Isolation and Identification of Korean type Streptococcus mutans (한국형 Streptococcus mutans의 분리 및 동정)

  • S. mutans known as a causative causative agent of dental caries was isolated from a carious lesion of Korean in the present study. The physiological, biochemical characteristics and polysaccharide pattern of these isolates were compated with those of four laboratory strains ; AHT(a), FA-1F(b), LM7(e), and OMZ65(g). One hundred strains of oral streptococci were isolated from dental caries sites of Korean (one male and one female). Among these, 3 strains were identified as S. mutans. These strains were able to grow on selective media MS, MST, MSP, MSP1, MSB, MSBT and were stained dark pink when sprayed with solutions of mannitol and TTC. So, these strains were called strain 108, 110, and 120, respectively. Strain 108, 110, and 120 were bacitracin resistnt. As these strains contained particularly hippurate hydrolysis enzyme, they were distinguished from laboratory strains. Apart from laboratory strains, the strain 108 was not capable of fermenting lactose, the strain 110 was not able to ferment sorbitol, inulin, melibiose, raffinose and the strain120 was incapable of fermenting inulin, raffinose. All fractions of extracellular and ecll bound polysaccharide of the strain108, 110, and 120 were consisted of more glucan than fructan. Aside from laboratory strains, the isolated strains were composed of more water-insoluble glucans related adherence on solid surface than water-soluble. According to these results, the strain108, 110, and 120 had native characteristecs of S. mutans, but they were different from laboratory strains in some characteristics. Therefore, each of them was given a name to S. mutans KHC108, KHC110, and KHC120, respcetively.

The Distribution Characteristics and Contamination of Heavy Metals in Soil from Dalcheon Mine (달천광산 토양 내 중금속의 존재형태 및 오염도)

  • The geochemical partitioning of arsenic in contaminated soils from a of wet land and tailing of the abandoned mine is examined. Chemical analysis and sequential extraction method by ultrasound-sonication extraction are applied to investigate the mobility and chemical existence conditions of arsenic as well as heavy metals. The results of this study showed that heavy metals concentration of tailings showed as a following order: Fe > As > Cu > Pb > Cr. The highest metal concentration was recognized in samples less than $63\;{\mu}m$ fraction in their particle sizes. Exchangeable and carbonate fractions in soil samples showed following Cu > As > Pb > Fe > Cr for tailings, and Fe > Pb > Cu > As > Cr for reservoir soils, respectively. Arsenic was bound as exchangeable fraction in tailings and its concentration appeared higher than those of the other metals. Thus, As can be easily dispersed into soil and water environments. The obtained results can be used to design soil remediation plan in the study area and require further detailed study to investigate severe environmental pollution of surface water as well as rivers with respect to heavy metals in terms of speciation analysis of toxic elements such as As and Cr.

Behavior of the herbicide metolachlor in carps (제초제 metolachlor의 잉어체내 행적)

  • In order to investigate the behavior of the herbicide metolachlor [2-chloro-6'-ethyl-N-(2-methoxy-1-methylethyl)aceto-O-toluidide] in fish, carps (Cyprinus carpio L.) were exposed to the herbicide at $LC_{10}$(1.93 mg/L) for 4 days. Metolachlor dissolved in water was absorbed rapidly into carps to mark the maximum concentration 6 hours after exposure. The amounts of the $^{14}C$-metolachlor and its metabolites absorbed in gall were much higher than those in the other parts, strongly suggesting that biliary excretion involving enterohepatic recirculation could be an important route for the elimination of metolachlor. The $^{14}C$-radioactivity distributed into aqueous phase fraction in test water and in carp extract was increased in time-dependent manner. Extraction rate of $^{14}C$ absorbed in carp tissues was decreased remarkably up to 6 hours after exposure, suggesting that the possible polar metabolites of metolachlor were transformed into the conjugates to form non-extractable bound residues.

