of Chinese cabbage (Brassica campestris L. ssp. pekinensis cv Seoul). Among the various combinations of naphthalene acetic acid (NAA) and 6-benzyladenine (BA) tested, the best shoot induction medium for cotyledon, with 2.67 shoots per explants, contained $2.0mg{\cdot}L^{-1}$ NAA, $1.0mg{\cdot}L^{-1}$ BA and $16.7mg{\cdot}L^{-1}$ $AgNO_3$. The shoot induction medium with $1.0mg{\cdot}L^{-1}$ NAA, $5.0mg{\cdot}L^{-1}$ BA and $16.7mg{\cdot}L^{-1}$ $AgNO_3$, was best for shoot induction from hypocotyl explants, with 1.87 shoots per explants. After shoot induction, regenerated shoots were excised and rooted on rooting medium. Rooted plantlets were then hardened in the high humidity growth chamber and transplanted to pots, and then grown in the greenhouse. Regenerated plants appeared phenotypically normal and there were no changes in chromosome number.

Evaluation of Phosphorus Acid Treatment as a Growth Stimulant for Red pepper (Capsicum annuum L.), Cucumber (Cucumis sativus L.), and Kimchi cabbage (Brassica campestris L. ssp. pekinensis) in the Bed Soil Environment (상토 환경에서 고추(Capsicum annuum L.), 오이(Cucumis sativus L.) 및 배추(Brassica campestris L. ssp. pekinensis)에 대한 생장촉진제로서 아인산 처리의 평가)

  • This study was conducted to evaluate the effect of phosphorus acid (H3PO3) addition to the horticultural bed soil on the initial growth of red pepper (Capsicum annuum L. cv.), cucumber (Cucumis sativus L. cv.), and kimchi cabbage (Brassica campestris L. ssp. pekinensis (Lour.) Rupr. cv.). The stem heights of red pepper and cucumber were 46.1% and 23.0% greater in the 50 mg/L of phosphorus acid treatment than the untreated (control). Further, the stem diameter of pepper and cucumber were 48.7% and 23.0% greater in the 50 mg/L of phosphorus acid treatment than the control. In addition, the number of kimchi cabbage leaves was 47.5% greater in the 50 mg/L of phosphorus acid treatment than the control. The dry weights of red pepper, cucumber and kimchi cabbage were 72.9%, 16.5%, and 30.4% heavier in the 50 mg/L than the control, respectively. Cations (K, Ca, and Mg) and total phosphorus (T - P) were quantitatively analyzed for these three horticultural crops. The concentrations of K, Ca, and Mg, and T - P were higher in the 50 mg/L of phosphorus acid than the control, respectively. Based on the results obtained in this study, it appears that treatment of phosphorus acid in horticultural bed soil enhanced the growth of red pepper, cucumber and Kimchi cabbage.

Time-based Expression Networks of Genes Related to Cold Stress in Brassica rapa ssp. pekinensis (배추의 저온 스트레스 처리 시간대별 발현 유전자 네트워크 분석)

  • Plants can respond and adapt to cold stress through regulation of gene expression in various biochemical and physiological processes. Cold stress triggers decreased rates of metabolism, modification of cell walls, and loss of membrane function. Hence, this study was conducted to construct coexpression networks for time-based expression pattern analysis of genes related to cold stress in Chinese cabbage (Brassica rapa ssp. pekinensis). B. rapa cold stress networks were constructed with 2,030 nodes, 20,235 edges, and 34 connected components. The analysis suggests that similar genes responding to cold stress may also regulate development of Chinese cabbage. Using this network model, it is surmised that cold tolerance is strongly related to activation of chitinase antifreeze proteins by WRKY transcription factors and salicylic acid signaling, and to regulation of stomatal movement and starch metabolic processes for systemic acquired resistance in Chinese cabbage. Moreover, within 48 h, cold stress triggered transition from vegetative to reproductive phase and meristematic phase transition. In this study, we demonstrated that this network model could be used to precisely predict the functions of cold resistance genes in Chinese cabbage.

Glucosinolate and isothiocyabate contents according to processing of Kimchi cabbage (Brassica rapa L. ssp. pekinensis) (배추의 가공에 따른 glucosinolates 및 isothiocyanates 함량 변화)

  • This study examined the total glucosinolate (GSL) and isothiocyanate (ITC) contents according to different processing conditions; fresh Kimchi cabbage (Brassica rapa L. ssp. pekinensis), salted Kimchi cabbage and kimchi (storage temperature $4^{\circ}C$ and $20^{\circ}C$) using two different cultivars (Bomatnorang and Chunkwang). Four GSL peaks representing gluconapin, glucobrassicanapin, glucobrassicin and 4-methoxyglucobrassicin were detected in Kimchi cabbage by HPLC and HPLC/MS analysis. The total GSL contents of fresh Kimchi cabbage of Bomatnorang and Chunkwang were $21.37{\pm}1.06{\mu}g/g$ dry weight (DW) and $20.96{\pm}3.33{\mu}g/g$ DW, respectively. After salting, the total GSL contents of salted Kimchi cabbage decreased by 39% and 52% in Bomatnorang and Chunkwang, respectively. Finally, the total GSL contents of kimchi after storage at $20^{\circ}C$ decreased by 83% and 56% in Bomatnorang and Chunkwang, respectively. The extracted ITC contents were analyzed by GC/MS. Three ITC peaks were detected in Kimchi cabbage representing 2-phenylethyl ITC, 3-butenyl ITC and 4-pentenyl ITC. The 2-phenylethyl ITC levels increased during the salting process but this generally fell during storage at $20^{\circ}C$ as kimchi. The 3-butenyl ITC levels of Kimchi cabbage according to processing decreased rapidly due to salting and then decreased slowly during storage as kimchi. The 4-pentenyl ITC of Kimchi cabbage was lost during the salting process. The results for the change in GSL and ITC contents during the kimchi making process will be used in the food industry.

