EMS-induced Mutagenesis for C18 Unsaturated Fatty Acids in Rapeseed (Brassica napus L.)

  • Rapeseed (Brassica napus L.) oil with high oleic acid content is of great interest for both food and non-food uses. The 'Tamla' variety, characterized by oleic acid content of approximately 69%, was treated with 1% ethyl methane sulfonate (EMS) to induce mutations in the fatty acid biosynthesis pathway. $M_1$ plants were selfed and subsequent generations ($M_2$, $M_3$, and $M_4$ mutants) were analyzed to identify mutants having increased levels of oleic acid. $M_2$ mutants showed oleic acid content ranging from 13.5% to 76.9% with some mutants (TR-458 and TR-544) having up to 74.7% and 76.9% oleic acid, which was an increase of nearly 5% and 7%, respectively, compared to untreated cv 'Tamla'. We selected two $M_3$ mutants with >75% oleic acid content. One mutant (TR-458-2) had increased oleic acid (75.9%) and decreased linoleic acid (12.5%) and linolenic acid (4.4%) contents. The other (TR-544-1) showed increased oleic acid content (75.7%) and decreased linoleic acid (13.5%) and linolenic acid (3.3%) contents. The accumulation or reduction of oleic acid content in the selected $M_4$ mutants was also accompanied by a simultaneous decrease or increase in linoleic and linolenic acid contents. The high-oleic lines could be utilized further in breeding programs for improvement of rapeseed oil quality.

Functionality Changes of Rapeseed Protein upon Proteolysis (유채단백질의 Proteolysis에 의한 기능성 변화)

  • purified rapeseed(Brassica napus var. Youngsan) protein was hydrolyzed by pronase. The hyrolysate protein was investigated for the some physicochemical and functional properties. UV and intrinsic fluorescence spectra of the hydrolysate showed the maximum absorption at 274nm and 360nm respectively. Intensity of yellow color decreased in the process of hydrolysis and the surface hydrophobicity decreased up to fourfold. The main bands of hydrolysate by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) were observed at 14,000 to 12,000 dalton molecular weight. Solubilities of hydrolyzed protein increased by 10~15% compared to those of unhydrolyzed protein at acidic pH. In the hydrolysate, while absorption of both water and oil, foam expansion and emulsion stability were increased, absolute viscosity, heat coagulation, calcium coagulation, foam stability and emulsion activity were decreased.

The Hydrolysis Conditions of Rapeseed Protein by Pronase (유채단백질의 단백효소에 의한 가수분해 조건)

  • Optimum conditions for enzymatic hydrolysis of purified rapeseed(Brassica napus var. Youngsan) protein were investigated. Pronase showed higher activity in the hydrolysis of rapeseed protein than that of alcalase and neutrase. Preheated treatment of the rapeseed protein decreased the activity of pronase. The degree of hydrolysis of rapeseed protein was greater in distilled water than in phosphate buffer solution. Degree of hydrolysis was reached in steady state after 1 hr. Optimum conditions of the hydrolysis of the rapeseed protein were $40^{\circ}C$ in reaction temperaturem pH 8.0 in substrate solution, 1/100 (w/w) in the ratio of enzyme to substrate and 1% (w/v) in substrate concentration for pronase, respectively. At the optimum hydrolysis conditions, Km value was 3.48% (w/v).

Effect of Sulfate Supply Level on Sulfate Assimilation in Different Oilseed Rape Cultivars (유채 품종별 황 공급수준이 황산염 동화에 미치는 영향)

  • To determine sulfate uptake and assimilation of various rape cultivars (Brassica napus L.) under different S levels, the activity of ATP sulfurylase, ${SO_4}^{2-}$ uptake and glutathione (GSH) concentrations were measured in different age of leaves. In this study, ten rape cultivars (Mokpo, Tamra, Youngsan, Naehan, Saturnin, Akela, Mosa, Capitol, Pollen, and Colosse) showed various sulfate uptake and assimilation capacity in response to inefficient sulfate supply. Under inadequate sulfate conditions, a greater ATP sulfurylase activity in young leaves was observed in all cultivars compared to that of old leaves. In addition, GSH concentration was considerably increased in young leaves when S supply was declined from 2.0 to 0.2 mM. These results suggested that rape cultivars differ in their capacity to utilize sulfate under limited S conditions.