Removal of Geosmin by Combined Treatment of PAC and Intermediate Chlorination in the Conventional WTP (표준정수처리공정에서 분말활성탄과 중간염소를 이용한 지오스민 저감방안)

  • In this study, the effective treatment reducing geosmin and dosage of PAC was suggested when taste & odor compounds flow in. The removal efficiency of geosmin was evaluated with types of chlorination. In case of low geosmin concentration below 25 ng/L, removal efficiency of geosmin was estimated at 46% by combined treatment of pre and intermediate-chlorination. But, in the exclusive intermediate-chlorination treatment, removal efficiency of geosmin was increased to 57%. In the medium geosmin concentration (25~79 ng/L), removal efficiency of geosmin was estimated at 59% by combined treatment. But, in the exclusive intermediate-chlorination treatment, removal efficiency of geosmin was increased to 87%. When high geosmin concentration above 80 ng/L flows in, removal efficiency of geosmin was estimated at 69% by combined treatment. However, in the exclusive intermediate-chlorination treatment, removal efficiency of geosmin was increased to 95%. Then, the exclusive intermediate-chlorination has an important effect on removal of geosmin. After correlation of geosmin concentration and dosage of PAC was analyzed, the coefficient of determination was estimated at 0.96. And, the proper PAC dosage chart was proposed. Also, at a initial occurrence of geosmin, when the combined treatment by intermediate-chlorination and PAC was applied, particle-bound geosmin should be removed continuously. Finally, it is proved that the combined treatment was effective to remove the geosmin by threshold levels as well as to reduce the dosage of PAC.

A Study on the Movement Distribution of Common Grey Mullet, Mugil cephalus in Funnel Net Fishing Ground of the Yeosu Coastal Sea (여수 연안 승망 어장에서 숭어의 이동 분포에 관한 연구)

  • In order to find out the environmental factors influencing movement of common grey mullet, Mugil cephalus in funnel net fishing ground of the Dolsan-do, Yeosu southern sea area, the oceanographic factor such as the water temperature, isobath and tidal current were observed respectively, the water temperature was compared with the amount of common grey mullet caught by funnel net. Also, to investigate the movement direction of common grey mullet in same sea area, 160 common grey mullets of body length 22 to 51cm caught at funnel nets of the Dolsan-do southern sea area were marked and then released at 5 positions in 5 times. The results obtained are summarized as follows : 1. The water temperature at the funnel net fishing ground of Dolsan-do in 2002 was ranged from 6.9 to 27.4$^{\circ}C$. The water temperature was displayed a maximum value in August to increase from March and a minimum value in February of the ensuing year to decrease from September. The catches of gray mullet caught by funnel net were generally abundant from March to September, but decreased sharply from October. The optimum range of water temperature for the funnel nets fishing was situated between 15.0 to 25.0$^{\circ}C$. 2. The isobath from 6m to 13m in coast sea set up funnel nets were densely distributed and the depth more than 14m of isobath were widely spreaded to the open sea at Dolsan-do southern sea area. 3. The tidal current of the coast sea set up funnel nets flowed southward and northward along the coast ato ebb and flood tide respectively. The direction of tidal current to the open sea was southeast at ebb tide with the mean speed 43cm/sec, but northwest at flood with the mean speed 25cm/sec. 4. The recapture rate through the experiment duration showed 9.4%. The recapture rate in Gyedong area was very high value with 33.3% as compared with others. The movement of common grey mullet in Dolsan-do southern sea area trended toward a inner bay and north bound mainly.

The Antioxidant Ability and Nitrite Scavenging Ability of Plant Extracts (식물체 추출물의 항산화성 및 아질산염 소거작용)

  • The plant extracted from Nameko, Gallic, Green tea, Allspice, Polygonum multiflorum, Schizandra chinensis, Armeniacae and Pine needle were utilized to investigate the effects of extracts on free radical reaction, lipid oxidation and nitrite scavenging ability. The pH of ethanol extracts showed a higher than that of hot water extracts, among of which were showed the lowest pH 3.0 in Schizandra chinensis. The important factor of lipid oxidation were $Fe^{2+}$ ion and active oxygen, in which were bound by plant extracts in case of $Fe^{2+}$ ion existed. However, the hydroxyl radical scavenging ability of extracts were lowed, compared to extracts reacted with $Fe^{2+}$ ion. Among of them, the hydroxyl radical scavenging ability of Nameko and Pine needle extracts had a lower TBARS value than those of control. The iron content of extracts were less than 2.0 mg/100 g, but the total iron content of Schizandra chinensis extracts were 6.8 mg/100 g. The ethanol extracts of pine needle were higher than those of hot water extracts on the basis of $Fe^{2+}$ ion content. The ascorbic acid content of green tea showed 14.3 mg/100 g in hot water extracts and 16.7 mg/100 g in ethanol extracts. Electron donating ability of extracts showed more than 50%, except Nameko and allspices, which were higher in ethanol extracts than those of hot water extracts. The superoxide dismutase(SOD)-like activity of green tea showed 85.3% and 63.5% in hot water and ethanol extracts, respectively. The nitrite scavenging ability of green tea was the most effective in both extracts.

