Basic reproduction number of African swine fever in wild boars (Sus scrofa) and its spatiotemporal heterogeneity in South Korea

  • Background: African swine fever (ASF) is a hemorrhagic fever occurring in wild boars (Sus scrofa) and domestic pigs. The epidemic situation of ASF in South Korean wild boars has increased the risk of ASF in domestic pig farms. Although basic reproduction number (R0) can be applied for control policies, it is challenging to estimate the R0 for ASF in wild boars due to surveillance bias, lack of wild boar population data, and the effect of ASF-positive wild boar carcass on disease dynamics. Objectives: This study was undertaken to estimate the R0 of ASF in wild boars in South Korea, and subsequently analyze the spatiotemporal heterogeneity. Methods: We detected the local transmission clusters using the spatiotemporal clustering algorithm, which was modified to incorporate the effect of ASF-positive wild boar carcass. With the assumption of exponential growth, R0 was estimated for each cluster. The temporal change of the estimates and its association with the habitat suitability of wild boar were analyzed. Results: Totally, 22 local transmission clusters were detected, showing seasonal patterns occurring in winter and spring. Mean value of R0 of each cluster was 1.54. The estimates showed a temporal increasing trend and positive association with habitat suitability of wild boar. Conclusions: The disease dynamics among wild boars seems to have worsened over time. Thus, in areas with a high elevation and suitable for wild boars, practical methods need to be contrived to ratify the control policies for wild boars.

The Cryoprotective Effect on Frozen-thawed Boar Semen of Egg Yolk Low Density Lipoproteins

  • In order to protect the spermatozoa against cold shock, hen egg yolk is widely used as a cryoprotective agent in semen freezing extenders for domestic animals. The protective action of yolk is largely presumed to be due to low density lipoproteins (LDL). The effects of LDL on sperm quality of bull and northern pike (Esox lucius) after freezing-thawing have been reported, but no study has been made to evaluate the effect of LDL on boar sperm motility and other characteristics. The experiment was carried out to investigate the effect of LDL on the freezing of boar sperm in 0.25 ml straws. The aim was to evaluate the quality of boar spermatozoa cryopreserved in the presence of LDL. Motility of semen cryopreserved in LDL was analyzed and compared to semen cryopreserved with Tris-citric acid-glucose (TCG) and Tris-citric acid-fructose (TCF), two basic freezing extenders containing egg yolk. Similarly, acrosome and plasma membrane integrity were also evaluated and compared to semen cryopreserved with TCG and TCF. Analysis of sperm quality after freeze-thaw showed that the motility, acrosome and plasma membrane integrity were improved with LDL in the extender, as compared to the TCG and TCF. The highest post-thaw integrity of acrosome and plasma membrane and motility were obtained with 9% LDL (w/v). Consequently, the optimum LDL concentration in the extender was 9%. It is also suggested that the concentration of LDL addition is important for the effect on boar sperm protection during freezing and thawing. The percentage of motile spermatozoa was significantly higher after freezing in 9% LDL than in TCG and TCF 54.4% versus 30.4% and 30.1% (p<0.05), respectively. The integrity of acrosome and plasma membrane were also significantly higher at 70.3% and 50.5% respectively with semen frozen in 9% LDL extender compared to TCG at 37.8% and 30.3% and TCF at 36.4% and 29.9%, respectively (p<0.05),. In conclusion, we propose that extender containing LDL extracted from hen egg yolk could be used as a cryoprotective media with a better efficiency than TCG and TCF. LDL improved boar semen quality, allowing better spermatozoa motility, acrosome and plasma membrane integrity after the freeze-thaw process. Furthermore, we found out that the extender with 9% LDL concentration significantly enhanced motility, acrosome and plasma membrane integrity of boar sperm after freezing and thawing.

