Purpose: The purpose of this study was to investigate the effects of a 12-week Tai Chi on the bone mineral density and bone metabolic markers in postmenopausal women Methods: Data were collected from March to July, 2009. Fifty postmenopausal women were recruited for the study. Twenty two women were allocated to experimental group, and 28 to control group. The experimental group underwent Tai Chi exercise twice a week for twelve weeks. The control group was only notified with results of bone mineral density and bone metabolic markers. Bone mineral density was measured by using of DTX-200 (Osteometer MediTech, Hawthorne, CA, USA) at distal radius site and bone metabolic markers were measured by radioimmunoassay method. Collected data were analyzed by t-test, $X^2$-test, and Mann-Whitney test. Results: After 12 weeks of treatment, the Tai Chi group showed a significant difference in bone mineral density compared to control group but no significant effect on osteocalcin and deoxypyridinoline level. Conclusion: Our results suggest that 12 weeks of Tai chi may delay bone loss in postmenopausal women.
BACKGROUND/OBJECTIVES: Women's bone health status is closely related with environmental factors and lifestyle factors. The purpose of this study is to evaluate the dietary risks of osteoporosis and osteopenia for Korean postmenopausal women. SUBJECTS/METHODS: The data from 1,433 subjects from the 2010 KNHANES were used and divided into three groups: normal, osteopenia, and osteoporosis group using bone mineral density (BMD). Nutrient intakes and food intake frequency were evaluated. Logistic regression analysis was applied to determine the odds ratios for osteoporosis and osteopenia. RESULTS: The RNI percentage of each nutrient and food intake frequency from 12 food groups decreased as bone mineral density status deteriorated. Risk for osteoporosis of low calcium (Ca) intake, under the EAR, showed an odds ratio of 2.13(95% CI; 1.26-3.61, P < 0.05). Higher intake frequency showed preventive effect from osteoporosis compared to lower intake frequency in such food group as dairy products (ORs 0.40, CI 0.21-0.75), beans (ORs 0.49, CI 0.29-0.83), seaweeds (ORs 0.55, CI 0.32-0.94), fish (ORs 0.56, CI 0.32-0.98), and fruits (ORs 0.42, CI 0.23-0.79) after adjusting for age. CONCLUSION: To prevent osteoporosis in later life, sufficient Ca intake and more frequent intakes of foods containing Ca such as dairy products, beans, fish, seaweeds, and fruits, which help in Ca absorption, should be stressed for Korean postmenopausal women.
An oral environment is constantly exposed to environmental factors and microorganisms. The periodontal ligament (PDL) fibroblasts within this environment are subject to bacterial infection and allergic reaction. However, how these condition affect PDL fibroblasts has yet to be elucidated. PDL fibroblasts were isolated from healthy donors. We examined using reverse transcription-polymerase chain reaction and measuring the intracellular $Ca^{2+}$ concentration ($[Ca^{2+}]_i$). This study investigated the receptors activated by exogenous bacterial pathogens (Lipopolysaccharide and peptidoglycan) and allergens (German cockroach extract and house dust mite) as well as these pathogenic mediators-induced effects on the intracellular $Ca^{2+}$ signaling in human PDL fibroblasts. Moreover, we evaluated the expression of pro-inflammatory cytokines (interleukin (IL)-$1{\beta}$, IL-6, and IL-8) and bone remodeling mediators (receptor activator of NF-${\kappa}B$ ligand and osteoprotegerin) and intracellular $Ca^{2+}$-involved effect. Bacterial pathogens and allergic mediators induced increased expression of pro-inflammatory cytokines, and these results are dependent on intracellular $Ca^{2+}$. However, bacterial pathogens and allergic mediators did not lead to increased expression of bone remodeling mediators, except lipopolysaccharide-induced effect on receptor activator of NF-${\kappa}B$ ligand expression. These experiments provide evidence that a pathogens and allergens-induced increase in $[Ca^{2+}]_i$ affects the inflammatory response in human PDL fibroblasts.