Study on the Processing and Compositions of Salted and Dried Mullet Roe (영암산 염건 숭어알의 가공과 조성에 관한 연구)

  • The salted-dried mullet(Mugil japonicus) roe is a kind of traditional food particulary in the area of Young-am gun, Chunnam province. This study was conducted to conform the scientific processing conditions and to evaluate the nutritional quality and changes of major components during storage times. The manufacturing method was that the fresh roe was salted for about 20 hours for the preparation of salted-dried roe, washed by clean waters, drained, shaped a flat piece with 1.2cm thickness by pressing, and spreaded sesame oils on the surface of the salted roe periodically during wind drying for 20 days. The dried roe was blanched in heated water$(80^{\circ}C/3min)$ and packaged the dried product for storages. The fractional compositions of free lipid of wind dried roe were 40% of neutral lipids, 12% of glycolipids and 9% of phospholipids and those of bound lipids were 13% of neutral lipids. 10% of glycolipids and 13% of phospholipids respectively. The major fatty acids of the roe were $C_{16:0}$, $C_{18:0}$, $C_{18:1}$, $C_{18:2}$ and $C_{20:0}$ which was consisted of free and bound lipids in wind drying method during processing and storages. Total amino acids were 99.87g/100g and major amino acids were Glu, Pro, Leu, Lys and CySH and the protein score was average 155% and the chemical score was average 109%. Free amino acids was 1,376mg% that had 50.61% of Pro and the major kinds of those were Tyr and CySH.

Effect of N-3, N-6 Fatty Acid and d-Limonene Treatment on Membrane Lipid Composition and Protein Kinase C Activity in Experimental Rat Hepatocarcinogenesis (쥐의 간 발암과정에서 N-3, N-6 지방산 섭취 및 d-Limonene 투여가 생체막 지질조성 및 Protein Kinase C 활성도에 미치는 영향)

  • This study was done to investigate the effects of n-3, n-6 fatty acid and d-limonene on the hepatic membrane lipid composition, protein kinase C (PKC) and glutathione S-transferase (GST) activities in experimental rat hepatocarcinogenesis. Sprague-Dawley female rats were fed with two different types of dietary oil for 20 weeks. Corn oil (CO) and sardine oil (SO) were used at 15% by weight as a source of n-6 and n-3 fatty acid, respectively. One week after feeding, rats were intraperitoneally injected twice with a dose of diethylnitrosamine (DEN, 50 mg/kg body weight) and after 1 week 0.05% phenobarbital (PB) was provided with drinking water. Membrane fractional lipid composition showed that the content of cholesterol was higher in 50 group than CO group and also significantly decreased by d-limonene. The content of phospholipid was increased by carcinogen treatment but not affected by dietary oils or d-limonene. Membrane C/PL molar ratio was significantly decreased by d-limonene or carcinogen treatment in 50 groups but not in CO groups. Fatty acid composition was changed by dietary oils but not by carcinogen treatment or d-limonene. Cytosolic PKC activity was not significantly different by dietary oils, d-limonene or carcinogen treatment. However, membrane PKC activity was significantly increased by carcinogen treatment and decreased by d-limonene. Cytosolic GST activity was affected by d-limonene or carcinogen treatment in all dietary groups. These data indicate that dietary oils, d-limonene and carcinogen treatment can not change much membrane phospholipid composition. But membrane C/PL molar ratio was changed by carcinogen treatment and d -limonene although the effect was different between dietary oils. Therefore, it is suggested that different dietary oils and d-limonene can somewhat modulate the changes of membrane fluidity and activities of membrane bound enzymes like membrane associated PKC during carcinogenesis.