Association Study Analysis of Cluster-of-Differentiation Antigen 9 (CD9) Gene Polymorphism (g.358A>T) for Duroc Boar Post-thawed Semen Motility and Kinematic Characteristics

  • Cryopreservation of boar semen is continually researched in reproductive technologies and genetic resource banking in breed conservation. For evaluating the boar semen quality, sperm motility (MOT) is an important parameter because the movement of spermatozoa indicates active metabolism, membrane integrity and fertilizing capacity. Various researches have been trying to improve the quality of semen post-thawed in boar. Recently, polymorphism (g.358A>T) of cluster-of-differentiation antigen 9 (CD9) gene reported to be significant association with MOT. Also, CD9 gene was expressed in the male germ line stem cells is crucial for sperm-egg fusion, and was therefore selected as candidate gene for boar semen. This study was conducted to evaluate the pig SNP (g.358A>T) of CD9 gene as a positional controlling for semen parameters of post-thawed boar semen. To results, the g.358A>T SNP of the CD9 gene was significantly associated with the traits such as MOT, curve linear velocity, straight line velocity, average path velocity and amplitude of lateral head displacement. Particularly, the g.358A>T SNP significantly has the highest association with MOT and animals with AA genotype (p<0.001). Therefore, we suggest that the g.358A>T in the intron 6 region of the porcine CD9 may be used as a molecular marker for Duroc boar Post-thawed semen quality, although its functional effect was not defined yet.

장기 카메라트래핑 모니터링 자료를 활용한 도시근교의 고라니 및 멧돼지 일주기성 연구 (Diel Activity Patterns of Water Deer (Hydropotes inermis) and Wild Boar (Sus scrofa) in a Suburban Area Monitored by Long-term Camera-Trapping)

  • This study aimed to figure out diel activity patterns of water deer and wild boar, whose habitat is disturbed by human activity in a suburban area. Photographic data of water deer and wild boar collected by 18 camera traps from January 2021 to November 2022 was categorized into four diel periods (dawn, day, dusk, and night), and converted into frequencies per 100 CTD (Camera Trap-Days) for seasonal and spatial diel activity pattern analysis. Water deer exhibited intense nocturnality in fall, and wild boar did in fall and winter. During the fall, water deer's night-time capture rate was the highest (61.9%), and day-time photographic rate was the lowest (13.9%) among other seasons. During the fall and winter, wild boar's night-time photographic rate was relatively high (77.6%, 78.0%, respectively), and day-time photographic rate was relatively low (0.7%, 1.8%) in comparison with other seasons. In spatial diel activity analysis, both water deer and wild boar showed a tendency to avoid humans, especially near trails during day-time hours. The day-time photographic rate of water deer in trail-near group was 12.1% and trail-far group was 24.6%. The day-time photographic rate of wild boar was 3.4%, 5.7%, respectively. Because the diel activity patterns of wildlife reflect human disturbance, this study provides useful ecological information for developing appropriate management plans for the coexistence of water deer and wild boar in suburban areas.

The Effect of Wet Pad and Forced Ventilation House on the Reproductive Performance of Boar

  • There were two trials involved in the experiment. Trial 1: the trial was conducted on two Taiwan Sugar Corporation (TSC) pig farms. One was located in the north of Taiwan and the other was located in the south. Both farms had wet pad and forced ventilation (WPFV) and conventional open design (COD) boar and sow houses. There were 12 Duroc boars, age ranging from 12-24 months. Half of them (6 boars) were raised in a WPFV pig house, and the other half were kept in a COD house. Semen was collected at 5-day intervals from May $1^{st}$ to the end of October. Sixteen sows (2-8 parity) were served by artificial insemination each week from the beginning of May to the end of Oct. These sows were checked for heat from 18 days to 25 days after insemination. Trial 2: there were four MPFV boar houses involved in the test. Two houses were located in the north of Taiwan, and the other two houses were located in the south. The test was conducted from January 2000 to December 2001. The total number of serviced sows by MPFV-housed boars was 35,105 head and for COD-housed boars 103,065 head. The results showed that the total semen volume, density of sperm, total sperm per ejaculate, sperm motility and morphological abnormality were significantly better (p<0.01) for boar raised in WPFV house than for COD houses. Average sperm motility in June and July was lower than for the other months. Morphological abnormality was higher during May, June and July. Although the results did not reach a significant level, the average value showed that the total volume of boar semen was higher in the north than for the south. The total semen volume production of boar raised in WPFV was higher than for boars raised in COD house, reaching a significant level only in summer. Boars kept in WPFV house had higher total sperm number than boars kept in COD house, reaching a significant level in spring (p<0.05), summer (p<0.01), and fall (p<0.05) but not in winter (p>0.05). Boars raised in WPFV house had significantly higher sperm motility than boars in COD house during spring (p<0.001), summer (p<0.001), fall (p<0.01) and winter (p<0.05). The average farrowing rate and piglets born alive were higher for boars in WPFV house than for boars in COD house, but neither reached a significant level (p>0.05). The present experiment shows that WPFV house can improve the reproduction performance of boars.

