Clinical, hematological, and pathohistological findings of cattle with bovine leukocyte adhesion deficiency (BLAD) (우백혈구유착결손증(牛白血球癒着缺損症)의 임상(臨床), 혈액(血液) 및 병리조직소견(病理組織所見))

  • During the period from April 1991 to July 1992 clinical, hematological, and pathohistological findings of Holstein-friesian calves 47 with bovine leukocyte adhesion deficiency(BLAD, immunologically ascertained), which were referzed to the clinic for diseases of cattle, veterinary school, Hannover, were described. Most cases show poor body condition, rough and dry in haircoat, salivation, gingivitis, reduction of gingiva and alveolar bone, exposing the incisors' necks, loss of teeth, phlegmonous subcutaneous swellings, ulcerated tongue, recurang fever, coughing, dyspnea, pharyngeal and laryngeal stertor, periodical diarrhea, impaired swallowing, placid and less painsensitive. Relevant laboratory findings are persistent leucocytosis(with more than 30,000 up to 150,000 cells per $mm^3$ of blood), marked neutrophilia(without "shift to the left"), hyperproteinemia, and hypergammaglobulinemia. At post-mortem the carcass of BLAD-affected calves is usually emaciated. All lymphnodes of the respiratory and gastrointestinal tract appear markedly activated(swollen). Lesions in the mouth(gingivitis, defective dentition, pulpitis/alveolar paraodontitis, ulcerated tongue), throat and larynx(inflammation/ulceration), and lungs(pneumonic foci) correspond to the clinical symptoms seen on the living animal. There may be ulcers on the prestomachal mucosa, hyperemia of the intestinal mucosa with hyperplasia of Peyer's patches, ulceration and/or intramural abscesses. The spleen shows follicular hyperplasia. Microscopically, both myeloand erythropoesis are markedly activated in the bone marrow ; capillaries in many organs show leucocytostasis.

Effects of different calcium-silicate based materials on fracture resistance of immature permanent teeth with replacement root resorption and osteoclastogenesis

  • Objectives: This study evaluated the effects of Biodentine (BD), Bio-C Repair (BCR), and mineral trioxide aggregate (MTA) plug on the fracture resistance of simulated immature teeth with replacement root resorption (RRR) and in vitro-induced osteoclastogenesis. Materials and Methods: Sixty bovine incisors simulating immature teeth and RRR were divided into 5 groups: BD and BCR groups, with samples completely filled with the respective materials; MTA group, which utilized a 3-mm apical MTA plug; RRR group, which received no root canal filling; and normal periodontal ligament (PL) group, which had no RRR and no root canal filling. All the teeth underwent cycling loading, and compression strength testing was performed using a universal testing machine. RAW 264.7 macrophages were treated with 1:16 extracts of BD, BCR, and MTA containing receptor activator of nuclear factor-kappa B ligand (RANKL) for 5 days. RANKL-induced osteoclast differentiation was assessed by staining with tartrate-resistant acid phosphatase. The fracture load and osteoclast number were analyzed using 1-way ANOVA and Tukey's test (α = 0.05). Results: No significant difference in fracture resistance was observed among the groups (p > 0.05). All materials similarly inhibited osteoclastogenesis (p > 0.05), except for BCR, which led to a lower percentage of osteoclasts than did MTA (p < 0.0001). Conclusions: The treatment options for non-vital immature teeth with RRR did not strengthen the teeth and promoted a similar resistance to fractures in all cases. BD, MTA, and BCR showed inhibitory effects on osteoclast differentiation, with BCR yielding improved results compared to the other materials.

In Vitro Evaluation of Shear Bond Strengths of Zirconia Cerami with Various Types of Cement after Thermocycling on Bovine Dentin Surface (지르코니아 표면 처리와 시멘트 종류에 따른 치면과의 전단 결합 강도 비교 연구)

