The wound healing is very complex biological processing including inflammatory, reepithelialization and matrix construction. According to the biological systematic category, the ability of the healing is very different. Generally healing ability of the lower animal group has been known more excellent compared to its higher group. Therefore, lower animals have been used as the experimental model to explore the mechanism of the wound healing or repair. To verify histochemical characteristics of the wound healing, we have used skin of the frog (Bombina orientalis) as known common amphibian. At day 1, 10, and 16, the mucous substance was very actively synthesized and strong positive by PAS and Alcian blue (pH 2.5). Day 10 after wounding, margin of the wound was gradually strong positive by PTAH staining for detection of collagen synthesis. At 3 to 6 hour and day 23 to 27, we have found the cell division was active through the MG-P staining, in which the concentration and division of DNA in nucleus was green to deep blue color.
The histochemical characteristics of the cutaneous granular glands and its fine structure of the Korean fire bellied toad, Bombino orientalis Boulenger the only species inhabitated in Korea, were studied with light and electron microscopes. By the granular size and histochemical properties, the granular glands were subdivided into two types. The type I glands have small secretory granules (3.6 $\mu$ m), whereas the type II glands contain relatively larger granules (7.5 $\mu$ m). These two types of glands were simple alveolar gland, and they were commonly composed of inner glandular epithelial cells and outermost myoepithelial cells. By the histochemical reactions, these two secretory granules were not responsed to Alcian blue, but weakly positive reaction was observed in type I granular glands by the PAS reaction. And by the scanning electron microscopic observation, extemal morphology of the opening site of the gland duct was resembled to the stomata of the plant leaves. Especially the epithelial cells of the type I glands had fine reticular surface whereas, type II glands had large vesicular surface. These types of the glands conimonly had larger cavities than those of mucous glands, and forming the multinucleated unicellulr structure (syncytium) at the glandular epithelial layers. Secretory granules of these glands were originated from the rough endoplasmic reticulum of the glandular epithelial cells, and by the maturation level of the granule, morphological diversities appeared.
The effect of skin extracts of Korean amphibian, poisonous snake and fresh-water fish were determined for their caerulein-like action on rabbit gall bladder strips. The isolated gall bladder strips were prepared according to the technique described by Amer and Becvar(1969). The strips were placed in a bath containing 100ml of Locke-Ringer solution maintained at $38^{\circ}C$. Oxygen was continuously bubbled through the solution. The tension of the muscle strip was initially adjusted to 0.7g. The contractile response was measured isometrically by a force-displacement transducer connected to a polygraph. In this rabbit gall bladder strip caerulein produced contraction of CCK-PZ type. The skin extract of Korean amphibian also elicited similar contraction as caerulein, which extracted from Australian amphibian, Hyla caerulea, by Erspamer et al. The calculated amount was approximately $2{\mu}g$ caerulein per gram of skin tissue in Korean amphibian and the potency was about 1/200 of that seen in Australian amphibian. The contraction of gall bladder strip by our amphibians occurs in decreasing order; Rana Nigromaculata coreana Okada, Rana nigromaculata Hallowell, Hyla arborea japonica Gunther and Bombina orientalis Boulenger. The skin extracts of poisonous snake and fresh-water fish produced no caerulein-like activity.
A cellulosse acetate electrophoretic survey of Korean Anura has revealed the presence of diverse lactate and malate dehydrogenase (LDH and MDH) isozymes. The pattern of LDH and MDH isozymes in the tissues of the central nervous system of the six species of Anura examined are species specific and differ from those of mammals and birds. Both Rana nigromaculata nigromaculata and Rana nigromaculata coreana have two molecular forms of LDH and MDH, respectively, with almost the same pattern. Whole brain homogenate of Rana temporaria shows also a maximum of only two LDH isozymes. Both Bufo bufo asiaticus and Bombina orientalis have five molecular forms of LDH with an entirely different spacing on the zymograms, whereas Rana rugosa has three. Two molecular forms of MDH are present in all animals examined and one band is shown in olfactory lobe and mixture of cerebellum and medulla oblongata of Rana nigromaculata nigromaculata.