Effect of Air Circulation Velocity on the Rate of Lumber Drying in a Small Compartment Wood Drying Kiln (소형 목재인공건조실에 있어서 공기순환속도가 목재건조율에 미치는 영향)

This study indicates that above the fiber saturation point the drying rate can be increased with increasing the velocity of the air circutation, i.e., the drying rate of sample boards is proportional to the air velocity, but below the fiber saturation point, the effect of the velocity of air circulation is very low as shown in Figs. 1 and 2. 2. Under the controlled temperature and humidity in the kiln, the more the sample boards have moisture, the higher drying rate of it can be obtained. In other words, this means that even though in the case of drying various moisture content of wood, at the final drying stage, approximately the same percentage of moisture content of wood can be secured by employing the higher velocity of air circulation. 3. This study shows that the rate of drying in kiln changes distinctly at the fiber saturation point, i, e., above the fiber saturation point, the drying curve shows concave aginst the X axsis, but below the fiber saturation point, in the range from 30 percent of moisture content to 20 percent of moisture content, the curve shows convex as shown in Fig. 3. As the drying progresses, however, the drying curve shows concave again below 20 percent of moisture content. This means that inflection point of drying curve may be located clearly at the fiber saturation point, i.e., 30 percent of moisture content. As mentioned above, the 30 percent of moisture content of wood at which the inflectional point appears can be recognized as a critical point, i. e., the fiber saturation point at which all free water was removed from wood. The existence of inflectional point indicates that the evaporation of hygroscopic water in a cell wall is more difficult than the evaporation of free water in a cell cavity and the minor space of cell wall. The convex curve in the range of moisture content from 30 percent to 20 percent means that the evaporation of capillary condensed water has a tendency of the same rates of drying approximately, but as approaching to the 20 percent of moisture, the transfusion of moisture from wood becomes difficult because of having less moisture in cell wall. Below 20 percent of moisture content, the drying curve shows concave again, which means that it is difficult to remove the moisture located nearer to the surface of cellulose molecules and the surface bound water. These relations were revealed in Fig. 4. In comparison AC curve which does not have the two inflection points with BD curve which has two inflection points, i.e., Band D, they are mentioned already, by existence of the inflection points, the curve BD shows that the change of drying rate in the interval from 20 percent of moisture content to 30 percent of moisture content is not greater than in the case of the curve AC in the same interval. At the inflection point of 30 percent of moisture content, it can be noticed that the changing of the drying rate is very conspicuous. This phenomenon also can be recognized, as it is noticed by the Fig. 3, the drying rate from green to 30 percent of moisture content is very great. But the inclination of the curve is very slow from 30 percent of moisture content to 20 percent of moisture content, i.e., the inclination of the curve becomes almost horizontal lines. The Landscape Components Illustrated in Tea-drinking Pictures of the Joseon Dynasty (조선시대 다화(茶畵)에 나타나는 경관요소)

  • The purpose of this research is to look into the tea-drinking pictures of the Joseon Dynasty in order to find the unique landscape traits of tea-drinking spaces of the era. A number of tea-drinking pictures were drawn during the Joseon Dynasty period and in most cases, the names of the painters are stated as well. These factors make tea-drinking pictures a seminal source that illustrates the landscape traits of the last Dynasty of Korea. For those pictures contain the main components of landscape such as the tastes of 'Yangban'(noblemen), natural characters, and space traits. Since tea was first introduced in Korea during the Three Kingdom Period, tea-drinking culture has a long history in the country. There have been various studies about tea and many study results have been presented. Few research, however, have looked into tea culture from the point of landscape. Under the premise that tea-drinking pictures of Joseon Dynasty can be a cultural code that explains Korean tea-drinking culture, this thesis looked into those pictures from various angles and analyzed them in order to elucidate the attributes of scenery components of tea-drinking spaces that Koreans have forged and developed, and following results could be found. The Landscape components illustrated in tea-drinking pictures of the Joseon Dynasty can be divided into the element of architecture, the element of water, stone and plants. First, for the element of architecture, it was found that tea-drinking took place in anywhere in Korea without specific tea-drinking buildings or gardens unlike the case of Japan. This has to do with traits of Koreans who were not bound by formality and truly loved nature. Second, for the element of water, water contributed to making harmonious landscape. Third, for the element of stone, it was clear that stone had a practical role in providing comfortable place for lying and enjoying tea-drinking and scenery. Fourth, plants made elegant figure in the landscape and were planted in accordance with their inner meaning. Tea-drinking pictures of the Joseon Dynasty elucidates that when it comes to tea-drinking, Koreans were not obsessed with a formality of tea-drinking, or a set of tea ceremony, which is profoundly different with Japanese, and Koreans did not put a limit on a place for tea-drinking because for Koreans any place they sat could be a great place for tea-drinking.