  • State of problem : The use of zirconium oxide all-ceramic material provides several advantages, including a high flexural strength(>1000MPa) and desirable optical properties, such as shading adaptation to the basic shades and a reduction in the layer thickness. Along with the strength of the materials, the cementation technique is also important to the clinical success of a restoration. Nevertheless, little information is available on the effect of different surface treatments on the bonding of zirconium high-crystalline ceramics and resin luting agents. Purpose : The aim of this study was to test the effects of surface treatments of zirconium on shear bond strengths between bovine teeth and a zirconia ceramic and evaluate differences among cements Material and methods : 54 sound bovine teeth extracted within a 1 months, were used. They were frozen in distilled water. These were rinsed by tap water to confirm that no granulation tissues have left. These were kept refrigerated at $4^{\circ}C$ until tested. Each tooth was placed horizontally at a plastic cylinder (diameter 20mm), and embedded in epoxy resin. Teeth were sectioned with diamond burs to expose dentin and grinded with #600 silicon carbide paper. To make sure there was no enamel left, each was observed under an optical microscope. 54 prefabricated zirconium oxide ceramic copings(Lava, 3M ESPE, USA) were assigned into 3 groups ; control, airborne-abraded with $110{\mu}m$ $Al_2O_3$ and scratched with diamond burs at 4 directions. They were cemented with a seating force of 10 ㎏ per tooth, using resin luting cement(Panavia $F^{(R)}$), resin cement(Superbond $C&B^{(R)}$), and resin modified GI cement(Rely X $Luting^{(R)}$). Those were thermocycled at $5^{\circ}C$ and $55^{\circ}C$ for 5000 cycles with a 30 second dwell time, and then shear bond strength was determined in a universal test machine(Model 4200, Instron Co., Canton, USA). The crosshead speed was 1 mm/min. The result was analyzed with one-way analysis of variance(ANOVA) and the Tukey test at a significance level of P<0.05. Results : Superbond $C&B^{(R)}$ at scratching with diamond burs showed the highest shear bond strength than others (p<.05). For Panavia $F^{(R)}$, groups of scratching and sandblasting showed significantly higher shear bond strength than control group(p<.05). For Rely X $Luting^{(R)}$, only between scratching & control group, significantly different shear bond strength was observed(p<.05). Conclusion : Within the limitation of this study, Superbond $C&B^{(R)}$ showed clinically acceptable shear bond between bovine teeth & zirconia ceramics regardless of surface treatments. For the surface treatment, scratching increased shear bond strength. Increase of shear bond strength by sandblasting with $110{\mu}m$ $Al_2O_3$ was not statistically different.

  • The purpose of this study was to evaluate the adaptability of light-cured glass ionomer cement to cavity walls. Class V cavities were prepared on the labial surfaces of extracted bovine incisor teeth. The cavities were restored with Fuji II as self-cured glass ionomer cement and Fuji II LC, Vitremer as light-cured glass ionomer cement. Fluorescent markers (fluoreceine and rhodamin B) were incorperated into liquid and primer for a better image of microscopic observation. Restored teeth were sectioned by longitudinal and labiolingual direction. The adaptability at the tooth-restoration interface was assessed incisally, axially and cervically by confocal scanning laser microscope. Following results were obtained : 1. Chemical-cured glass iomomer cement restoration showed close adaptation on the all of the cavity walls, but, cracks formed within the cement. 2. Light-cured glass ionomer cement restoration was well adapted to the cavity walls, but showed crack in the cement adjacent to axial dentinal wall. 3. There' was no significant difference in adaptability between two light-cured glass ionomer cement restorations.

  • This study evaluated the possibility of the 3-dimensional attachment of human periodontal ligament fibroblasts to a periodntally involved root surface after an EDTA treatment in vitro. The human PDL fibroblasts were isolated from the middle third of the root of periodontally healthy teeth extracted for orthodontic reasons. The cells were cultured in a medium containing Dulbecco's modified Eagle medium supplemented with 10% fetal bovine serum at $37^{\circ}C$ in humidified air containing 5% $CO_2$. Eight single-rooted teeth were obtained from patients diagnosed with periodotitis. After scaling and root planing, four teeth were etched with 24% ethylenediaminetetracetic acid (EDTA) for two minutes (Experimental group). The other four teeth were not treated with EDTA and were used as the control group. The human PDL fibroblasts were placed in the total root surface and cultured for 4 weeks. The teeth were fixed in 2.5% glutaraldehyde in PBS before preparation for the scanning electron microscopy (SEM) examination. The human PDL fibroblasts showed a healthy morphology on the root surfaces treated with EDTA (Experimental group) and a relatively unhealthy appearance on the treated root surfaces (Control group). This suggests that EDTA favorably affects the 3-dimensional attachment of human PDL fibroblasts cultured on the root surfaces. which may play an important role in periodontal healing and regeneration.

Clinical and Histopathological Study Using Platelet Rich Plasma and Bone Grafts in Extraction Sockets (발치와의 혈소판 농축 혈장과 골 이식술 시행시 골 형성에 대한 임상 및 조직병리학적 연구)

  • The socket preservation procedure was a simple and effective technique, and has better prognosis for implantation. The socket preservation usually used barrier membrane in combination with/without alloplastic bone materials. A recently study had shown that a regenerative therapy to tooth extraction utilizing growth factors made better results. Platelet-rich plasma was clinically easy method that acquired the growth factors, and is known that accelerated new bone formation and mineralization of bone graft materials. The purpose of this study was to evaluate clinical and histopathologic results which occur following socket preservations using platelet-rich plasma and bovine bone powder. Twelve patients who required extraction of one or more teeth for implantation at the department of periodontics in Dankook University Dental Hospital were selected. Extraction sockets were treated by using platelet-rich plasma and bovine bone powder. 3 months later, we observed clinical and histopathological results as follows: 1. Internal vertical measurement was an average of 7.33mm preoperatively and statistically significantly decreased to an average of 1.42mm postoperatively(p<0.05). 2. External vertical measurement was an average of 3.33mm preoperatively and decreased to an average of 2.75mm postoperatively; therefore there was no significant difference. 3. Horizontal measurement was an average of 7.75mm preoperatively and statistically decreased to an average of 6.08mm postoperatively(p<0.05). 4. Osteocyte-like cells and new bone formation connected with bovine bone grafts were observed in histopathologic examination. This study implied that platelet-rich plasma and bovine bone powder grafts were effective treatment for socket preservation and regeneration of severe bony defect made by implantation failure.