Two isozymic forms of LDH and MDH exist in both Rana nigromaculata and Rana nigromaculata coreana, respectively, with almost the same pattern. LDH of Bombina orientalis has five isozymic forms, and that of Rana temporaria ornativentris contains four isozymes. Bufo sp. has 3 to 5, and Rana rugosa has 3 to 4 isozymic bands according to the tissues. MDH's of all animals have two isozymic forms with different spacing on the zymograms.
The present investigation was performed to purify bombesin-like immunoreactivity (BBS-LI) from the skin of frogs, B. orientalis inhabiting Korea. For extraction of BBS-LI, the fresh skin of 360 g from frogs was immersed in 1,800 ml of 100% methanol and then kept at $4^{\circ}C$ for 5 days. BBS-LI was partially purified by liquid chromatography using an alkaline alumina column followed by a Sephadex G-10 column. BBS-LI was further purified by using sequential HPLC of reversed phase C18 preparation, gel permeation, SP-ion exchange and reversed phase C18 analysis. BBS-LI in fractions of each step was monitored by radioimmunoassay for which bombesin antiserum with a titer of 1 : 188,800 was raised in a guinea pig. Eventually, two different BBS-LI were successfully purified and each BBS-LI showed the following character. 1) BBS-LI was well separated into two peaks in SP-ion exchange HPLC. One (BBS-LI-K1) bound to the column while the other (BBS-LI-K2) did not. 2) BBS-LI-K1, 73.8% of total BBS-LI, was not differentiated from synthetic bombesin in reversed phase C18 analytical and gel permeation HPLC. 3) BBS-LI-K2, 26.2% of total BBS-LI, eluted later than synthetic bombesin in reversed phase C18 analytical HPLC, but it eluted with a retention time identical to that of synthetic bombesin in gel permeation HPLC. 4) The two forms of BBS-LI and synthetic bombesin identically stimulated gastrin release and pancreatic exocrine secretion including volume, protein output and amylase output in anesthetized rats. It is concluded from the above results that the skin of B. orientalis contains two different forms of BBS-LI which are very identical to bombesin immunologically and biologically. In comparison with synthetic bombesin containing 14 amino acid residues, the major form shows quite similar pattern in all HPLC used in the present study, but the minor form exhibits quite different pattern in SP-ion exchange and reversed phase C18 analytical HPLG.
Amphibian biodiversity, community structure and habitats were surveyed in the Mt. Myungji area from April 2002 to March 2003. During the survey period 2orders, 4 families, 7 species of amphibians were observed. Bombina orientalis (DI =45.1%) was dominant and Bufo stejnegeri (DI = 12.8%), Rana rugosa and Rana dybowskii (DI = 9.9% each) Rana nigromaculata and Rana huanrenensis (DI 1.4% each), and Onycodactylus fisheri (DI = 7.0%) followed. Frequency of occurrence of O. fisheri and B. stejnegeri was largely different according to the altitude and human residence in their habitats.R. nigromaculata and R. huanrenensis showed low frequency of occurrence (<10%) suggesting the decrease in habitation density in this area. The species richness (R'), general diversity (H'), and evenness (E') of amphibians in this area were 0.98, 2.12, and 0.76, respectively, suggesting relatively healthy condition of amphibian community in this area.