Purpose: Modeling has been used for elucidating the mechanism of complex biosystems. In spite of importance and uniqueness of adolescent calcium (Ca) metabolism characterized by a threshold Ca intake, its regulatory mechanism has not been covered and even not proposed. Hence, this study aims at model-based proposing potential mechanisms regulating adolescent Ca metabolism. Methods: Two different hypothetic mechanisms were proposed. The main mechanism is conceived based on Ca-protein binding which induces renal Ca filtration, while additional mechanism assumed that active renal Ca re-absorption regulated Ca metabolism in adolescents. Mathematical models were developed to represent the proposed mechanism and simulated them whether they could produce adolescent Ca profiles in serum and urine. Results: Simulation showed that both mechanisms resulted in the unique behavior of Ca metabolism in adolescents. Based on the simulation insulin-like growth factor-1 (IGF-1) is suggested as a potential regulator because it is related to both growth, a remarkable characteristic of adolescence, and Ca metabolism including absorption and bone accretion. Then, descriptive modeling is employed to conceptualize the hypothesized mechanisms governing adolescent Ca metabolism. Conclusions: This study demonstrated that modeling is a powerful tool for elucidating an unknown mechanism by simulating potential regulatory mechanisms in adolescent Ca metabolism. It is expected that various analytic applications would be plausible in the study of biosystems, particularly with combination of experimental and modeling approaches.
The purpose of this study was to examine the effects of dietary calcium supplementation and exercise on bone mineral density and bone mineral content of growing female rats. The exercise and control group were fed a diet containing 0.5% calcium and Ca supplementation group were fed a diet containing 1.0% calcium diet. The exercise group ran on a rodent treadmill (speed of 15m/min for 30 min) three days per week during the 3-week study period. Bone mineral density (BMD) and bone mineral content (BMC) of spine and femur were determined by using dual energy x-ray absorptiometry (FIXI-mus, GE Lunar Radiation Cooperation, Madison, WI, USA). The exercise group had significantly greater (6.25%) spine BMD compared to the nonexercise group and the exercise group had but not significantly greater spine BMC (7.1%) compared to nonexercisers. Femur BMD and BMC divided by the rats final body weight appears to have a higher BMD (7.5%) and BMC (4.5%) in the exercise group, which indicates that exercise had a positive influence on femur bone mineral density and bone mineral content. The supplementation of calcium did not significantly affect spine and femoral BMC and BMD for the 3 weeks experimental period. It can be concluded that when calcium intake meets the recommended, exercise is beneficial for acquisition of spine bone mineral density in young growing female rats. An experiment was conducted to study the requirement of calcium (Ca) and non-phytin phosphorus (NPP) in commercial broilers during starter phase. Seven hundred and twenty day-old Vencob male broiler chicks were randomly distributed into 144 stainless steel battery brooders, 5 birds in each. Four levels each of Ca (6, 7, 8, and 9 g/kg) and NPP (3, 3.5, 4, and 4.5 g/kg diet) were fed in a factorial design in a corn-soya basal diet. Levels of dicalcium phosphate and oyster shell grit were adjusted to obtain the desired levels of Ca and NPP. Each diet was fed ad libidum to chicks in 9 battery brooders from one d to 21 d of age. Body weight gain and feed intake were depressed (p<0.01) by increasing the dietary Ca level (8 and 9 g/kg) at lower levels of NPP (3 and 3.5 g/kg diet). The growth depression observed at lower NPP level was alleviated by reducing the Ca content to 6 g/kg diet. The tibia ash content and tibia breaking strength increased with increase in both Ca (>6 g/kg) and NPP (>3 g/kg) levels. The leg abnormality score decreased (p<0.01) with increase in NPP content in the diet at all levels of Ca tested. The serum Ca and inorganic P levels were increased with increase in the level of the respective mineral in the diet, but the serum concentration of Ca and P were inversely related to the level of NPP and Ca, respectively /kg diet. In general, the excretion of macro minerals (Ca, and P), and micro minerals {zinc (Zn), manganese (Mn), iron (Fe), and copper (Cu)} was significantly lower at lower levels of Ca and NPP tested (6 and 3 g/kg diet, respectively). The mineral excretion increased with increase in dietary Ca and NPP levels, more conspicuously at the disproportionate ratio of these minerals (>2:1, Ca and NPP). Similarly, the retention of Zn, Mn, and Fe in liver was significantly higher (p<0.01) at lower levels of Ca and NPP tested. Results from this study indicate that the commercial broilers do not require more than 3 g NPP and 6g Ca/kg diet during starter phase (up to 21 d of age) for optimum weight gain, feed efficiency and utilization of Ca, P, Zn, Mn, Fe and Cu. However, the requirements of these minerals for optimum bone mineralization were higher than the levels suggested above.