An experimental study on the effect of children's range beverage on bovine enamel (시판중인 어린이음료가 치아 표면의 탈회에 미치는 영향)

  • The purpose of this study was to observe the demineralization effects of children's range beverage on bovine enamel. 15 bovine teeth without dental caries were divided into 4 experimental groups (n=12) and control group (n=3). All samples immersed in beverages maintained 48 hours by $37^{\circ}C$ in incubate respectively. The surface microhardness values(${\Delta}VHN$) between before and after 48 hours treatment decreased and DIGNOdent value increased because enamel surface were corroded by experimentation beverage. Especially, there was statistical significantly difference in the carbonated drink. Morphological changes by scanning electron microscope were indicated that children's range beverage cause significant enamel demineralization effects on bovine tooth surface among five beverages.

  • The purpose of this study is to investigate shear bond strengths of $Peak^{(R)}$ Universal Bond (Ultradent, USA) containing 0.2% chlorhexidine in bovine dentin. Total of 30 bovine teeth were divided into three groups, 10 teeth each. Before comparing and evaluating shear bond strength, in group I, $Adper^{TM}$ Single Bond Universal (3M ESPE, USA) was applied, in group II, processing with $Consepsis^{(R)}$ (Ultradent, USA) was followed by applying $Adper^{TM}$ Single Bond Universal, and in group III, $Peak^{(R)}$ Universal Bond was applied and filled with $Filtek^{TM}$ Z-350 XT (3M/ESPE, USA) shade B3. As a result, processing with $Consepsis^{(R)}$ after acid etching showed no statistically significant influence on shear bond strength of dentin (p > 0.05). The shear bond strength of with or without $Consepsis^{(R)}$ on $Adper^{TM}$ Single Bond Universal and that of $Peak^{(R)}$ Universal Bond showed statistically significant difference (p < 0.05).

Effect of fluoride varnishes on the surface hardness of bovine teeth under demineralization/remineralization cycling

  • We investigated whether fluoride varnishes recover the hardness of bovine teeth under 20 days of demineralization/remineralization cycling. The fluoride varnish groups (two commercial fluoride varnishes [V-varnish (Vericom, Korea) and CavityShield (3M ESPE, USA)] and an experimental fluoride varnish including 5 wt.% NaF were compared with a control group without fluoride varnish. Vickers hardness was measured at baseline, 3 days after immersion in caries-inducing solution, 24 hours after application of a fluoride varnish, and after 10 and 20 days of demineralization/remineralization cycling. Afterward, tooth surfaces were observed by scanning electron microscope. After fluoride varnish application and the cycling 10 and 20 days, the experimental varnish group showed the highest hardness, while the CavityShield and the control groups demonstrated the lowest hardness. The experimental varnish group recovered the hardness of the baseline at 24 hours after application of the varnish, while it was recovered after 20 days of the cycling in case of the V-varnish. However, the CavityShield and the control groups did not recover the hardness even after 20 days of the cycling. The experimental fluoride varnish with fast recovery in the hardness of the baseline can be used as an effective fluoride varnish to resist demineralization and to facilitate remineralization.

The effect of different fluoride application methods on the remineralization of initial carious lesions

  • Objectives: The purpose of this study was to assess the effect of single and combined applications of fluoride on the amount of fluoride release, and the remineralization and physical properties of enamel. Materials and Methods: Each of four fluoride varnish and gel products (Fluor Protector, FP, Ivoclar Vivadent; Tooth Mousse Plus, TM, GC; 60 Second Gel, A, Germiphene; CavityShield, CS, 3M ESPE) and two fluoride solutions (2% sodium fluoride, N; 8% tin(ii) fluoride, S) were applied on bovine teeth using single and combined methods (10 per group), and then the amount of fluoride release was measured for 4 wk. The electron probe microanalysis and the Vickers microhardness measurements were conducted to assess the effect of fluoride application on the surface properties of bovine teeth. Results: The amount of fluoride release was higher in combined applications than in single application (p < 0.05). Microhardness values were higher after combined applications of N with FP, TM, and CS than single application of them, and these values were also higher after combined applications of S than single application of A (p < 0.05). Ca and P values were higher in combined applications of N with TM and CS than single application of them (p < 0.05). They were also increased after combined applications of the S with A than after single application (p < 0.05). Conclusions: Combined applications of fluoride could be used as a basis to design more effective methods of fluoride application to provide enhanced remineralization.
