  • Objective: The neuroanatomical studies on the acupoints(Waiguan(SJ5), Neiguan(Pe6), Sanyinjiao(SP6) and Xuanzhong(GB39)) projecting to the brain area related to dimentia using the pseudorabies virus (PRV-Ba strain) in the mouse was described. Methods: The common locations of the brain projecting to the Waiguan, Neiguan, Sanyinjiao and Xuanzhong following injection of PRV-Ba were histochemically observed. The results were as follows Results : 1. PRV-Ba labeled areas in medulla oblongata, pons and midbrain were similar to 4 acupoints, theses areas were related to autonomic center. 2. PRV-Ba labeled areas in diencephalon and cebrebrum were differently labeled according to the acupoints. 3. CNS labeled areas in Waiguan were dense labeled in CA1-3 area of hippocampus, amygdaloid nucleus, insular cortex, parietal cortex, entorhinal cortex, perirhinal cortex, dorsal endopiriform cortex, piriform cortex, amygdalopiriform transition and bed n. of stria terminalis. 4. CNS labeled areas in Neiguan were dense labeled in insular cortex, amygdaloid nucleus, parietal cortex, entorhinal cortex, perirhinal cortex, dorsal endopiriform cortex, piriform cortex, amygdalopiriform transition and bed n. of stria terminalis. 5. CNS labeled areas in Sanyinjiao were dense labeled in CA1-3 of hippocampus, suprachiasmatic n., dorsal endopiriform cortex, piriform cortex and bed n. of stria terminalis. 6. CNS labeled areas in Xuanzhong were dense labeled in suprachiasmatic n., dorsal endopiriform cortex and piriform cortex. Conclusions : Following these results, labeled acupoints in brain areas related to dimentia are Waiguan and Neiguan. Common labeled areas are amygdaloid n., entorhinal cortex, amygdaopiriform transition, bed n. stria terminalis and perirhinal cortex.

간정격과 사관혈 침 치료의 우울 행동 개선 효과 및 뇌신경 반응성 분석 연구 (Antidepressant Effect of Liver Tonification and Four Gate Acupuncture Treatments and Its Brain Neural Activity)

  • Objectives : We aimed to identify the antidepressant effect of liver tonification acupuncture treatment (ACU (LT); KI10, LR8, LU8, LR4) and four gate acupuncture treatment (ACU (FG); LI4, LR3) and its brain neural activity in the normal and chronic restraint stress (CRS)-induced mouse model. Methods : Firstly, normal mice were given ACU (LT) or ACU (FG) and the c-Fos expressions in each brain region were analyzed to examine brain neural activity. Secondly, CRS was administered to mice for 4 weeks, then ACU (LT) or ACU (FG) was performed for 2 weeks. The depression-like behavior was evaluated using open field test (OFT) before and after acupuncture treatment. Then, the c-Fos expressions in each brain region were analyzed to examine brain neural activity. Results : In normal mice, ACU (FG) regulated brain neural activities in the hypothalamus, hippocampus, and periaqueductal gray. ACU (LT) changed more brain regions in the prefrontal cortex, insular cortex, striatum, and hippocampus, including those altered by ACU (FG). In CRS-induced model, ACU (LT) alleviated depression-like behavior more than ACU (FG). Also, brain neural activities in the motor cortex area 2 (M2), agranular ventral part and piriform of insular cortex (AIV and Pir), and cornu ammonis (CA) 1 and CA3 of hippocampus were changed by ACU (LT), and those of AIV and CA3 were also changed by ACU (FG). As in normal mice, ACU (LT) resulted in changes in more brain regions, including those altered by ACU (FG) in CRS model. M2, Pir, and CA1 were only changed by ACU (LT) in depression model, suggesting that these brain regions reflect the specific effect of ACU (LT). Conclusions : ACU (LT) relieved depression-like behavior more than ACU (FG), and this acupuncture effect was associated with modulation of brain neural activities in the motor cortex, insular cortex, and hippocampus.

Regional Heterogeneity of Morphological Changes in Cultured Rat Astrocytes

  • We examined astrocyte regional heterogeneity in their morphological changes in response to various stimuli. Astrocytes were cultured from six different neonatal rat brain regions including cerebral cortex, hippocampus, cerebellum, mid brain, brain stem and hypothalamus. Astrocyte stellation was induced by serum deprivation and the maximum stellation in different regional astrocytes was achieved after 2 h. After 24 h, in all astrocyte cultures, the level of stellation returned to their original level. Cerebellar or hypothalamic astrocytes were the most or the least sensitive, respectively, to serum deprivation. The order of maximum sensitivity to serum deprivation among different regional astrocytes was: cerebellum>mid $brain{\ge}hippocampus,\;brain\;stem{\ge}cerebral$ cortex>hypothalamus. Isoproterenol-induced astrocyte stellation was also examined in different regional astrocytes, and similar order of maximum sensitivity as in serum deprivation was observed. Next a possible developmental effect on astrocyte morphological changes was examined in cerebral cortex and cerebellum astrocytes cultured from postnatal day 1 (P1), P4 and P7 rat brains. A much higher sensitivity of cerebellum astrocytes to serum deprivation as well as isoproterenol treatment was consistently observed in P1, P4 and P7-derived astrocytes compared to cerebral cortex astrocytes. The present study demonstrates different regional astrocytes maintain different levels of morphological plasticity in vitro.