임신전 불임 수퇘지 종부 및 무정자정액 주입에 의한 미경산돈의 산자수 증가에 관한 연구 (Studies on Increase of Litter Size by Infusion with Sterile Boar and Their Semen before Gestation in Gilts)

  • 본 연구는 초임전 임신항원인자로 정소상체제거 및 정관절제 수퇘지를 종부하거나 무정자 정액을 사용하였을 때 미경산돈의 번식향상 효과를 조사하기 위하여 수행하였다. 임신항원인자의 주입에 따른 번식성적을 조사하기 위하여 7∼8개월령의 교잡종 미경산돈을 160두 공시하였다. 시험돈은 4개의 시험군으로 나누었으며, 처리는 A처리구는 정소상체제거 교배, B처리구는 무정자 정액, C 처리구는 정관절제 교배, D처리구는 무처리하여 각구당 40두를 배 치 하여 완전임의배치법으로 하였다. 미경산돈의 초임시 총산자수는 A, B, C 및 D 처리구가 각각 10.05, 10.44, 11.63 및 9.97두로 가장 많은 처리구는 정관절제교배구인 C처리구였다(P<0.05). 비경산돈의 생존산자수는 총산자수와 유사하였다. 특히 C처리구는 10.70두로 9.12 및 9.11 두인 A군과 무처리보다 많았다(P<0.05). 반면 정관절제군과 무정자 처리구간에는 총산자수 및 생존산자수에서 차이가 없었다. 한편 미경돈의 혈중 progesterone 농도는 교배후 6일에서 C처리구가 다른 처리구보다 유의성 있게 높았다. 그러나 8일 이후에는 다른 처 리구보나 낮았다. 한편 혈중 cortisol농도는 처리간에 통계적인 유의성이 없었다. 이상의 시험결과를 볼 때 임신전 정관절제 수퇘지나 무정자정액을 항원으로 주입으로 미경산돈의 산자수 증가에 의한 번식효율을 증진을 확인할 수 있었다. 미경산돈의 산자수 증가를 위한 항원으로 정관절제 수퇘지나 무정자정액을 자궁에 주입하는 방법이 효과가 있다고 할 수 있다.

  • To evaluate the fertilizing capacity of domestic animal spermatozoa by hamster test, semen were collected from 15 boars(Duroc, Landrace, and Yorkshire) and 2 mixed dogs which had been proved to be fertile in the past then, the semen were preserved in BWW medium at $4^{\circ}C$ or $18^{\circ}C$ for about 20 hours and coincubated with zona-free hamster ova for 5 hours. The ova were stained by lacmoid and examined under phase contrast microscope to investigate the rates of sperm binding to the ova, penetration and formation of a male pronucleus, and the numbers of both bound and penetrated sperm per ovum. Both the semen preserved at $18^{\circ}C$ for about 20 hours and that treated by swim up procedure showed considerably higher rates of sperm binding and penetration as well as higher number of penetrated sperm than that preserved at $4^{\circ}C$ for about 20 hours, respectively(p<0.01). Motility of boar sperm at insemination was from 40 to 90% and no difference in hamster test was obtained according to different degree of sperm motility. Abnormality in morphology of boar sperm at insemination was from 6 to 45% and no difference in hamster test was obtained according to different degree of sperm abnormality. The sperm concentrations of $7{\times}10^7$ and $7{\times}10^6$ showed considerably higher rates of sperm binding and penetration as well as higher number of bound sperm than that of $7{\times}10^4$ (p<0.01) along with the same higher results than that of $7{\times}10^5$(0<0.05), respectively. Boar sperm showed considerably higher rates of sperm binding and penetration as well as higher numbers of both bound and penetrated sperm than dog sperm, when both semen were treated by BWW+heparin medium and swim up procedure, respectively. These results indicated that fertile boar sperm showed considerably lower rates in the results of hamster test, when preserved at $4^{\circ}C$ for about 20 hours and in lower concentration of sperm than when preserved at $18^{\circ}C$ for about 20 hours and in higher concentration of sperm, respectively, and at the same time considerably higher results than fertile dog sperm, consequently to prove that hamster test would be of great value in assaying the fertilizing capacity of boar sperm.