This study explored the effect of calcium levels and/or ovariectomy on bone metabolism using female Sprague-Dawley weanling rats as a model . Rats received a low (0.1%) calcium diet for 8 weeks. The rats were then divided into three subgroups that were fed 0.1% ,0.5% and 1.5% calcium diets for 8 weeks after operation. The results of this experiment indicate that body weight gin was higher in ovariectomy groups than in sham groups regardless of calcium level and food intake. Serum Ca and P concentrations were of normal level regardless of calcium level and ovariectomy. Estrogen concentration was low in the ovariectomized group. Serum alkaline phophatase activity and urinary hydroxyproline have been used as markers of bone formation and resorption. These values were increased in ovariectomized groups. The weight, length and breaking force of femur were not significantly different between the groups. Ash, Ca, P and total lipid contents in femur and lumbar were decreased in the groups fed low calcium . Mg content was decreased in the ovariectomy and total protein content was not affected by calcium level and ovariectomy. The effect groups of ovarectomy on calcium contents of bone was more prominent in lumbar than in femur. In conclusion, though low calcium intakes during growth period may retard the attainment of peak bone mass, calcium supplementation after this period increased bone growth and mineral contents, but not significant effect in three calcium levels. Furthermore, calcium intake was shown to have a greater influence on the mineral contents of femur than of lumbar, and removal of endogenous estrogen production by ovariectomy was shown to be more deleterious on the ash and calcium contents of the lumbar than of femur.
We studied the effects of soy isoflavone supplements on bone metabolism marker (serum osteocalcin, urinary deoxypyridinoline) and urinary mineral excretion (urinary Ca, Mg, Zn) in 47 postmenopausal women. There were 24 participants in the treatment group and 23 in the control group. The treatment group consumed isoflavone extract capsules daily (which contained 90 mg of soy isoflavones) for 12 weeks. The study compared before and after isoflavone intake in the following areas: Physical examination, diet survey, bone metabolism marker and urinary mineral excretion. The average age of the treatment group was 64.6 years and that of the control group was 66.5 years. There were no significant differences between the two groups in terms of height, weight and body mass index. Both groups maintained a regular diet pattern in terms of their average daily nutrient intake. There were no significant differences between the treatment group (23.9 mg) and the control group (25.4 mg) in terms of daily isoflavone intake based on diet. The analysis of bone metabolism marker changes in the treatment group after 12 weeks of taking the isoflavone supplements demonstrated significant differences in the following: Serum osteocalcin (13.7 ng/mL in befor versus 6.8 ng/mL in after) and urinary deoxypyridinoline (5.9 nmol/mmol Cr in befor versus 4.5 nmol/mmol Cr in after). The subjects in the treatment group showed no significant difference in urinary Ca excretion. But the subjects showed a significant difference in urinary Mg (131.9 mg/day in befor versus 115.6 mg/day in after) and Zn (400.5 $\mu\textrm{g}$/day in befor versus 310.2 $\mu\textrm{g}$/day in after) excretion in the isoflavone treatment group at the levels of p<0.001, p<0.01, respectively. No changes were made in the intake of minerals. The composition of serum osteocalcin and urinary deoxypyridinoline, and indicators of bone metabolism, including the excretion Mg and Zn, significantly decreased. As a result, bone mineral loss was lessened. (Korean J Nutrition 36(5): 476~482, 2003)
The purpose of this study was to examine whether soybean protein isolate with rich isoflavones which was the same amount as used for earlier study prevents bone induced by ovarian hormone deficiency. Forty female Sprague-Dawley rats (body weight 210 $\pm$ 5 g) were divided into two groups, ovariectomy (OVX) and sham surgery groups, which were each randomly divided into two subgroups that were fed casein and soybean protein isolate with rich isoflavones (isoflavones 3_4 mg/g protein). The diets were fed for 9 weeks after operation. The results of this study indicate that body weight gain and food intake were higher in OVX groups than in SHAM groups regardless of diets. Weight gain were higher in soy isolate groups than in casein groups. Serum Ca concentration was lower in OVX rats than in SHAM rats when fed casein diet. Within the OVX groups, serum Ca concentration was higher in rats fed soy isolate diet than in rats fed casein diet. Serum ALP were higher in OVX groups than in SHAM groups. Within the OVX, soy isolate groups, indices of serum osteocalcin, were a trend for greater than those of the OVX casein group. Crosslink value were increased in ovariectomy groups. Spine BMD and femur BMD of ovariectomy groups were significantly lower than SHAM groups. However, soy isolate with rich isoflavones group in ovariectomy groups, spine BMC and femur BMC were significantly increased after 9 weeks. In conclusion, the soy protein with rich isoflavones was beneficial for bone in ovareiectomized rats and the results were similar to the findings of our previous study which showed a beneficial effect on bone in growing rats.