Injury of the Thalamocortical Pathway Between the Mediodorsal Nuclei and the Prefrontal Cortex in a Patient with Traumatic Brain Injury

  • Purpose: Traumatic brain injury (TBI) refers to brain damage caused by external forces or trauma. TBIs can vary in severity and result from accidents, falls, sports injuries, assaults, or other forms of physical trauma. The prefrontal cortex (PFC) is known have roles in various cognitive functions. We report on a patient with traumatic brain injury who showed prefrontal symptoms after injury of thalamocortical connections between mediodorsal nuclei (MD) of thalamus and PFC. Methods: A 54-year-old, male patient suffered a TBI as a result of a heavy object falling on his head. After onset of TBI, he showed typical symptoms of prefrontal lobe injury, including personality changes, memory impairment, and general cognition problem. The thalamocortical connections between MD and PFC (ventrolateral prefrontal cortex (VLPFC), dorsolateral prefrontal cortex (DLPFC), and obrbitofrontal cortex (OFC)) were reconstructed using diffusion tensor tractography. In terms of fractional anisotropy value, the right thalamocortical connections to the OFC were significantly lower than those of control subjects. Results: The value of mean diffusivity in the right thalamocortical connections to the DLPFC was significantly higher than that of control subjects. By contrast, both VLPFC and left OFC showed significant decrement in the tract volume of thalamocortical connections compared with that of control subjects. Conclusion: We reported on a patient who showed cognitive and neuropsychiatric impairment due to global injury of the thalamocoritcal connections between MD and PFC following TBI.

Effects of the Bee Venom Herbal Acupuncture on the Neurotransmitters of the Rat Brain Cortex

  • In order to study the effects of bee venom herbal acupuncture on the neurotransmitters of the rat brain cortex, herbal acupuncture with the bee venom group and normal saline group was performed bilaterally on the point corresponding to LI 4 of the rat. The average optical density of the neurotransmitters from the cerebral cortex was analyzed 30 minutes after the herbal acupuncture with immunohistochemical methods. The results were as follows: 1. The density of NADPH-diaphorase in the bee venom group was increased significantly at the motor cortex, visual cortex, auditory cortex, cingulate cortex, retrosplenial cortex, and perirhinal cortex, compared to the normal saline group. 2. The average optical density of vasoactive intestinal peptide in the bee venom group had significant changes at the insular cortex, retrosplenial cortex, and perirhinal cortex, compared to the normal saline group. 3. The average optical density of neuropeptide-Y in the bee venom group increased significantly at the visual cortex and cingulate cortex, compared to the normal saline group.

An EEG-based Brain Mapping to Determine Mirror Neuron System in Patients with Chronic Stroke during Action Observation

  • Purpose: The aim of this study was to compare EEG topographical maps in patients with chronic stroke after action observation physical training. Methods: Ten subjects were recruited from a medical hospital. Participants observed the action of transferring a small block from one box to another for 6 sessions of 1 minute each, and then performed the observed action for 3 minutes, 6 times. An EEG-based brain mapping system with 32 scalp sites was used to determine cortical reorganization in the regions of interest (ROIs) during observation of movement. The EEG-based brain mapping was comparison in within-group before and after training. ROIs included the primary sensorimotor cortex, premotor cortex, superior parietal lobule, inferior parietal lobule, superior temporal lobe, and visual cortex. EEG data were analyzed with an average log ratio in order to control the variability of the absolute mu power. The mu power log ratio was in within-group comparison with paired t-tests. Results: Participants showed activation prior to the intervention in all of the cerebral cortex, whereas the inferior frontal gyrus, superior frontal gyrus, precentral gyrus, and inferior parietal cortex were selectively activated after the training. There were no differences in mu power between each session. Conclusion: These findings suggest that action observation physical training contributes to attaining brain reorganization and improving brain functionality, as part of rehabilitation and intervention programs.