돼지 액상정액 보존을 위한 Lactose-Egg Yolk와 $B\ddot{u}tschwiler$ 희석액의 비교 (Comparision of Preservation of Liquid Boar Semen between Lactose-Egg Yolk and $B\ddot{u}tschwiler$ Diluents)

  • 본 연구는 돼지 액상정액을 인공수정용 100ml 플라스틱 병에 보존하면서 Lactose-Egg yolk 희석액과 B tschwiler 희석액 간에 보존 온도별 차이를 조사하고, Lactose-Egg yolk 희석액에서의 글리세롤 농도의 효과를 조사하여 돼지 액상정액을 좀더 장기간 사용할 수 있는 방법을 찾고자 실시하였다. 돼지 액상정액을 5$^{\circ}C$ 냉장고에 보존하면서조사한 바에 의하면, 37$^{\circ}C$에서 0.5 및 2시간 배양 후의 정자 운동성은 전체 보존기간동안 Lactose-Egg yolk 희석액이 B tschwiler 희석액보다 유의하게 (P,0.05) 높게 나타났고, 정상첨체비율은 두 희석액간에 차이가 없었다. 돼지 액상정액을 15$^{\circ}C$에 보존하면서 조사한 바에 의하면, 3일부터 7일 보존시까지 정자 운동성과 정상첨체비율에 있어서 B tschwiler 희석액이 Lactose-Egg yolk 희석액보다 유의하데 높게 나타났다. Lactose-Egg yolk 희석액을 이용한 돼지 액상정액의 글리세롤 농도의 효과에 있어서는 최종 글리세롤 농도가 0, 1, 3 및 5%보다 2%일 때 가장 높은 정자 운동성과 정상첨체비유을 나타내었다. 분만율, 복당 생존자돈수 그리고 출생시 평균 생시체중은 Lactose-Egg yolk 희석액과 B tschwiler 희석액간에 차이가 없었다. 이상의 연구 결과를 종합해 볼 때 Lactose-Egg yolk 희석액은 5$^{\circ}C$에서 B tschwiler 희석액은 15$^{\circ}C$에서 6~7일 동안 돼지 액상정액을 보존할 수 있었다.

GIS와 로지스틱 회귀분석을 이용한 멧돼지 서식지 모형 개발 (Wild Boar (Sus scrofa corranus Heude ) Habitat Modeling Using GIS and Logistic Regression)