Changes in Composition and Content of Flavonoids by Processing Type in Rapeseed (Brassica napus) Flowers (유채꽃 가공유형별 플라보노이드 조성 및 함량 변화)

  • BACKGROUND: Increased value added by rapeseed (Brassica napus) by-product and the development of a usable rapeseed functional tea. METHODS AND RESULTS: To develop a usable rapeseed functional tea, the total flavonoid content in the varieties Youngsan, Tammi, Tamra, Naehan, Hanra, Mokpo No. 68, and Mokpo No. 111 was investigated. Effect of three treatments, i.e., drying, leaching, and roasting, on flavonoid contents or flower was tested using multiple processing methods per treatment. Total flavonoid content decreased under the various drying methods, confirming that flavonoid content is heat-dependent. This finding was more pronounced for freezing and oven-drying (15.3 and 13.8 mg/g DW, respectively), with a 10% difference in the total flavonoid content between the two methods. Under leaching conditions, the flavonoid content decreased with increasing treatment time. Notably, roasting methods did not result in loss of flavonoid content. The total flavonoid content in the rapeseed varieties decreased in the following order: Youngsan, Tammi, Tamra, Naehan, Hanra, Mokpo No. 68, and Mokpo No. 111. CONCLUSION: The flavonoid content in rapeseed flower was higher in Youngsan than in the other varieties, under processing conditions such as freeze-drying, leaching at $90^{\circ}C$ for 5 min, and roasting.

Partitioning of Carbon and Nitrogen Reserves During Winter Adaptation and Spring Regrowth I. Effects of temperature on growth, total content of nitrogen and non-structureal carbohydrate in forage rape(Brassica napus L.) (저장탄수화물과 질소의 월동성과 재생활력에 대한 이용성 I. 저온처리가 유채 ( Brassica napus L. ) 의 생육 , 질소 및 비구조성 탄수화물의 총 함량에 미치는 영향)

  • The objective of this study is to obtain the basic data for investigating the effects of organic reserves on winter survial or regrowth yield. Dry matter, nitrogen and non-structural carbohydrate content of plants grown under $5^{\circ}C$ or $20^{\circ}C$ of culture temperature during 25 days were investigated. The dry matter content of leaves and roots were significantly reduced under $5^{\circ}C$ compared with $20^{\circ}C$culture condition. Comparing with the dry matter per plant under $20^{\circ}C$, those in leaves and roots under $5^{\circ}C$ decreased to 25% and 10%, respectively, after 25 days of temperature treatment. Total nitrogen content in leaves under $20^{\circ}C$ and $5^{\circ}C$ increased to 68% and 39% compared to the initial lenel(day O), respectively, during 25 days after temperature treatment, Nitrogen content in roots highly increased under 5 C while there was a little change under $20^{\circ}C$ condition. The nitrogen contents in roots under $5^{\circ}C$ and $20^{\circ}C$ were 39.0 and 30.8mgJg DM, respectively, after 25 days of temperature treatment. Total contents of soluble carbohydrate in both leaves and roots under $5^{\circ}C$ were higher than those under $20^{\circ}C$ condition. After 25 days of temperature treatment under$5^{\circ}C$ , their contents in leaves and roots were 1.4 and 2.0 times higher than those of under $20^{\circ}C$ condition. Stach atent in roots under $20^{\circ}C$ was less changed, while thatof under $5^{\circ}C$ greatly increased from 64.8 to 178.7mglg DM duling 25 days. 'Ihese results clearly showed that an accumulation of both nitrogen and non-structural carbohydrate in the plants occured under low temperature condition.e condition.

  • The objective of this study is to obtain the basic data for investigating the potentiality of continuous utili~ation (first cutting in the late fall and regrowth yield in next spring) of forage rape seeded in fall. Fresh yield and the content of nitrogenous compound in leaves and roots were measured during a growth period. Fresh yield accumulation was very low between the late fall(470kg/10a) and the early regrowth period( 1.070kg /IOa). Total nitrogen content in leaves until the wintering period was decreased from 4.71% to 3.70%. while that in roots slightly increased during this period. The highest content(4.84%) in roots was observed in the early regrowth period, and then rapidly decreased as growth advanced. Protein-N was the largest pool of nitrogenous compound in leaves and roots through entire growth period, Its content in leaves decreased until the wintering period (Feb. 4), and then increased until the bolting stage(Apr. 10). Protein-N in roots highly accumulated from the late fall(11.1mg/gDM) to the early regrowth period(l6.9mg/gDM), and then decreased until the early tlowering stage. The content of amino acid-N in leaves showed a little change with a range from 5.7 to 8.5mgIgDM during entire growth period, while that in roots rapidly decreased from early regrowth period. The content of $NO_3$-N decreased from 7.0 to 4.3mglgDM in leaves, while increased from 0.9 to 2.3mg/gDM in roots from the late fall to the wintering period. 7he content of $NH_4$-N was lower and less varillble than other nitrogen compound during entire growth period. The results clearly showed that protein-N was the main storage form and highly accumulated in roots of overwintering forage rape.