Effect of Aqueous Extracts on the Callus Growth, Seed Germination, and Growth of Some Plants and Antibacterial Activity by Equisetum arvense L. (쇠뜨기(Equisetum arvense L.)의 수용추출액이 몇가지 식물의 캘러스 생장, 종자발아, 유묘생장 및 항균활성에 미치는 영향)

  • To investigate phytotoxic substances in Equisetum arvense, the aqueous extracts(25 and 8$0^{\circ}C$) or the freeze-drying matter from aqueous extracts($25^{\circ}C$) of E. arvense were tested at different concentrations for biological activities on callus induction and growth, seed germination, seedling growth and antibacterial function. Callus induction and growth of Oryza sativa and Brassica campestris ssp. pekinensis were inhibited by the aqueous extracts at 8$0^{\circ}C$ than at $25^{\circ}C$ and the higher concentrations. Callus growth of four receptor species was inhibited in order of Platycodon grandiflorum, Sesamum indicum, Brassica campestris ssp. pekinensis and Oryza sativa. Seed germination of Ο. sativa, S. indicum, and B. campestris ssp. pekinensis was not affected at low concentration, but it was proportionally inhibited by the higher concentrations. The greatest inhibition of seed germination was 28.3% compared to control, when 2,000$\mu\textrm{g}$/ml of freeze-drying matter was applied to B. campestris ssp. pekinensis. Shoot growth was stimulated by 500$\mu\textrm{g}$/ml of freeze-drying matter, but it was inhibited by the higher concentrations. Root growth was significantly inhibited compare to control at all concentrations. Antibacterial activity of freeze-drying matter didn't showed against Xantomonas oryzae and Eschrichia coli, but a small clear zone was formed by 500$\mu\textrm{g}$ of freeze-drying matter against Erwinia carotovora ssp. carotovora.

배추 청방약근${\times}$무우 울산재래의 속간잡동에 대한 세포유전학적 연구

  • Intergeneric crossings between the inbred line of Brassica pekinensis var. Chongbangkokun and inbred line of Raphanus sativus var. Oolsanjaelae were made using Brassica as female plant, and obtained two individuals of intergeneric hybrids. Morphological characters of the two F1 hybrids are mostly intermediate of the two parental species, and their somatic chromosome number is 19. Meiotic behaviors of the parental plants were normal, while those of F1 plants were extremely irregular. The mean pairing frequencies per cell of hybrids are 1.09II+16.811(examined in May), and 1.76II+15.47i(examined in June). Majority of the pollen grain of parent species are normal, whereas those of F1's are invariably abortive. No seed setting was obtained in the hybrid plants left in the open field.

The Effect of Simulated Acid Rain on the Growth of Important Crops (인공 산성비가 중요 농작물의 생육에 미치는 영향)

  • The experiment was performed to elucidate the effect of simulated acrid rain on growth of the seven crops(caucus carota vu sativa DC. , Fagopyrum esculenkm Moench, Brassica cmpespis subsp. napus var. pekinensis Makino , Raphuus satlws vu. hortensis for. acnMormis Makino, Brassica dbogjabra Bailey, Caphslcum mum L., and Perilla frutescens Britton). The pH levels of simulated acid rain ranged pH 3.1, 3.6, 4.1, 4.6, 5.1 and 5.6. The germination 10 each crop was influenced from stimulated acrid rain except buckwheat and kale. A general decrease of growth in crops was observed with Increasing pH concentrations. The pattern of soluble protein was observed a tendency to decrease from acidic pH eradlents. According to acidity, total DNA contents of each crop was showed a definite reduction. In conclusion, plant growth was stimulated decreasln and the chanties of total Protein Patterns and BNA contents extracted from leaves trended with stimulated acrid rain was showed seriously.

Nimbya scirpicola Causing Brown Spot of Bayonet-Gras (Scirpus maritimus) (매자기에 갈색무늬병(가칭)을 일으키는 Nimbya scirpicola)

  • A brown leaf and stem spot disease of bayonet-grass (Scirpus maritimus) was epidemic in reclaimed paddy fields of Chunbuk province, Korea. A fungal pathogen was repeatedly isolated from the necrotic lesions of the bayonet-grass and identified as Nimbya scirpicola. The pathogen induced disease symptoms only in bayonet-grass but not in 8 other plants tested; Brassica pathogen induced disease symptoms only in bayonet-grass but not in 8 other plants tested; Brassica compestris subsp. napus var. pekinensis, Cucumis sativus, Glycine max, Hordeum vulgare, Lycopersicon esculentum, Oryza sativa, Sesamum indicum and Triticum aestivum. The fungus has potential to be developed as a mycoherbicide.

  • Penicillium sp. PS-113 showed no toxicity against loach(Misgurnus mizolepis) and monnow(Oryzias latipes), a kind of biological indicator, for 96 hours at the concentrations of $1.0\;{\times}\;10^8$ conidia/ml inoculum. Additionally, it showed no toxicity against several plants such as Chinese cabbage(Brassica campestris subsp. napus var. pekinensis), mung bean(Phaseolus radiatus), etc. during their germination at the same concentration of inoculum.