  • 보호동물사에 대한 서식지 분포정보는 개발압력이 심한 우리나라에서는 서식지 관리에 있어 매우 중요하다. 본 연구는 GIS와 로지스틱 회귀분석을 이용하여 멧돼지의 서식지 적합성모형을 개발하여 서식지 분포도를 작성하고 본 연구의 결과를 토대로 현재 우리나라의 멸종위기종과 야생동물보호종들의 서식지 관리를 위한 기초를 마련하고자한다. 서식지 모형을 구현하기 위한 연구내용 및 방법은 다음과 같다. 첫째, 멧돼지의 서식지 특성을 파악하기 위하여 GIS 자료를 구축하였다. 멧돼지의 위치파악을 위해 원격무선측정기법과 GPS를 이용하였다 둘째, 멧돼지의 서식지 이용과 유용성에 영향을 미치는 환경인자를 파악하기위해 원격무선측정기법과 GPS를 이용하였다. 둘째, 멧돼지의 서식지 이용과 유용성에 영향을 미치는 환경인자를 파악하기 위해 χ²검정(Chi-square test)을 실시하였다. 마지막으로, 최적 서식지의 분포를 예측하기위한 서식지 적합성모형 개발을 위해 로지스틱 회귀분석을 이용하였다. 본 연구의 결과는 다음과 같다. 첫째, 멧돼지의 서식지 이용특성 분석의 결과, 성별 그리고 활동유형별에서는 뚜렷한 차이를 볼 수 없으나, 계절별 그리고 서식지 유형별에서는 서식지 이용특성의 차이가 뚜렷한 것을 볼 수있었다. 둘째, 서식지 유용성 분석의 결과 멧돼지의 서식환경에 영향을 미치는 자연적인 환경요인으로 표고, 향, 임상 그리고 영급이 유의성이 있는 인자로 나타났다. 반면에, 경사, 능선/계곡 , 물 그리고 일사량의 경우 통계적 유의성이 적어 전반적으로 서식지를 결정하는 환경인자라고 볼 수 없었다. 마지막으로, 개발된 서식지 적합성모형의 결과 예측확률 0.5를 기준으로 전제서식지는 75.00%, 커버서식지는 84.26% 의 높은 분류정확도를 나타내었다. 모형검증의 결과 대상지 내부의 경우 전체서식지가 75.00%로 나타나 본 모형이 신뢰성이 있는 것으로 판단되었다.

Hamster test를 이용한 가축정자의 수정능력 검정 II. 정액배지 및 X-선조사가 돼지정자의 Hamster test에 미치는 영향 (Assessment of the Fertilizing Capacity of Domestic Animal Spermatozoa by Hamster Test II. Effects of incubation medium and X-ray irradiation on hamster test for boar spermatozoa)

  • To assay the fertilizing capacity of domestic animal spermatozoa by hamster test, semen were collected from 13 boars(Duroc. Landrace and Yorkshire) which had been proved to be fertile in the past. then, were preserved in BWW medium or in raw state at 18$^{\circ}C$ or at room temperature. The preserved semen were given each different treatment according to the experimental design and coincubated with zona-free hamster ova for 5 hours. The ova were stained by lacmoid and examined under phase contrast microscope to investigate the rates of ova bound with sperm(sperm binding). ova penetrated by sperm(penetration) and formation of a male pronucleus(pronucleus formation) and also numbers of both bound and penetrated sperm per ovum. Between BWW and TBM medium for boar sperm. no difference in the results of hamster test was obtained. The boar spermatozoa in BWW medium, BWW with caffeine, BWW with heparin, and BWW with both caffeine and heparin showed no difference in the results of hamster test. The boar spermatozoa in BWW medium containing both calcium and RSA showed considerably higher rates of sperm binding, penetration and pronucleus formation as well as higher numbers of both bound and penetrated sperm than those not containing calcium with or without BSA( p<0.01) and also the same results higher than that containing calcium without BSA( p< 0.05). The boar spermatozoa irradiated by X-ray(70 KVP, 20mA) for 3 seconds. then, maintained at 18$^{\circ}C$ for 18 hours showed considerably lower rate of sperm binding than all the other groups including the control and X-ray groups irradiated by smaller dose or maintained for shorter period(p<0.01), and also showed lower number of bound sperm than the other groups(p<0.01, p<0.05). All the control groups of both raw and diluted sperm in BWM medium showed higher rates of sperm binding, penetration and pronucleus formation as well as higher number of penetrated sperm than all the X-ray groups irradiated for 3 seconds(70KVP, 20mA) and maintained for either 3 or 18 hours (p<0.01, p<0.05). At the same time the control groups of diluted sperm showed considerably higher rates of sperm penetration and pronucleus formation than the control group of raw sperm( p<0.01). These results indicates that fertile boar sperm showed considerably lower rates In the results of hamster test, when incubated in the medium without calcium and irradiated by X-ray than when incubated in the medium with calcium and not irradiated by X-ray, respectively, to prove consequently that hamster test would be of great value in assaying the fertilizing capacity of boar spermatozoa.