  • The objective of this study is to obtain the basic data for investigating the potentiality of continuous utilization (first cutting in the late fall and regrowth yield in next spring) of forage rape seeded in fall. Non-structural carbohydrates(NSC) in leaves and roots of forage rape seeded on Oct. 3 were analyzed during a growth period. The greatest change in NSC content was observed between the wintering and the early spring period. The total content of soluble sugar in leaves and roots highly increased from the late fall(Nov. 7) to the wintering period (Feb. 4), and then rapidly decreased on the early regrow^ period(Mar. 31). The contents of fructose and glucose were relatively lower, and their quantitative change also was smaller than those of other sugars through entire growth period. The highest accumulation of sucrose occured hum the late fall to wintering period, and then greatly decreased in the early regrowth period. Sucrose content in roots was 2.3 times higher than that of leaves in wintering period. Starch was the largest pool of NSC and its content in leaves and roots showed a similar pattern with that of sucrose through entire growth period. Starch contents in leaves and roots were 38mg and 189mgl gDM in the late fall(Nov. 7), 187 and 497mg/gDM(Feb. 4) in the wintering period and 69 and 79mglgDM(Mar. 28) in the early regrowth period, respectively. The results clearly showed that the main reserve forms of NSC are starch and sucrose, and that they are hlghly stored in roots in overwintering forage rape.

Comparison of Sulfur Utilization among 10 Different Genotypes of Rape (Brassica napus) (다양한 유채품종에서의 황의 이용 효율 비교)

  • To investigate the sulfate utilization efficiency in different rape (Brassica napus) cultivars, sulfate uptake are analyzed under complete S-supply level (2.0mM ${SO_4}^{2-}$). This study used ten different genotypes of rape (Mokpo, Tamra, Youngsan, Naehan, Saturnin, Akela, Mosa, Capitol, Pollen and Colosse). For comparison of ${SO_4}^{2-}$ uptake among 10 cultivars, leaf number, leaf length and width, root length was also observed. Leaf length and width in all cultivars less variable among the cultivars examined. The longest root was shown in Saturnin (36.3 cm). ${SO_4}^{2-}$ uptake in Saturnin, Youngsan and Mokpo was significantly higher whereas that of Mosa and Pollen was relativety lower. Saturnin and Mokpo which have a high ${SO_4}^{2-}$ uptake exhibited a high ${NO_3}^-$ uptake.

Physicochemical and Functional Properties of Rapeseed Protein upon Purification Conditions (유채박 단백질의 정제조건에 따른 이화학적 성질과 기능성에 관한 연구)

  • We investigated the physicochemical and functinal properties of rapeseed(Brassica napus var. Youngsan) protein prepared by combining various soluvent and purification procedures, such as ultrafiltration (UF) concentration and acid-washin. The lightness value(YCIE) of each protein was greadully improved and its hydrophobicity increased by the degree of purification. The analysis of each protein by sodium dodecyl sulfate-plyacrylamide gel electrophoresis (SDS-PAGE) had nine bands revealed without difference, the considerable portion of which were of $1.96~1.59{\times}10^4$ dalton molecular weight. The content of amino acid increased a little more in the other processed proteins than in the control, and decreased considerably in the proteins extracted by the mixed solvent of 1% sodium hexametaphosphate(SHMP) and 0.25M ethlene diamine tetraacetic acid (EDTA). The better proteins were purified, the lower the kinematic viscosities were in their values. The water absorption and foaming properties were scarcely different according to the processes. The oil absorption and the emulsion activity index normally increased according to the degree of purification. The properties of heat coagulation revealed high values only in the proteins processed by EDTA while they showed considerably low values the other proteins.
