Background: The aim of the study was to investigate any associations between benign prostate hyperplasia (BPH) and single nucleotide polymorphisms (SNPs) in the VDR gene (FokI, BsmI, ApaI and Taq${\alpha}$I loci) and the CYP17 gene (MspA1I locus), as well as TA repeat polymorphism in SRD5A2 gene among Lebanese men. Materials and Methods: DNA extracted from blood of 68 subjects with confirmed BPH and 79 age-matched controls was subjected to PCR/PCR-restriction fragment length polymorphism analysis. The odds ra=tio (OR) of having a genotype and the relative risk (RR) of developing BPH for having the genotype were calculated and the alleles were designated risk-bearing or protective. Results: Our data indicated that the A and B alleles of the VDR ApaI and BsmI SNPs were highly associated with increased risk of BPH (p=0.0168 and 0.0002, respectively). Moreover, 63% of the controls compared to 43% of the subjects with BPH were homozygous for none of the risk-bearing alleles (p=0.0123) whereas 60% of the controls and 28% of the subjects with BPH were homozygous for two or more protective alleles (p<0.0001). Conclusions: For the first time, our study demonstrated that ApaI and BsmI of the VDR gene are associated with risk of BPH among Lebanese men. Our study also indicated that overall polymorphism profile of all the genes involved in prostate physiology could be a better predictor of BPH risk.

In this study, the marker compounds of Curcumae Rhizoma (CR) were simultaneously quantified by high-performance liquid chromatography equipped with a photodiode array detector and the anti-inflammatory effects of CR extract and marker compounds in human benign prostatic hyperplasia epithelial-1 (BPH-1) cell lines were investigated. The marker components (4S,5S)-(+)-germacrone-4,5-epoxide, furanodienone, and germacrone, were separated on Gemini $C_{18}$ columns ($250mm{\times}4.6mm$, $5{\mu}m$) at $40^{\circ}C$ by using a gradient of two mobile phases eluting at 1.0 mL/min. Prostaglandin $E_2$ ($PGE_2$) levels in Human BPH-1 cells were determined with an ELISA kit. The coefficients of determination in a calibration curve of each analyte were all 0.9997. The limits of detection and quantification of the three compounds were $0.10-0.32{\mu}g/mL$ and $0.30-0.98{\mu}g/mL$, respectively. The content of three compounds, (4S,5S)-(+)-germacrone-4,5-epoxide, furanodienone, and germacrone, in the CR sample were found to be 5.79 - 5.92 mg/g, 4.72 - 4.86 mg/g, and 1.06 - 1.09 mg/g, respectively. Regarding pharmacological activity against benign prostatic hyperplasia, CR and its components significantly suppressed $PGE_2$ levels of BPH-1 cells. The established analysis method will help to improve quality assessment of CR samples and related products. In addition, CR and its components exhibit antiinflammatory activity in BPH-1 cells, suggesting the inhibitory efficacy of these compounds against the pathogenesis of BPH.

Comamonas testosteroni (formerly Pseudomonas sp.) NCIMB 10643 can grow on biphenyl and alkylbenzenes $(C_2-C_7)$ via 3-substituted catechols. Thus, to identify the genes encoding the degradation, transposon-mutagenesis was carried out using pAG408, a promoter-probe mini-transposon with a green fluorescent protein (GFP), as a reporter. A mutant, NT-1, which was unable to grow on alkylbenzenes and biphenyl, accumulated catechols and exhibited an enhanced expression of GFP upon exposure to these substrates, indicating that the gfp had been inserted in a gene encoding a broad substrate range catechol 2,3-dioxygenase. The genes (2,826 bp) flanking the gfp cloned from an SphI-digested fragment contained three complete open reading frames that were designated bphCDorfl. The deduced amino acid sequences of bphCDorfl were identical to 2,3-dihydroxybiphenyl 1,2-dioxygenase (BphC), 2-hydroxy-6-oxo-6-phenylhexa-2,4-dienoate hydrolase (BphD), and OrfI, respectively, that are all involved in the degradation of biphenyl/4-chlorobiphenyl (bph) by Ralstonia oxalatica A5. The deduced amino acid sequence of the orfl revealed a similarity to those of outer membrane proteins belonging to the OmpW family. The introduction of the bphCDorfl genes enabled the NT-l mutant to grow on aromatic hydrocarbons. In addition, PCR analysis indicated that the DNA sequence and gene organization of the bph operon were closely related to those in the bph operon from Tn4371 identified in strain A5. Furthermore, strain A5 was also able to grow on a similar set of alkylbenzenes as strain NCIMB 10643, demonstrating that, among the identified aromatic hydrocarbon degradation pathways, the bph degradation pathway related to Tn4371 was the most versatile in catabolizing a variety of aromatic hydrocarbons of mono- and bicyclic benzenes.