Normal and Disordered Formation of the Cerebral Cortex : Normal Embryology, Related Molecules, Types of Migration, Migration Disorders

  • The expansion and folding of the cerebral cortex occur during brain development and are critical factors that influence cognitive ability and sensorimotor skills. The disruption of cortical growth and folding may cause neurological disorders, resulting in severe intellectual disability and intractable epilepsy in humans. Therefore, understanding the mechanism that regulates cortical growth and folding will be crucial in deciphering the key steps of brain development and finding new therapeutic targets for the congenital anomalies of the cerebral cortex. This review will start with a brief introduction describing the anatomy of the brain cortex, followed by a description of our understanding of the proliferation, differentiation, and migration of neural progenitors and important genes and molecules that are involved in these processes. Finally, various types of disorders that develop due to malformation of the cerebral cortex will be discussed.

전침자극이 Spontaneously Hypertensive Rat의 대뇌겉질, 뇌줄기, 소뇌 부위의 Nitric Oxide Synthase 신경세포에 미치는 영향 (Difference in NOS between 2 Hz and 100 Hz EA in cerebral cortex, brain stem and cerebellum of spontaneously hypertensive rats)

  • Background and Objetive : The aim of this study was to investigate the effect of various electroacupuncture stimulation on NADPH-diaphorase in cerebral cortex, brain stem, cerebellum of spontaneously hypertensive rats. Materials and Methods : We evaluated the changes of NADPH-d-positive neurons using a histochemical method. The staining intensity of NADPH-d-positive neurons was assessed in a quantitative fashion using a microdensitometrical method based on optical density by means of an image analyzer. Results and Conculsion : The average optical density of NADPH-d-positive neurons of 100 Hz (bipolar square wave 0.2 ms duration and 100 Hz frequency) electroacupuncture treatment group significantly increased in most cortical areas comparison between the manual acupuncture and 2 Hz (bipolar square wave 0.2 ms duration and 2 Hz frequency) electroacupuncture groups. In the brain stem, the optical density of NADPH-d-positive neuron at only superficial gray layer of the superior colliculus area was same as cerebral cortex. We conclude that the morphological evidence for NADPH-d-positive neurons may be have regional change in cerebral cortex brain stem and cerebellum according to various electroacupuncture stimulations.

유머와 기쁨에 따른 뇌 활성화와 말초의 피부전기 반응 (Neural Correlates and Electrodermal Activity Produced by Humor and Joy)

  • 본 연구에서는 뇌의 반응과 말초신경계의 변화를 통하여 유머와 기쁨의 긍정적인 정서의 구분이 가능한가를 밝히고자 하였다. fMRI와 말초신경계 반응 측정 실험 각각을 수행하는 동안, 유머와 기쁨정서를 유발하는 동영상 자극(2분)이 제시되었고, 실험이 끝난 후 심리적인 평가도 함께 수행되었다. fURI 실험은 boxcar design으로 한 session 내에 두 block으로 구성되었다. 말초신경계 반응실험에서는 안정상태와 정서상태에서의 피부전기 반응을 측정하였다. fMRI 결과, 유머자극과 기쁨자극 제시 시 공통적으로 precentral Cortex, temporal Cortex, precuneus 가 활성화 되었고, 유머자극은 기쁨자극에 비하여 우측 middle temporal cortex, 우측 inferior frontal cortex, 좌측 middle frontal cortex 에서 큰 활성화를 보였다. 피부전기 반응(EDA) 분석 결과 두 정서 모두 안정상태에 비하여 유의하게 증가하였고, 유머자극은 기쁨자극에 비하여 피부전도수준(SCL)과 피부전도반응의 수(NSCR)에서 유의하게 증가하는 것으로 나타났다.

Characterization of Multiple Synaptic Boutons in Cerebral Motor Cortex in Physiological and Pathological Condition: Acrobatic Motor Training Model and Traumatic Brain Injury Model

  • Multiple synaptic boutons (MSBs) have been reported to be synapse with two or more postsynaptic terminals in one presynaptic terminal. These MSBs are known to be increased by various brain stimuli. In the motor cortex, increased number of MSB was observed in both acrobat training (AC) model and traumatic brain injury (TBI) model. Interestingly one is a physiological stimuli and the other is pathological insult. The purpose of this study is to compare the connectivity of MSBs between AC model and TBI model in the cerebral motor cortex, based on the hypothesis that the connectivity of MSBs might be different according to the models. The motor cortex was dissected from perfused brain of each experimental animal, the samples were prepared for routine transmission electron microscopy. The 60~70 serial sections were mounted on the one-hole grid and MSB was analyzed. The 3-dimensional analysis revealed that 94% of MSBs found in AC model synapse two postsynaptic spines from same dendrite. But, 28% MSBs from TBI models synapse two postsynaptic spines from different dendrite. This imply that the MSBs observed in motor cortex of AC model and TBI model might have different circuits for the processing the information.