내분비계 장애물질 (Endocrine Disruptors: EDs)이란 ‘내분비 기능에 변화를 일으켜, 생체 또는 그 자손의 건강에 위해한 영향을 미치는 외인성 물질’로서 세계야생생물보호기금 (World Wildife Fund: WWF)의 목록에는 67 여종, 일본 후생성에서는 142 여종의 물질을 내분비계 장애물질로 분류하고 있다. 이렇게 분류된 내분비계장애물질 가운데에는 많은 종류의 농약이 포함되어 있으며, 이들이 자연상태계에 미치는 영향에 대한 많은 보고가 있다. 양서류는 먹이연쇄의 중위포식자로 내분비계 장애물질의 순환 및 생채축적 회로에서 중요한 위치를 갖는다. 또한 농경지나 계류 등에 서식하며 산란하는 습성이 있다. 이러한 서식지는 농약 등에 노출받기 쉬우므로 환경오염평가 대상동물로 중요하다. 본 연구에서는 국내에서 사용되고 있는 제초제, 살충제 등이 양서류 발생에 미치는 영향을 평가하고, 이들 물질에 의한 노출을 평가할 수 있는 biomarker 유전자를 발굴하고자 하였다. 우리나라 전역에 많이 분포하는 무당개구리(Bombina orientalis)의 초기 배아발생 과정에서 이들 농약을 처리하였을 때, 물질의 종류에 따라 정도의 차이는 있으나 농도 의존적으로 배아 및 올챙이의 치사율이 높게 나타났다. 또한 농약은 배아 및 올챙이의 기형을 유발하였다. 기형의 종류는 농약에 따라 서로 상이한 형태로도 나타났으나, 특히 몸통휨 또는 꼬리휨등의 척추골 기형이 많이 나타났다. 환경 위해성 평가 biomarker 유전자로서 골격계 형성에 관여하는 Sox9 유전자를 선택하였다. 무당개구리에서 처음으로 Sox9 유전자를 동정하였으며, 농약을 처리한 실험군에서의 Sox9 유전자의 발현 양상은 대조군에 비하여 높게 나타났다. 본 연구결과 생태계에서 내분비계 장애물질의 위해성 평가를 위해 양서류 초기배아의 치사 및 기형의 정도를 관찰하는 방법이 유효할 것으로 사료된다. 또한 야생동물의 biomarker 유전자를 발굴과 이를 이용한 위해성 평가는 더욱 정밀한 평가법으로 유용할 것이다.
The census of Amphibia fauna in Tokyusan National Park was carried out during April 10, 1993 and August 8 - August 9, 1993 at the areas of construction field of golf course in Muju Resort and the Chibong. There were 6 species of 5 genera on the 4 families, 2 orders of Amphibia in Tokyusan National Park including Hynobius leechii. Onychodactylus fischeri, and Bufo bufo gargarizans which were announced the specific wild animals with the object of conserving by The Ministry of Environment. At the area of construction field of golf course in Muju Resort, there were 5 species of Amphibia observed as Hynobius leechii, Bombina orientalis, Bufo bufo gargarizans, Rana nigromaculata, and Rana dybowskii. Onychodactylus fischeri and Rana dyhowskii were observed at the Chibong area. The Chibong inwhich was observed 9 relatively large number of individuals of Onychodactylus fischeri was revealed as an environmentally sound habitat to Onychodactylus fischeri. Whereas in the construction field of golf course, there were intended to diminishing species and population size rapidly by progress with construction works. Therefore the large construction works such as golf couse and ski slopes which were concerning to deteriorate the natural environment should be restrain in National Park for animal diversity conservative.

The authors observed the ultrastructure of the mucous glandular epithelial cells in the amphibian skin by mean of electron microscope. The specimens from the experimental animals were fixed in 2.5% glutaraldehyde-oaraformaldehyde fixative in phosphate buffer at pH 7.2 prior to fixation in 1% osmium tetroxide, dehydrated in graded ethanol and acetone, embedded in Epon 812 mixture, and sectioned with Sorvall MT-2 ultramicrotome. The ultrasections were contrasted with uranyl acetate and lead citrate and observed with a JEOL-100B electron microscope. The results were as follows: 1. The cutaneous mucous glands in amphibia consisted of the glandular epithelial and the myoepithelial cells. 2. Several different cells in ultrastructure were observed in the mucous glandular epithelium of the adult amphibian skin. a. The dark and the light cells were observed in Hynobius leechi. b. The mitochondria-rich and the round secretory granule-containing cells were observed in Bombina orientalis. c. The round secretory granule-containing and the foam-like granule mass-containing cells were observed in Kaloula tornieri. d. The cutaneous mucous gland of Rana nigromaculata were divided into two types: A and B-type glands. In the A-type mucous gland, the mitochondria-rich and the round secretory granule-containing cells and in the B-type mucous gland, the mitochondria-rich, the secretory granule-containing and the ER-rich cells were observed. 3. Based upon the above findings, the authors infer that the mucous granular epithelium of the amphibian skin consists of the mitochondria-rich undifferentiated, the secetory granule-containing and mature, and the ER-rich evacuated cells.