Rice plant extracts of brown planthopper (BPH) resistant rice varieties, Jangseongbyeo (JSB) and Hwacheongbyeo (HCB) at different growth stages (seedling, tillering, heading and ripening) were sequentially fractioned using hexane, ethyl ether, ethyl acetate, butanol, and distilled water. The extracts were applied to BPH susceptible rice variety, Dongjjnbyeo (DJB), to investigate the insecticidal and repellent effects against BPH. BPH insecticidal effects were not clearly observed with almost all of the extract fractions obtained from both JSB and HCB varieties for 12 h, whereas the ethyl ether and hexane extract fractions showed about 10 to 30% of BPH mortality in 24 to 48 h of application periods. An effective BPH repellent activity was found with the applications of ethyl ether extract fractions obtained from JSB variety. The extract fractions obtained from HCB variety did not show any different repellence among the various fractions. The BPH repellent effects of the extract fractions obtained at different growth stages of either JSB or HCB varieties did not show any correlations. The effect of ethyl ether fraction on BPH repellent was continually increased by 30 h after treatment and thereafter decreased. In addition, the first sub-fraction separated by a flash column chromatography eluted with chloroform:methanol (9:1, v/v) from the BPH effective ethyl ether faction in JSB variety might be meaningful to repel BPH from BPH susceptible target rice plants. The results indicated that the ethyl ether fraction obtained from JSB was higher in repellent activity than in insecticidal activity, and suggesting that there might be specific substance(s) in the first sub-fraction (sF1) of the ethyl ether fraction in JSB that could provide repellent activity against BPH.

Leptin is a type of adipokine mainly produced by adipocytes and reported to be overproduced in prostate cancer. However, it is not known whether it stimulates the proliferation of prostate cells. In this study, we investigated whether benign prostatic hyperplasia epithelial cells (BPH-1 cells) infected with Trichomonas vaginalis induced the proliferation of prostate cells via a leptin signaling pathway. To investigate the effect of crosstalk between adipocyte leptin and inflamed epithelial cell in proliferation of prostate cells, adipocytes 3T3-L1 cells were incubated in conditioned medium of BPH-1 cells infected with T. vaginalis (T. vaginalis-conditioned medium, TCM), and then the adipocyte-conditioned medium (ATCM) was identified to cause proliferation of prostate cells. BPH-1 cells incubated with live T. vaginalis released pro-inflammatory cytokines, and conditioned medium of these cells caused migration of adipocytes. When prostate stromal cells and BPH-1 cells were incubated with adipocyte conditioned medium containing leptin, their growth rates increased as did expression of the leptin receptor (known as OBR) and signaling molecules such as JAK2/STAT3, Notch and survivin. Moreover, blocking the OBR reduced this proliferation and the expression of leptin signaling molecules in response to ATCM. In conclusion, our findings show that inflamed BPH-1 cells infected with T. vaginalis induce the proliferation of prostate cells through leptin-OBR signaling. Therefore, it is likely that T. vaginalis contributes to prostate enlargement in BPH via adipocyte leptin released as a result of inflammation of the prostate.
Some attempts were made to investigate the biological characteristics of the brown planthopper(BPH), Nilaparvata lugens St${\aa}$l, biotypes in terms of perference in feeding oviposition on rice field, egg and nymphal periods, egg hatchability, emergemce ratio, growth index, adult lingevity and gecundity fed on 60-day-old rece cultivaes with different resistance genes. Feeding and oviposition perference of the three BPH biotypes on Dongjinbyeo with no resistance gene were shown very higher than on rice cultivars with resistance genes. Those of biotype-2 on Cheongcheongbyeo with bph 1 gene and bitype-3 on Milyang 63 with bph 2 gene were relatively high, however, they were still remarkably lower than those of Dongjibyeo. The egg and nymphal periods of the three BPH biotyes on Milyang 23 were shorter than on the other rice cultivars. The periods of biotype-2 on Cheongcheongbyeo and biotype-3 on Milyang 63 were as short as those of the three BPH biotypes on Milyang 23. The egg hachability, emergence ratio, and growth index of the three BPH bioyoes on Milyang 23 were higher than on the other rice cultivars and those of biotype-2 on Cheongcheongbyeo and biotyoe-3 on Milyang 63 were as high as on Milyang 23. The female adult longevity of the BPH biotype-1 on Milyang 23 was longer than on the other cultivaes, and that of biotype-2 on Cheongcheongbyeo and bioty-3 an Milyang 63 were as ling as on Milyang 23. The number of egg laid by a female of the three BPH biotypes were greatly increased on Milyang 23. Those of biotype-2 on Cheongcheongbyeo and biotype-3 an Milyang 63 were also greatly increased.
Houttuynia cordata Thunberg has been studied for a variety of pharmacological actions in traditional oriental medicine. In this study, we investigated the effects of Houttuynia cordata ethanol extract (HCE) on benign prostatic hyperplasia (BPH) models induced by castration and testosterone propionate (TP) injection. Thirty rats were divided into six groups. One group was used as a normal control, and the other groups were castrated and had intraperitoneal injections of TP for 14 days to induce BPH. A positive control group was given daily doses of finasteride (5 mg/kg) to the BPH model. Rats administered HCE (0.5, 1 or 2 mg/kg) instead of finasteride were compared with controls as experimental groups. There was no statistical significance in terms of prostate weight based on 100 g of body weight. The concentrations of 5-α reductase and dehydroxytestosteronre (DHT) were determined via ELISA tests, and there was a significant decrease in all experimental groups. The 0.5 mg/kg HCE group had the lowest level of 5-α reductase, and the 2 mg/kg HCE group had the lowest level of DHT. In the histopathological observation of prostates, the control and the 2 mg/kg HCE groups had normal cell shapes and no swelling. However, in the negative control group and the 1 mg/kg HCE group, the cells were swollen, and the gap between the cells was narrowed. In particular, in the 0.5 mg/kg HCE group, some cells were bursting. Therefore, the administration of more than 2 mg/kg of HCE is suitable to protect against BPH.
The effects of 1 wt.% N-benzylpyrazinium hexafluoroantimonate (BPH) as a thermal latent initiator and blend compositions composed of 0, 5, 10, 20 and 40 wt.% of phenol-novolac resin to epoxy resin were investigated in terms of cure kinetics, thermal stabilities and rheological properties. Thermal latent properties of BPH were measured from the conversion as a function of reaction temperature on a dynamic DSC. This cationic BPH system turned out to be an effective thermal latent initiator in the epoxy-phenol curing system. And the increase of phenol-novolac resin concentration led to the decrease in the latent temperature and to the increase of cure activation energy ($E_a$) of the blend system. The thermal stability and activation energy ($E_t$) for decomposition, gel-time and activation energy ($E_c$) for cross-linking from rheometer increased within the composition range of 20~40 wt.% of phenol-novolac resin. This implies that the three-dimensional cross-linking may take place among hydroxyl group within phenol resin, epoxide ring within epoxy resin and BPH.
Cure behavior and thermal stability of the different ratio of diglycidylether of bisphenol A(DGEBA)/trimethylolpropane triglycidylether(TMP) epoxy blends initiated by 1 wt % N-benzylpyrazinium hexafluoroantimonate (BPH) as a cationic latent catalyst were studied using DSC and TGA, respectively. Latent properties were performed by measurement of the conversion as a function of temperature using dynamic DSC. Dynamic DSC thermograms of DGEBA/TMP blends revealed that the weak peak was formed by complex formation between the hydroxyl groups in DGEBA and BPH, and between epoxides and BPH in low temperature ranges. The strong peak was considered as an exothermic reaction by the formation of three-dimensional network in high temperature ranges. Isothermal DSC revealed that the reaction rate of the blends was found to be higher than that of the neat TMP. The thermal stabilities in the cured resins were increased with increasing the DGEBA content. These results could be interpreted in terms of the stable aromatic structure, existence of hydroxyl group and high molecular weight of DGEBA.
In this work, two thermal latent catalysts, i.e., N-benzylpyrazinium hexafluoroantimonate (BPH) and benzyl-2,5-dimethylpyrazinium hexafluoroantimonate (BDPH), were synthesized. The cure behaviors and thermal stabilities of diglycidylether of bisphenol A (DCEBA) epoxy resins initiated by 1 wt.% of the catalysts were investigated by DSC, NIR, TCA, and DMA Latent properties of the catalysts were examined by conversion of epoxy resins using NIR from $100^{\circ}C$ to $180^{\circ}C$ From the resultes of near-IR, DGEBA/BPH system showed higher conversion than that of DGEBA/BDPH system. The thermal stabilities of DGEBA/BDPH system based on the initial decomposition temperature (IDT) and integral procedural decomposition (IPDT) were relatively lower than those of DCEBA/BPH system. These could be attributed to the hindered structure of BDPH, resulting in decreasing the thermal stability in the DGEBA/BDPH system.
